METHODS: pGe...AIM: To observe the inhibition of hepatitis B virus replication and expression by transfecting vector-based small interference RNA (siRNA) pGenesil-HBV X targeting HBV X gene region into HepG2.2.15 cells. METHODS: pGenesil-HBV X was constructed and trans- fected into HepG2.2.15 cells via lipofection. HBV antigen secretion was determined 24, 48, and 72 h after trans- fection by time-resolved immunofluorometric assays (TRFIA). HBV replication was examined by fluorescence quantitative PCR, and the expression of cytoplasmic viral proteins was determined by immunohistochemistry. RESULTS: The secretion of HBsAg and HBeAg into the supernatant was found to be inhibited by 28.5% and 32.2% (P < 0.01), and by 38.67% (P < 0.05) and 42.86% (P < 0.01) at 48 h and 72 h after pGenesil-HBV X transfection, respectively. Immunohistochemical stain- ing for cytoplasmic HBsAg showed a similar decline in HepG2.2.15 cells 48 h after transfection. The number of HBV genomes within culture supernatants was also sig- nifi cantly decreased 48 h and 72 h post-transfection as quantifi ed by fluorescence PCR (P < 0.05). CONCLUSION: In HepG2.2.15 cells, HBV replication and expression is inhibited by vector-based siRNA pGenesil- HBV X targeting the HBV X coding region.展开更多
BACKGROUND Tuberculosis(TB)is a widespread infectious disease,with an incidence that is increasing worldwide.Cutaneous TB(CTB)occurs rarely,accounting for less than 1%of all TB cases.Due to the clinical presentation a...BACKGROUND Tuberculosis(TB)is a widespread infectious disease,with an incidence that is increasing worldwide.Cutaneous TB(CTB)occurs rarely,accounting for less than 1%of all TB cases.Due to the clinical presentation and diagnostic difficulties,CTB is often clinically neglected and misdiagnosed.CASE SUMMARY A 32-year-old man underwent several debridement surgeries and skin flap transplantation after trauma.The wound remained unhealed,accompanied by sinus formation.According to empirical judgment,T-cell spot of TB test,and bacterial culture of pyogenic fluids,he was diagnosed with CTB due to infection with exogenous Mycobacterium tuberculosis.A comprehensive anti-TB regimen that included isoniazid,rifampicin,ethambutol,and pyrazinamide was applied.The sinus was filled with a hydrophilic fiber-containing silver dressing,and woundprotecting sponges were applied to part of the wound.The wound healed after 40 d.No ulceration was found within 2 mo after discharge;further follow-up will be conducted.CONCLUSION A non-healing wound may be caused by TB infection.Comprehensive treatment of CTB is effective.展开更多
BiOCl nanostructures including microspheres,microflowers,microplates,and nanoplates,have been synthesized by a simple solvothermal method using bismuth nitrate and sodium chloride as raw materials without adding any a...BiOCl nanostructures including microspheres,microflowers,microplates,and nanoplates,have been synthesized by a simple solvothermal method using bismuth nitrate and sodium chloride as raw materials without adding any additives.Structure and morphology of the products were characterized by powder X-ray diffraction,scanning electron microscopy,and transmission electron microscopy.The results indicated that the as-prepared microspheres and microflowers were composed of nanosheets.Although with different shape and lateral size,the nanoplates and microplales were all single-crystalline plates with exposed {001) facets.It was found that the volume ratio of polyethylene glycol 400 and H_2O in the solvent played a key role in the morphology of the products,and the possible growth mechanism was also discussed.The photocatalytic measurements indicated that the BiOCl samples exhibit good photocatalytic properties towards Rhodamine B.展开更多
