Background:To explore the effective chemical constituents of Feiduqing formula for prevention and treatment of coronavirus disease 2019(COVID-19).Methods:The compounds and action targets of twelve herbal medicines in Feiduqing formula were collected via Traditional Chinese Medicine Systems Pharmacology Database and Analytic Platform.The genes corresponding to the targets were queried through the UniProt database.The“herbal medicine-ingredient-target”network was established by Cytoscape software.The Gene Ontology function enrichment analysis and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were performed by Database for Annotation,Visualization and Integrated Discovery.Molecular docking was used to analyze the binding force of core active compounds of Feiduqing formula with PTGS2,HSP90AA1,SARS-CoV-23CL hydrolase and angiotensin converting enzyme II(ACE2).Results:The“herbal medicine-ingredient-target”network included 434 nodes and 1948 edges,including 222 components such as quercetin,kaempferol,luteolin,etc.The key targets are PTGS2,HSP90AA1,PTGS1,ESR1,AR,NOS2,etc.Gene Ontology function enrichment analysis revealed 2530 items,including RNA polymerase II-specific,response to oxidative stress,transcription factor activity,etc.Kyoto Encyclopedia of Genes and Genomes pathway enrichment screened 169 signal pathways,including Human cytomegalovirus infection,Kaposi sarcoma-associated herpesvirus infection,Hepatitis B,Hepatitis C,IL-17,TNF,etc.The results of molecular docking showed that quercetin,luteolin,β-sitosterol,stigmasterol and other core active compounds have a certain degree of affinity with PTGS2,HSP90AA1,SARS-CoV-23CL hydrolase and ACE2.Conclusion:The active compounds of Feiduqing formula may have a therapeutic effect on COVID-19 pneumonia through the action on PTGS2,HSP90AA1,SARS-CoV-23CL hydrolase and ACE2,and regulating many signaling pathways. 展开更多

Effects of scalp acupoint cluster needling on learning and memory ability and inflammatory mediators in hippocampal CA1 region of rats with Alzheimer's disease

Objective:To observe the effect of scalp acupoint cluster needling on learning and memory ability and inflammatory mediators in CA1 area of hippocampus in rats with Alzheimer's disease(AD).Methods:A total of 40 SPF Wistar rats were randomly divided into blank group,sham operation group,model group and scalp acupoint cluster needling group,with 10 rats in each group.The AD model was duplicated by injecting Abeta 1-42 solution into bilateral hippocampus in model group and scalp acupoint cluster needling group.The sham operation group was injected with the same amount of sterile hydrogen peroxide,while the blank group was not treated.The scalp acupoint cluster needling group was given acupuncture intervention from 2 d after model establishment.The blank group,sham operation group and model group were only bundled and fixed in the dorsal position once a day for 14 consecutive days.After treatment,the learning and memory ability of AD rats was tested by Morris water maze test,the contents and activities of Ach,ChAT and AcE in hippocampal CA1 area were detected by ELISA,and the expressions of IL-6,IL-10 protein and mRNA in hippocampal CA1 area were detected by immunohistochemistry and RT-PCR,respectively.Results:Ach content,ChAT activity,IL-10 protein and mRNA expression in hippocampus were significantly lower than those in blank group,while AchE activity and IL-6 protein and mRNA expression were significantly higher than those in blank group.Compared with the model group,scalp acupoint cluster needling could significantly reduce the escape latency time,increase the number of crossing platforms and effective residence time,increase the content of Ach,the activity of ChAT,the expression of IL-10 protein and mRNA in hippocampus,and decrease the activity of AchE and the expression of IL-6 protein and mRNA.Conclusion:Controlling the expression of inflammatory factors and promoting the content of Ach in hippocampal CA1 region of AD rats can improve the learning and memory ability of AD rats. 展开更多

Objective:To investigate the effects of acupuncture on endoplasmic reticulum stress and apoptosis in hippocampal neurons of rats with Alzheimer's disease(AD).Methods:A total of 36 Wistar rats were randomly divided into the blank group(the control),model group,sham-operated group,and acupuncture group,with 9 rats in each group.The AD was replicated by injection of beta-amyloid protein 1-42 solution into the bilateral hippocampal CA1 area in the model group and acupuncture group.The sham-operated group was injected with the same amount of sterile hydrogen peroxide while the control group was not treated.Rats in the acupuncture group were treated with cluster needling of scalp point therapy from 2 days after model establishment once a day for 14 consecutive days,while rats in the control group,model group,and sham-operated group were bundled and fixed in the dorsal position.After treatment,the morphology of hippocampal CA1 neurons was observed by HE staining,the apoptotic level of hippocampal CA1 neurons was detected by Tunel method,the expression of Abeta 1-42 and glucose-regulated protein 78(GRP78)in hippocampal CA1 region was detected by immunohistochemistry,and the levels of GRP78 and Caspase-12 proteins in hippocampal CA1 region were detected by Western blot.Results:Compared with the blank group,the neurons in the hippocampal CA1 area in the model group decreased significantly,and the apoptotic rate,the expression of Abeta 1-42,GRP78 and Caspase-12 protein increased significantly(P<0.01).Compared with the model group,the morphology of hippocampal CA1 neurons in the acupuncture group was significantly improved,the number of neurons was significantly increased,the apoptotic rate,the expression of Abeta 1-42,GRP78 and Caspase-12 protein were significantly reduced,and the acupuncture group was significantly improved(P<0.01).Conclusions:Scalp acupoint cluster needling therapy improves the morphology of hippocampal CA1 neurons and inhibits the accumulation of Abeta 1-42 by inhibiting endoplasmic reticulum stress and cell apoptosis. 展开更多
