AIM:To investigate the potential role of Active Chinese mistletoe lectin-55 (ACML-55) in tumor immune surveillance. METHODS: In this study, an experimental model was established by hypodermic inoculating the colon cancer cell line CT26 (5 × 105 cells) into BALB/c mice. The experimental treatment was orally administered with ACML-55 or PBS, followed by the inoculation of colon cancer cell line CT26. Intracellular cytokine staining was used to detect IFN-γ production by tumor antigen specific CD8+ T cells. FACS analysis was employed to profi le composition and activation of CD4+, CD8+, γδ T and NK cells. RESULTS: Our results showed, compared to PBS treated mice, ACML-55 treatment signifi cantly delayed colon cancer development in colon cancer -bearingBalb/c mice in vivo. Treatment with ACML-55 enhanced both Ag specifi c activation and proliferation of CD4+ and CD8+ T cells, and increased the number of tumor Ag specific CD8+ T cells. It was more important to increase the frequency of tumor Ag specific IFN-γ producing-CD8+ T cells. Interestingly, ACML-55 treatment also showed increased cell number of NK, and γδT cells, indicating the role of ACML-55 in activation of innate lymphocytes. CONCLUSION: Our results demonstrate that ACML-55 therapy can enhance function in immune surveillance in colon cancer-bearing mice through regulating both innate and adaptive immune responses. 展开更多

Objective: To investigate the effects of montelukast sodium combined with budesonide on pulmonary function, inflammatory factors and immunoglobulin in children with asthma. Methods: A total of 100 children with asthma treated in our hospital from May 2013 to November 2016 were randomly divided into control group and observation group. In the control group, budesonide inhalation therapy was given. On the basis of the control group, montelukast sodium treatment was given, pulmonary function, inflammatory factors and immunoglobulin levels in the two groups before and after treatment were measured. Results:Compared with before treatment, the levels of FEV1%, FEF50%, FEF25% and PEF% in the two groups were significantly increased, those in the observation group were higher than in control group;compared with before treatment, IL-10 levels were significantly increased, TNF-α and IL-8 levels were significantly decreased in the two groups after treatment, moreover those in the observation group IL-10 levels were significantly higher than the control group, TNF-α and IL-8 were significantly lower than those in the control group, the difference was statistically significant;Compared with before treatment, IgA and IgM levels were significantly increased in both groups after treatment, IgE levels were significantly decreased, and IgA and IgM levels in the observation group were significantly higher than in the control group, IgE levels were significantly lower than the control group, the difference was statistically. Conclusion: Montelukast sodium combined with budesonide can effectively reduce the inflammation level of children with asthma, improve lung function, enhance immunity, the effect is significant, it is worth further clinical application. 展开更多

Meniscus is a wedge-shaped fibrocartilaginous tissue,playing important roles in maintaining joint stability and function.Meniscus injuries are difficult to heal and frequently progress into structural breakdown,which then leads to osteoarthritis.Regeneration of heterogeneous tissue engineering meniscus(TEM)continues to be a scientific and translational challenge.The morphology,tissue architecture,mechanical strength,and functional applications of the cultivated TEMs have not been able to meet clinical needs,which may due to the negligent attention on the importance of microenvironment in vitro and in vivo.Herein,we combined the 3D(three-dimensional)-printed gradient porous scaffolds,spatiotemporal partition release of growth factors,and anti-inflammatory and anti-oxidant microenvironment regulation of Ac2-26 peptide to prepare a versatile meniscus composite scaffold with heterogeneous bionic structures,excellent biomechanical properties and anti-inflammatory and anti-oxidant effects.By observing the results of cell activity and differentiation,and biomechanics under anti-inflammatory and anti-oxidant microenvironments in vitro,we explored the effects of anti-inflammatory and anti-oxidant microenvironments on construction of regional and functional heterogeneous TEM via the growth process regulation,with a view to cultivating a high-quality of TEM from bench to bedside. 展开更多
