目的 :α viniferin ,vaticanol(B ,CandG)和hopeaphenol是白黎芦醇的低聚体 ,具有抗肿瘤和抗氧化等活性 ,而这些化学保护作用是与其抗炎作用有联系。本文研究旨在了解这些化合物对小鼠骨髓分化肥大细胞 (BMMC)组织胺、肿瘤坏死因子和... 目的 :α viniferin ,vaticanol(B ,CandG)和hopeaphenol是白黎芦醇的低聚体 ,具有抗肿瘤和抗氧化等活性 ,而这些化学保护作用是与其抗炎作用有联系。本文研究旨在了解这些化合物对小鼠骨髓分化肥大细胞 (BMMC)组织胺、肿瘤坏死因子和白三烯介质释放的影响 ,并观察对细胞外信号调节激酶 (ERK)的激活作用。方法 :分离小鼠骨髓细胞 ,培养 4~ 5周 (RPMI 16 4 0 ,IL 310ng/ml) ,抗 DNPIgE致敏 ,以DNP BSA( 30ng/ml)刺激释放反应。在非免疫刺激释放实验中 ,A2 3187作为刺激剂。结果 :所试化合物均表现出不同程度的抑制BMMC释放反应的作用。这些化合物对IgE刺激的TNF α和LTs释放表现出显著的抑制作用 ,而对IgE刺激的组织胺释放反应 ,只有vaticanolB和vaticanolC具有抑制作用 ( 10 0μmol/L)。在A2 3187介导的非免疫刺激释放反应中 ,vaticanol(B ,C和G)和hopeaphenol对组织胺释放表现出明显的对抗作用 ,大多数所试化合物有效地抑制了TNF α和LTs的释放 ,唯vaticanolC和vaticanolG对LTs的释放无明显影响。α viniferin和vaticanolC有效地抑制了IgE刺激的ERK酶的激活。结论 :α viniferin ,vaticanol(B ,C和G)和hopeaphenol能有效地抑制肥大细胞的炎性介质的释放反应 。 展开更多

BACKGROUND Double-balloon endoscopic retrograde cholangiography (DB-ERC) is widely performed for biliary diseases after reconstruction in gastrointestinal surgery,but there are few reports on DB-ERC after hepatectomy or living donor liver transplantation (LDLT).AIM To examine the success rates and safety of DB-ERC after hepatectomy or LDLT METHODS The study was performed retrospectively in 26 patients (45 procedures) who underwent hepatectomy or LDLT (liver operation:LO group) and 40 control patients (59 procedures) who underwent pancreatoduodenectomy (control group).The technical success (endoscope reaching the choledochojejunostomy site),diagnostic success (performance of cholangiography),therapeutic success(completed interventions) and overall success rates,insertion and procedure(completion of DB-ERC) time,and adverse events were compared between these groups.RESULTS There were no significant differences between LO and control groups in the technical [93.3%(42/45) vs 96.6%(57/59),P=0.439],diagnostic [83.3%(35/42) vs83.6%(46/55),P=0.968],therapeutic [97.0%(32/33) vs 97.7%(43/44),P=0.836],and overall [75.6%(34/45) vs 79.7%(47/59),P=0.617] success rates.The median insertion time (22 vs 14 min,P <0.001) and procedure time (43.5 vs 30 min,P=0.033) were significantly longer in the LO group.The incidence of adverse events showed no significant difference [11.1%(5/45) vs 6.8%(4/59),P=0.670].CONCLUSION DB-ERC after liver operation is safe and useful but longer time is required,so should be performed with particular care. 展开更多

It is very important to control any risk of instability of embankment during vacuum construction, the simulation vacuum preloading method using tri-axial apparatus is proposed to predict the behavior of soft soil improvement in the laboratory, as well as to make this method become familiar and easier in the future. The tri-axial apparatus is used instead of the large-scale one, which has been performed by Bergado (1998) and Indaratna (2008). The tri-axial test on small size specimen can be carried out in one week compared to the large-scale apparatus takes one month for big specimen. In addition, the lateral deformation as well as the shear strength increase with time can determine accurately. 展开更多

Phenytoin(5,5-diphenylhydantoin;DPH) induces expression of cytochromes P450(CYPs). Interactions between DPH and tacrolimus suggested that the persistence of CYP induction after discontinuation of DPH is dependent on the history of administration and dosing period of DPH. However, the relationship between the duration of DPH administration and expression of CYPs in the liver and small intestine of rats is not known. Alterations in levels of P-glycoprotein(P-gp;MDR1;ABCB1) as well as CYPs cause drug interactions in the small intestine. We examined the effects of the duration of DPH administration on expression of CYPs and P-gp in the liver and small intestine of rats. Rats were treated with DPH(100 mg/kg,peroral(p.o.) twice a day(b.d.)) for 2, 4, 8, and 16 d. mRNA levels of CYPs and P-gp were examined using the total RNA extracted from the liver and duodenum 2 h and 24 h after the final administration of DPH. CYP3 A activities were determined using microsomes. DPH administration for 2 d and 4 d markedly increased m RNA levels of CYPs such as CYP3 A1, CYP3 A2,CYP2 B1, and CYP2 B2 in the liver. A relatively long duration of DPH administration(8 d and16 d) resulted in abolition of the induction of hepatic CYP but increased CYP3 A activities were maintained. These results suggest that the duration of DPH administration could be an important determinant of hepatic CYP induction. 展开更多

Acute respiratory failure after surgery for aortic dissection is a serious complication that has been associated with increased mortality and morbidity. Veno-venous (VV) extracorporeal membrane oxygenation (ECMO) is reported as one of the treatments for this life-threatening complication. A 78-year-old male patient was admitted to our hospital for Stanford type A acute aortic dissection. He underwent emergency replacement of the ascending aorta, after which he developed acute lung injury. It was difficult to maintain oxygenation with a respirator mask. Therefore, VV-ECMO was initiated on postoperative day 1. The oxygenation gradually improved, and VV-ECMO was continued until postoperative day 13. On postoperative day 25, mechanical ventilation was withdrawn. The patient was discharged from the hospital on postoperative day 149. We report a case of successful treatment of acute lung injury with VV-ECMO initiated after surgery for aortic dissection. VV-ECMO can be considered as a treatment option for severe acute lung injury after surgery for aortic dissection. 展开更多

Green fluorescent protein (GFP) is a protein that emits green fluorescence when exposed to a radiation of ultraviolet wavelength range, even without the addition of substrate and cofactor. Because of such characteristics, the usage of GFP is widespread in both in vivo and in vitro applications. In addition, recent advances in biotechnology have enabled GFP to be expressed in various hosts, including bacteria, yeast, plants, animals, and even living-cells, for multiple purposes. Currently, GFP is a subject of great interest in the analytical sciences, especially in immunoassays for qualitative and quantitative analyses, when it is fused with an antibody because of the high sensitivity of GFP and antigen-binding specificity of antibodies. Recently the fluobody, which is a fusion protein of GFP with single-chain variable fragment antibody (scFv), has become a useful tool in various fields. In a shale gas and oil reservoir,hydrocarbon fluids are stored in organic nanopores with sizes on the order of~1-100 nm.The adsorption,selectivity,and phase behavior of hydrocarbons in the nanopores are crucial for estimating the gas-in-place and predicting the productivity.In this study,to understand the characteristics of the phase behavior of multicomponent hydrocarbon systems in shale reservoirs,the phase behavior of a CH_(4)/n-C_(4)H_(10)binary mixture in graphite nanopores was investigated by Grand Ca-nonical Monte Carlo(GCMC)molecular simulation.The method for determining the dew-point pressure and bubble-point pressure in the nanopores was explored.The condensation phenomenon was observed owing to the difference in the adsorption selectivities of the hydrocarbon molecules on the nanopore surfaces,and hence the dew-point pressure(and bubble-point pressure)of hydrocarbon mixtures in the nanopores significantly shifted.The GCMC simulations reproduced both the higher and lower bubble-point pressures in nanopores in previous studies.This work highlights the crucial role of the selec-tivity in the phase behavior of hydrocarbons in nanopores. 展开更多
