阿仑膦酸钠对去卵巢小鼠骨质量及脏器病理学的影响

目的:探究阿仑膦酸钠对去卵巢小鼠骨质量和脏器病理学的影响。方法:将30只SPF级C57雌性小鼠随机分为3组,每组10只,分别为假手术组(Sham),去卵巢组(OVX)和去卵巢+阿仑膦酸钠组(ALN),ALN组每周阿仑磷酸钠400μg/kg皮下注射2次,Sham组和OV... 目的:探究阿仑膦酸钠对去卵巢小鼠骨质量和脏器病理学的影响。方法:将30只SPF级C57雌性小鼠随机分为3组,每组10只,分别为假手术组(Sham),去卵巢组(OVX)和去卵巢+阿仑膦酸钠组(ALN),ALN组每周阿仑磷酸钠400μg/kg皮下注射2次,Sham组和OVX组每周等体积的生理盐水皮下注射2次,给药12周后取材,分别观察脏器系数、主要脏器病理学切片、骨组织病理学切片、分析micro CT、L4椎体生物力学和血清生化指标。结果:Sham组子宫系数(0.0054±0.0007)显著高于OVX组(0.0026±0.0009)和ALN组(0.0025±0.0007)小鼠的子宫系数,差异具有统计学意义(P<0.05),主要脏器未发现明显的病变和毒副作用。骨组织脱钙切片ALN组与OVX组相比骨小梁结构致密,脂肪细胞较少。micro CT结果显示,ALN组的Tb.BMD、Tb.N、Tb.Th和Tb.BV/TV与OVX组相比,均有显著性增加,但Tb.Sp数值显著性减少,差异具有统计学意义(P<0.05)。L4椎体生物力学中ALN组的弹性模量(50.29±13.43)和最大载荷数(29.83±4.92)显著高于OVX组的弹性模量(14.77±3.12)和最大载荷数(11.57±3.18),差异具有统计学意义(P<0.05)。ALN组血清中骨形成指标OCN和PINP与OVX组相比都呈上升趋势,骨吸收指标TRACP-5b和CTX-I都呈下降趋势,差异具有统计学意义(P<0.05)。结论:阿仑膦酸钠可能是通过增加骨密度、改善骨微结构、提高骨强度、促进骨形成、抑制骨吸收从而提高骨质量,并且对脏器没有明显的毒副作用。 展开更多

Objective:To investigate the effect of alendronate on bone mass and organ pathology of ovariectomized mice.Methods:Thirty SPF grade C57 female mice were randomly divided into three groups(n=10):Sham operation group(Sham),ovariectomized group(OVX)and ovariectomized+alendronate group(ALN).The sodium alendronate was injected subcutaneously at 400μg/kg twice a week in the ALN group.The equal volume of normal saline was injected subcutaneously twice a week in the SHAM group and OVX group.After 12 weeks of drug administration,the samples were taken.The organ coefficients,main organ pathological sections,and bone histopathological sections were observed,and the micro CT,L4 biomechanics and serum biochemical indicators were analyzed.Results:The uterine coefficient of Sham group was(0.0054±0.0007)significantly higher than that of OVX group(0.0026±0.0009)and ALN group(0.0025±0.0007),and the difference was statistically significant(P<0.05).No obvious lesions or toxic or side effects were observed in the main organs.Compared with the OVX group,the ALN group with decalcified sections of bone tissue had compact trabecular structure and fewer adipocytes.Micro-CT results showed that the Tb.BMD,Tb.N,Tb.Th and Tb.BV/TV values of the ALN group were significantly increased compared with those of the OVX group,but the Tb.Sp value was significantly decreased,and the difference was statistically significant(P<0.05).In L4 vertebral body biomechanics,the elastic modulus(50.29±13.43)and maximum load number(29.83±4.92)of ALN group were significantly higher than those of OVX group(14.77±3.12)and maximum load number(11.57±3.18),and the difference was statistically significant(P<0.05).Compared with the OVX group,the serum OCN and PINP indicators of bone formation in the ALN group were increased,while the bone resorption indicators TRACP-5b and CTX-I were decreased,with statistical significance(P<0.05).Conclusion:Alendronate sodium improves bone quality by increasing bone density,improving bone microstructure,increasing bone strength,promoting bone formation and inhibiting bone resorption,without obvious toxic and side effects on organs. 展开更多

Physical oceanography is now entering the eddy-resolving era.Eddies are commonly referred to the so-called mesoscale or submesoscale eddies;by definition,they have horizontal scales from 1 to 500 km and vertical scales from meters to hundreds of meters.In one word,the ocean is a turbulent environment;thus,eddy motions are one of the fundamental aspects of oceanic circulation.Studies of these eddies,including observations,theory,laboratory experiments,and parameterization in numerical models,will be the most productive research frontiers for the next 10 to 20 years,although we have made great efforts to collect data about eddies in the ocean;thus far,we know very little about the three-dimensional structure of these eddies and their contributions to the oceanic general circulation and climate.Therefore,the most important breakthrough may come from observations and physical reasoning about the fundament aspects of eddy structure and their contributions to ocean circulation and climate. 展开更多

In this paper,adiabatic density surface,neutral density surface and potential density surface are compared.The adiabatic density surface is defined as the surface on which a water parcel can move adiabatically,without changing its potential temperature and salinity.For a water parcel taken at a given station and pressure level,the corresponding adiabatic density surface can be determined through simple calculations.This family of surface is neutrally buoyant in the world ocean,and different from other surfaces that are not truly neutrally buoyant.In order to explore mixing path in the ocean,a mixing ratio m is introduced,which is defined as the portion of potential temperature and salinity of a water parcel that has exchanged with the environment during a segment of migration in the ocean.Two extreme situations of mixing path in the ocean are m=0(no mixing),which is represented by the adiabatic density curve,and m=1,where the original information is completely lost through mixing.The latter is represented by the neutral density curve.The reality lies in between,namely,0<m<1.In the turbulent ocean,there are potentially infinite mixing paths,some of which may be identified by using different tracers(or their combinations)and different mixing criteria.Searching for mixing paths in the real ocean presents a great challenge for further research. 展开更多

目的:探讨按揉“肾俞”穴对自然衰老大鼠腰椎间盘退变及相关疼痛的影响。方法:将16只12月龄大鼠随机分为衰老组和推拿组,每组8只;另8只2周龄大鼠作为青年组对照。推拿组接受6个月的按揉“肾俞”穴干预,其他组只做单纯抓取。干预结束后,... 目的:探讨按揉“肾俞”穴对自然衰老大鼠腰椎间盘退变及相关疼痛的影响。方法:将16只12月龄大鼠随机分为衰老组和推拿组,每组8只;另8只2周龄大鼠作为青年组对照。推拿组接受6个月的按揉“肾俞”穴干预,其他组只做单纯抓取。干预结束后,对所有大鼠进行缩足阈值测试与旷场试验,以评估其行为学改变。用X线摄像观察大鼠腰椎并测量椎间盘高度指数(DHI),番红-固绿染色观察大鼠腰椎间盘病理改变,PCR法检测大鼠髓核组织端粒酶活性,免疫荧光法检测大鼠背根神经节(DRG)中胶质纤维酸性蛋白(GFAP)表达,Western blot法检测大鼠DRG中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)蛋白表达。结果:与青年组比较,衰老组大鼠的机械痛阈值和运动能力显著下降(P<0.01),DHI显著降低(P<0.01),腰椎间盘表现出严重的退变表型,髓核组织端粒酶活性显著降低(P<0.01),DRG组织中GFAF、IL-6、TNF-α蛋白表达水平显著增高(P<0.01)。与衰老组比较,推拿组大鼠的机械痛阈值和运动能力显著上升(P<0.01,P<0.05),DHI显著提高(P<0.01),腰椎间盘的退变表型得到了一定程度改善,髓核组织端粒酶活性显著提高(P<0.01),DRG组织中GFAF、IL-6、TNF-α蛋白表达水平显著降低(P<0.01)。结论:按揉“肾俞”穴可能通过延缓老年大鼠腰椎间盘组织衰老,改善其结构和退变程度,从而抑制其相邻DRG中卫星胶质细胞活化并降低炎症因子水平,进而缓解相关疼痛症状。 展开更多
