金莲花多糖提取工艺优化及其抗氧化活性评价

目的优化金莲花多糖提取工艺,并分析测试金莲花多糖抗氧化活性。方法采用回流提取法提取金莲花多糖,通过单因素考察料液比、提取温度、提取时间3个因素对金莲花多糖提取率的影响,并在此基础上通过正交试验设计对提取工艺进行优化。以DPP... 目的优化金莲花多糖提取工艺,并分析测试金莲花多糖抗氧化活性。方法采用回流提取法提取金莲花多糖,通过单因素考察料液比、提取温度、提取时间3个因素对金莲花多糖提取率的影响,并在此基础上通过正交试验设计对提取工艺进行优化。以DPPH自由基、ABTS^(+)自由基、羟基自由基、超氧阴离子清除能力和总还原力为指标评价金莲花多糖的抗氧化活性。结果金莲花多糖的最佳提取工艺为料液比1∶25、提取温度80℃、提取时间2 h,在此条件下金莲花多糖的提取率为1.350%±0.125%。体外抗氧化实验结果表明,金莲花多糖其DPPH自由基、ABTS^(+)自由基、羟基自由基和超氧阴离子自由基清除率IC_(50)分别为0.161、0.079、0.006、4.216 mg·mL^(-1),且当浓度为4.0 mg·mL^(-1)时,总还原力为2.78。结论优化的金莲花多糖提取工艺提取率较高且获得的金莲花多糖具有较好的抗氧化活性,对金莲花多糖的进一步开发利用提供了参考依据。 展开更多

Objective: To determine the effect of flurbiprofen combined with prednisolone on interleukin-6 in elderly surgery patients. Methods: In this double-blind randomized controlled study, patients aged 65 to 80 who were undergoing spinal fusion surgery for disc herniation were administered flurbiprofen 100 mg (P group, flurbiprofen group), prednisolone 0.6 mg/kg (D group, prednisolone group), prednisolone 0.6 mg/kg plus flurbiprofen 100 mg (P + D group, flurbiprofen + prednisolone group) or normal saline (S group, saline group) 15 minutes before the induction of anesthesia. Plasma samples were collected before surgery (T0) and on day 1 (T1), day 2 (T2) and day 3 (T3) following surgery. At the same time, systemic inflammatory response syndrome (SIRS) was assessed by SIRS criteria. The levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and C-reactive protein (CRP) for collected samples were measured. Results: Other groups had significantly lower levels of IL-6, CRP and occurrence of SIRS than S group (p < 0.05). Compared to groups P and D, the levels of IL-6 and CRP in P + D group were significantly lower on T1 (p < 0.05). Peak levels of IL-6 in all groups were presented on T1 (p 0.05). The levels of CRP within three days were significantly different but did not show peak levels (p > 0.05). Conclusion: Compared to prednisolone or flurbiprofen, combining flurbiprofen with prednisolone in elderly surgery patients led to an increased suppression of IL-6. 展开更多

西葫芦杂聚果胶的制备及其降血糖活性初探

为了促进西葫芦的深度开发,本研究对西葫芦杂聚果胶进行了分析和降血糖活性初探。采用酸碱联合处理提取分离西葫芦果胶,分别得到酸提果胶(ZPA)和残渣碱提果胶(ZPB)两个组分,分析了两个组分的得率、组成和结构性质。构建了熊蜂高脂饮食... 为了促进西葫芦的深度开发,本研究对西葫芦杂聚果胶进行了分析和降血糖活性初探。采用酸碱联合处理提取分离西葫芦果胶,分别得到酸提果胶(ZPA)和残渣碱提果胶(ZPB)两个组分,分析了两个组分的得率、组成和结构性质。构建了熊蜂高脂饮食导致的糖尿病模型,初步探究西葫芦杂聚果胶ZPA和ZPB的降血糖活性。结果表明,ZPB的得率是ZPA的2.45倍,ZPA的平均分子量为70297Da,多分散系数为1.149,ZPB的平均分子量为126170Da,多分散系数为1.677,表明ZPB的分子分布范围较大,组成更为复杂。ZPA和ZPB的单糖组成均含有阿拉伯糖、半乳糖、半乳糖醛酸、葡萄糖醛酸、鼠李糖、甘露糖6种单糖,ZPB还含有葡萄糖、核糖2种单糖,可推测ZPA和ZPB均属于含有RG-I结构的杂聚果胶。经计算ZPA的RG-I结构较少但侧链长度较长,ZPB的RG-I结构多但侧链长度较短。利用熊蜂糖尿病模型发现ZPA和ZPB均能显著降低熊蜂血液葡萄糖和海藻糖水平,初步表明西葫芦杂聚果胶具有一定的降血糖活性。 展开更多

由滑坡等失稳地质体快速堆积并堵塞河道形成的堰塞坝溃决后会对下游人民生命财产安全造成极大威胁。建立快速准确的溃决参数预测模型,是制定有效防灾减灾措施的必要条件。本文收集了75例包含坝高、库容、溃口峰值流量、坝体冲蚀系数等... 由滑坡等失稳地质体快速堆积并堵塞河道形成的堰塞坝溃决后会对下游人民生命财产安全造成极大威胁。建立快速准确的溃决参数预测模型,是制定有效防灾减灾措施的必要条件。本文收集了75例包含坝高、库容、溃口峰值流量、坝体冲蚀系数等参数的堰塞坝案例,基于均方根误差、回归相关系数、百分比偏差三个指标对具有代表性的土石坝和堰塞坝参数模型进行评估。通过回归分析建立新的堰塞坝溃口深度预测模型及溃口峰值流量预测模型,将新建模型与现有模型对比,在实际案例中验证了模型的适用性。研究结果表明:现有模型中单参数、双参数及土石坝模型对于堰塞坝溃决参数预测效果不佳,考虑坝体易蚀性的参数模型具有较高准确性,本文所建新模型预测效果有所提升,使用范围更广,预测结果可为堰塞坝的防灾减灾措施提供参考。 展开更多

中西医结合方案对慢性阻塞性肺疾病D组稳定期的临床疗效研究 被引量:13

目的评价中西医结合方案治疗慢性阻塞性肺疾病(COPD)D组稳定期患者的临床疗效。方法选择2016年12月至2018年12月在成都中医药大学附属医院咳喘专病门诊或急诊科门诊就诊的180例阳虚血瘀、痰浊蕴肺型COPD D组稳定期患者作为研究对象。按... 目的评价中西医结合方案治疗慢性阻塞性肺疾病(COPD)D组稳定期患者的临床疗效。方法选择2016年12月至2018年12月在成都中医药大学附属医院咳喘专病门诊或急诊科门诊就诊的180例阳虚血瘀、痰浊蕴肺型COPD D组稳定期患者作为研究对象。按照随机数字表法以1:1比例分为常规西医治疗组和中西医结合治疗组,每组各90例。常规西医治疗组给予西医内科治疗;中西医结合治疗组在西医内科治疗基础上给予三拗汤合瓜萎薤白半夏汤合桔梗汤加味白附片、生晒参、丹参。两组均连续治疗6个月。比较两组患者治疗前后中医证候量化积分、肺功能指标[用力肺活量占预计值的百分比(FVC%)、第1秒用力呼气容积占预计值的百分比(FEV1%)]以及圣·乔治呼吸问卷(SCRQ)评分。结果①中医证候量化积分:治疗前,两组间各维度中医证候量化积分以及总积分比较差异均无统计学意义。治疗6个月后,中西医结合治疗组喘息、咳嗽、咳痰、畏寒肢冷、舌象症状积分以及中医证候量化总积分均较治疗前明显降低,且明显低于同期常规西医治疗组[喘息症状积分(分):5.56±2.12比6.93±1.54,咳嗽症状积分(分):6.00±1.43比6.84±1.33,咳痰症状积分(分):4.87±2.34比6.01±1.86,畏寒肢冷症状积分(分):5.53±1.59比8.25±1.38,舌象积分(分):4.58±1.78比7.23±1.58,中医证候量化总积分(分):41.69±3.56比50.66±4.45.均P<0.05]。②肺功能指标:治疗前、两组间FVC%和FEV1%比较差异均无统计学意义。治疗6个月后,两组FVC%均较治疗前明显降低,FEV1%均较治疗前明显升高,且中西医结合治疗组FEV1%明显高于常规西医治疗组(%:55.66±7.12比51.23±9.61.P<0.05)。③SCRQ评分:治疗前、两组间SCRQ评分比较差异无统计学意义。治疗6个月后,两组SCRQ评分均较治疗前明显降低,且中西医结合治疗组SCRQ评分明显低于常规西医治疗组(分:42.64±4.98比52.67±5.25.P<0.05)。结论该中西医结合治疗方案能够改善COPD D组稳定期患者的临床症状,肺功能和生活质量。 展开更多

Soil alkali-hydrolyzable nitrogen, which is sensitive to N fertilization rate, is one of the indicators of soil nitrogen supplying capacity. Two field experiments were conducted in Dongtai(120°19″ E, 32°52″ N), Jiangsu, China in 2009 and Dafeng(120°28″ E, 33°12″ N), Jiangsu province, China in 2010. Six nitrogen rates(0, 150, 300, 375, 450, and 600 kg ha^(-1)) were used to study the effect of N fertilization rate on soil alkali-hydrolyzable nitrogen content(SAHNC), subtending leaf nitrogen concentration(SLNC), yield, and fiber quality. In both Dongtai and Dafeng experiment station, the highest yield(1709 kg ha^(-1)), best quality(fiber length 30.6 mm, fiber strength 31.6 c N tex^(-1), micronaire 4.82), and highest N agronomic efficiency(2.03 kg kg^(-1)) were achieved at the nitrogen fertilization rate of 375 kg ha^(-1). The dynamics of SAHNC and SLNC could be simulated with a cubic and an exponential function,respectively. The changes in SAHNC were consistent with the changes in SLNC. Optimal average rate(0.276 mg day^(-1)) and duration(51.8 days) of SAHNC rapid decline were similar to the values obtained at the nitrogen rate of 375 kg ha^(-1)at which cotton showed highest fiber yield, quality, and N agronomic efficiency. Thus, the levels and strategies of nitrogen fertilization can affect SAHNC dynamics. The N fertilization rate that optimizes soil alkali-hydrolyzable nitrogen content would optimize the subtending leaf nitrogen concentration and thereby increase the yield and quality of the cotton fiber. 展开更多

Faced with hundreds of thousands of news articles in the news websites,it is difficult for users to find the news articles they are interested in.Therefore,various news recommender systems were built.In the news recommendation,these news articles read by a user is typically in the form of a time sequence.However,traditional news recommendation algorithms rarely consider the time sequence characteristic of user browsing behaviors.Therefore,the performance of traditional news recommendation algorithms is not good enough in predicting the next news article which a user will read.To solve this problem,this paper proposes a time-ordered collaborative filtering recommendation algorithm(TOCF),which takes the time sequence characteristic of user behaviors into account.Besides,a new method to compute the similarity among different users,named time-dependent similarity,is proposed.To demonstrate the efficiency of our solution,extensive experiments are conducted along with detailed performance analysis. 展开更多

BACKGROUND Acute urinary retention(AUR)is rare during pregnancy.CASE SUMMARY We report on three pregnant women with AUR between the 10^(th) and 18^(th) wk of gestation.Case 1 was first diagnosed as urinary tract infection and developed a urinary tract infection due to urinary retention caused by urethral obstruction.Case 2 had a history of previous abdominal surgery for pelvic tuberculosis,leading to severe adhesions and a persistent retroverted uterus.In case 3,healthcare providers focused on the patient’s gastrointestinal symptoms and did not investigate her inability to void.Case 1 required manual disimpaction of the uterus and the knee-chest position.The other cases required immediate catheterization.The condition resolved in cases 1 and 2;these patients had normal pregnancies.Case 3 had severe complications at the time of consultation,leading to an abortion.CONCLUSION Retroverted uterus is the most common cause of AUR.Prompt recognition and diagnosis are required.Clinicians should be aware of the risk factors,etiology,and clinical presentation of AUR in the first and second trimester of pregnancy. 展开更多

Rupture model of the 2013 M_W 6.6 Lushan (China) earthquake constrained by a new GPS data set and its effects on potential seismic hazard 被引量:1

Vertical records are critically important when determining the rupture model of an earthquake, especially a thrust earthquake. Due to the relatively low fitness level of near-field vertical displacements, the precision of previous rupture models is relatively low, and the seismic hazard evaluated thereafter should be further updated. In this study, we applied three-component displacement records from GPS stations in and around the source region of the 2013 MW6.6 Lushan earthquake to re-investigate the rupture model.To improve the resolution of the rupture model, records from both continuous and campaign GPS stations were gathered, and secular deformations of the GPS movements were removed from the records of the campaign stations to ensure their reliability. The rupture model was derived by the steepest descent method(SDM), which is based on a layered velocity structure. The peak slip value was about 0.75 m, with a seismic moment release of 9.89 × 10^(18) N·m, which was equivalent to an M_W6.6 event. The inferred fault geometry coincided well with the aftershock distribution of the Lushan earthquake. Unlike previous rupture models, a secondary slip asperity existed at a shallow depth and even touched the ground surface. Based on the distribution of the co-seismic ruptures of the Lushan and Wenchuan earthquakes, post-seismic relaxation of the Wenchuan earthquake, and tectonic loading process, we proposed that the seismic hazard is quite high and still needs special attention in the seismic gap between the two earthquakes. 展开更多

BACKGROUND Pancreatic ductal adenocarcinoma(PDAC)is frequently diagnosed and treated in advanced tumor stages with poor prognosis.More effective screening programs and novel therapeutic means are urgently needed.Recent studies have regarded tight junction protein claudin 18.2(CLDN18.2)as a candidate target for cancer treatment,and zolbetuximab(formerly known as IMAB362)has been developed against CLDN18.2.However,there are few data reported thus far related to the clinicopathological characteristics of CLDN18.2 expression for PDAC.AIM To investigate the expression of CLDN18.2 in PDAC patients and subsequently propose a new target for the treatment of PDAC.METHODS The Cancer Genome Atlas,Genotype-Tissue Expression,Gene Expression Omnibus,and European Genome-phenome Archive databases were first employed to analyze the CLDN18 gene expression in normal pancreatic tissue compared to that in pancreatic cancer tissue.Second,we analyzed the expression of CLDN18.2 in 93 primary PDACs,86 para-cancer tissues,and 13 normal pancreatic tissues by immunohistochemistry.Immunostained tissues were assessed applying the histoscore.subsequently,they fell into two groups according to the expression state of CLDN18.2.Furthermore,the correlations between CLDN18.2 expression and diverse clinicopathological characteristics,including survival,were investigated.RESULTS The gene expression of CLDN18 was statistically higher(P<0.01)in pancreatic tumors than in normal tissues.However,there was no significant correlation between CLDN18 expression and survival in pancreatic cancer patients.CLDN18.2 was expressed in 88(94.6%)of the reported PDACs.Among these tumors,50(56.8%)cases showed strong immunostaining.The para-cancer tissues were positive in 81(94.2%)cases,among which 32(39.5%)of cases were characterized for strong staining intensities.Normal pancreatic tissue was identified solely via weak immunostaining.Finally,CLDN18.2 expression significantly correlated with lymph node metastasis,distant metastasis,nerve invasion,stage,and survival of PDAC patients,while there was no correlation between CLDN18.2 expression and localization,tumor size,patient age and sex,nor any other clinicopathological characteristic.CONCLUSION CLDN18.2 expression is frequently increased in PDAC patients.Thus,it may act as a potential therapeutic target for zolbetuximab in PDAC. 展开更多

Seven fractions were isolated from the ethyl acetate extract of Prinsepia utilis Royle by silica gel column chromatography. The antibacterial activities determined by disc diffusion method in vitro, indicated that the first and fourth fractions showed better antibacterial activity than the other fractions, while the sixth and seventh fractions did not showed any antibacterial activity. The diameters of the inhibition zone of first and fourth fractions were greater than 10 mm against three standard strains (Staphylococcus aureus ATCC25923, Escherichia coli ATCC44102 and Salmonella sp. CMCC(B)50041) at the concentration of 20 mg/ml. The first fraction was then repeatedly recrystallized in acetone to yield a white snowflake-like compound A, the inhibition zones of which were 14.03 mm, 11.54 mm and 12 mm, respectively. The Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were measured by broth dilution method at the concentrations ranging from 20 to 0.313 mg/mL. The MIC and MBC values of the first, fourth fractions and compound A were lower than that of oregano oil (positive control) against S. aureus ATCC25923. 展开更多

The complex heat of BF_3 –methanol was found to be 49.2 and 58.1 kJ/mol when the molar ratio of BF_3 to methanol was 1:2 and 1:1,respectively. In addition,the complex heat of BF_3 –anisole was also measured to test the apparatus error. The BF_3 –anisole result showed a calorimeter value of 53.1 kJ/mol with a system error of 2.3% as compared with the value reported in the literature. The mechanism of the reaction of BF_3 and methanol was interpreted based on our obtained results. This apparatus is useful and suitable for measuring the heat of other liquid–gas and liquid–liquid reactions. 展开更多
