BACKGROUND Intestinal ischemia reperfusion(I/R) injury is a serious but common pathophysiological process of many diseases, resulting in a high mortality rate in clinical practice. Ubiquitin-specific protease 22(USP22) acts as regulator of cell cycle progression, proliferation, and tumor invasion. Depleted USP22 expression has been reported to contribute to arrested cell cycle and disrupted generation of differentiated cell types in crypts and villi. However, the role of USP22 in intestinal damage recovery has not been investigated. Therefore, elucidation of the underlying mechanism of USP22 in intestinal I/R injury may help to improve the tissue repair and patient prognosis in clinical practice.AIM To investigate the role of USP22 in intestinal cell proliferation and regeneration after intestinal I/R injury.METHODS An animal model of intestinal I/R injury was generated in male Sprague-Dawley rats by occlusion of the superior mesenteric artery followed by reperfusion.Chiu's scoring system was used to grade the damage to the intestinal mucosa. An in vitro model was developed by incubating rat intestinal epithelial IEC-6 cells in hypoxia/reoxygenation conditions in order to simulate I/R in vivo. siRNA and overexpression plasmid were used to regulate the expression of USP22. USP22,Cyclin D1, and proliferating cell nuclear antigen(PCNA) expression levels were measured by Western blot analysis and immunohistochemistry staining. Cell survival(viability) and cell cycle were evaluated using the Cell Counting Kit-8and flow cytometry, respectively.RESULTS USP22 expression was positively correlated with the expression levels of PCNA and Cyclin D1 both in vivo and in vitro, which confirmed that USP22 was involved in cell proliferation and intestinal regeneration after intestinal I/R injury. Decreased levels of Cyclin D1 and cell cycle arrest were observed in the USP22 knockdown group(P < 0.05), while opposite results were observed in the USP22 overexpression group(P < 0.05). In addition, increased expression of USP22 was related to improved intestinal pathology or IEC-6 cell viability after I/R or hypoxia/reoxygenation. These results suggested that USP22 may exert a protective effect on intestinal I/R injury by regulating cell proliferation and facilitating tissue regeneration.CONCLUSION USP22 is correlated with promoting intestinal cell proliferation and accelerating intestinal tissue regeneration after intestinal I/R injury and may serve as a potential target for therapeutic development for tissue repair during intestinal I/R injury. 展开更多

It accounts for less than 1% of all gastric cancers. In this report, we describe our pathological findings along with a review of the literature to improve our understanding of the disease and reduce misdiagnosis, as well as to provide evidence for its treatment and prognosis. A 49-year-old male patient was admitted to our hospital (Dalian Municipal Central Hospital, Dalian, China) with a complaint of epigastric pain that had persisted for half a month. Physical examination, regular laboratory blood tests, and computed tomography revealed no obvious abnormalities. Gastroscopy revealed ulcers in the lower part of the stomach, and pathological assessment revealed adenocarcinoma. Radical gastrectomy was performed, and the folinic acid, fluorouracil, oxaliplatin (FOLFOX) chemotherapy regimen was administered postoperatively. Pathological assessment of the mass revealed a protruding tumor measuring 1.5 × 1.5 × 0.7 cm in the lower part of the stomach. The tumor infiltrated through the full wall of the stomach. This was confirmed by immunohistochemical (IHC) staining for cytokeratin (CK)(+), villin (-), p63 (++), and high-molecular-weight CK (+++). The patient remains alive with no recurrence more than seven years after surgery. Primary GASC is a rare malignant neoplasm. The diagnostic criteria for GASC mainly depend on the clinical, radiographic, and histopathological findings. Pathological assessment and IHC staining can be utilized to confirm the diagnosis. Radical gastrectomy plus postoperative chemotherapy containing the FOLFOX regimen is effective for treating GASC and might contribute to long-term survival. 展开更多

Small cell carcinoma of the gastric remnant:a case report

Here, we report a case and review the literature in order to improve the diagnosis and treatment of SCC of the gastric remnant.Methods We report a case of SCC of the gastric remnant in a 71-year-old male Chinese patient who presented with epigastric pain, acid regurgitation, and belching and who underwent Billroth II gastrectomy more than 38 years ago.Results Physical examination showed no obvious abnormalities. Laboratory data were within normal limits, except for anemia. Pathology of the mass showed a protruded tumor measuring 5.0 × 5.0 × 2.5 cm at the anastomotic edge of the gastric remnant that infiltrated through the full wall of the stomach; this was confirmed by immunohistochemical staining for cytokeratin [CK(-)], leukocyte common antigen(LCA)(+), synaptophysin(+), CD56(+), and Ki-67(+ > 50%).Conclusion SCC of the gastric remnant is extremely rare, although the pathology, symptoms, diagnosis, treatment, and prognosis of SCC are similar to those of gastric SCC. Although the standard treatment of SCC of the gastric remnant remains unclear, effective surgical resection and subsequent multiagent chemotherapy should be performed for long-term survival. Our case shows the efficacy of tegafurgimeracil-oteracil-potassium capsule chemotherapy. Examination of a large series is required to determine the optimal treatment strategy for SCC of the gastric remnant. 展开更多

In this report, we describe our pathological findings along with a review of the literature to improve our understanding of the disease and prevent misdiagnosis, as well as to provide evidence for its treatment and prognosis.Methods A 68-year-old male patient was admitted to our hospital(Dalian Municipal Central Hospital, Dalian, China) complaining of upper abdominal pain. Physical examination and regular laboratory blood tests showed no obvious abnormalities. A contrast-enhanced computed tomography scan displayed a 3.0 cm × 3.0 cm mass in the gastric antrum, but no metastasis was found in the adjacent organs.Results A radical gastrectomy was performed and postoperative histopathology showed a moderately differentiated adenocarcinoma with partial hepatoid adenocarcinoma.Conclusion Hepatoid adenocarcinoma of the stomach is a rare malignant neoplasm. The diagnostic criteria for hepatoid adenocarcinoma of the stomach depends mainly on the clinical, radiographic, and histopathological findings. Pathomorphology and immunohistochemical staining can be utilized to confirm the diagnosis. 展开更多

Abdominal wall metastasis of gastric cancer(GC)is a very rare occurrence in the clinic setting.We recently diagnosed and treated a patient with abdominal wall metastasis of GC and we hope to provide some helpful guidance on the clinical diagnosis and treatment of this disease.A 49-year-old male patient with GC was admitted to our hospital(Dalian Municipal Central Hospital,Dalian,China)complaining of left upper abdominal wall mass.Physical examination and regular laboratory blood tests showed no obvious abnormalities.Ultrasound and CT of the abdomen showed a subcutaneous solid mass in the abdominal wall.Radical gastrectomy was performed on February 27,2019,six months after it was first noticed by the patient.Pathological examination and immunohistochemistry showed GC with abdominal metastasis.Postoperative radiotherapy or chemotherapy was not pursued after the second operation and no recurrence or metastasis has been noted so far.GC with abdominal metastasis is very rare and can be easily missed or misdiagnosed.For metastasis to a single site in the abdominal wall,surgical resection,which is recommended,may improve patient outcomes. 展开更多
