Objective:To explore the effectiveness and safety of cardiac rehabilitation qigong exercise in stable coronary artery disease(CAD)patients undergoing phaseⅢrehabilitation.Methods:This was a randomized controlled tria...Objective:To explore the effectiveness and safety of cardiac rehabilitation qigong exercise in stable coronary artery disease(CAD)patients undergoing phaseⅢrehabilitation.Methods:This was a randomized controlled trial.A total of 59 stable CAD patients undergoing phaseⅢrehabilitation treated in Sports Medicine Hospital from March 2017 to September 2017 were enrolled after meeting the inclusion criteria and then divided into cardiac rehabilitation qigong exercise group(n=30)receiving a 12-week intervention and control group(n=29).All participants were assessed at baseline and at 12-week intervention for the primary outcome,that was treadmill test parameter and the secondary outcomes including physical fitness,body composition,bone mineral density,and cardiac ultrasound B-mode imaging.Results:There were no significant differences in baseline demographics between the two groups.After a 12-week cardiac rehabilitation qigong exercise intervention,compared with the control group,ΔV02(initial 1352.63±340.95 vs 12 weeks 1594.57±467.14)vs(initial 1363.83±322.90 vs 12 weeks 1323.76±318.92)(P=.003),ΔVO2/kg(initial 21.23±3.56 vs 12 weeks 24.75±5.11)vs(initial 21.01±3.71 vs 12 weeks 20.35±3.66)(P=.002),ΔMETS(initial 6.19±1.12 vs 12 weeks 7.16±1.60)vs(initial 6.00±1.19 vs 12 weeks 5.86±1.23)(P=.001),ΔVO2/HR(P=.027),ΔSV(P=.014),ΔOUES(P=.012),Δhand-grip strength(P=.002),Δflexibility(P=.001),Δbalance(P=.002),ΔT-score(P=.042),ΔBQI(P=.018).However,Δresting systolic blood pressure(P=.004)andΔresting diastolic pressure(P=.012)decreased in the cardiac rehabilitation qigong exercise group.Conclusion:Cardiac rehabilitation qigong exercise can improve cardiopulmonary aerobic capacity,physical fitness,bone mineral density in patients with stable CAD,suggesting that certain effect and safety for stable CAD patients undergoing phaseⅢrehabilitation can be obtained.展开更多

It can satisfy several desirable characteristics for Io T systems. To reduce communication pressure from Io T devices, data aggregation is a good candidate. However, data processing in MEC may suffer from many challenges, such as unverifiability of aggregated data, privacy-violation and fault-tolerance. To address these challenges, we propose PVF-DA: privacy-preserving, verifiable and fault-tolerant data aggregation in MEC based on aggregator-oblivious encryption and zero-knowledge-proof. The proposed scheme can not only provide privacy protection of the reported data, but also resist the collusion between MEC server and corrupted Io T devices. Furthermore, the proposed scheme has two outstanding features: verifiability and strong fault-tolerance. Verifiability can make Io T device to verify whether the reported sensing data is correctly aggregated. Strong fault-tolerance makes the aggregator to compute an aggregate even if one or several Io Ts fail to report their data. Finally, the detailed security proofs are shown that the proposed scheme can achieve security and privacy-preservation properties in MEC.展开更多

Interstitial pores are defined to describe fluid flow and particle movement. The proposed model couples multiphase fluid flow with particle movement to simulate the thermodynamics of gas hydrate dissociation triggered by sharp temperature rises. Hydrates respond quickly to temperature rise in low-permeability sediments. Dissociation causes pore pressure to rise rapidly to equilibrium then steadily increase above equilibrium pressure. Lower permeability sediment builds up greater excess pore pressure as the dissipation of pore pressure is constrained.展开更多

The results have demonstrated that the different concentration of 5-Aza has the influence on proliferation rate of different generations of BMSCs, the second generation BMSCs was superior to the sixth, and tenth generation in proliferation capacity after being induced, and 5-Aza had some influence on proliferation capacity. Rat BMSCs could be differentiated into cardiomyocytes-like cells, which have a good biocompatibility with acellular bovine pericardium, and myocardium-like tissue could be engineered with BMSCs and acellular bovine pericardium in vitro. In conclusion, BMSCs could be induced and differentiated into cardiomyocytes-like cells in vitro. Different generations and different 5-Aza density have influence on the rate of increase of BMSCs, and the engineered myocardium-like tissue could be constructed with BMSCs and acellular bovine pericardium in vitro.展开更多

The metabolite transport inhibition of tumor cells holds promise to achieve anti-tumor efficacy.Herein,we presented an innovative strategy to hinder the delivery of metabolites through the in-situ besieging tumor cell...The metabolite transport inhibition of tumor cells holds promise to achieve anti-tumor efficacy.Herein,we presented an innovative strategy to hinder the delivery of metabolites through the in-situ besieging tumor cells with polyphenolic polymers that strongly adhere to the cytomembrane of tumor cells.Simultaneously,these polymers underwent self-crosslinking under the induction of tumor oxidative stress microenvironment to form an adhesive coating on the surface of the tumor cells.This polyphenol coating effectively obstructed glucose uptake,reducing metabolic products such as lactic acid,glutathione,and adenosine triphosphate,while also causing reactive oxygen species to accumulate in the tumor cells.The investigation of various tumor models,including 2D cells,3D multicellular tumor spheroids,and xenograft tumors,demonstrated that the polyphenolic polymers effectively inhibited the growth of tumor cells by blocking key metabolite transport processes.Moreover,this highly adhesive coating could bind tumor cells to suppress their metastasis and invasion.This work identified polyphenolic polymers as a promising anticancer candidate with a mechanism by impeding the mass transport of tumor cells.展开更多

Natural killer(NK)cells are found in lymphoid and non-lymphoid organs.In addition to important roles in immune surveillance,some NK cells contribute to angiogenesis and circulatory regulation.The uterus of early pregn...Natural killer(NK)cells are found in lymphoid and non-lymphoid organs.In addition to important roles in immune surveillance,some NK cells contribute to angiogenesis and circulatory regulation.The uterus of early pregnancy is a non-lymphoid organ enriched in NK cells that are specifically recruited to placental attachment sites.In species with invasive hemochorial placentation,these uterine natural killer(uNK)cells,via secretion of cytokines,chemokines,mucins,enzymes and angiogenic growth factors,contribute to the physiological change of mesometrial endometrium into the unique stromal environment called decidua basalis.In humans,uNK cells have the phenotype CD56^(bright) CD16^(dim) and they appear in great abundance in the late secretory phase of the menstrual cycle and early pregnancy.Gene expression studies indicate that CD56^(bright) CD16^(dim) uterine and circulating cells are functionally distinct.In humans but not mice or other species with post-implantation decidualization,uNK cells may contribute to blastocyst implantation and are of interest as therapeutic targets in female infertility.Histological and genetic studies in mice first identified triggering of the process of gestation spiral arterial modification as a major uNK cell function,achieved via interferon(IFN)-c secretion.During spiral arterial modification,branches from the uterine artery that traverse the endometrium/decidua transiently lose their muscular coat and ability to vasoconstrict.The expression of vascular markers changes from arterial to venous as these vessels dilate and become low-resistance,high-volume channels.Full understanding of the vascular interactions of human uNK cells is difficult to obtain because endometrial time-course studies are not possible in pregnant women.Here we briefly review key information concerning uNK cell functions from studies in rodents,summarize highlights concerning human uNK cells and describe our preliminary studies on development of a humanized,pregnant mouse model for in vivo investigations of human uNK cell functions.展开更多

The first step in the study of tissue lymphoid cells is their isolation in intact and viable form appropriate for establishment of in vitro culture systems. For reasons of simplicity, cell purity, cell yields and various purposes, lymphocytes obtained from different tissues in different labs were subjected to diverse protocols. To fully elucidate the nature of the local immune system as well as to adequately study the innate role of lymphocytes in liver, intestine, lung and uterus, we briefly reviewed the characterization of resident lymphocytes, and additional information on those cells from non-lymphoid tissues by using the recommended operation procedure was also presented.展开更多

Within the pregnant decidualizing uterus, co...Successful pregnancy and long-term, post-natal maternal and offspring cardiac, vascular and metabolic health require key maternal cardiovascular adaptations over gestation. Within the pregnant decidualizing uterus, coordinated vascular, immunological and stromal cell changes occur. Considerable attention has been given to the roles of uterine natural killer (uNK) cells in initiating decidual spiral arterial remodeling, a process normally completed by mid-gestation in mice and in humans. However, leukocyte roles in much earlier, region specific, decidual vascular remodeling are now being defined. Interest in immune cell-promoted vascular remodeling is driven by vascular aberrations that are reported in human gestational complications such as infertility, recurrent spontaneous abortion, preeclampsia (PE) and fetal growth restriction. Appropriate maternal cardiovascular responses during pregnancy protect mothers and their children from later cardiovascular disease risk elevation. One of the earliest uterine responses to pregnancy in species with hemochorial placentation is stromal cell decidualization, which creates unique niches for angiogenesis and leukocyte recruitment. In early decidua basalis, the aspect of the implantation site that will cradle the developing placenta and provide the major blood vessels to support mature placental functions, leukocytes are greatly enriched and display specialized properties. UNK cells, the most abundant leukocyte subset in early decidua basalis, have angiogenic abilities and are essential for normal early decidual angiogenesis. The regulation of uNK cells and their roles in determining maternal and progeny cardiovascular health over ore^nancv and PostPartum are discussed.展开更多

High-confidence function...Wolfiporia cocos(F. A. Wolf) has been praised as a food delicacy and medicine for centuries in China. Here, we present the genome and transcriptome of the Chinese strain CGMCC5.78 of W. cocos. High-confidence functional prediction was made for 9277 genes among the 10,908 total predicted gene models in the W. cocos genome. Up to 2838 differentially expressed genes(DEGs)were identified to be related to sclerotial development by comparing the transcriptomes of mycelial and sclerotial tissues. These DEGs are involved in mating processes, differentiation of fruiting body tissues, and metabolic pathways. A number of genes encoding enzymes and regulatory factors related to polysaccharide and triterpenoid production were strikingly regulated. A potential triterpenoid gene cluster including the signature lanosterol synthase(LSS) gene and its modified components were annotated. In addition, five nonribosomal peptide synthase(NRPS)-like gene clusters,eight polyketide synthase(PKS) gene clusters, and 15 terpene gene clusters were discovered in the genome. The differential expression of the velevt family proteins, transcription factors,carbohydrate-active enzymes, and signaling components indicated their essential roles in the regulation of fungal development and secondary metabolism in W. cocos. These genomic and transcriptomic resources will be valuable for further investigations of the molecular mechanisms controlling sclerotial formation and for its improved medicinal applications.展开更多

We investigated the functional character of human dNK cells during ...Decidual natural killer (dNK) cells express an array of activation receptors to regulate placental immunity and development during early pregnancy. We investigated the functional character of human dNK cells during the first and second trimester of gestation and the interaction between dNK and trophoblast cells. Although the frequency of CD56+CD16-dNK among the total CD45+ leukocytes did not change over this period, the expression of the activating receptors, NKp80 and NKG2D, was greatly upregulated. We observed a significantly higher number of extravillous trophoblast cells in proximity to the dNK cells in the first trimester in comparison with the second trimester decidua. NKG2D expression by first trimester dNK cells was decreased when co-cultured with the HTR-8 trophoblast cell line. In the second trimester, functional markers of dNK activation, i.e., angiogenic factor production (e.g., vascular endothelial growth factor, interleukin-8, interferon-gamma), remained stable despite an increase in NKp80 or NKG2D surface expression. Furthermore, the degranulation capacity of dNK cells, as assessed by CD107a, was decreased in the second trimester. We suggest that in the first trimester, trophoblast-dNK interactions generate a population of dNK cells with a suppressed activating phenotype. In the second trimester, the loss of trophoblast-dNK interactions led to the inhibition of dNK cell function, although their activating receptor expression was increased. We speculate that during pregnancy, two mechanisms operate to modulate the dNK cell activation:suppression of activating receptor levels in the first trimester by trophoblasts and disengagement of receptor-ligand coupling in the second trimester.展开更多
A constrained back propagation neural network(C-BPNN)model for standard penetration test based soil liquefaction assessment with global applicability is developed,incorporating existing knowledge for liquefaction trig...A constrained back propagation neural network(C-BPNN)model for standard penetration test based soil liquefaction assessment with global applicability is developed,incorporating existing knowledge for liquefaction triggering mechanism and empirical relationships.For its development and validation,a comprehensive liquefaction data set is compiled,covering more than 600 liquefaction sites from 36 earthquakes in 10 countries over 50 years with 13 complete information entries.The C-BPNN model design procedure for liquefaction assessment is established by considering appropriate constraints,input data selection,and computation and calibration procedures.Existing empirical relationships for overburden correction and fines content adjustment are shown to be able to improve the prediction success rate of the neural network model,and are thus adopted as constraints for the C-BPNN model.The effectiveness of the C-BPNN method is validated using the liquefaction data set and compared with that of several liquefaction assessment methods currently adopted in engineering practice.The C-BPNN liquefaction model is shown to have improved prediction accuracy and high global adaptability.展开更多

The results have demonstrated that the different concentration of 5-Aza has the influence on proliferation rate of different generations of BMSCs, the second generation BMSCs was superior to the sixth, and tenth generation in proliferation capacity after being induced, and 5-Aza had some influence on proliferation capacity. Rat BMSCs could be differentiated into cardiomyocytes-like cells, which have a good biocompatibility with acellular bovine pericardium, and myocardium-like tissue could be engineered with BMSCs and acellular bovine pericardium in vitro. In conclusion, BMSCs could be induced and differentiated into cardiomyocytes-like cells in vitro. Different generations and different 5-Aza density have influence on the rate of increase of BMSCs, and the engineered myocardium-like tissue could be constructed with BMSCs and acellular bovine pericardium in vitro.

文摘Medicinal plants are a valuable source of essential medicines and herbal products for healthcare and disease therapy.Compared with chemical synthesis and extraction,the biosynthesis of natural products is a very promising alternative for the successful conservation of medicinal plants,and its rapid development will greatly facilitate the conservation and sustainable utilization of medicinal plants.Here,we summarize the advances in strategies and methods concerning the biosynthesis and production of natural products of medicinal plants.The strategies and methods mainly include genetic engineering,plant cell culture engineering,metabolic engineering,and synthetic biology based on multiple“OMICS”technologies,with paradigms for the biosynthesis of terpenoids and alkaloids.We also highlight the biosynthetic approaches and discuss progress in the production of some valuable natural products,exemplifying compounds such as vindoline(alkaloid),artemisinin and paclitaxel(terpenoids),to illustrate the power of biotechnology in medicinal plants.

文摘The metabolite transport inhibition of tumor cells holds promise to achieve anti-tumor efficacy.Herein,we presented an innovative strategy to hinder the delivery of metabolites through the in-situ besieging tumor cells with polyphenolic polymers that strongly adhere to the cytomembrane of tumor cells.Simultaneously,these polymers underwent self-crosslinking under the induction of tumor oxidative stress microenvironment to form an adhesive coating on the surface of the tumor cells.This polyphenol coating effectively obstructed glucose uptake,reducing metabolic products such as lactic acid,glutathione,and adenosine triphosphate,while also causing reactive oxygen species to accumulate in the tumor cells.The investigation of various tumor models,including 2D cells,3D multicellular tumor spheroids,and xenograft tumors,demonstrated that the polyphenolic polymers effectively inhibited the growth of tumor cells by blocking key metabolite transport processes.Moreover,this highly adhesive coating could bind tumor cells to suppress their metastasis and invasion.This work identified polyphenolic polymers as a promising anticancer candidate with a mechanism by impeding the mass transport of tumor cells.

文摘As an invisible“endocrine organ”,gut microbiota is widely involved in the regulation of nervous system,endocrine system,circulatory system,and digestive system.It is also closely related to host health and the occurrence of many chronic diseases.Relevant literature shows that high temperature,low temperature,and highaltitude hypoxia may have negative effects on commensal microorganisms.The stimulation of exercise may aggravate this reaction,which is related to the occurrence of exercise-induced fever and gastrointestinal and respiratory diseases.The intervention of probiotics can alleviate the above problems to a certain extent.Therefore,this paper takes exercise in a special environment as the starting point,deeply analyses the intervention effect and potential mechanism of probiotics,and provides the theoretical basis and reference for follow-up research and application of probiotics in sports science.

文摘Natural killer(NK)cells are found in lymphoid and non-lymphoid organs.In addition to important roles in immune surveillance,some NK cells contribute to angiogenesis and circulatory regulation.The uterus of early pregnancy is a non-lymphoid organ enriched in NK cells that are specifically recruited to placental attachment sites.In species with invasive hemochorial placentation,these uterine natural killer(uNK)cells,via secretion of cytokines,chemokines,mucins,enzymes and angiogenic growth factors,contribute to the physiological change of mesometrial endometrium into the unique stromal environment called decidua basalis.In humans,uNK cells have the phenotype CD56^(bright) CD16^(dim) and they appear in great abundance in the late secretory phase of the menstrual cycle and early pregnancy.Gene expression studies indicate that CD56^(bright) CD16^(dim) uterine and circulating cells are functionally distinct.In humans but not mice or other species with post-implantation decidualization,uNK cells may contribute to blastocyst implantation and are of interest as therapeutic targets in female infertility.Histological and genetic studies in mice first identified triggering of the process of gestation spiral arterial modification as a major uNK cell function,achieved via interferon(IFN)-c secretion.During spiral arterial modification,branches from the uterine artery that traverse the endometrium/decidua transiently lose their muscular coat and ability to vasoconstrict.The expression of vascular markers changes from arterial to venous as these vessels dilate and become low-resistance,high-volume channels.Full understanding of the vascular interactions of human uNK cells is difficult to obtain because endometrial time-course studies are not possible in pregnant women.Here we briefly review key information concerning uNK cell functions from studies in rodents,summarize highlights concerning human uNK cells and describe our preliminary studies on development of a humanized,pregnant mouse model for in vivo investigations of human uNK cell functions.
文摘In steady-state conditions, the number and distribution of lymphocyte populations are under homeostatic control. New lymphocytes are continuously produced in primary and secondary lymphoid organs and then achieve immune-competence within different tissues, and they must challenge with resident cells for survival. The first step in the study of tissue lymphoid cells is their isolation in intact and viable form appropriate for establishment of in vitro culture systems. For reasons of simplicity, cell purity, cell yields and various purposes, lymphocytes obtained from different tissues in different labs were subjected to diverse protocols. To fully elucidate the nature of the local immune system as well as to adequately study the innate role of lymphocytes in liver, intestine, lung and uterus, we briefly reviewed the characterization of resident lymphocytes, and additional information on those cells from non-lymphoid tissues by using the recommended operation procedure was also presented.
文摘Successful pregnancy and long-term, post-natal maternal and offspring cardiac, vascular and metabolic health require key maternal cardiovascular adaptations over gestation. Within the pregnant decidualizing uterus, coordinated vascular, immunological and stromal cell changes occur. Considerable attention has been given to the roles of uterine natural killer (uNK) cells in initiating decidual spiral arterial remodeling, a process normally completed by mid-gestation in mice and in humans. However, leukocyte roles in much earlier, region specific, decidual vascular remodeling are now being defined. Interest in immune cell-promoted vascular remodeling is driven by vascular aberrations that are reported in human gestational complications such as infertility, recurrent spontaneous abortion, preeclampsia (PE) and fetal growth restriction. Appropriate maternal cardiovascular responses during pregnancy protect mothers and their children from later cardiovascular disease risk elevation. One of the earliest uterine responses to pregnancy in species with hemochorial placentation is stromal cell decidualization, which creates unique niches for angiogenesis and leukocyte recruitment. In early decidua basalis, the aspect of the implantation site that will cradle the developing placenta and provide the major blood vessels to support mature placental functions, leukocytes are greatly enriched and display specialized properties. UNK cells, the most abundant leukocyte subset in early decidua basalis, have angiogenic abilities and are essential for normal early decidual angiogenesis. The regulation of uNK cells and their roles in determining maternal and progeny cardiovascular health over ore^nancv and PostPartum are discussed.

文摘Wolfiporia cocos(F. A. Wolf) has been praised as a food delicacy and medicine for centuries in China. Here, we present the genome and transcriptome of the Chinese strain CGMCC5.78 of W. cocos. High-confidence functional prediction was made for 9277 genes among the 10,908 total predicted gene models in the W. cocos genome. Up to 2838 differentially expressed genes(DEGs)were identified to be related to sclerotial development by comparing the transcriptomes of mycelial and sclerotial tissues. These DEGs are involved in mating processes, differentiation of fruiting body tissues, and metabolic pathways. A number of genes encoding enzymes and regulatory factors related to polysaccharide and triterpenoid production were strikingly regulated. A potential triterpenoid gene cluster including the signature lanosterol synthase(LSS) gene and its modified components were annotated. In addition, five nonribosomal peptide synthase(NRPS)-like gene clusters,eight polyketide synthase(PKS) gene clusters, and 15 terpene gene clusters were discovered in the genome. The differential expression of the velevt family proteins, transcription factors,carbohydrate-active enzymes, and signaling components indicated their essential roles in the regulation of fungal development and secondary metabolism in W. cocos. These genomic and transcriptomic resources will be valuable for further investigations of the molecular mechanisms controlling sclerotial formation and for its improved medicinal applications.
文摘Decidual natural killer (dNK) cells express an array of activation receptors to regulate placental immunity and development during early pregnancy. We investigated the functional character of human dNK cells during the first and second trimester of gestation and the interaction between dNK and trophoblast cells. Although the frequency of CD56+CD16-dNK among the total CD45+ leukocytes did not change over this period, the expression of the activating receptors, NKp80 and NKG2D, was greatly upregulated. We observed a significantly higher number of extravillous trophoblast cells in proximity to the dNK cells in the first trimester in comparison with the second trimester decidua. NKG2D expression by first trimester dNK cells was decreased when co-cultured with the HTR-8 trophoblast cell line. In the second trimester, functional markers of dNK activation, i.e., angiogenic factor production (e.g., vascular endothelial growth factor, interleukin-8, interferon-gamma), remained stable despite an increase in NKp80 or NKG2D surface expression. Furthermore, the degranulation capacity of dNK cells, as assessed by CD107a, was decreased in the second trimester. We suggest that in the first trimester, trophoblast-dNK interactions generate a population of dNK cells with a suppressed activating phenotype. In the second trimester, the loss of trophoblast-dNK interactions led to the inhibition of dNK cell function, although their activating receptor expression was increased. We speculate that during pregnancy, two mechanisms operate to modulate the dNK cell activation:suppression of activating receptor levels in the first trimester by trophoblasts and disengagement of receptor-ligand coupling in the second trimester.

文摘A constrained back propagation neural network(C-BPNN)model for standard penetration test based soil liquefaction assessment with global applicability is developed,incorporating existing knowledge for liquefaction triggering mechanism and empirical relationships.For its development and validation,a comprehensive liquefaction data set is compiled,covering more than 600 liquefaction sites from 36 earthquakes in 10 countries over 50 years with 13 complete information entries.The C-BPNN model design procedure for liquefaction assessment is established by considering appropriate constraints,input data selection,and computation and calibration procedures.Existing empirical relationships for overburden correction and fines content adjustment are shown to be able to improve the prediction success rate of the neural network model,and are thus adopted as constraints for the C-BPNN model.The effectiveness of the C-BPNN method is validated using the liquefaction data set and compared with that of several liquefaction assessment methods currently adopted in engineering practice.The C-BPNN liquefaction model is shown to have improved prediction accuracy and high global adaptability.
