γ-辐照对指状青霉的抑制作用及在椪柑果实保鲜中的应用 被引量:1

本实验探究强度为0.25、0.50、0.75及1.00kGy的γ-射线对供试菌指状青霉的生长、培养液相对电导率、菌丝体总糖与可溶性蛋白含量、苹果酸脱氢酶与琥珀酸脱氢酶活性大小的影响;同时考察了不同强度的γ-辐照对椪柑果实失重率、呼吸强度、... 本实验探究强度为0.25、0.50、0.75及1.00kGy的γ-射线对供试菌指状青霉的生长、培养液相对电导率、菌丝体总糖与可溶性蛋白含量、苹果酸脱氢酶与琥珀酸脱氢酶活性大小的影响;同时考察了不同强度的γ-辐照对椪柑果实失重率、呼吸强度、可溶性固形物、有机酸及抗坏血酸含量的影响。结果表明:γ-辐照可以有效的抑制指状青霉的生长,抑制率和辐照强度呈正相关;同时,γ-辐照能增加菌丝体细胞膜渗透性,降低菌丝体总糖与可溶性蛋白的含量以及与能量代谢的相关酶活性,从而打乱指状青霉正常的生理代谢。此外,不同强度的γ-辐照对椪柑果实的采后生理代谢也有不同程度的影响,在所有的辐照强度中,0.5kGy的γ-辐照降低椪柑失重率、呼吸强度,同时抑制可溶性固形物、有机酸及抗坏血酸含量降低的综合效果最佳。 展开更多

为了开发天然柑橘类果实保鲜剂,研究了竹醋液和壳聚糖对采后椪柑的主要致病菌-指状青霉菌的抑制作用,以及对贮藏期椪柑采后生理的影响。结果显示:竹醋液和壳聚糖对指状青霉菌均有抑制作用,最小抑菌浓度为竹醋液1.5%、壳聚糖0.8%。在椪... 为了开发天然柑橘类果实保鲜剂,研究了竹醋液和壳聚糖对采后椪柑的主要致病菌-指状青霉菌的抑制作用,以及对贮藏期椪柑采后生理的影响。结果显示:竹醋液和壳聚糖对指状青霉菌均有抑制作用,最小抑菌浓度为竹醋液1.5%、壳聚糖0.8%。在椪柑采后贮藏中,用竹醋液、壳聚糖以及二者复合物分别处理果实,以清水处理为对照,通过测定椪柑采后生理生化指标来考察竹醋液壳聚糖复合涂膜对椪柑的保鲜效果。结果表明:与对照相比,各个处理都能够降低采后椪柑的病果率、失重率和丙二醛含量,延缓可溶性固形物、可滴定酸以及Vc含量的下降,以及诱导过氧化物酶活性上升。其中竹醋液壳聚糖复合涂膜效果优于二者单独使用。 展开更多

A case of Meropenem as a novel antibacterial agent to suppress and eliminate Agrobacterium tumefaciens in the Agrobacterium-mediated transformation of orchid protocorm-like bodies (PLBs) has been reported in this article. The in vitro activities of meropenem and four comparator antibacterial agents against three Agrobacterium tumefaciens strains, LBA4404, EHA101, and GV3101, were assessed. In addition, the effect of meropenem on the growth of Dendrobium phalaenopsis PLBs was determined. Compared with other commonly used antibiotics (including ampicillin, carbenicillin, cefotaxime, and cefoperazone), meropenem showed the highest activity in suppressing all tested A. tumefaciens strains (minimum inhibitory concentration [MIC] 〈 0.5 mg L^-1, which is equal to minimum bactericidal concentration [MBC]). Meropenem, at all tested concentrations, except for 10 mg L^-1 concentration, had little negative effect on the growth of orchid tissues. The A. tumefaciens strain EHA101 in genetic transformation with vector plG121Hm in infected PLBs of the orchid was visually undetectable after a two-month subculture in 1/2 MS medium with 50 mg L^-1 meropenem and 25 mg L^-1 hygromacin. The expression and incorporation of the transgenes were confirmed by GUS histochemical assay and PCR analysis. Meropenem may be an alternative antibiotic for the effective suppression of A. tumefaciens in genetic transformation. 展开更多

Gliadin, the major storage protein in endosperm, affects grain quality in spring wheat by its content and composition. Eighteen cultivars differing in HMW-GS were used in the study to approach the accumulation pattern of gliadin fractions α, β, γ, ω and regulation of three kinds of nitrogen source. The results showed that the content of gliadin in grains increased gradually along with the process of grain-filling, but the accumulation intensity and final content differed evidently among cultivars with different HMW-GS composition. Of all the subunit types used here, cultivars with subunits 7+9 and 2+12 had smaller accumulation intensity and lower final content. During grain-filling, 4 gliadin fractions had the same increase trend, but differed in accumulation course. The dynamic trends of gliadin accumulation were similar in different nitrogen treatments whose effects on initial amount, accumulation intensity and final level of accumulation varied with cultivars. Of three nitrogen fertilizer types, the amide-form nitrogen source was better to the formation and accumulation of gliadin as well as its four fractions. 展开更多
