Enhanced tolerance to drought in transgenic rice plants overexpressing C_4 photosynthesis enzymes 被引量:7

Maize-specific pyruvate orthophosphate dikinase(PPDK) was overexpressed in rice independently or in combination with the maize C4-specific phosphoenolpyruvate carboxylase(PCK). The wild-type(WT) cultivar Kitaake and transgenic plants were evaluated in independent field and tank experiments. Three soil moisture treatments,well-watered(WW), moderate drought(MD) and severe drought(SD), were imposed from 9d post-anthesis till maturity. Leaf physiological and biochemical traits, root activities,biomass, grain yield, and yield components in the untransformed WT and two transgenic rice lines(PPDK and PCK) were systematically studied. Compared with the WT, both transgenic rice lines showed increased leaf photosynthetic rate: by 20%–40% under WW, by45%–60% under MD, and by 80%–120% under SD. The transgenic plants produced 16.1%,20.2% and 20.0% higher grain yields than WT under the WW, MD and SD treatments,respectively. Under the same soil moisture treatments, activities of phosphoenolpyruvate carboxylase(PEPC) and carbonic anhydrase(CA) in transgenic plants were 3–5-fold higher than those in WT plants. Compared with ribulose-1,5-bisphosphate carboxylase, activities of PEPC and CA were less reduced under both MD and SD treatments. The transgenic plants also showed higher leaf water content, stomatal conductance, transpiration efficiency, and root oxidation activity and a stronger active oxygen scavenging system than the WT under all soil moisture treatments, especially MD and SD. The results suggest that drought tolerance is greatly enhanced in transgenic rice plants overexpressing C4photosynthesis enzymes. This study was performed under natural conditions and normal planting density to evaluate yield advantages on a field basis. It may open a new avenue to droughttolerance breeding via overexpression of C4enzymes in rice. 展开更多

Effects of heat treatment on microstructure and mechanical properties of Mg-6Zn-4Sm-0.4Zr alloy

A new rare earth magnesium alloy(Mg-6 Zn-4 Sm-0.4 Zr, wt.%) was prepared by permanent mould casting. The microstructure and mechanical properties of the alloy sample in as-cast and various heat treatment situations were characterized with an optical microscope(OM), X-ray diffractometer(XRD), scanning electron microscope(SEM) equipped with energy dispersive spectroscope(EDS), transmission electron microscope(TEM) and mechanical tests at room temperature, respectively. The experimental results show that the as-cast alloy mainly consists of α-Mg, eutectic Mg_2Zn_3, MgZnSm and Mg_(41)Sm_5. These eutectic phases with continuous or semicontinuous morphology principally distribute along grain boundaries. Almost all the eutectic compounds dissolve in α-Mg and the grains have no obvious growth trend after optimum solution treatment at 490 °C for 18 h. Meanwhile, the ultimate tensile strength(UTS) of 229 MPa and elongation(EL) to rupture of 9.78% can be achieved through the optimal solution treatment, which increase by 37 MPa and 57.74%, respectively, compared with that of the as-cast alloy. Further aging treatments at 200 °C for different durations lead to the conspicuous increment of mechanical properties and prominent age-hardening response. Peak-aged alloy(treated at 200 °C for 12 h) reveals better mechanical properties(UTS 258 MPa, EL 9.42%, hardness 73.4 HV) compared with the same alloy treated in other aging conditions, which is mainly ascribed to precipitated Mg_2Zn_3 and MgZn_2 phases. Fracture analysis demonstrates that the as-cast alloy belongs to inter-granular and cleavage fracture patterns, while the solutionized alloy(treated at 490 °C for 18 h) reveals trans-granular and quasi-cleavage fracture modes. For the peak-aged alloy, the fracture pattern obeys the mixture of trans-granular and cleavage modes. 展开更多

Much Improved Water Use Efficiency of Rice under Non-Flooded Mulching Cultivation 被引量:6

Water shortage is increasingly limiting the luxury use of water in rice cultivation. In this study, non-flooded mulching cultivation of rice only consumed a fraction of the water that was needed for traditional flooded cultivation and largely maintained the grain yield. We also investigated the growth and development of rice plants and examined grain yield formation when rice was subjected to non-flooded mulching cultivation. One indica hybrid rice combination was grown in a field experiment and three cultivation methods, traditional flooding (TF), non-flooded straw mulching cultivation (SM) and non-flooded plastic mulching cultivation (PM), were conducted during the whole season. Grain yield showed that there was no significant difference between SM and TF rice, but the grain yield of SM cultivation was significantly higher than that of PM. The tiller numbers were inhibited in the early stage under non-flooded mulching cultivation, but the situation was reversed at the later period. Both SM and PM rice reduced dry matter accumulation of shoot, but increased root dry weight, enhanced the remobilization of assimilates from stems to grains and increased the harvest index. During the middle and later grain filling period, mulched plants showed a faster decrease in chlorophyll concentrations, photosynthetic rates of flag leaves and root activity than TF rice, indicating that non-flooded mulching cultivation enhanced plant senescence. In comparison, SM treatment produced higher grain yield and, more dry matter accumulation and panicle numbers than the PM treatment. The overall results suggest that high yield of non-flooded mulching cultivation of rice can be achieved with much improved irrigaUonal water use efficiency. 展开更多
