The shear stressresistance of cultured adult human endothelial cells was investigated on 6mm polytetrafluoroethylene vascular grafts (made in China). Endothelial cells were sodded onto pretreated vascular grafts at a high density. Grafts were then cultivated for 9 days to enable the maturation of thecytoskeleton, before they were exposed to pulsatile flow simulating the flow patterns and the wallshear forces of the small srtery. After 1 hour of per fusion,a cell loss of 11% in grafts pretreatedwith 20μ g/ ml fibronectin and of 12% in grafts pretreated with 5μ g /ml fibronectin coated human platelet- poor- plasma (p>0. 50 n = 10). Therefore we conclude that the endothelial celllining on vascular grafts pretreated with suitable substrates can form a shear stress-resistant endothelial cell monolayer on polytetrafluoroethylene vascular grafts. 展开更多
