Rose(Rosa hybrida)is one of most famous ornamental plants in the world,and its commodity value largely depends on its flower color.However,the regulatory mechanism underlying rose flower color is still unclear.In this...Rose(Rosa hybrida)is one of most famous ornamental plants in the world,and its commodity value largely depends on its flower color.However,the regulatory mechanism underlying rose flower color is still unclear.In this study,we found that a key R2R3-MYB transcription factor,RcMYB1,plays a central role in rose anthocyanin biosynthesis.Overexpression of RcMYB1 significantly promoted anthocyanin accumulation in both white rose petals and tobacco leaves.In 35S:RcMYB1 transgenic lines,a significant accumulation of anthocyanins occurred in leaves and petioles.We further identified two MBW complexes(RcMYB1-RcBHLH42-RcTTG1;RcMYB1-RcEGL1-RcTTG1)associated with anthocyanin accumulation.Yeast one-hybrid and luciferase assays showed that RcMYB1 could active its own gene promoter and those of other EBGs(early anthocyanin biosynthesis genes)and LBGs(late anthocyanin biosynthesis genes).In addition,both of the MBW complexes enhanced the transcriptional activity of RcMYB1 and LBGs.Interestingly,our results also indicate that RcMYB1 is involved in the metabolic regulation of carotenoids and volatile aroma.In summary,we found that RcMYB1 widely participates in the transcriptional regulation of ABGs(anthocyanin biosynthesis genes),indicative of its central role in the regulation of anthocyanin accumulation in rose.Our results provide a theoretical basis for the further improvement of the flower color trait in rose by breeding or genetic modification.展开更多
Drug adulteration and contamination are serious threats to human health therefore,their accurate monitoring is very important.Allopurinol(Alp)and theophylline(Thp)are commonly used drugs for the treatment of gout and ...Drug adulteration and contamination are serious threats to human health therefore,their accurate monitoring is very important.Allopurinol(Alp)and theophylline(Thp)are commonly used drugs for the treatment of gout and bronchitis,while their isomers hypoxanthine(Hyt)and theobromine(Thm)have no effect and affect the efficacy of the drug.In this work,the drug isomers of Alp/Hyt and Thp/Thm are simply mixed withα-,β-,γ-cyclodextrin(CD)and metal ions and separated using trapped ion mobility spectrometry-mass spectrometry(TIMS-MS).TIMS-MS results showed that Alp/Hyt and Thp/Thm isomers could interact with CD and metal ions and form corresponding binary or ternary complexes to achieve their TIMS separation.Different metal ions and CDs showed different separation effect for the isomers,among which Alp and Hyt could be successfully distinguished from the complexes of[Alp/Hyt+γ-CD+Cu–H]^(+)with separation resolution(RP–P)of 1.51;whereas Thp and Thm could be baseline separated by[Thp/Thm+γ-CD+Ca–H]^(+)with RP–P of 1.96.Besides,chemical calculations revealed that the complexes were in the inclusion forms,and microscopic interactions were somewhat different,making their mobility separation.Moreover,relative and absolute quantification was investigated with an internal standard to determine the precise isomers content,and good linearity(R^(2)>0.99)was obtained.Finally,the method was applied for the adulteration detection where different drugs and urine were analyzed.In addition,due to the advantages of fast speed,simple operation,high sensitivity,and no chromatographic separation required,the proposed method provides an effective strategy for the drug adulteration detection of isomers.展开更多

In this study, Cas12a was applied to construct plant cytosine base editors(CBEs). The main elements of Cas12aCBEs were engineered and their efficiency was evaluated in stably transformed rice cells. An optimized ttCas12a-hyA3Bctd editor, consisting of a LbCas12a variant carrying catalytic inactive D832A and temperature-tolerance D156R double mutations, a truncated human APOBEC3B deaminase, a human RAD51 single-stranded DNA-binding domain, and double copies of UGI, outperformed other Cas12aCBEs in base editing efficiency. In T0transgenic rice plants, ttCas12a-hyA3Bctd edited an average of42.01% and a maximum of 68.75% of lines at six genomic targets. A-to-G conversions were generated in rice by an adenine base editor with a similar architecture to the optimized CBE. Our results provide preliminary evidence for the feasibility of robust and efficient plant Cas12a base editing systems, which could be useful for precise crop breeding.展开更多

Wax apple(Syzygium samarangense)is an economically important fruit crop with great potential value to human health because of its richness in antioxidant substances.Here,we present a haplotype-resolved autotetraploid ...Wax apple(Syzygium samarangense)is an economically important fruit crop with great potential value to human health because of its richness in antioxidant substances.Here,we present a haplotype-resolved autotetraploid genome assembly of the wax apple with a size of 1.59 Gb.Comparative genomic analysis revealed three rounds of whole-genome duplication(WGD)events,including two independent WGDs after WGT-γ.Resequencing analysis of 35 accessions partitioned these individuals into two distinct groups,including 28 landraces and seven cultivated species,and several genes subject to selective sweeps possibly contributed to fruit growth,including the KRP1-like,IAA17-like,GME-like,and FLACCA-like genes.Transcriptome analysis of three different varieties during flower and fruit development identified key genes related to fruit size,sugar content,and male sterility.We found that AP2 also affected fruit size by regulating sepal development in wax apples.The expression of sugar transport-related genes(SWEETs and SUTs)was high in‘ZY’,likely contributing to its high sugar content.Male sterility in‘Tub’was associated with tapetal abnormalities due to the decreased expression of DYT1,TDF1,and AMS,which affected early tapetum development.The chromosome-scale genome and large-scale transcriptome data presented in this study offer new valuable resources for biological research on S.samarangense and shed new light on fruit size control,sugar metabolism,and male sterility regulatory metabolism in wax apple.展开更多

Objective:To explore the mechanism of circRNA-vgll3 in osteogenic differentiation of human bone marrow mesenchymal stem cells.Methods:BMSCs cells were transfected with circRNA-vgll3,and divided into circRNA-vgll3 high...Objective:To explore the mechanism of circRNA-vgll3 in osteogenic differentiation of human bone marrow mesenchymal stem cells.Methods:BMSCs cells were transfected with circRNA-vgll3,and divided into circRNA-vgll3 high-level group,circRNA-vgll3 low-level group,and negative control group(circRNA-vgll3 not transfected)according to the amount of transfection.The proliferation and apoptosis of BMSCs osteoblasts in each group were analyzed,and the alkaline phosphatase(ALP)activity,type I collagen gray value,bone morphogenetic protein 2(BMP-2),Runx2 protein,and mRNA expression levels were detected.Results:The circRNA-vgll3 low-level group had a significant inhibitory effect on the proliferation of BMSCs osteoblasts,and the apoptosis rate of the circRNA-vgll3 low-level group was significantly higher than that of the circRNA-vgll3 high-level group(P<0.05);ALP activity,type I collagen gray value,BMP-2,Runx2 protein,and mRNA expression levels in the high-level circRNA-vgll3 group were significantly higher than those in the low-level circRNA-vgll3 group,and the difference was statistically significant(P<0.05).Conclusion:Overexpression of circRNA-vgll3 can promote the osteogenic differentiation ability of BMSCs,while low expression of circRNA-vgll3 can inhibit the osteogenic differentiation ability of BMSCs.The main mechanism of action is that circRNA-vgll3 can affect osteogenic differentiation by regulating the Runx2 protein.展开更多

The effects of exogenous gibberellic acid (GA(3)) and abscisic acid (ABA) on the total respiratory rate, percentages of total respiratory rate contributed by respiratory pathways [Embden-Meyerhof- Parnas Pathway (EMP)...The effects of exogenous gibberellic acid (GA(3)) and abscisic acid (ABA) on the total respiratory rate, percentages of total respiratory rate contributed by respiratory pathways [Embden-Meyerhof- Parnas Pathway (EMP), Pentose Phosphate Pathway (PPP), and Tricarboxylic Acid Cycle (TCA)], and conversion of starch to soluble sugars in the buds of black currants during secondary bud burst were investigated to determine the relationship between respiratory rates and secondary bud burst. 'Adelinia', a black currant cultivar that is prone to secondary bud burst after the first harvest, was used in this study. Mature bushes of Adelinia were sprayed with 30 mg/L GA(3) and 50 mg/L ABA to manipulate bud burst. The results showed that exogenous applications of GA(3) and ABA had opposite effects on bud respiratory rate. Generally, GA(3) treatment increased the total respiratory rate and respiratory rate of the TCA and PPP, and the respiratory rates after GA(3) treatment were higher than those of control. While ABA treatment mostly decreased the total respiratory rate and the respiratory rate of TCA and PPP in buds in comparison to control. In terms of the percentage of the three respiratory rates in comparison to the total respiratory rate, GA(3) treatment significantly increased the percentage of TCA and PPP respiratory rate in comparison to the control (P < 0.01), whereas ABA decreased the rates. GA(3) significantly increased the content of soluble sugars and decreased the starch content, while the starch content in buds after ABA treatment was significantly higher than that of the control. All results showed that PPP is a critical process for the second bud burst in black currants. While the EMP-TCA pathway is still dominant in bud respiration, provides a series of basic materials and energy (ATP). The conversion of starch to soluble sugars is essential for bud burst. Thus, we conclude that an energy shortage is a main contributor in ABA inhibition of the secondary bud burst of black currants.展开更多
BACKGROUND Oral intake is dependent on the gastric ability to accommodate the food bolus.Comparatively,neonates have a smaller gastric capacity than adults and this may limit the volume of their milk intake.Yet,we pre...BACKGROUND Oral intake is dependent on the gastric ability to accommodate the food bolus.Comparatively,neonates have a smaller gastric capacity than adults and this may limit the volume of their milk intake.Yet,we previously reported that the newborn rat gastric milk volume is greatest after birth and,when normalized to body weight,decreases with postnatal age.Such age-dependent changes are not the result of intake differences,but greater gastric accommodation and reduced emptying rate.AIM Hypothesizing that breastmilk-derived adiponectin is the factor regulating gastric accommodation in neonates,we comparatively evaluated its effects on the rat fundic muscle tone at different postnatal ages.METHODS In freshly dispersed smooth muscle cells(SMC),we measured the adiponectin effect on the carbachol-induced length changes.RESULTS Adiponectin significantly reduced the carbachol-stimulated SMC shortening independently of age.In the presence of the inhibitor iberiotoxin,the adiponectin effect on SMC shortening was suppressed,suggesting that it is mediated via largeconductance Ca^(2+)sensitive K^(+)channel activation.Lastly,we comparatively measured the newborn rat gastric milk curd adiponectin content in one-and twoweek-old rats and found a 50%lower value in the latter.CONCLUSION Adiponectin,a major component of breastmilk,downregulates fundic smooth muscle contraction potential,thus facilitating gastric volume accommodation.This rodent’s adaptive response maximizes breastmilk intake volume after birth.展开更多

文摘Drug adulteration and contamination are serious threats to human health therefore,their accurate monitoring is very important.Allopurinol(Alp)and theophylline(Thp)are commonly used drugs for the treatment of gout and bronchitis,while their isomers hypoxanthine(Hyt)and theobromine(Thm)have no effect and affect the efficacy of the drug.In this work,the drug isomers of Alp/Hyt and Thp/Thm are simply mixed withα-,β-,γ-cyclodextrin(CD)and metal ions and separated using trapped ion mobility spectrometry-mass spectrometry(TIMS-MS).TIMS-MS results showed that Alp/Hyt and Thp/Thm isomers could interact with CD and metal ions and form corresponding binary or ternary complexes to achieve their TIMS separation.Different metal ions and CDs showed different separation effect for the isomers,among which Alp and Hyt could be successfully distinguished from the complexes of[Alp/Hyt+γ-CD+Cu–H]^(+)with separation resolution(RP–P)of 1.51;whereas Thp and Thm could be baseline separated by[Thp/Thm+γ-CD+Ca–H]^(+)with RP–P of 1.96.Besides,chemical calculations revealed that the complexes were in the inclusion forms,and microscopic interactions were somewhat different,making their mobility separation.Moreover,relative and absolute quantification was investigated with an internal standard to determine the precise isomers content,and good linearity(R^(2)>0.99)was obtained.Finally,the method was applied for the adulteration detection where different drugs and urine were analyzed.In addition,due to the advantages of fast speed,simple operation,high sensitivity,and no chromatographic separation required,the proposed method provides an effective strategy for the drug adulteration detection of isomers.

文摘Base editors of the Cas9 system have been widely used for precise nucleotide substitution in crops. In this study, Cas12a was applied to construct plant cytosine base editors(CBEs). The main elements of Cas12aCBEs were engineered and their efficiency was evaluated in stably transformed rice cells. An optimized ttCas12a-hyA3Bctd editor, consisting of a LbCas12a variant carrying catalytic inactive D832A and temperature-tolerance D156R double mutations, a truncated human APOBEC3B deaminase, a human RAD51 single-stranded DNA-binding domain, and double copies of UGI, outperformed other Cas12aCBEs in base editing efficiency. In T0transgenic rice plants, ttCas12a-hyA3Bctd edited an average of42.01% and a maximum of 68.75% of lines at six genomic targets. A-to-G conversions were generated in rice by an adenine base editor with a similar architecture to the optimized CBE. Our results provide preliminary evidence for the feasibility of robust and efficient plant Cas12a base editing systems, which could be useful for precise crop breeding.

文摘Mining the core value of Industrial Internet of Things(IIoT)data safely and reducing the risk of malicious attacks are the inherent requirements of industrial data visualization.Visualization technology has become the main tool for data aggregation,mining and analysis of IIoT data through graphical representation.However,visualization technology still has two shortcomings in big data calculation and analysis scenarios.On the one hand,visual results will lead to the disclosure of sensitive privacy.On the other hand,most visualization tools can't provide an interactive framework for users to select the suitable solutions.To address these problems,we present an open accessible Visual framework based on Differential Privacy theory(VisDP),which provides Multi-index Quantitative comprehensive Evaluation technology(MQE)for data mining results.Considering the advantages of interactive mechanism,VisDP provides rich optional schemes,including the operating web,calling API and the downloading SDK.Finally,we verify the availability and privacy of MQE through mathematical proofs,analyze the hospital medical waste detection system that actually applies the framework,and the experimental results have showed the effectiveness and practicality of the proposed platform.

文摘Wax apple(Syzygium samarangense)is an economically important fruit crop with great potential value to human health because of its richness in antioxidant substances.Here,we present a haplotype-resolved autotetraploid genome assembly of the wax apple with a size of 1.59 Gb.Comparative genomic analysis revealed three rounds of whole-genome duplication(WGD)events,including two independent WGDs after WGT-γ.Resequencing analysis of 35 accessions partitioned these individuals into two distinct groups,including 28 landraces and seven cultivated species,and several genes subject to selective sweeps possibly contributed to fruit growth,including the KRP1-like,IAA17-like,GME-like,and FLACCA-like genes.Transcriptome analysis of three different varieties during flower and fruit development identified key genes related to fruit size,sugar content,and male sterility.We found that AP2 also affected fruit size by regulating sepal development in wax apples.The expression of sugar transport-related genes(SWEETs and SUTs)was high in‘ZY’,likely contributing to its high sugar content.Male sterility in‘Tub’was associated with tapetal abnormalities due to the decreased expression of DYT1,TDF1,and AMS,which affected early tapetum development.The chromosome-scale genome and large-scale transcriptome data presented in this study offer new valuable resources for biological research on S.samarangense and shed new light on fruit size control,sugar metabolism,and male sterility regulatory metabolism in wax apple.

文摘The covers of booklets and books in folk documents primarily serve to protect the pages.Owing to long-term storage limitations,a considerable number of book covers have suffered varying degrees of damage.Following the principles of restoration,a comparative analysis and restoration of folk document covers were conducted,selecting four different types of carriers from the Taihang Mountain Documents,ranging from the Qing dynasty to the Republican Era.These carriers included hemp,mulberry bark,and machinemade paper,and cotton blue cloth.Each cover type was matched with an appropriate restoration paper,and different methods were employed during the restoration process.Through restoration,the previously damaged document covers can continue to fulfill their role in protecting the books,thereby extending the lifespan of these four folk documents.
文摘Objective:To explore the mechanism of circRNA-vgll3 in osteogenic differentiation of human bone marrow mesenchymal stem cells.Methods:BMSCs cells were transfected with circRNA-vgll3,and divided into circRNA-vgll3 high-level group,circRNA-vgll3 low-level group,and negative control group(circRNA-vgll3 not transfected)according to the amount of transfection.The proliferation and apoptosis of BMSCs osteoblasts in each group were analyzed,and the alkaline phosphatase(ALP)activity,type I collagen gray value,bone morphogenetic protein 2(BMP-2),Runx2 protein,and mRNA expression levels were detected.Results:The circRNA-vgll3 low-level group had a significant inhibitory effect on the proliferation of BMSCs osteoblasts,and the apoptosis rate of the circRNA-vgll3 low-level group was significantly higher than that of the circRNA-vgll3 high-level group(P<0.05);ALP activity,type I collagen gray value,BMP-2,Runx2 protein,and mRNA expression levels in the high-level circRNA-vgll3 group were significantly higher than those in the low-level circRNA-vgll3 group,and the difference was statistically significant(P<0.05).Conclusion:Overexpression of circRNA-vgll3 can promote the osteogenic differentiation ability of BMSCs,while low expression of circRNA-vgll3 can inhibit the osteogenic differentiation ability of BMSCs.The main mechanism of action is that circRNA-vgll3 can affect osteogenic differentiation by regulating the Runx2 protein.
文摘This paper studies the time-fractional Korteweg-de Vries (KdV) equations with Caputo-Fabrizio fractional derivatives. The scheme is presented by using a finite difference method in temporal variable and a local discontinuous Galerkin method (LDG) in space. Stability and convergence are demonstrated by a specific choice of numerical fluxes. Finally, the efficiency and accuracy of the scheme are verified by numerical experiments.
文摘In this paper we investigate a class of impulsive differential equations with Dirichlet boundary conditions. Firstly, we define new inner product of <img src="Edit_890fce38-e82b-4f36-be40-9d05e8119b88.png" width="40" height="17" alt="" /> and prove that the norm which is deduced by the inner product is equivalent to the usual norm. Secondly, we construct the lower and upper solutions of (1.1). Thirdly, we obtain the existence of a positive solution, a negative solution and a sign-changing solution by using critical point theory and variational methods. Finally, an example is presented to illustrate the application of our main result.

文摘The effects of exogenous gibberellic acid (GA(3)) and abscisic acid (ABA) on the total respiratory rate, percentages of total respiratory rate contributed by respiratory pathways [Embden-Meyerhof- Parnas Pathway (EMP), Pentose Phosphate Pathway (PPP), and Tricarboxylic Acid Cycle (TCA)], and conversion of starch to soluble sugars in the buds of black currants during secondary bud burst were investigated to determine the relationship between respiratory rates and secondary bud burst. 'Adelinia', a black currant cultivar that is prone to secondary bud burst after the first harvest, was used in this study. Mature bushes of Adelinia were sprayed with 30 mg/L GA(3) and 50 mg/L ABA to manipulate bud burst. The results showed that exogenous applications of GA(3) and ABA had opposite effects on bud respiratory rate. Generally, GA(3) treatment increased the total respiratory rate and respiratory rate of the TCA and PPP, and the respiratory rates after GA(3) treatment were higher than those of control. While ABA treatment mostly decreased the total respiratory rate and the respiratory rate of TCA and PPP in buds in comparison to control. In terms of the percentage of the three respiratory rates in comparison to the total respiratory rate, GA(3) treatment significantly increased the percentage of TCA and PPP respiratory rate in comparison to the control (P < 0.01), whereas ABA decreased the rates. GA(3) significantly increased the content of soluble sugars and decreased the starch content, while the starch content in buds after ABA treatment was significantly higher than that of the control. All results showed that PPP is a critical process for the second bud burst in black currants. While the EMP-TCA pathway is still dominant in bud respiration, provides a series of basic materials and energy (ATP). The conversion of starch to soluble sugars is essential for bud burst. Thus, we conclude that an energy shortage is a main contributor in ABA inhibition of the secondary bud burst of black currants.

文摘BACKGROUND Oral intake is dependent on the gastric ability to accommodate the food bolus.Comparatively,neonates have a smaller gastric capacity than adults and this may limit the volume of their milk intake.Yet,we previously reported that the newborn rat gastric milk volume is greatest after birth and,when normalized to body weight,decreases with postnatal age.Such age-dependent changes are not the result of intake differences,but greater gastric accommodation and reduced emptying rate.AIM Hypothesizing that breastmilk-derived adiponectin is the factor regulating gastric accommodation in neonates,we comparatively evaluated its effects on the rat fundic muscle tone at different postnatal ages.METHODS In freshly dispersed smooth muscle cells(SMC),we measured the adiponectin effect on the carbachol-induced length changes.RESULTS Adiponectin significantly reduced the carbachol-stimulated SMC shortening independently of age.In the presence of the inhibitor iberiotoxin,the adiponectin effect on SMC shortening was suppressed,suggesting that it is mediated via largeconductance Ca^(2+)sensitive K^(+)channel activation.Lastly,we comparatively measured the newborn rat gastric milk curd adiponectin content in one-and twoweek-old rats and found a 50%lower value in the latter.CONCLUSION Adiponectin,a major component of breastmilk,downregulates fundic smooth muscle contraction potential,thus facilitating gastric volume accommodation.This rodent’s adaptive response maximizes breastmilk intake volume after birth.

文摘Objective The SYNTAX score(Synergy between Percutaneous Coronary Intervention with Taxus and Cardiac Surgery,SS)is now broadly accepted to guide and predict mortality of complex coronary artery disease(CAD)patients undergoing PCI or CABG.Recently,the clinical SYNTAX score which is a combination of age,ejection fraction,decrease in creatinine clearance and classic SYNTAX score,has been found to be superior in predicting adverse outcomes compared with the SYNTAX scores.Whether the clinical SS could help to determine which patients could be managed medically has never been explored.We sought to investigate the prognostic impact of the SS on long-term clinical outcomes in patients with three vessels disease undergoing medical therapy only.

文摘Dear Editor,Accurately labeling proteins in living plant cells has long been a challenge and can be addressed by targeted insertion of tag sequences in a given locus.Recent optimized plant prime editors(PEs)enable efficient programmable installation of small insertions or deletions,including insertions of short sequences(Li et al.,2022a,2022b;Jiang et al.,2022;Xu et al.,2022;Zong et al.,2022;Zou et al.,2022).To investigate whether prime editing can be used to tag endogenous proteins in rice,we made use of the enpPE2 system described in our previous report(Li et al.,2022b).
