Intracellular accumulation of tau inhibits autophagosome formation by activating TIA1-amino acid-mTORC1 signaling

Background:Autophagy dysfunction plays a crucial role in tau accumulation and neurodegeneration in Alzheimer’s disease(AD).This study aimed to investigate whether and how the accumulating tau may in turn affect autophagy.Methods:The primary hippocampal neurons,N2a and HEK293T cells with tau overexpression were respectively starved and treated with vinblastine to study the effects of tau on the initiating steps of autophagy,which was analysed by Student’s two-tailed t-test.The rapamycin and concanamycin A were employed to inhibit the mammalian target of rapamycin kinase complex 1(mTORC1)activity and the vacuolar H+-ATPase(v-ATPase)activity,respectively,which were analysed by One-way ANOVA with post hoc tests.The Western blotting,co-immunoprecipitation and immunofuorescence staining were conducted to gain insight into the mechanisms underlying the tau effects of mTORC1 signaling alterations,as analysed by Student’s two-tailed t-test or One-way ANOVA with post hoc tests.The autophagosome formation was detected by immunofuorescence staining and transmission electron microscopy.The amino acids(AA)levels were detected by high performance liquid chromatography(HPLC).Results:We observed that overexpressing human full-length wild-type tau to mimic AD-like tau accumulation induced autophagy deficits.Further studies revealed that the increased tau could bind to the prion-related domain of T cell intracellular antigen 1(PRD-TIA1)and this association significantly increased the intercellular level of amino acids(Leucine,P=0.0038;Glutamic acid,P=0.0348;Alanine,P=0.0037;Glycine,P=0.0104),with concordant upregulation of mTORC1 activity[phosphorylated eukaryotic translation initiation factor 4E-binding protein 1(p-4EBP1),P<0.0001;phosphorylated 70 kD ribosomal protein S6 kinase 1(p-p70S6K1),P=0.0001,phosphorylated unc-51-like autophagyactivating kinase 1(p-ULK1),P=0.0015]and inhibition of autophagosome formation[microtubuleassociated protein light chain 3 II(LC3 II),P=0.0073;LC3 puncta,P<0.0001].As expected,this tau-induced deficit of autophagosome formation in turn aggravated tau accumulation.Importantly,we also found that blocking TIA1 and tau interaction by overexpressing PRD-TIA1,downregulating the endogenous TIA1 expression by shRNA,or downregulating tau protein level by a small proteolysis targeting chimera(PROTAC)could remarkably attenuate tau-induced autophagy impairment.Conclusions:Our findings reveal that AD-like tau accumulation inhibits autophagosome formation and induces autophagy deficits by activating the TIA1/amino acid/mTORC1 pathway,and thus this work reveals new insight into tau-associated neurodegeneration and provides evidence supporting the use of new therapeutic targets for AD treat-ment and that of related tauopathies. 展开更多

目的:研究柚皮素(Nar)及其磷脂复合物(NPC)对叔丁基氢过氧化物(t-BHP)诱导产生的氧化损伤人视网膜色素上皮细胞(ARPE-19细胞)的保护作用及其作用机制。方法:根据文献最佳制作工艺制备NPC。将体外培养的ARPE-19细胞分为溶媒组(用DMSO培养... 目的:研究柚皮素(Nar)及其磷脂复合物(NPC)对叔丁基氢过氧化物(t-BHP)诱导产生的氧化损伤人视网膜色素上皮细胞(ARPE-19细胞)的保护作用及其作用机制。方法:根据文献最佳制作工艺制备NPC。将体外培养的ARPE-19细胞分为溶媒组(用DMSO培养)、模型组(用200μmol/L t-BHP干预)、Nrf2-siRNA组(针对Nrf2基因进行细胞转染)、柚皮素组(200μmol/L柚皮素培养基预处理后加入200μmol/L t-BHP)、NPC组(200μmol/L NPC培养基预处理后加入200μmol/L t-BHP)、Nrf2-siRNA+柚皮素组(200μmol/L柚皮素预处理后Nrf2基因干扰,再加入200μmol/L t-BHP)、Nrf2-siRNA+NPC组(200μmol/L NPC预处理后Nrf2基因干扰,再加入200μmol/L t-BHP)。检测各组细胞内活性氧、丙二醛、超氧化物歧化酶、总抗氧化能力水平,分别采用RT-PCR和Western blot法检测各组细胞内血红素加氧酶-1(HO-1)、醌氧化还原酶-1(NQO-1)、谷氨酸半胱氨酸连接酶(GCL)和核因子E2相关因子2(Nrf2)的表达水平。结果:NPC较柚皮素能明显降低ARPE-19细胞内丙二醛、活性氧的含量,提高细胞内超氧化物歧化酶、总抗氧化能力水平。柚皮素和NPC预保护ARPE-19细胞后,Nrf2、HO-1、NQO-1和GCL mRNA的相对表达量和蛋白表达水平均较模型组和Nrf2-siRNA组升高。柚皮素组与NPC组,Nrf2-siRNA+柚皮素组与Nrf2-siRNA+NPC组细胞内4种基因和蛋白相对表达水平均有差异。Nrf2-siRNA+柚皮素组与Nrf2-siRNA组Nrf2、HO-1、NQO-1蛋白表达无显著差异。Nrf2-siRNA+NPC组与Nrf2-siRNA组4种蛋白表达均有差异,NPC作用明显强于柚皮素。结论:柚皮素磷脂复合物能明显提高细胞内抗氧化能力,降低氧化水平,其通过激活Nrf2/ARE抗氧化应激通路上调Nrf2及其下游抗氧化酶和Ⅱ相解毒酶的表达对氧化损伤的ARPE-19细胞起到更好的保护作用。 展开更多

目的通过口服红花逍遥片联合炎症肿块内注射曲氨耐德治疗肉芽肿乳腺炎,分析控制炎症表现的临床及影像学相关指标,探讨红花逍遥片在中西医结合治疗肉芽肿乳腺炎中的价值。方法选取2015年3月—2018年3月石家庄市第四医院住院治疗的肉芽肿... 目的通过口服红花逍遥片联合炎症肿块内注射曲氨耐德治疗肉芽肿乳腺炎,分析控制炎症表现的临床及影像学相关指标,探讨红花逍遥片在中西医结合治疗肉芽肿乳腺炎中的价值。方法选取2015年3月—2018年3月石家庄市第四医院住院治疗的肉芽肿乳腺炎患者266例作为研究对象。按随机数字表法分为研究组和对照组,每组133例。对照组仅予以炎症肿块内注射曲氨耐德注射液50 mg,2次/周,连续1个月;研究组在对照组的基础上,口服红花逍遥片1.56 g,3次/d,连续治疗1个月。比较两组治疗后疼痛评分、下肢红斑、肿块体积、彩超影像学回声变化、彩超血流变化、穿刺脓血情况、C反应蛋白下降情况等相关指标变化。结果治疗后研究组疼痛评分与对照组比较,差异有统计学意义(P<0.05);研究组治疗后下肢红斑消退时间、肿块体积缩小程度与对照组比较,差异有统计学意义(P<0.05),研究组低于对照组;研究组治疗后炎症肿块彩超影像学回声均匀程度、炎症肿块彩超血流信号减少程度与对照组比较,差异有统计学意义(P<0.05),研究组优于对照组;研究组治疗后炎症肿块穿刺抽出的脓血黏稠度、C反应蛋白水平与对照组比较,差异有统计学意义(P<0.05),研究组低于对照组。结论红花逍遥片口服联合炎症肿块内注射曲氨耐德治疗肉芽肿乳腺炎,控制临床炎症的各项指标研究组均优于对照组,表明红花逍遥片在中西医结合治疗肉芽肿乳腺炎时,其疏肝理气、降解乳络瘀血、痰核、湿热的功效可以大大提高西医治疗功能,值得临床推广。 展开更多

新发糖尿病与胰腺癌的相关临床研究

背景胰腺癌(pancreatic carcinoma,PCa)是发生于胰腺外分泌腺的恶性肿瘤,起病隐匿,恶性程度高,5年生存率低于1%,早期临床症状常常不典型,且缺少特异的检查手段,发现时大多已进展至疾病的中晚期,预后较差.大量研究表明糖尿病(diabetes me... 背景胰腺癌(pancreatic carcinoma,PCa)是发生于胰腺外分泌腺的恶性肿瘤,起病隐匿,恶性程度高,5年生存率低于1%,早期临床症状常常不典型,且缺少特异的检查手段,发现时大多已进展至疾病的中晚期,预后较差.大量研究表明糖尿病(diabetes mellitus,DM)与PCa密切相关,但两者因果关系不甚明了.本研究试图对PCa患者进行回顾性分析,以期能发现PCa的早期诊断线索.目的探讨PCa与新发DM的关系.方法以463例PCa患者为PCa组,以同期非肿瘤、非消化系统、非激素代谢异常相关疾病的658例患者作为对照组,分析比较PCa组与对照组患者DM的发病年龄和病程.结果PCa组新发DM患者平均患病年龄70.09岁(95%CI:69.00-71.18),2型DM患者平均患病年龄58.68岁(95%CI:57.84-59.53),差异有显著统计学意义(P<0.01).将PCa组患者按DM病程分为七组.PCa组DM病程<1年、DM病程2-1年及>10年者DM的患病率显著高于对照组(19.87%vs 1.52%,P<0.01)(6.48%vs 1.67%,P<0.05)(2.16%vs 0.91%,P<0.05).结论新发DM可能是PCa早期诊断的线索.临床医生对于2年内新发诊断DM或短时间血糖波动较大的老年患者要高度警惕PCa患病的可能性. 展开更多

Objective To observe the application of continuous renal replacement therapy(CRRT) and heparin anticoagulation in patients with HFRS, and to explore a more suitable anticoagulant strategy. Methods Eighty-five severe-type patients(severe group) and 71 critical-type patients(critical group) were enrolled in this study. The frequency of CRRT was compared between the two groups; the frequency of CRRT treated with and without heparin anticoagulation and the frequency of hemorrhage and channel blood clotting induced by the two anticoagulant strategies were observed. Results The frequency of CRRT in the critical group was higher than that in the severe group(P < 0.001). The frequency of CRRT initiated during the overlapping phases in the critical group was significantly higher than that of the severe group(P = 0.032). The total times of CRRT was 103, and 70 of them were treated with heparin anticoagulation. The frequencies of hemorrhage induced by heparin anticoagulation and no heparinization were 16 and 0, respectively, and the frequencies of channel blood clotting were 2 and 4, respectively. Conclusions CRRT has been used extensively in the critical-type patients with HFRS. The heparin anticoagulation and no anticoagulant strategies should be used more rationally in patients treated with CRRT, according to the clinical characteristics of the disease. 展开更多

Effective suppression of beta oscillation in Parkinsonian state via a noisy direct delayed feedback control scheme

This work explores the function of the noisy direct delayed feedback(NDDF)control strategy in suppressing the pathological oscillations in the basal ganglia(BG)with Parkinson’s disease(PD).Deep brain stimulation(DBS)alleviates the PD state fantastically.However,due to its unclear mechanism and open-loop characteristic,it is challenging to further improve its effects with lower energy expenditure.The noise stimulus performs competitively in alleviating the PD state theoretically,but it cannot adapt to the neural condition timely and automatically due to its open-loop control scheme.The direct delayed feedback(DDF)control strategy is able to disturb excessive synchronous effectively.Therefore,the NDDF control strategy is proposed and researched based on a BG computational model,which can reflect the intrinsic properties of the BG neurons and their connections with thalamic neurons.Simulation results show that the NDDF control strategy with optimal parameters is effective in removing the pathological beta oscillations.By comparison,we find the NDDF control strategy performs more excellent than DDF in alleviating PD state.Additionally,we define the multiple-NDDF control strategy and find that the multiple-NDDF with appropriate parameters performs better than NDDF.The obtained results contribute to the cure for PD symptoms by optimizing the noise-induced improvement of the BG dysfunction. 展开更多

SrLi_(2)Ti_(6)O_(14)(SLTO)coated with different amount of ZrO_(2)was successfully prepared.The as-obtained composites are stacked by a series of particles with a pure phase structure and a good crystallinity.Furthermore,ZrO_(2)coating not only enhances the structural stability of the materials but also facilitates the diffusion of lithium through the SEI film.As a result,the redox polarization was reduced,and the reversibility of the electrochemical reaction was enhanced.Particularly,SLTO-ZrO_(2)-2 sample delivers a high initial lithiation capacity of 283.6 mA h g^(-1),and the values maintain at 251.7,228.0,207.4,175.3,and 147.7 mA h g^(-1)at the current densities of 0.13,0.26,0.54,1.31,and 2.62 A g^(-1),respectively.Our experiment also confirmed that SLTO materials coated with ZrO_(2)are suitable for high power density applications,and the lithiation specific energy efficiency of SLTO-ZrO_(2)-2 is 200%as high as that of pure SLTO at a power density of 1257 W kg^(-1). 展开更多

BACKGROUND An intrauterine device(IUD)is a commonly used contraceptive among women in China.It is widely used because it is safe,effective,simple,economic,and reversible.Among the possible complications,an ectopic IUD in the bladder is rare.It occurs insidiously,has a long course,is associated with a high risk for injury,and is difficult to treat.CASE SUMMARY A 44-year-old woman was admitted for repeated episodes of urinary frequency,urgency,and dysuria over three months.Laboratory tests revealed significantly elevated urine leukocytes and bacteria.Urine culture suggested colonization with Enterococcus faecalis.Abdominal computed tomography images suggested an abnormally positioned IUD that was protruding into the bladder.Cystoscopy revealed a metallic foreign body with multiple stones on its surface in the left posterior bladder wall.The foreign body measured approximately 1 cm.Hysteroscopy revealed the arm of a V-type metal IUD embedded in the middle and upper sections of the anterior wall of the cervical canal.The majority of the IUD was located in the uterine cavity.Cystoscopy was performed,and a holmium laser was utilized to break the stones attached to the portion of the IUD in the bladder.The IUD was then removed through hysteroscopy.CONCLUSION Ectopic IUDs in the bladder can be diagnosed with thorough imaging and safely removed through cystoscopy or hysteroscopy. 展开更多

BACKGROUND Hematuria is one of the most common clinical symptoms for urologists and is typically observed in urinary system tumors,prostate hyperplasia,and urinary stone disease.Hematuria due to vesical varices is very rare,and only a few cases have been reported since 1989.We report the first case of vesical varices due to portal hypertension with aberrant development and functioning of the genitourinary system along with the complete diagnosis and treatment process.CASE SUMMARY This patient was a 53-year-old man with a history of aberrant development of the genitourinary system and hepatitis B-associated cirrhosis.He was admitted to the emergency department with severe hematuria and bladder clot tamponade.Many abnormally dilated blood vessels were found surrounding the bladder in the pelvis by color Doppler ultrasound,contrast-enhanced computed tomography,and three-dimensional visualization technology.It was difficult to perform transurethral cystoscopy and hemostasis in this patient,so we performed open surgical bladder exploration for hemostasis and surgical devascularization around the bladder.CONCLUSION Urologists should improve the understanding of the pathophysiology,clinical manifestations,diagnosis,and treatment of vesical varices.This case may be presented as a reference for the diagnosis and management of severe hematuria due to vesical varices. 展开更多

Objective:To investigate the neuroprotective effect and mechanism of Acteoside(AC)on brain injury in epileptic rats.Methods:30 Sprague-Dawlay rats(SPF grade,healthy males,about 200g)were randomly divided into epilepsy group,AC group and blank control group.The epileptic group and the AC group were given 28 days of intraperitoneal injection of pentaerythritazine(PTZ),and the remaining groups were given normal saline injection.The changes of electroencephalogram(EEG),learning and memory ability,pathological changes of brain tissue,superoxide dismutase(SOD)and malondialdehyde(MDA)were observed in all rats.Results:Compared with the epilepsy group,the AC group showed significantly reduced degeneration and necrosis of nerve cells in brain tissue,significantly improved learning and memory ability,and the EEG was dominated by wavelet and slow wave,replacing the typical spike and spike waves.SOD increased and MDA decreased in oxygen radical(P<0.05).Conclusions:Epilepsy are closely related to Oxidative stress.AC can play a protective role in the brain by improving EEG,learning and memory ability,regulating changes in SOD and MDA content,and inhibiting the generation of oxygen free radicals. 展开更多

Zn_(3)V_(2)O_(8) was considered as a promising anode material for lithium-ion battery(LIB),because of its high theoretical specific capacity,environmental friendliness,and ease of availability.However,the large volume change and low electronic conductivity of Zn_(3)V_(2)O_(8)in repeated charge/discharge cycles have severely limited its applications.To solve the above issues,hierarchical Zn_(3)V_(2)O_(8) microspheres assembled by two-dimensional(2D)nanosheets were successfully synthesized,and carbon nanotubes(CNTs)were further introduced to cross-link the Zn_(3)V_(2)O_(8) microspheres.The interconnected nature of the three-dimensional(3D)conducting network and the special hierarchical morphology were beneficial for improving the stability and conductivity of the composite,leading to the reduction of the impedance and a significant improvement of the electrochemical performance.The reversible capacity of the as-prepared composite can achieve 1049.5mAh·g^(-1)at a current density of 0.2 A·g^(-1)with a capacity retention of~81%after 100 cycles.It is suggested that morphology modulation coupled with interconnecting CNT network is an effective method to boost the electrochemical performance of the anode materials for lithium-ion batteries. 展开更多

目的:探讨人参皂苷Rg1对高血糖所致心肌损害的保护作用及其机制。创新点:使用糖尿病大鼠为实验对象,探讨三种浓度的人参皂苷Rg1对糖尿病心肌损伤的保护作用及其机制,检测其是否具有浓度依赖性。方法:将60只Wistar大鼠随机分组,其中空白... 目的:探讨人参皂苷Rg1对高血糖所致心肌损害的保护作用及其机制。创新点:使用糖尿病大鼠为实验对象,探讨三种浓度的人参皂苷Rg1对糖尿病心肌损伤的保护作用及其机制,检测其是否具有浓度依赖性。方法:将60只Wistar大鼠随机分组,其中空白对照组10只,另50只给予高脂高糖饲养,4周后腹腔注射40 mg/kg链脲佐菌素(STZ)。成功制备糖尿病大鼠模型40只,再随机分为糖尿病模型组,糖尿病大鼠+低剂量人参皂苷Rg1(10 mg/(kg·d)),糖尿病大鼠+中剂量人参皂苷Rg1(15 mg/(kg·d)),糖尿病大鼠+高剂量人参皂苷Rg1(20 mg/(kg·d))。12周后处死大鼠,取血测定空腹血糖、总胆固醇(TC)、甘油三酯(TG)、心肌酶及氧化应激水平,留取心肌组织使用透射电镜观察心肌细胞超微结构改变,应用TUNEL法检测心肌细胞凋亡,免疫组化检测细胞凋亡相关蛋白半胱氨酸天冬氨酸蛋白酶3(CASP3)和Bcl-x L的表达。结论:人参皂苷Rg1对糖尿病大鼠糖脂代谢无明显影响,人参皂苷Rg1可降低糖尿病大鼠血清肌钙蛋白(c Tn I)和肌酸激酶同工酶(CK-MB)水平,改善心肌细胞超微结构,减少心肌细胞凋亡,降低大鼠血清和心肌组织中丙二醛(MDA)含量,提高超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH)水平,降低凋亡蛋白CASP3的表达,同时提高Bcl-x L蛋白表达。总之,人参皂苷Rg1能显著保护糖尿病大鼠心肌损伤,其机制可能与其抗氧化及抗细胞凋亡作用有关。 展开更多

Transition-metal oxides have attracted increased attention in the application of high-performance lithium ion batteries(LIBs), owing to its higher reversible capacity,better structural stability and high electronic conductivity.Herein, CoWO4 nanoparticles wrapped by reduced graphene oxide(CoWO4–RGO) were synthesized via a facile hydrothermal route followed by a subsequent heat-treatment process. When evaluated as the anode of LIB, the synthetic CoWO4–RGO nanocomposite exhibits better Li^+ storage properties than pure CoWO4 nanostructures synthesized without graphene oxide(GO). Specifically, it delivers a high initial specific discharge capacity of1100 mAh·g^-1 at a current density of 100 mA·g^-1, and a good reversible performance of 567 mAh·g^-1 remains after the 100th cycle. Moreover, full battery using CoWO4–RGO as anode and commercial LiCoO2 powder as cathode was assembled, which can be sufficient to turn on a 3 V,10 mW blue light emitting diode(LED). The enhanced electrochemical performance for lithium storage can be attributed to the three-dimensional(3D) structure of the CoWO4–RGO nanocomposite, which can accommodate huge volume changes, and synergetic effect between CoWO4 and reduced graphite oxide(RGO) nanosheets,including an increased conductivity, shortened Li^+ diffusion path. 展开更多

Impaired brain glucose metabolism is an early indicator of Alzheimer’s disease(AD);however,the fundamental mechanism is unknown.In this study,we found a substantial decline in isocitrate dehydrogenase 3β(IDH3β)levels,a critical tricarboxylic acid cycle enzyme,in AD patients and AD-transgenic mice’s brains.Further investigations demonstrated that the knockdown of IDH3βinduced oxidation-phosphorylation uncoupling,leading to reduced energy metabolism and lactate accumulation.The resulting increased lactate,a source of lactyl,was found to promote histone lactylation,thereby enhancing the expression of paired-box gene 6(PAX6).As an inhibitory transcription factor of IDH3β,the elevated PAX6 in turn inhibited the expression of IDH3β,leading to tau hyperphosphorylation,synapse impairment,and learning and memory deficits resembling those seen in AD.In AD-transgenic mice,upregulating IDH3βand downregulating PAX6 were found to improve cognitive functioning and reverse AD-like pathologies.Collectively,our data suggest that impaired oxidative phosphorylation accelerates AD progression via a positive feedback inhibition loop of IDH3β-lactate-PAX6-IDH3β.Breaking this loop by upregulating IDH3βor downregulating PAX6 attenuates AD neurodegeneration and cognitive impairments. 展开更多
