Glabridin is the main ingredient of hydrophobic fraction in licorice extract and has been shown to have anti-melanogenesis activity in skins.However,the underlying mechanism(s)remain not completely understood.The aim of this study is thus to elucidate the possible mechanisms related to the melanogenesis suppression by glabridin in cultured B16 murine melanoma cells and in UVA radiation induced hyperpigmentation model of BALB/c mice as well.Molecular docking simulations revealed that between catalytic core residues and the compound.The treatment by glabridin significantly downregulated both transcriptional and/or protein expression of melanogenesis-related factors including melanocyte stimulating hormone receptor(MC1R),microphthalmia-associated transcription factor(MITF),tyrosinase(TYR),TYR-related protein-1(TRP-1)and TRP-2 in B16 cells.Both PKA/MITF and MAPK/MITF signaling pathways were found to be involved in the suppression of melanogenesis by glabridin in B16 cells.Also in vivo glabridin therapy significantly reduced hyperpigmentation,epidermal thickening,roughness and inflammation induced by frequent UVA exposure in mice skins,thus beneficial for skin healthcare.These data further look insights into the molecular mechanisms of melanogenesis suppression by glabridin,rationalizing the application of the natural compound for skin healthcare. 展开更多

复合多肽延缓小鼠皮肤衰老的功效研究

通过细胞增殖、抗氧化、抗炎以及促进胶原蛋白4个细胞实验研究了棕榈酰三肽-5,五肽-3,大豆多肽,类蛇毒肽和六肽-115种多肽的抗衰能力和复合后的效果提升作用,采用UVB联合D-半乳糖皮下注射造模方式研究了5种多肽复合后的皮肤抗衰老作用... 通过细胞增殖、抗氧化、抗炎以及促进胶原蛋白4个细胞实验研究了棕榈酰三肽-5,五肽-3,大豆多肽,类蛇毒肽和六肽-115种多肽的抗衰能力和复合后的效果提升作用,采用UVB联合D-半乳糖皮下注射造模方式研究了5种多肽复合后的皮肤抗衰老作用。结果表明,5种多肽具有一定的促细胞增殖、抗氧化、抗炎及促胶原蛋白合成能力,且5种多肽复合后具有显著的效果提升作用,复合多肽能够显著改善和缓解UVB联合D-半乳糖皮下注射引起的小鼠皮肤表皮增厚、胶原较少、弹性蛋白减少情况,并且能够显著抑制造模引起的IL-1,IL-6,TNF-α表达上调,抑制MMP3,MMP9,β-半乳糖苷酶的表达。以上结果表明5种多肽复合能够通过缓解UVB联合D-半乳糖皮下注射造模造成的表皮增厚、炎症、细胞外基质流失及衰老相关酶表达增加来发挥皮肤抗衰老作用。 展开更多

研究天山雪莲多糖对特异性皮炎(AD)小鼠的作用及调控机制。外用2,4-二硝基氟苯(DNFB)反复诱导建立昆明小鼠AD模型,40只小鼠,每组8只,随机分为:空白组、模型组、阳性泼尼松龙组和天山雪莲多糖剂量组(质量浓度为100和500μg/mL)。记录各... 研究天山雪莲多糖对特异性皮炎(AD)小鼠的作用及调控机制。外用2,4-二硝基氟苯(DNFB)反复诱导建立昆明小鼠AD模型,40只小鼠,每组8只,随机分为:空白组、模型组、阳性泼尼松龙组和天山雪莲多糖剂量组(质量浓度为100和500μg/mL)。记录各组小鼠皮损情况,检测小鼠经皮失水率,观察皮损组织的病理学,ELISA法测定天山雪莲多糖高剂量组皮损组织中IL-6,IL-4,IL-13,Ig E,TNF-a,NOD 1,TLR-2,CRAMPS的水平。相对于模型组,外用天山雪莲多糖处理小鼠皮肤特异性皮炎湿疹样得到明显改善,经皮失水率显著降低,HE染色病理图片的表皮层厚度显著降低,皮肤组织中Th2相关的细胞因子IL-6,IL-4,IL-13和Ig E的表达均显著下降(P<0.01),同时TLR受体调控相关的因子NOD 1,TLR-2,TNF-α的蛋白表达均显著降低(P<0.01),CRAMPS含量显著升高(P<0.05)。结果表明,天山雪莲多糖通过调控TLR-2和Th2相关细胞因子的表达,改善了AD模型小鼠的皮损,为天山雪莲多糖在预防和治疗特异性皮炎制剂中的应用提供数据支持。 展开更多

In recent years,the abuse of chemical fertilizers has caused numerous environmental problems,such as soil acidification and compaction.Replacing chemical fertilizers with organic fertilizers can effectively alleviate these problems.However,the effects of alternative organic fertilizers remain unclear.To explore the effects of organic fertilizer substitution on rice yield and paddy soil physicochemical properties and bacterial community structure,we conducted a 5-year experiment using different proportions of organic fertilizer substitution in a double-cropping rice field in Jiangxi,China.Our results showed that replacing chemical fertilizers with organic fertilizers can reduce soil acidification,increase soil organic matter content,nutrient contents,and enzyme activities,improve soil physicochemical properties and microbial community,and enhance soil metabolism.Appropriate organic substitution also had positive effects on rice production.These findings enhance our understanding of the effects of different alternative organic fertilization methods and have important theoretical significance for the promotion of the use of organic fertilizers in the future. 展开更多
