Colloidal gold was treated by a light-sensitive si...A novel sensitive electrochemical immunoassay with colloidal gold as the antibody labeling tag and subsequent signal amplification by silver enhancement is described. Colloidal gold was treated by a light-sensitive silver enhancement system which made silver deposit on the surface of colloidal gold(form Au/Ag core-shell structure), followed by the release of the metallic silver atoms anchored on the antibody by oxidative dissolution of them in an acidic solution and the indirect determination of the dissolved Ag + ions by anodic stripping voltammetry(ASV) at a carbon fiber microelectrode. The electrochemical signal is directly proportional to the amount of analyte(goat IgG) in the standard or a sample. The method was evaluated by means of a noncompetitive heterogeneous immunoassay of immunoglobulin G(IgG) with a concentration as low as 0.2 ng/mL. The high performance of the method is related to the sensitive ASV determination of silver(Ⅰ) at a carbon fiber microelectrode and to the release of a large number of Ag + ions from each silver shell anchored on the analyte(goat IgG).展开更多

Methods: Fifty B...To investigate the effect of arsenic trioxide (As2O3 )with three different concentration groups on the distribution and recruitment of dendritic cells(DCs) in the conducting airways of asthmatic mice. Methods: Fifty BALB/c mice were divided into 5 groups at random: control group, asthmatic group, therapeutic groups with low dose(1 mg/kg), moderate dose( 5 mg/lqg) and high dose( 10 mg/kg) of As2O3. The immunohistochomistry, scanning electron microscope and computerized image analysis were applied to detect airway DCs, respectively. Results: We demonstrated from the control mice that all intraepithelial NLDC-145 DCs throughout the respiratory tree cotdd be accounted for a network of cells with dendritic cell morphology, and the density of DCs varied from(500±50) cells/ram^2 epithelial surface in the large airways, to(60±10)cells/mm^2 epithelial surface in the small airways(P<0.05).The DCs characteristic of spiny and long processes were impressively observed by the scanning electron microscope. Comtmred with the control mlce,the density,but not the distribution of NLDC-145 DCs was significantly upregulated in the conducing airways of asthmatic mice(P<0.01) .Compared with the asthmatic mice, the densilies, but not the distributions of NLDC-145 DCs were significantly down regulated in the conducting airways of the thempeutic groups with three varying concentrations of As2O3(P<0.01 )while there were no statistical differences between each other(P>0.05).Conchsion : Our findings suggest that it might be an important therapeutic mechanism of As2 03 to downregulate not the distribution but the density of DCs in the conducting airways of asthmatic mice,and low dose of As203 has potential value in treating asthma.展开更多

Methods: Thirty patients undergoing gastric carcinectomy were randomly divi...Objective: To study the effects of allogeneic blood transfusion on polarized Th cell in gastric carcinoma patients during perioperative time. Methods: Thirty patients undergoing gastric carcinectomy were randomly divided into three groups with 10 patients in each group: group A received no allogeneic blood; group B received leukodepleted blood; group C received allogeneic whole blood during perioperative time. The polarized rates of Th cell were measured by flow cytometry. Results: The counts of Thl in group A decreased significantly on the 2nd postoperation, but returned to the baseline levels on the 5th postoperative day. The Thl counts and Th1/Th2 ratio decreased significantly in group B and group C after blood transfusion (P<0.05). The Th2 counts increased significantly in group B and C postoperative time. On the 10th postoperative day the Th1 ,Th2 cell counts and Th1/Th2 ratio returned to preoperative level in group B, but remained low in group C. As compared with that in group A, the Th1 cell counts, Th1/Th2 ratio reduced and Th2 cell counts increased markedly in group C than in group B(P<0.05). Conclusion: Perioperative blood transfusion contributes to the immunosuppression, which is more serious after whole blood transfusion than leukodepleted blood. In perioperative period component blood transfusion is superior to whole blood transfusion. The Th cells polarized to Th2 cells may be one of the causes of immunosuppression in patients received allogeneic blood transfusion.展开更多
