Plasmonic nanostructure-mediated photothermal therapy(PTT) has proven to be a promising approach for cancer treatment, and new approaches for its effective delivery to tumor lesions are currently being developed. This study aimed to assess macrophage-mediated delivery of PTT using radioiodine-124-labeled gold nanoparticles with crushed gold shells(124I-Au@AuCBs) as a theranostic nanoplatform.124I-Au@AuCBs exhibited effective photothermal conversion effects both in vitro and in vivo and were efficiently taken up by macrophages without cytotoxicity. After the administration of 124I-Au@AuCB-labeled macrophages to colon tumors, intensive signals were observed at tumor lesions, and subsequent in vivo PTT with laser irradiation yielded potent antitumor effects. The results indicate the considerable potential of 124I-Au@AuCBs as novel theranostic nanomaterials and the prominent advantages of macrophage-mediated cellular therapies in treating cancer and other diseases. 展开更多

Hot plate forming using a cell-typed die is a process for forming a large thick plate with a spherical shape for the manufacture of a large spherical LNG tank.Cell-typed upper and lower dies made of a framework of steel plates fitted to make a grid pattern are used in this process, and an air-cooling device is separately installed inside the lower die.A finite element analysis (FEA) technique was developed, which included hot forming, air flow, cooling and thermal deformation analysis for the hot plate forming process using the cell-typed die.Further, the convective and interface heat transfer coefficients were used to reproduce analytically the effects of the cooling device in the hot plate forming analysis.A small-scale model test of the process was conducted to verify the FEA technique. The analysis results show that the curvature of the final plate agrees well with that of the designed experiment within a maximum relative error of 0.03% at the corner of the plate. 展开更多

Age-related macular degeneration(AMD)causes blindness due to loss of retinal pigment epithelium(RPE)and photoreceptors(PRs),which comprise the two outermost layers of the retina.Given the small size of the macula and the importance of direct contact between RPE and PRs,the use of scaffolds for targeted reconstruction of the outer retina in later stage AMD and other macular dystrophies is particularly attractive. We developed microfabricated,honeycomb-patterned,biodegradable poly(glycerol sebacate)(PGS)scaffolds to deliver organized, adjacent layers of RPE and PRs to the subretinal space.Furthermore,an optimized process was developed to photocure PGS,shortening scaffold production time from days to minutes. The resulting scaffolds robustly supported the seeding of human pluripotent stem cell-derived RPE and PRs,either separately or as a dual celllayered construct. These advanced, economical, and versatile scaffolds can accelerate retinal cell transplantation efforts and benefit patients with AMD and other retinal degenerative diseases. 展开更多
