The potential mechanism of Isodon suzhouensis against COVID-19 via EGFR/TLR4 pathways

Corona Virus Disease 2019(COVID-19)has brought the new challenges to scientific research.Isodon suzhouensis has good anti-inflammatory and antioxidant stress effects,which is considered as a potential treatment for COVID-19.The possibility for the treatment of COVID-19 with I.suzhouensis and its potential mechanism of action were explored by employing molecular docking and network pharmacology.Network pharmacology and molecular docking were used to screen drug targets,and lipopolysaccharide(LPS)induced RAW264.7 and NR8383 cells inflammation model was used for experimental verification.Collectively a total of 209 possible linkages against 18 chemical components from I.suzhouensis and 1194 COVID-19 related targets were selected.Among these,164 common targets were obtained from the intersection of I.suzhouensis and COVID-19.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enriched 582 function targets and 87 target proteins pathways,respectively.The results from molecular docking studies revealed that rutin,vitexin,isoquercitrin and quercetin had significant binding ability with 3 chymotrypsin like protease(3CLpro)and angiotensin converting enzyme 2(ACE2).In vitro studies showed that I.suzhouensis extract(ISE)may inhibit the activation of PI3K/Akt pathway and the expression level of downstream proinflammatory factors by inhibiting the activation of epidermal growth factor receptor(EGFR)in RAW264.7 cells induced by LPS.In addition,ISE was able to inhibit the activation of TLR4/NF-κB signaling pathway in NR8383 cells exposed to LPS.Overall,the network pharmacology and in vitro studies conclude that active components from I.suzhouensis have strong therapeutic potential against COVID-19 through multi-target,multi-pathway dimensions and can be a promising candidate against COVID-19. 展开更多

Ferritin has good thermal stability,resistance to certain acids and bases,and targeting,and has broad application prospects in the synthesis of gold nanostars(AuNS).In this study,we screened monodisperse AuNS with uniform particle size and morphology through a one-step synthesis method and coupled the synthesized AuNS with oyster ferritin(GF1).The results showed that the surface plasmon resonance(SPR)peaks of the coupled GF1@AuNS changed signifi cantly,and the changes in infrared spectra and potential confirmed the success of the synthesis,while the microscopic morphology showed an increase in particle size and surface peak coverage.Furthermore,GF1@AuNS does not induce cell death in the 100µmol/L range,is highly stable in physiological environments,and exhibits good X-ray attenuation in micro-computed tomography.Due to the unique functional activity of ferritin and AuNS,GF1@AuNS has potential applications in food detection and drug development in the future. 展开更多

Slip distribution inversion of seismic sub-fault dip iteration using gradient based optimizer algorithm

This paper considers setting different dips for different sub-faults to fit the actual rupture situation based on the fault rupture of the 2013 Lushan M_(S)7.0 earthquake.Meanwhile,combined with the coseismic GNSS data of the Lushan earthquake,the source parameters and sliding distribution of the Lushan earthquake fault are inversed.Firstly,we use the gradient based optimizer(GBO)in nonlinear inversion to obtain the source parameters of this seismic fault.The inversion results indicate that the strike of the fault is 206.52°,the dip is 44.10°,the length is 21.92 km,and the depth is 12.79 km.To refine the sliding distribution of the seismic fault,the seismic fault is divided into 3×3 sub-faults.Then,we fix the central sub-fault dip of 44.10°;the dip of other sub-faults is obtained by iteration.After that,the model is further divided into a fault layer model composed of 23×19 sub fault slices,and using the Matlab fitting function is used to fit the dip of the 23×19 sub faults.Finally,the Lushan seismic fault plane is established as a shovel structure with steep upper and gentle lower,steep south and gentle north.The slip distribution inversion results indicate that the depth of the slip peak is 13 km,the corresponding maximum slip momentum is 0.67 m,the seismic moment is 1.10×10^(19)N·m and the corresponding moment magnitude is MW6.66.The results above are consistent with the research results of seismology. 展开更多

Understanding the doping evolution from a Mott insulator to a superconductor probably holds the key to resolve the mystery of unconventional superconductivity in copper oxides. To elucidate the evolution of the electronic state starting from the Mott insulator, we dose the surface of the parent phase Ca_(2)CuO_(2)Cl_(2) by depositing Rb atoms, which are supposed to donate electrons to the CuO_(2) planes underneath. We successfully achieved the Rb sub-monolayer thin films in forming the square lattice. The scanning tunneling microscopy or spectroscopy measurements on the surface show that the Fermi energy is pinned within the Mott gap but close to the edge of the charge transfer band. In addition, an in-gap state appears at the bottom of the upper Hubbard band(UHB), and the Mott gap will be significantly diminished. Combined with the Cl defect and the Rb adatom/cluster results, the electron doping is likely to increase the spectra weight of the UHB for the double occupancy. Our results provide information to understand the electron doping to the parent compound of cuprates. 展开更多

BACKGROUND The development of tumor is closely linked to inflammation.Therefore,targeting molecules involved in inflammation may be effective in predicting cancer prognosis.Transarterial chemoembolization(TACE)holds significant therapeutic significance in addressing hepatocellular carcinoma(HCC).At present,no studies have evaluated the predictive value ofγ-glutamyl transferase to albumin ratio(GAR)on the prognosis of HCC undergoing TACE.AIM To explore the potential prognostic significance of the GAR in individuals undergoing TACE for HCC.METHODS A total of 1231 patients from seven hospitals in China were randomized into a training cohort(n=862)and a validation cohort(n=369).To establish inde pendent prognostic factors for overall survival(OS),we utilized multivariate and univariate Cox regression models.The best cut-off value of the GAR was determined with the X-tile software,with OS as the basis.Validations were performed using dual therapy cohort and triple therapy cohort.RESULTS X-tile software revealed a GAR threshold of 4.75 as optimal.Both pre-and post-propensity score matching analyses demonstrated that the median OS in the low-GAR group(<4.75)was notably longer compared to the high-GAR group(≥4.75),showing results of 26.9 vs 9.8 months(P<0.001)initially,and 18.1 vs 11.3 months(P<0.001)after match.Furthermore,multivariate analysis identified GAR≥4.75 as an independent prognostic factor(P<0.001).The receiver operating characteristic curves for the nomogram showed area under receiver operating characteristic curves of 0.741,0.747,and 0.708 for predicting 1-,2-,and 3-year survival,respectively.Consistent findings were reiterated in the two cohorts involving TACE in combination with targeted therapy and TACE in combination with targeted therapy and immunotherapy.Calibration curve and decision curve analyses substantiated the model’s relatively robust predictive capabilities.CONCLUSION Our study validates the effective prognostic capacity of the GAR-based nomogram for HCC patients undergoing TACE or TACE in combination with systemic therapy. 展开更多

目的:探究自体富血小板血浆(Platelet rich plasma,PRP)注射联合自体表皮移植及白芍总苷治疗稳定期白癜风的疗效及对美学效果的影响。方法:回顾性分析2020年6月-2023年1月衡水市第二人民医院收治的205例稳定期白癜风患者的临床资料,按... 目的:探究自体富血小板血浆(Platelet rich plasma,PRP)注射联合自体表皮移植及白芍总苷治疗稳定期白癜风的疗效及对美学效果的影响。方法:回顾性分析2020年6月-2023年1月衡水市第二人民医院收治的205例稳定期白癜风患者的临床资料,按治疗方法及入院时间的不同分成对照组(100例,2020年6月-2021年9月)和实验组(105例,2021年10月-2023年1月)。对照组予以自体表皮移植术联合白芍总苷治疗,在此基础上实验组予以PRP注射治疗,时长为3个月。比较两组疗效、白斑改善情况[白癜风面积评分指数(Vitiligo area score index,VASI)、色素积分]、皮肤病生活质量指数(Dermatology life quality index,DLQI)、美学效果、免疫功能(CD4^(+)、CD8^(+)、CD4^(+)/CD8^(+))、氧化应激指标[超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)]、不良反应发生率和复发率的差异。结果:治疗后,实验组的总有效率高于对照组(P<0.05);治疗3个月及随访6个月后,两组DLQI评分均较治疗前降低,且实验组低于对照组(P<0.05),随访6个月后实验组的美学效果评分高于对照组(P<0.05);两组CD4^(+)、CD4^(+)/CD8^(+)、SOD、CAT水平均较治疗前升高,CD8^(+)、MDA水平均较治疗前降低,且实验组变化幅度优于对照组(P<0.05);两组VASI评分均治疗前降低,色素沉着积分较治疗前升高,且实验组变化幅度优于对照组(P<0.05);治疗期间两组不良反应发生率差异无统计学意义(P>0.05);随访6个月后实验组的疾病复发率低于对照组(P<0.05)。结论:PRP注射、自体表皮移植术联合白芍总苷治疗稳定性期白癜风患者的疗效显著,能有效提高免疫功能,减轻氧化应激反应,促进白斑消退,改善生活质量及美学效果,降低疾病复发率,且安全性高。 展开更多

为研究电子束退火对Li-N共掺杂Zn O薄膜性能的影响,首先利用溶胶-凝胶旋涂法在p型Si(111)衬底上制备Li-N共掺杂的Zn O前驱膜,然后用电子束对前驱膜进行退火。退火时,电子束加速电压10 k V,退火时间5 min,聚焦束流123 m A,束流为0.7~1.9 ... 为研究电子束退火对Li-N共掺杂Zn O薄膜性能的影响,首先利用溶胶-凝胶旋涂法在p型Si(111)衬底上制备Li-N共掺杂的Zn O前驱膜,然后用电子束对前驱膜进行退火。退火时,电子束加速电压10 k V,退火时间5 min,聚焦束流123 m A,束流为0.7~1.9 m A,最后得到Li-N共掺杂的Zn O薄膜。XRD谱分析表明,当束流高于1.5 m A之后,薄膜为六方Zn O和立方Zn O的混合多晶薄膜,且有金属Zn生成,导致薄膜有较强的绿光发射。SEM图片分析显示,薄膜的晶粒尺寸随束流增加而增大,当束流高于1.5 m A后,晶粒尺寸变化不大,约为60 nm。光致发光(PL)谱和激光拉曼谱的分析结果证实Li、N元素已掺入Zn O晶格中,PL谱中观察到Li元素掺杂引起的紫光发射,拉曼散射光谱中观察到N替代O位的缺陷振动模式。 展开更多

目的:探讨艾灸“关元”穴对衰老进程中大鼠子宫组织细胞周期相关蛋白CyclinD1、CDK4、E2F1、SOD和CAT含量的影响,进一步探究艾灸延缓子宫衰老的机制。方法:选取SPF级12月龄Wistar雌性大鼠14只,随机分为对照组与艾灸组,SPF级3月龄Wistar... 目的:探讨艾灸“关元”穴对衰老进程中大鼠子宫组织细胞周期相关蛋白CyclinD1、CDK4、E2F1、SOD和CAT含量的影响,进一步探究艾灸延缓子宫衰老的机制。方法:选取SPF级12月龄Wistar雌性大鼠14只,随机分为对照组与艾灸组,SPF级3月龄Wistar雌性大鼠7只作为青年组。其中艾灸组大鼠对“关元”穴进行20 min温和灸刺激,每日1次,连续治疗40 d;对照组和青年组大鼠进行同样的抓取与固定。实验结束后取大鼠子宫组织,以酶联免疫吸附测定(ELISA)法检测细胞周期相关蛋白CyclinD1、CDK4、E2F1、SOD和CAT含量。结果:与青年组比较,对照组大鼠子宫组织中SOD、CAT、CyclinD1、CDK4和E2F1含量降低,差异具有统计学意义(P<0.05);而与对照组比较,艾灸组上述蛋白含量明显上升,差异具有统计学意义(P<0.05);各组大鼠子宫组织中CyclinD1含量与SOD、CAT含量均无相关性(P>0.05)。结论:艾灸“关元”穴可上调衰老进程中大鼠子宫组织中SOD、CAT含量,提高其抗氧化能力;可调节子宫组织细胞周期相关蛋白CyclinD1、CDK4的表达,促进E2F1的释放,使细胞加速通过G1期,进而对抗细胞衰老。艾灸的抗衰老作用可能是通过以上两条途径分别进行的。 展开更多

【目的】明确微咸水灌溉条件下粉垄耕作土壤水分入渗规律及水盐运移特征。【方法】基于室内土箱试验,分析3种微咸水矿化度(0g/L(S1)、3g/L(S2)和5g/L(S3))和2种耕作(粉垄耕作(FL)和传统翻耕(FG))条件下的土壤水分入渗特征和水盐运移规... 【目的】明确微咸水灌溉条件下粉垄耕作土壤水分入渗规律及水盐运移特征。【方法】基于室内土箱试验,分析3种微咸水矿化度(0g/L(S1)、3g/L(S2)和5g/L(S3))和2种耕作(粉垄耕作(FL)和传统翻耕(FG))条件下的土壤水分入渗特征和水盐运移规律。【结果】水分平均入渗速率和垂直湿润锋推进速率随着微咸水矿化度的升高呈先增大后减小的变化趋势,水平湿润锋运移速度随着微咸水矿化度的升高而持续增加。FL处理下的土壤水分入渗速率、湿润锋推进速率相比FG处理有明显提升。Kostiakov模型可较好地拟合2种耕作措施下的土壤累积入渗量与入渗时间之间的关系(R^(2)>0.99)。灌溉后20 d,FL处理下的土壤含水率均小于相同微咸水矿化度下的FG处理;同一耕作措施下,土壤含水率随着微咸水矿化度的升高而增加。【结论】微咸水灌溉与粉垄耕作对土壤水分入渗、土壤湿润锋运移和土壤水盐再分布具有改善作用。与灌溉前相比,灌溉后20 d,FL处理下的土壤平均脱盐率为42.95%~55.98%,而FG处理下的土壤平均脱盐率为32.34%~43.29%。随着矿化度的增加,FL处理下的平均土壤电导率相比FG处理分别下降22.38%、18.54%和15.68%。 展开更多

According to the characteristics of comprehensive mechanized heading face, established the mathematical model of single-phase air flow with κ-ε two equations model, and have established κ-ε-θ-κp mathematic model to solve two-phase flow of gas and particles in dust space with eulerian-eulerian method and eulerian-lagrangian method. Numerical solution of gas-particle two-phase flow was put forward based on collocated grid SIMPLE algorithm. Moreover, numerical simulation of dust concentration in fully mechanized caving face was carded out by using Fluent software. Finally, when in forced-exhaust ventilation circumstance, drawer type fan drum have less dust absorption, and most of dust spread to the other site; the dust concentration is inversely proportional to the distance from tunneling head, and the dust concentration has already diffused to decrease below 102 mg/m3 at the position ofx=12 m. Dust are more focused on relative side(in the range about y from 0 to 2 meter) of roadway space of press-ventilated fan drum, especially between tunneling place and drawer type fan drum; the roadway with road header have a higher dust concentration. These conclusions provide reliable theory basis for the dust prevention in comprehensive mechanized heading face. 展开更多

为了研究欧洲温室型黄瓜农艺性状遗传多样性、亲缘关系,明确种质资源的特征特性,为欧洲温室型黄瓜新品种选育提供参考,对搜集并纯化的41份欧洲温室型黄瓜自交系进行了第一雌花节位、叶长、叶宽等18个农艺性状鉴定,并进行遗传多样性、相... 为了研究欧洲温室型黄瓜农艺性状遗传多样性、亲缘关系,明确种质资源的特征特性,为欧洲温室型黄瓜新品种选育提供参考,对搜集并纯化的41份欧洲温室型黄瓜自交系进行了第一雌花节位、叶长、叶宽等18个农艺性状鉴定,并进行遗传多样性、相关性和聚类分析。18个农艺性状间变异系数在13.36%~76.48%之间,变异系数最大的性状为第一雌花节位,达到76.48%;变异系数最小的性状为植株生长势,为13.36%、平均变异系数为27.78%。单瓜质量和单株产量均与第一雌花节位呈显著正相关;果皮颜色与果皮硬度呈显著正相关,与心腔比呈极显著负相关。在遗传相似系数(GS)为1.86时,分为三大类,第一类含有23份黄瓜种质资源,占供试材料的56%;第二类含有4份黄瓜种质资源;第三类含有14份黄瓜种质资源。41份欧洲温室型黄瓜材料遗传差异较大,有丰富的遗传多样性,具有较大的育种目标选择潜力,为欧洲温室型黄瓜新品种选育提供了优良资源。 展开更多

BACKGROUND Liver failure has high mortality and poor prognosis,and establishing new reliable markers for predicting its prognosis is necessary.Mucosal-associated invariant T(MAIT)cells are a novel population of innate-like lymphocytes involved in inflammatory liver disease,and their potential role in liver failure remains unclear.AIM To investigate alteration of circulating MAIT cells and assess its prognostic value in patients with hepatitis B virus(HBV)-related liver failure.METHODS We recruited 55 patients with HBV-related liver failure,48 patients with chronic hepatitis B and 40 healthy controls(HCs)from Nantong Third People’s Hospital Affiliated to Nantong University.Peripheral blood mononuclear cells were isolated,and the percentage and number of circulating MAIT cells were detected by flow cytometry.Plasma levels of interleukin(IL)-7,IL-12p70,IL-18 and interferon-αwere measured by Luminex assay.RESULTS Circulating MAIT cells were significantly decreased in HBV-related liver failure patients(percentage:2.00±1.22 vs 5.19±1.27%,P<0.0001;number:5.47±4.93 vs 84.43±19.59,P<0.0001)compared with HCs.More importantly,there was a significant reduction of MAIT cells in patients with middle/late-stage compared with early-stage liver failure.Circulating MAIT cells partially recovered after disease improvement,both in percentage(4.01±1.21 vs 2.04±0.95%,P<0.0001)and in cell count(17.24±8.56 vs 7.41±4.99,P<0.0001).The proportion(2.29±1.01 vs 1.58±1.38%,P<0.05)and number(7.30±5.70 vs 2.94±1.47,P<0.001)of circulating MAIT cells were significantly higher in the survival group than in the dead/liver transplantation group,and the Kaplan–Meier curve showed that lower expression of circulating MAIT cells(both percentage and cell count)predicted poor overall survival(P<0.01).Also,the levels of IL-12(20.26±5.42 pg/mL vs 17.76±2.79 pg/mL,P=0.01)and IL-18(1470.05±1525.38 pg/mL vs 362.99±109.64 pg/mL,P<0.0001)were dramatically increased in HBV-related liver failure patients compared with HCs.CONCLUSION Circulating MAIT cells may play an important role in the process of HBV-related liver failure and can be an important prognostic marker. 展开更多

目的探讨首发精神分裂症(FES)患者血清可溶性转铁蛋白受体(sTfR)及成纤维细胞生长因子22(FGF22)表达情况,分析二者与FES患者临床症状的关系并进行诊断价值分析。方法选取2021年3月至2023年2月在该院确诊的97例FES患者作为FES组,同期选... 目的探讨首发精神分裂症(FES)患者血清可溶性转铁蛋白受体(sTfR)及成纤维细胞生长因子22(FGF22)表达情况,分析二者与FES患者临床症状的关系并进行诊断价值分析。方法选取2021年3月至2023年2月在该院确诊的97例FES患者作为FES组,同期选取来该院体检的96例健康志愿者作为对照组。采用免疫透射比浊法检测sTfR水平,酶联免疫吸附试验(ELISA)检测FGF22水平,Spearman法分析FES患者血清中sTfR及FGF22水平与阳性与阴性病症量表(PANSS)评分及威斯康辛卡片分类测验(WCST)结果的相关性,受试者工作特征(ROC)曲线分析sTfR及FGF22水平对FES的临床诊断价值。结果两组在性别、年龄、体质量指数、受教育年限、饮酒史及吸烟史方面差异均无统计学意义(P>0.05),与对照组比较,FES组血清sTfR及FGF22水平均下降(P<0.05)。sTfR单独诊断FES的曲线下面积(AUC)为0.835,最佳截断值为4.606 mg/L,FGF22单独诊断FES的AUC为0.772,最佳截断值为208.333μg/L,二者联合诊断的AUC(0.921)大于sTfR单独诊断的AUC(Z=2.613,P=0.009)及FGF22单独诊断的AUC(Z=5.140,P<0.001);sTfR高水平组及FGF22高水平组的PANSS阳性症状评分、阴性症状评分、病理症状评分、总评分、WCST持续性错误数、错误应答数分别低于sTfR低水平组及FGF22低水平组(P<0.05),而WCST完成分类数、WCST正确应答数分别高于sTfR低水平组及FGF22低水平组(P<0.05);FES组中sTfR、FGF22水平与PANSS阳性症状评分、阴性症状评分、病理症状评分、总评分、WCST持续性错误数、WCST错误应答数均呈负相关(P<0.05),与WCST完成分类数及WCST正确应答数均呈正相关(P<0.05)。结论FES患者血清sTfR及FGF22水平下降,联合检测sTfR及FGF22水平对于FES的临床诊断具有重要意义。 展开更多

Developing wheat that acquires and uses phosphorus(P)more efficiently is a promising and low-cost solution for increasing grain yield and reducing P-related environmental impacts.The present study identified agronomic and physiological traits that contribute to genetic variation in the P acquisition,remobilization,and utilization efficiency of 11 wheat cultivars from southwest China grown in P-deficient purple lithomorphic soil(Olsen P=4.7)with balanced(75 kg P ha^(−1))and excess P(120 kg P ha^(−1))supplies.On average,soil P deficiency(–P)reduced root P uptake(17.0%–60.8%),P remobilization(33.9%–52.8%),dry mass yield(11.5%–39.2%),and grain yield(17.7%–54.4%).Balanced P(+P)increased grain yield via increased plant biomass rather than increased HI.–P increased phosphorus uptake efficiency(PUpE,4.5-fold),phosphorus utilization efficiency(PUtE,1.25-fold),and phosphorus use efficiency(PUE,5.4-fold)compared with those under+P,and PUtE explained most(58.1%–60.8%)of the genetic variation in PUE under both–P and+P.The high root P uptake of P-efficient cultivars under–P was regulated by root surface area and root length density in the 0–10 cm soil layer but not in the 10–20 and 20–40 cm soil layers,suggesting that a topsoil foraging strategy is a more economical approach than deeper root exploration for increasing P uptake.Root P uptake before anthesis and P remobilization after anthesis were critical for increasing the PUtE of wheat,given that P-efficient cultivars showed higher Pn(net photosynthetic rate)and sucrose levels than P-inefficient cultivars.Pn reduction by–P resulted from decreased Gs and Ci,and high evapotranspiration under+P increased shoot P%by increasing root P uptake.Genetic variation in the source-to-sink ratio was observed in consequence of a+P-induced allometric increase in sucrose in leaves and kernels.Owing to these beneficial effects,+P increased the kernel N and P yields of the 11 cultivars by 9.9%–52.4%and 12.3%–48.8%,respectively.The findings of this study could help improve wheat in future breeding efforts and P management by identifying desirable Pefficient phenotypes in P-deficient farming systems. 展开更多

This article reviews the research progress of rehabilitation treatment and nursing care of patients with neurogenic bladder after spinal cord injury,in order to provide reference for the rehabilitation treatment and nursing care of patients.We reviewed recent medical literature on patients with neurogenic bladder,focusing on neurogenic bladder caused by spinal cord injury.We analyzed 30 recent of publications in patients with neurogenic bladder after spinal cord injury,in addition to reviewing and evaluating the commonly used rehabilitation nursing methods for neurogenic bladder.Psychological counseling is a vital aspect which cannot be neglected in the process of neurogenic bladder rehabilitation.Hitherto,the commonly used drug and surgical treatments may have negatively impacted the mental health of patients in varying degrees.However,in clinical practice,applying intermittent catheterization in patients who have neurogenic bladder with spinal cord injury may help improve patients’life quality,mitigate psychological burden,and reduce negative emotions. 展开更多

To improve our understanding of the formation and evolution of the Moon, one of the payloads onboard the Chang'e-3 (CE-3) rover is Lunar Penetrating Radar (LPR). This investigation is the first attempt to explore the lunar subsurface structure by using ground penetrating radar with high resolution. We have probed the subsur- face to a depth of several hundred meters using LPR. In-orbit testing, data processing and the preliminary results are presented. These observations have revealed the con- figuration of regolith where the thickness of regolith varies from about 4 m to 6 m. In addition, one layer of lunar rock, which is about 330 m deep and might have been accumulated during the depositional hiatus of mare basalts, was detected. 展开更多

AIM:To investigate the alleviation of scutellarein(SN)against inner blood-retinal-barrier(iBRB)dysfunction in microglia cells stimulated by hyperglycemia and to elucidate the engaged mechanism.METHODS:Microglia BV2 cells were stimulated by using 25 mmol/L D-glucose.The same concentration of mannitol(25 mmol/L)was applied as an isotonic contrast.Real-time PCR,Western-blot assay and immunofluorescence staining assay was performed.The dysfunction of iBRB in vitro was detected by using transendothelial electrical resistance(TEER)assay.Additionally,the leakage of fluorescein isothiocyanate(FITC)-conjugated dextran(70 kDa)was detected.RESULTS:SN abrogated microglia BV2 cells activation and reduced the phosphorylated activation of extracellular signal-regulated protein kinase(ERK)1/2.SN also decreased the transcriptional activation of nuclear factorκB(NFκB)and the elevated expression of tumor necrosis factorα(TNFα),interleukin(IL)-6 and IL-1βin BV2 cells treated with D-glucose(25 mmol/L).SN attenuated iBRB dysfunction in human retinal endothelial cells(HRECs)or choroid-retinal endothelial RF/6 A cells when those cells were treated with TNFα,IL-1βor IL-6,or co-cultured with microglia cells stimulated by D-glucose.Moreover,SN restored the decreased protein expression of tight junctions(TJs)in TNFα-treated HRECs and RF/6 A cells.CONCLUSION:SN not only alleviate iBRB dysfunction via directly inhibiting retinal endothelial injury caused by TNFα,IL-1βor IL-6,but also reduce the release of TNFα,IL-1βand IL-6 from microglia cells by abrogating hyperglycemia-mediated the activation of microglia cells. 展开更多
