AIM:To evaluate the impact of alcohol dehydrogenase-2(ADH2) and aldehyde dehydrogenase-2(ALDH2) polymorphisms on esophageal cancer susceptibility in Southeast Chinese males.METHODS:Two hundred and twenty-one esophageal cancer patients and 191 healthy controls from Taixing city in Jiangsu Province were enrolled in this study.ADH2 and ALDH2 genotypes were examined by polymerase chain reaction and denaturing high-performance liquid chromatography.Unconditional logistic regression was used to calculate the odds ratios(OR) and 95% confi dence interval(CI) .RESULTS:The ADH G allele carriers were more susceptible to esophageal cancer,but no association was found between ADH2 genotypes and risk of esophageal cancer when disregarding alcohol drinking status.Regardless of ADH2 genotype,ALDH2G/A or A/A carriers had significantly increased risk of developing esophageal cancer,with homozygous individuals showing higher esophageal cancer risk than those who were heterozygous.A signif icant interaction between ALDH2 and drinking was detected regarding esophageal cancer risk;the OR was 3.05(95% CI:1.49-6.25) .Compared with non-drinkers carrying both ALDH2 G/G and ADH2 A/A,drinkers carrying both ALDH2 A allele and ADH2 G allele showed a signif icantly higher risk of developing esophageal cancer(OR = 8.36,95% CI:2.98-23.46) .CONCLUSION:Both ADH2 G allele and ALDH2 A allele significantly increase the risk of esophageal cancer development in Southeast Chinese males.ALDH2 A allele significantly increases the risk of esophageal cancer development especially in alcohol drinkers.Alcohol drinkers carrying both ADH2 G allele and ALDH2 A allele have a higher risk of developing esophageal cancer. 展开更多

AIM: To investigate associations between the RsaⅠ polymorphism of CYP2E1 and risk of colorectal cancer. METHODS: A case-control study was conducted with 315 colorectal cancer cases (105 colon, 210 rectal) and 439 population-based controls in Jiangsu Province of China. Genomic DNA samples were assayed for restriction fragment length polymorphisms in CYP2E1 by PCR amplification followed by digestion with Rsa Ⅰ. Information on smoking and alcohol drinking was collected using a questionnaire. Odds ratios (ORs) were estimated with an unconditional logistic model. RESULTS: The proportional distribution of the CYP2E1 RsaⅠc1/c1, c1/c2 and c2/c2 genotypes were 61.4%, 35.6% and 3.0% in controls, 60.6%, 33.7% and 5.8% in colon cancer cases, and 58.4%, 34.0% and 7.7% in rectal cancer cases, respectively. A significant difference was noted between controls and rectal cancer cases (P = 0.029), the c2/c2 genotype being associated with elevated OR (adjusted age, sex and status of the smoking and alcohol drinking) for rectal cancer (1.64, 95% CI, 1.12-2.41, vs c1 allele carriers), but not for colon cancer. In interaction analysis between the CYP2E1 RsaⅠgenotype and smoking and drinking habits, we found a significant cooperative action between the c2/c2 genotype and alcohol drinking in the sex-, age-adjusted ORs for both colon (4.74, 95% CI, 1.10-20.40) and rectal (5.75, 95% CI, 1.65-20.05) cancers. Among nonsmokers, the CYP2E1 RsaⅠc2/c2 genotype was also associated with elevated ORs in the two sites (1.95, 95% CI, 0.99-3.86 and 2.30, 95% CI, 1.32-3.99). CONCLUSION: The results of the present study suggest that the CYP2E1 c2/c2 genotype increases susceptibility to rectal cancer and the gene-environmental interactions between the CYP2E1 polymorphism and smoking or alcohol drinking exist for colorectal neoplasia in general. 展开更多

AIM:To assess midterm results of stapled transanal rectal resection(STARR)for obstructed defecation syndrome(ODS)and predictive factors for outcome.METHODS:From May 2007 to May 2009,75 female patients underwent STARR and were included in the present study.Preoperative and postoperative workup consisted of standardized interview and physical examination including proctoscopy,colonoscopy,anorectal manometry,and defecography.Clinical and functional results were assessed by standardized questionnaires for the assessment of constipation constipation scoring system(CSS),Longo’s ODS score,and symptom severity score(SSS),incontinence Wexner incontinence score(WS),quality of life Patient Assessment of Constipation-Quality of Life Questionnaire(PAC-QOL),and patient satisfaction visual analog scale(VAS).Data were collected prospectively at baseline,12 and 30 mo.RESULTS:The median follow-up was 30 mo(range,30-46 mo).Late postoperative complications occurred in 11(14.7%)patients.Three of these patients required procedure-related reintervention(one diverticulectomy and two excision of staple granuloma).Although the recurrence rate was 10.7%,constipation scores(CSS,ODS score and SSS)significantly improved after STARR(P<0.0001).Significant reduction in ODS symptoms was matched by an improvement in the PAC-QOL and VAS(P<0.0001),and the satisfaction index was excellent in 25(33.3%)patients,good in 23(30.7%),fairly good in 14(18.7%),and poor in 13(17.3%).Nevertheless,the WS increased after STARR(P=0.0169).Incontinence was present or deteriorated in 8(10.7%)patients;6(8%)of whom were new onsets.Univariate analysis revealed that the occurrence of fecal incontinence(preoperative,postoperative or new-onset incontinence;P=0.028,0.000,and 0.007,respectively)was associated with the success of the operation.CONCLUSION:STARR is an acceptable procedure for the surgical correction of ODS.However,its impact on symptomatic recurrence and postoperative incontinence may be problematic. 展开更多

AIM: To evaluate the relationship between drinking and polymorphisms of alcohol dehydrogenase 2 (ADH2) and/or aldehyde dehydrogenase 2 (ALDH2) for risk of colorectal cancer (CRC) in Chinese males. METHODS: A case-control study was conducted in 190 cases and 223 population-based controls. ADH2 Arg47His (G-A) and ALDH2 Glu487Lys (G-A)genotypes were identified by PCR and denaturing high-performance liquid chromatography (DHPLC). Information on smoking and drinking was collected and odds ratio (OR) was estimated. RESULTS: The ADH2 A/A and ALDH2 G/G genotypes showed moderately increased CRC risk. The age- and smoking-adjusted OR for ADH2 A/A relative to G/A and G/G was 1.60 (95% CI=1.08-2.36), and the adjusted OR for ALDH2 G/G relative to G/A and A/A was 1.79 (95% CI=1.19-2.69). Signif icant interactions between ADH2, ALDH2 and drinking were observed. As compared to the subjects with ADH2 G and ALDH2 A alleles, those with ADH2 A/A and ALDH2 G/G genotypes had a signif icantly increased OR (3.05, 95% CI= 1.67-5.57). The OR for CRC among drinkers with the ADH2 A/A genotype was increased to 3.44 (95% CI= 1.84-6.42) compared with non-drinkers with the ADH2 G allele. The OR for CRC among drinkers with the ALDH2 G/G genotype was also increased to 2.70 (95% CI= 1.57-4.66) compared with non-drinkers with the ALDH2 A allele. CONCLUSION: Polymorphisms of the ADH2 and ALDH2 genes are significantly associated with CRC risk. There are also signifi cant gene-gene and gene- environment interactions between drinking and ADH2 and ALDH2 polymorphisms regarding CRC risk in Chinese males. 展开更多

The Hekoulinchang Sn-Pb-Zn-Ag polymetallic deposit(20000 t Sn at 0.27%,236 t Ag at 122.89 g/t,15000 t Pb at 0.84%,and 38000 t Zn at 1.43%)is located in the Wandashan Terrane of the easternmost segment of the Central Asian Orogenic Belt.The timing of Sn-Pb-Zn-Ag polymetallic mineralization remains unclear due to a lack of precise isotope dating directly conducted on ore minerals.The authors herein report that the LA-ICP-MS U-Pb ages of cassiterite and zircon from the granite porphyry in the Hekoulinchang Sn-Pb-Zn-Ag polymetallic deposit are 101.4±7.9 Ma and 115.4±1.0 Ma,respectively,indicating that Sn mineralization and magmatism occurred during the Early Cretaceous.The granite porphyry belongs to the subalkaline series peraluminous I-type granites that are depleted in Nb,Ta,and Ti and enriched in Rb,Th,U,and Pb.TheεHf(t)values of the granite porphyry range from 0.9 to 7.4,with an average of about 5.6 and two-stage model ages(T_(DM2))of 705–1116 Ma,with an average age of 819 Ma.The ε_(Nd)(t)values of the apatites are–1.60–0.45,with an average of–0.9,and two-stage model ages(T_(DM2))of 872–1040 Ma,with an average age of 983 Ma.The Nd-Hf isotope data indicate that the magma may have been derived from the partial melting of juvenile crustal material. 展开更多

Based on the current indoor natural ventilation and lighting in the space of traditional residential buildings,this paper starts from the passive-design optimization of the spatial natural ventilation and lighting,and makes quantitative evaluation on the quality of current interior natural ventilation and lighting for two typical residential buildings by three indexes,including wind speed,static wind area ratio and satisfaction ratio about minimum lighting coefficient. Based on that, this paper conducts the passive design optimization, and establishes the quantitative association and reevaluation among the passive reformation design, natural ventilation,and lighting environmental quality,proposing the general strategy for the existing residential buildings to respond to the passive reformation design of the natural ventilation and lighting. The special reconstruction of core functionary space of integration of "the living room + dining room + partial space"is researched,and the redesign for the optimization and replacement of both indoor and outdoor enclosure parts is explored,which is expected to provide practical exploration on the strategies for passive construction of spatial natural environmental quality within a large number of highly-energy-consumed residential buildings in China,as well as the green design of residential buildings. 展开更多

Piloti is commonly used in tropical and subtropical climate zones to get high wind velocity and create shadowed areas in order to optimize the living environment of residential blocks,but there are few studies to reveal the influence of piloti on the radiant environment of residential blocks systematically. Taking the city of Guangzhou as an example,using 3-D Unsteady State Heat Balance Radiation Calculation Method,this paper shows that the mean radiant temperature( MRT) under piloti area increases with the increase of piloti ratio,and especially when piloti ratio is equal to 100%,the MRT increase trend becomes sharp. The MRT of exposed area decreases with the increase of piloti ratio,especially when piloti ratio reaches 100%,the decrease trend of MRT becomes sharp,which offers the reference for the study on piloti design in subtropical climate zones and further research on living environment by CFD simulation in residential blocks. 展开更多

We cloned the complete coding sequences of porcine Gpr3,Gpr6,and Gpr12 genes.Further,on the basis of their high levels of sequence similarity,these genes are identified as a subfamily of G protein-coupled receptors.These putative protein sequences also showed high sequence identity with other mammalian orthologs,including several highly conserved motifs.A wide expression of the Gpr3 gene in pigs was observed through tissue distribution analysis by reverse transcriptase-polymerase chain reaction(RT-PCR)and real-time PCR,specially in the brain,pituitary,fat,liver and oocyte,where its strong expression was observed.The Gpr3 gene was found to be located on chromosome 6 and a single exon coded for the entire open-reading frame.Expression of porcine Gpr3 in HEK293 cells resulted in constitutive activation of adenylate cyclase(AC)similar in amplitude to that produced by fully stimulated Gs-coupled receptors.Moreover,sphingosine 1-phosphate(S1P)could increase AC activation via the constitutively active Gpr3 receptor.When a Gpr3-green fluorescent protein(GFP)construct was expressed in HEK293 cells,GFP-labeled Gpr3 protein was shown to be localized in the plasmalemma and subcellular membranes.After S1P treatment,agonist-mediated internalization could be visualized by confocal microscopy.In short,our findings suggest the porcine Gpr3,Gpr6,and Gpr12 genes as a subfamily of G protein-coupled receptors,and porcine Gpr3 was a constitutively active G protein-coupled receptor.Constitutive activation of AC and agonist-mediated internalization of Gpr3 receptor could be modulated by the S1P,suggesting that S1P might act as an activator for porcine Gpr3 receptor. 展开更多
