类伸展蛋白(Leucine-Rich Repeats Extensins,LRX)是一类细胞壁嵌合蛋白,其N端包含一个LRR(leucine-rich repeats)结构域,C端含Extensins结构域。研究表明,LRX基因家族在拟南芥(Arabidopsis thaliana)花粉萌发和花粉管生长过程中具有重... 类伸展蛋白(Leucine-Rich Repeats Extensins,LRX)是一类细胞壁嵌合蛋白,其N端包含一个LRR(leucine-rich repeats)结构域,C端含Extensins结构域。研究表明,LRX基因家族在拟南芥(Arabidopsis thaliana)花粉萌发和花粉管生长过程中具有重要作用,而水稻(Oryza sativa L.)LRX基因家族是否在调控花粉发育方面具有保守的生物学功能尚不清楚。本研究首先进行了生物信息学分析,结果显示,水稻LRX基因家族包括8个成员,OsPEX3、OsLRX3、OsLRX5位于水稻第1号染色体;OsLRX1、OsLRX3、OsLRX2、OsPEX1和OsPEX2分别位于第2、第5、第6、第11和第12号染色体,其中OsPEX1基因在花粉中高表达,暗示OsPEX1可能参与了花粉发育调控。为此,本研究采用RNAi技术进一步研究了OsPEX1基因对花粉发育的影响。结果表明,OsPEX1基因的RNAi转基因植株花粉败育,结实率仅为10%-30%。qRT-PCR分析显示,这些RNAi转基因植株OsPEX1基因表达量显著低于野生型,而且其表达量越低花粉育性亦随之降低。上述研究结果表明,水稻OsPEX1基因是水稻花粉发育的重要基因,该基因的克隆和功能分析有助于进一步阐明水稻花粉发育调控的分子遗传学机制。 展开更多

Dear editor,We have read with great interest the recent article“Comparison of different versions of the quick sequential organ failure assessment for predicting in-hospital mortality of sepsis patients:A retrospective observational study”.[1]They showed that the performance of the procalcitonin-enhanced qSOFA(PqSOFA)was similar to that of the qSOFA,and the lactate-enhanced qSOFA(LqSOFA)and the modified qSOFA(MqSOFA)were superior to the qSOFA for predicting in-hospital mortality.Further,they suggested that the MqSOFA should be a candidate for the qSOFA in the emergency department(ED).We congratulate the authors for their contributions.However,we has several concerns about this research. 展开更多

Many papers have been published that solve UC through different methods. Articles that systematically summarize UC problems’ progress in order to update researchers interested in this field are needed. Because of its promising performance, stochastic programming (SP) has become increasingly researched. Most papers, however, present SP’s UC solving approaches differently, which masks their relationships and makes it hard for new researchers to quickly obtain a general idea. Therefore, this paper tries to give a structured bibliographic survey of SP’s applications in UC problems. 展开更多

急性缺血性脑卒中患者并发深静脉血栓风险预测列线图模型构建与评价

目的探讨急性缺血性脑卒中(AIS)患者并发深静脉血栓(DVT)的危险因素,并构建并发DVT的预测模型。方法纳入2018年1月-2023年1月于南京大学医学院附属鼓楼医院全科医学科接受治疗的455例AIS患者的临床资料,将患者是否并发DVT分为DVT组和非... 目的探讨急性缺血性脑卒中(AIS)患者并发深静脉血栓(DVT)的危险因素,并构建并发DVT的预测模型。方法纳入2018年1月-2023年1月于南京大学医学院附属鼓楼医院全科医学科接受治疗的455例AIS患者的临床资料,将患者是否并发DVT分为DVT组和非DVT组,收集患者的临床资料,采用Logistic回归分析明确AIS患者并发DVT的独立影响因素,将获得的独立影响因素纳入构建列线图预测模型,采用R软件中C指数、受试者工作曲线(ROC)及校准曲线评价AIS患者并发DVT风险的列线图模型效能。结果本院全科医学科接受治疗的455例AIS患者中,并发DVT的患者45例,占比9.89%。两组临床资料比较中,性别、体质量指数(BMI)、病程、高血压、吸烟史、饮酒史、收缩压、舒张压、空腹血糖(FBG)、总胆固醇(TC)、红细胞计数(RBC)、白细胞计数(WBC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)以及低密度脂蛋白胆固醇(LDL-C)对比差异无统计学意义(P>0.05);DVT组的年龄、高血糖、美国国立卫生院卒中量表(NIHSS)评分、卧床时间、血小板计数以及纤维蛋白原(FIB)均>非DVT组,DVT组的D-二聚体和凝血酶时间(TT)均<非DVT组,差异有统计学意义(^(均)P<0.05)。经Logistics回归分析显示年龄大、高血糖、NIHSS评分高、卧床时间长、血小板计数多、FIB水平高、D-二聚体水平高以及TT短均是AIS患者并发DVT的独立影响因素(^(均)P<0.05)。绘制ROC曲线,结果显示,年龄、高血糖、NIHSS评分、卧床时间、血小板计数、FIB、D-二聚体和TT的曲线下面积(AUC)值均>0.700,说明上述指标对于AIS患者并发DVT具有较好的预测价值。基于以上影响因素建立列线图风险模型,验证结果显示,校准曲线的C-index值为0.850,说明该列线图模型具备较好的区分度,ROC曲线建模组和验证组的AUC分别为0.850和0.901,说明该列线图模型具有良好的预测能效和判别能力。结论基于AIS患者并发DVT的独立危险因素构建的列线图预测模型,能直观的预测AIS患者并发DVT的概率。 展开更多

目的分析1株人源奇异变形杆菌CA151922的2个耐药相关基因岛SGI1和SXT/R391的基因结构和耐药基因。方法聚合酶链式反应(PCR)检测SGI1和SXT/R391基因岛特异性整合酶基因,采用二代测序方法测定CA151922基因组。分别以SGI1和SXT/R391基因岛... 目的分析1株人源奇异变形杆菌CA151922的2个耐药相关基因岛SGI1和SXT/R391的基因结构和耐药基因。方法聚合酶链式反应(PCR)检测SGI1和SXT/R391基因岛特异性整合酶基因,采用二代测序方法测定CA151922基因组。分别以SGI1和SXT/R391基因岛的参考序列,从基因组序列中提取基因岛片段并组装。在线预测并注释开放读码框(ORFs)。基因岛的同源序列通过在线工具BLASTn获得,采用cd-hit获得非冗余的同源基因,利用R语言构建同源基因的聚类进化关系树。利用微量肉汤稀释法进行耐药表型检测。结果奇异变形杆菌CA151922同时含有SGI1(SGI1-B)和SXT/R391(ICEPmi Jpn1)2个耐药相关基因岛。SGI1-B和ICEPmi Jpn1与已知序列相似性分别为97%~99%和96%~99%。SGI1-B和ICEPmi Jpn1携带的耐药基因种类有所差异,但两者携带不同的编码β-内酰胺酶的基因。CA151922为多耐药菌株,耐药达18种,与基因岛耐药基因有关的耐药表型为氨苄西林和磺胺类药物,提示菌株耐药表型不完全由该耐药岛内的耐药基因决定。结论首次在奇异变形杆菌中发现,同时携带SGI1和SXT/R391耐药相关基因岛,增加了菌株耐药的复杂性和严重性,提示应加强对多耐药菌株耐药相关基因岛的监测。 展开更多

Long-term and stable preservation of bacteriophages is of crucial importance.Although many efforts have been made in the past decades to explore the influence of external factors on bacteriophage preservation,there is still little understanding,and a systematic description is lacking.In this study,we explored the influence of different factors on the preservation of lytic bacteriophage VP3,one of the typing bacteriophages of Vibrio cholerae O1 biotype El Tor,and attempted to optimize its preservation.We examined external factors,including temperature,solution,and cryoprotectant,in stable cooling/freezing conditions or alternate cooling/freezing and thawing.We found that whether in Luria-Bertani(LB)medium or SM buffer,in terms of 20-week stable cooling or freezing,−20℃ was the most damaging while 4℃,−80℃,and−196℃ were protective.Thirteen cycles of alternate cooling/freezing and thawing caused a loss in the survival rates of bacteriophages.The addition of cryoprotectant,glycerol(30%,w/v)or dimethyl sulfoxide(DMSO,10%,w/v)significantly improved the survival rates of bacteriophages preserved at−20℃.However,at 4℃,−80℃,and−196℃,the cryoprotectant effect was only slightly positive or even harmful.In summary,for bacteriophage VP3,the best preservation method is to directly preserve the bacteriophage stocks in LB medium at−80℃ or−196℃ instead of storing them in SM buffer or adding cryoprotectant.Our results provided insights into the external influencing factors on bacteriophage VP3 during preservation at low temperature and can be applied to the optimization of bacteriophage preservation in the future. 展开更多

This paper aims to provide an efficient and straightforward structural form-finding method for designers to extrapolate component forms during the conceptual stage.The core idea is to optimize the classical method of structural form-finding based on principal stress lines by using parametric tools.The traditional operating process of this method relies excessively on the designer’s engineering experience and lacks precision.Meanwhile,the current optimization work for this method is overly complicated for architects,and limitations in component type and final result exist.Therefore,to facilitate an architect’s conceptual work,the optimization metrics of the method in this paper are set as simplicity,practicality,freedom,and rapid feedback.For that reason,this paper optimizes the method from three aspects:modeling strategy for continuum structures,classification processing of data by using the k-nearest neighbor algorithm,and topological form-finding process based on stress lines.Eventually,it allows architects to create structural texture with formal aesthetics and modify it in real time on the basis of structural analysis results.This paper also explores a comprehensive application strategy with internal force analysis diagramming to form-finding.The finite element analysis tool Karamba3D verifies the structural performance of the form-finding method.The performance is compared with that of the conventional form,and the comparison results show the practicality and potential of the strategy in this paper. 展开更多
