目的:评价麻黄附子细辛汤治疗病态窦房结综合征的疗效与安全性,为临床实践提供循证参考.方法:采用Cochrane系统评价方法,通过计算机检索和手工检索,全面收集麻黄附子细辛汤治疗病态窦房结综合征的所有语种随机对照试验,检索时间为200... 目的:评价麻黄附子细辛汤治疗病态窦房结综合征的疗效与安全性,为临床实践提供循证参考.方法:采用Cochrane系统评价方法,通过计算机检索和手工检索,全面收集麻黄附子细辛汤治疗病态窦房结综合征的所有语种随机对照试验,检索时间为2000年至2017年01月,由2名评价者按照纳入和排除标准独立选择文献、提取资料、评价质量、交叉核对并进行方法学质量评估,若遇分歧,通过讨论或者第三名研究者协助解决,对纳入文献采用RevMan5.3软件进行Meta分析.结果:共纳入7篇文献,共612例患者,麻黄附子细辛汤(治疗组)358例,对照组254例.纳入的7篇文献均报告了有效率,治疗组有效率(86.9%)高于对照组(70.1%),差异具有统计学意义[RR=1.25,95%CI(1.15,1.37),P〈0.00001];纳入的文献中有5篇报告了不良反应,治疗组不良反应发生率(17.0%)明显低于对照组(49.8%),差异具有统计学意义[RR=0.23,95%CI(0.06,0.93),P=0.04].结论:现有临床研究提示,麻黄附子细辛汤治疗病态窦房结综合征疗效明显且未见明显不良反应;受纳入文献质量限制,以上结论有待开展更多高质量随机对照试验予以验证. 展开更多

A method for isolating rat cardiomyocytes by acute enzymatic hydrolysis

Method: Our method was divided into five steps: preparation, heart ext raction, perfusion, mechanical dissociation and pu rifica tion and r ecalcifica ti on. Fir stly, make a preparation, including solutions, equipment and so on. Secondly, after anesthesia satisfactorily, open the chest of rat and take the heart out. Thirdly, transect the aorta and hang the aorta onto the Langendroff perfusion system such that the tip is just distal to the aortic valve. For th, perfuse the hea rt with diges tion solution for 17-20 min afte r Ca2+ free tyrode solution for 5 min. Finally, remove the heart from the system quickly and dissociate mechanically in the KB solution and recalcification after filter. Result: Though the microscope, good-state cardiomyocytes (clear stripes, strong sense of three-dimensional, stick well) can be observed. Conclusion: Those cells can be used to undergo medical electrophysiological experiment, including the study of iron channel. 展开更多

Effects of hypoxia on sodium current in rat cardiomyocytes

本文旨在研究缺氧对大鼠心肌细胞持续钠电流的影响,以探讨心肌细胞缺氧发生的离子通道机制。急性酶解法分离大鼠心肌细胞,一组未做处理,使用全细胞膜片钳技术记录钠电流;一组进行缺氧处理,使用全细胞膜片钳技术记录钠电流。结果显示:与... 本文旨在研究缺氧对大鼠心肌细胞持续钠电流的影响,以探讨心肌细胞缺氧发生的离子通道机制。急性酶解法分离大鼠心肌细胞,一组未做处理,使用全细胞膜片钳技术记录钠电流;一组进行缺氧处理,使用全细胞膜片钳技术记录钠电流。结果显示:与正常细胞比较,缺氧细胞钠电流形态和I-V曲线的形态轨迹特征不变;相对于正常细胞,缺氧细胞的I-V曲线上移明显,INa峰值电流相对正常细胞上调了15.68%(P<0.0001);正常与缺氧细胞激活电位均在-40mV左右,最大电流峰值对应刺激电压为-25mV。上述结果提示,缺氧情况下,大鼠心肌细胞持续钠电流增大。 展开更多

目的:探讨定悸复脉汤对室性心律失常大鼠心电图及Na^+-K^+-ATP酶的影响。方法:将健康雄性SD大鼠40只(200±20g)随机分为空白组、模型组、美托洛尔组及定悸复脉汤组,每组10只,均喂养普通饲料和自由饮水,各组均给予灌胃,各组灌胃量均... 目的:探讨定悸复脉汤对室性心律失常大鼠心电图及Na^+-K^+-ATP酶的影响。方法:将健康雄性SD大鼠40只(200±20g)随机分为空白组、模型组、美托洛尔组及定悸复脉汤组,每组10只,均喂养普通饲料和自由饮水,各组均给予灌胃,各组灌胃量均按体重计算。模型组给予尾静脉注射0.001%乌头碱30ug/kg,美托洛尔组按每天5.2mg/kg的规格灌服美托洛尔悬液,定悸复脉汤组按每天17.6g/kg剂量给予定悸复脉汤灌胃。给药2周后,通过生物机能实验系统BL-420F监测Ⅱ导联心电图曲线观察并且记录心电图的变化,然后取大鼠左心室,部分行心脏组织的Na^+-K^+-ATP酶检测。结果:(1)定悸复脉汤对室性心律失常大鼠心电图的影响:静脉推注乌头碱后,模型组VP、VT、VF发生率为100%,提示室性心律失常大鼠模型造模成功。(2)室性早搏(VP)、室性心动过速(VT)、室性颤动(VF)出现时间:与模型组比较,定悸复脉汤组和美托洛尔组可显著推迟VP、VT、VF出现时间,差异具有统计学意义(P<0.05),定悸复脉汤组与美托洛尔组在推迟VP、VT、VF出现时间方面作用相同,无明显差异(P>0.05)。(3)定悸复脉汤对大鼠室性心律失常心脏组织Na^+-K^+-ATP酶的影响:与空白组比较,模型组组织Na^+-K^+-ATP酶值明显降低,差异具有统计学意义(P<0.05)。与模型组比较,美托洛尔组和定悸复脉汤组的组织Na^+-K^+-ATP酶值升高,差异具有统计学意义(P<0.05);美托洛尔组与定悸复脉汤组组织Na^+-K^+-ATP酶值无明显差异(P>0.05)。结论:1.采用静脉推注乌头碱的方法可以成功制备室性心律失常大鼠模型。2.定悸复脉汤可延长乌头碱所致大鼠VP、VT、VF等心律失常的出现时间,其机制可能与快Na^+通道相关,可能通过抑制Na^+内流而降低快反应细胞自律性而防治心律失常。3.定悸复脉汤能保护心肌组织Na^+-K^+-ATP酶,具有保护心肌细胞,从而改善心肌代谢作用。 展开更多

Objective:To report a patient with acne by using the combined treatment of pricking cupping bloodletting and slide cupping.Methods:Pricking cupping bloodletting was prescribed at the points of Feishu(BL13)in the bladder meridian of foot-Taiyang and Dazhui(DU14)in governor vessel and slide cupping was prescribed at the points on the back in the bladder meridian of foot-Taiyang.Results:Clinical manifestations revealed that acne and scar were significantly reduced after two courses.Conclusion:Pricking cupping bloodletting combined with slide cupping can successfully relieve acne and promote skin healing,which may provide us a new idea for the non-drug therapy of acne. 展开更多

目的:通过肝阳上亢型肾性高血压大鼠的降压实验来探索“风药”(防风,白芷)在高血压病治疗中的降压增效作用及其作用机理.方法:筛选雄性SD大鼠,计算机随机分组后制备肝阳上亢型肾性高血压大鼠模型,建模成功后随机分组予以相应药液灌... 目的:通过肝阳上亢型肾性高血压大鼠的降压实验来探索“风药”(防风,白芷)在高血压病治疗中的降压增效作用及其作用机理.方法:筛选雄性SD大鼠,计算机随机分组后制备肝阳上亢型肾性高血压大鼠模型,建模成功后随机分组予以相应药液灌胃4周,然后腹主动脉取血,制备血清.采用化学比色法测定血清一氧化氮(NO)浓度,ELISA法测定血清内皮素(ET)、血栓素B2(TXB2)、6-酮-前列腺素(6-K-PGF1a)含量.结果:与模型对照组大鼠相比,风药组大鼠血压及血清ET与TXB2并没有明显降低(P〉0.05)血清NO与6-K-PGF1a并没有明显增高(P〉0.05),但是卡托普利组、天麻钩藤饮Ⅰ号组血压及血清ET与TXB2均明显降低(P〈0.05).同时,与模型对照组大鼠相比,卡托普利组、天麻钩藤饮Ⅰ号组血清NO与6-K-PGF1a均明显增高(P〈0.05).有趣的是,风药+天麻钩藤饮Ⅰ号联合治疗组的大鼠血压及血清ET与TXB2较风药组及天麻钩藤饮Ⅰ号单药组均明显降低(P〈0.05);同时,血清NO与6-K-PGF1a较风药组及天麻钩藤饮Ⅰ号单药组均明显升高(P〈0.05).结论:1.风药单独降压作用并不明显,但可以增强天麻钩藤饮Ⅰ号的降压作用;2.风药与天麻钩藤饮Ⅰ号联合增效机制可能与降低ET与TXB2及上调NO与6-K-PGF1a有关. 展开更多

目的:观察蛭龙活血通瘀胶囊拆方组分对大鼠血管NF-κB-P65、ICAM-1表达的影响,探讨蛭龙活血通瘀胶囊及其拆方组分对高血压血管重构的影响及作用机制.方法:手术组缩窄左肾动脉建立肾性高血压大鼠模型.分别给予相应药物1次/日灌胃,以同... 目的:观察蛭龙活血通瘀胶囊拆方组分对大鼠血管NF-κB-P65、ICAM-1表达的影响,探讨蛭龙活血通瘀胶囊及其拆方组分对高血压血管重构的影响及作用机制.方法:手术组缩窄左肾动脉建立肾性高血压大鼠模型.分别给予相应药物1次/日灌胃,以同体积生理盐水灌胃正常组、假手术组与模型组.灌胃4周后,取大鼠胸主动脉,依次固定、包埋、切片、HE染色,免疫组化的方法检测血管NF-κB-p65.结果:1.药物对高血压大鼠胸主动脉NF-κB蛋白表达的影响(免疫组化法):电镜下,棕黄色颗粒为NF-κB蛋白阳性表达,主要分布于血管中膜平滑肌.正常组及假手术组可见少量的棕黄色颗粒;模型组可见散在分布的大量表达棕黄色颗粒;经治疗后,卡托普利组、全方组表达较模型组明显减少,补气组、温通经络组、活血化瘀组表达较模型组有所减少.其中卡托普利组较全方组有所减少;补气组较活血化瘀组表达明显减少,与温通经络组表达相当.结论:蛭龙活血通瘀胶囊其对肾性高血压大鼠血管重构机制,可能与其降低血压、抗氧化应激、抗炎作用有关.各组方配伍后产生了显著的协同增效作用,其配伍后体现了中医药多靶点的治疗优势. 展开更多
