Objective:To investigate the effects of melatonin on renal inflammation,oxidative stress,apoptosis,as well as DNA and tissue damage in acrylamide-induced nephrotoxicity in rats.Methods:Fifty male rats were randomly divided into five groups.The control group received distilled water by gastric lavage for 11days and the acrylamide group was administered acrylamide(50 mg/kg,i.g.)for 11 days.The MEL10+ACR and MEL20+ACR groups received intraperitoneal melatonin 10 and 20 mg/kg,respectively,for 11 days,and acrylamide(50 mg/kg,i.g.)was administered 1h after melatonin injection.The MEL20 group was injected with melatonin(20 mg/kg)for 11 days.Kidney function tests were performed and biochemical and inflammatory parameters were determined.In addition,histopathological,immunohistochemical,and immunofluorescence examinations were carried out.Results:Melatonin significantly abated acrylamide-induced rise in serum urea and creatinine levels.Acrylamide caused oxidative stress,inflammation,apoptosis,as well as DNA and tissue damage in the kidneys.Melatonin treatment alleviated acrylamide-induced renal damage by exhibiting antioxidant,anti-inflammatory,and antiapoptotic effects.Moreover,melatonin significantly ameliorated acrylamide-caused histopathological changes in kidney tissue.Conclusions:Melatonin attenuates acrylamide-induced renal oxidative stress,inflammation,apoptosis,and DNA damage in rats. 展开更多

In this study,the effect of welding parameters on the microstructure and mechanical properties of the dissimilar resistance spot welded DP1000–QP1180 joints was investigated.Heat affected zone(HAZ)width of QP1180 side was smaller than that of DP1000 side.HAZ width and indentation depth increased with increasing welding current and welding time.The nugget size increased with increasing welding current whereas it increased at lower currents and decreased at higher currents with increasing welding time.The lowest hardness was on the DP1000 side.On the QP1180 side,the center of HAZ had the peak hardness.With increasing welding current,hardness values throughout the weld zone decreased and the tensile shear load increased.At lower welding currents,the welding time did not affect the tensile shear load.Tensile elongation decreased with the increase of welding time,whereas there is no relationship between the welding current and elongation.The spot-welded joints having higher strength exhibited a more ductile fracture characteristic. 展开更多

Objective:Active surveillance(AS)offers a strategy to reduce overtreatment and now is a widely accepted treatment option for low-risk prostate cancer.An ideal tool for risk-stratification would detect aggressive cancers and exclude such men from taking up AS in the first place.We evaluate if a combination of transperineal template biopsy with magnetic resonance imaging(MRI)-targeted biopsy identifies significant prostate cancer amongst men initially diagnosed with low-risk prostate cancer.Methods:This prospective,single-blinded study included men with low-risk prostate cancer(D’Amico’s Criteria)diagnosed on conventional transrectal ultrasound-guided biopsy.Patients first underwent multiparametric MRI of the prostate6 weeks after initial biopsy.Each suspicious lesion is mapped and assigned a Prostate Imaging Reporting and Data System(PIRADS)score.Template biopsy is first performed with the surgeon blinded to MRI findings followed by MRI-targeted biopsy using a robotic transperineal biopsy platform.Results:The age of the 19 men included is 65.4±4.9 years(mean±SD).Prostate specific antigen(PSA)at diagnosis and at the time of transperineal biopsy were comparable(7.3±1.7 ng/mL and 7.0±1.8 ng/mL,p Z 0.67),so were prostate volumes(34.2±8.9 mL and 32.1±13.4 mL,p Z 0.28).MRI-targeted biopsy had a higher percentage of cancer detection per core compared to template biopsy(11.7%vs.6.5%,p Z 0.02),this was more than 3 times superior for Gleason 7 disease(5.9%vs.1.6%,p<0.01).Four of 18(22.2%)patients with MRI lesions had significant disease with MRI-targeted biopsy alone.Three of 19 patients(15.8%)had significant disease with template biopsy alone.In combination,both techniques upclassified five patients(26.3%),all of whom underwent radical prostatectomy.Whole mount histology confirmed tumour location and grade.All six patients with PIRADS 5 lesions had cancer detected(66.6%significant disease).Conclusion:A combination of MRI-targeted and template biopsy may optimally risk-classify“low-risk”patients diagnosed on initial conventional transrectal ultrasonography(TRUS)prostate biopsy. 展开更多

Testicular metastasis is rare with the prostate being the most common site of primary cancer.We report a case of a 72-year-old man with castration-resistant prostate cancer(CRPC)and known metastases to bone and lymph nodes,who developed bilateral painful swollen testes 3 years after the initial diagnosis of prostate cancer.He had first presented with lower urinary tract symptoms(LUTS)with suspicious findings on digital rectal examination of the prostate,and an elevated serum prostate specific antigen(PSA)level of 129 ng/mL.Transrectal prostate biopsy revealed Gleason 4þ5 adenocarcinoma.Radiological staging showed locally advanced prostate cancer with extensive metastases to bone and pelvic and retroperitoneal lymph nodes.He was given hormonal therapy for over 2 years until progression to CRPC.Six months later he developed painful bilateral testicular swellings,and serum markers for testicular germ cell cancer were normal.Bilateral orchiectomy was performed,showing metastatic prostate cancer(Gleason 4þ5)on histology.One month postoperatively his PSA level dropped to 0.1 ng/mL from a presurgery level of 6.24 ng/mL. 展开更多

Sequence analysis of Turkish field strains of bovine torovirus shows unique amino acid changes in the partial M gene

Objective: To investigate the presence, prevalence and phylogenetic classification of bovine torovirus(BToV). Methods: Stool samples from 72 calves, which were negative for primary gastroenteritis agents, were examined with the nested PCR method by using BToV M genespecific primers. Results: BToV was detected in 12(16.7%) out of 72 samples. Phylogenetic analysis was performed using nucleotide and amino acid sequences. In the phylogenetic tree, European, American, Far East and Turkish strains were found to be divided into different branches. Interestingly, it was observed that Turkish strains were divided into two subgroups. Considering the amino acid sequences of these strains having differences at nucleotide level, the change at the 3rd amino acid of the partial M gene in Turkish strains has made Turkish strains different from all other strains. Similarly, the differences were observed in the 18th, 20th, 63rd and 93rd amino acids of the partial M gene only in Turkish field strains. Conclusions: This study revealed that Turkish strains of BToV constitute a separate phylogenetic group and can be divided into two subgroups. In addition, BToV was found to be a common pathogen causing diarrhea in calves in Turkey, and it is a necessity to consider BToV in cases of diarrhea with unknown cause. 展开更多

The aim of this study is analyzed in detail for better understanding of energy and power of an aero-engine. In this regard, this study presents energy equations were applied to the turbofan engine components. The engine has a thrust range of 82 to 109 kN. It consists of fan, axial low pressure compressor (LPC), axial high pressure compressor (HPC), an annular combustion chamber, high-pressure turbine (HPT) and low pressure turbine (LPT). The results show that power of the engine flow approaches a maximum value to be 82.85 MW in the combustor outlet, while minimum power is observed at LPC inlet with the value of 1.37 MW. Furthermore, important parameters of the engine are also analyzed from reverse-engineering method. It is expected that results of this study will be beneficial of power, cogeneration and aero-propulsive generation systems in similar environment. 展开更多
