The objective of this study was to determine the expression of the important vesicle trafficking- regulating factor Rab25 in human gastric cancer tissues, to analyze the correlation between Rab25 protein expression wi...The objective of this study was to determine the expression of the important vesicle trafficking- regulating factor Rab25 in human gastric cancer tissues, to analyze the correlation between Rab25 protein expression with gastric cancer occurrence and development, and to discuss the correlation of Rab25 protein expression with gastric cancer cell metastasis. The overall aim was to provide experimental evidence that can be used to design future biological treatments of human gastric cancer. Human gastric cancer tissue and the adjacent normal gastric tissue were surgically removed, and immunohistochemistry and Western blotting were used to detect Rab25 protein expression. The correlation between Rab25 protein expression with the development and pathological characteristics of gastric cancer was analyzed. Using RNAi, Rab25 expression was reduced in the gastric cancer cell line MGC80-3, and the changes in MGC80-3 cell invasiveness were then monitored. Immunohistochemistry showed that the Rab25 protein expression rates were 78.21% and 23.08% in gastric carcinoma and the adjacent normal gastric tissue, respectively. Immunohistochemistry and Western blot results showed that Rab25 protein expression in gastric cancer was significantly higher than in adjacent normal gastric tissues (t)〈0.01). Less differentiated gastric cancer cells had higher expression of Rab25 protein (P〈0.01). Gastric carcinomas from patients with a late pathological stage (Ⅲ-Ⅳ) had significantly higher Rab25 protein expression than early stage (Ⅰ-Ⅱ) patients (P〈0.01). Gastric carcinomas from patients with lymph node metastasis had significantly higher Rab25 protein expression than lymph node metastasis- free patients (P〈0.01). Gastric carcinomas from patients with distant metastases had significantly higher Rab25 protein expression than the distant metastasis-negative patients (P〈0.01). Rab25 protein expression in gastric cancer was not affected by the patients' sex, age, or tumor size (P〉0.05). MGCS0-3 cells transfected with Rab25 siRNA had significantly lower Rab25 protein expression (P〈0.01) and a significantly lower number of cells that passed through a Transwell chamber compared with non-transfected controls and the transfected control group (P〈0.01). Rab25 protein expression is associated with the development of gastric cancer, siRNA knockdown of Rab25 protein expression in MGC80-3 gastric cancer cells reduced MGC80-3 cell invasiveness and provided experimental evidence for potential future biological treatment strategies of human gastric cancer.展开更多
The Great Power Mentality possessed by the citizens is an important part of the country’s soft power and a necessary condition for China’s rise.Based on the operational measurement of the Great Power Mentality,this ...The Great Power Mentality possessed by the citizens is an important part of the country’s soft power and a necessary condition for China’s rise.Based on the operational measurement of the Great Power Mentality,this paper uses the data of the 2015"Public Social Awareness Survey"as a support to examine the current status of the Chinese people's Great Power Mentality and the characteristics of the Great Power Mentality in different groups.The main research conclusions are as follows:First,the Great Power Mentality can be specifically operated into three types:self-confidence,normal mentality,and responsibility mentality;second,overall,among the three Great Power Mentality of the public,the self-confidence has the highest score(3.93),the normal mentality is second(3.32),and the responsibility mentality is the lowest(2.67);third,whether it is self-confidence,normal mentality,or responsibility mentality,the ideological position has a significant impact on people’s Great Power Mentality.The"Left"people’s Great Power Mentality scores higher.展开更多

Here we report on reversible isomerization of solvatochromic donor-acceptor Stenhouse adducts(DASAs) in water through the confinement from dendritic oligoethylene glycols(OEGs). Dendronization of DASAs with dendritic OEGs affords them characteristic thermoresponsiveness. These dendronized DASAs spontaneously isomerize in water from hydrophobic linear state into hydrophilic cyclic state at room temperature due to the strong hydration. However, hydrophobic microenvironment through thermally dehydration and collapse of the dendritic OEGs at elevated temperatures confines hydration of the DASA moieties and mediates their interactions with the collapsed hydrophobic OEG domains, affording their isomerization recovery in water efficiently from the hydrophilic cyclic state into the hydrophobic linear state. The confinement-mediated reversible isomerization of DASA moieties in water can be repeated through alternative photo-irradiation and thermal dehydrations, exhibiting excellent fatigue resistance.展开更多
Recently,many efforts have been dedicated to construct artificial catalysts with enzyme-like activity.However,it is still a big challenge to endow artificial catalysts with specific substrate selectivity.In this study...Recently,many efforts have been dedicated to construct artificial catalysts with enzyme-like activity.However,it is still a big challenge to endow artificial catalysts with specific substrate selectivity.In this study,we developed a facile strategy to construct a MIL-53(Fe)-based nanocatalyst with designable selectivity in the degradation of oxytetracycline(OTC).Through the Fe–O–P conjunction,oxytetracycline aptamer(OA)can be easily anchored on MIL-53(Fe)to provide the specific site for OTC binding.We verified that the obtained MIL-53(Fe)-Apt nanocatalyst displayed enhanced catalytic ability in the degradation of OTC,whereas obvious suppression toward other substrate analogues.This performance therefore brings about an anticipated selectivity toward OTC.Moreover,we highlighted that the configuration of aptamers on MIL-53(Fe)can be modulated through varying conjunction mode.Structure–function analysis revealed that aptamer configuration affects the local concentration of substrate around catalytic site,which thus decides the catalytic performance toward OTC.This work presented a facile and promising strategy for developing artificial catalysts with designable selectivity.展开更多
We report the coordination assembly of the ferrocene-diphenylalanine(Fc-FF)with divalent copper ions(Cu^(2+))into metalpeptide assemblies(MPAs)with hierarchical spiral architectures.The MPA particles are composed of h...We report the coordination assembly of the ferrocene-diphenylalanine(Fc-FF)with divalent copper ions(Cu^(2+))into metalpeptide assemblies(MPAs)with hierarchical spiral architectures.The MPA particles are composed of helically organized nanofibers which can be correlated to the logarithmic spirals.The MPAs are hierarchically porous with abundant Fc and Cu2+active sites and show much higher catalytic activity than natural laccase toward the decolorization reaction.Moreover,a series of hierarchical structures of the MPAs can be synthesized by controlling the temperature and enantiomeric excess(ee).The peptide enantiomers with higher ee values will self-assemble into highly complex and ordered structures,which show higher surface area and porosity and thus enhanced catalytic activity compared with those assembled by peptides with lower ee values.The results provide new insights into the vital role of chirality in directing the self-assembly of biomolecules into highly ordered complex functional structures.展开更多
The present study investigated the effects of cow manure ratios mixed with maize stover,rice straw,and wheat stalk at 3,2,1(total solid based,TS-based),respectively,on methane production and microbial community struct...The present study investigated the effects of cow manure ratios mixed with maize stover,rice straw,and wheat stalk at 3,2,1(total solid based,TS-based),respectively,on methane production and microbial community structure during the anaerobic co-digestion process.Results showed cow manure co-digested with maize stover,wheat stalk,and rice straw at ratios of 2,1,and 3 had the highest cumulative methane yields(272.99,153.22167.73 mL/g volatile solid(VS),respectively)and better stability(e.g.pH,volatile fatty acids(VFAs)and their component).The main microbe evolution had a similar trend which was Petrimonas and Methanosaeta in the early digestion process(Days 0-7)and then evolved into Longilinea,Ruminofilibacter,and Methanosarcina with the progress of digestion,but the relative abundance of these microbes in each reactor was different.It was worth noting that Caldicoprobacter in cow manure to maize stover ratio of 2,and to rice straw ratio of three reactors had a relatively higher proportion than reactor of cow manure to wheat stalk ratio of 1,and Hydrogenophaga was the specific bacterium in cow manure to wheat stalk ratio of 1 reactor.In addition,Petrimonas showed positive relationship with VFAs and Longilinea was the opposite.Methanosaeta and Methanobacterium contributed the most during the peak period of methane production in cow manure and maize stover co-digested reactor,and showed positive relationship with acetic acid.However,Methanosarcina and Methanospirillum made a great contribution during the peak period of methane production in cow manure co-digested with wheat stalk and rice straw reactors.These findings could provide further information on the application of cow manure co-digested with crop wastes.展开更多

Alternat...Because glycidyl(Gly) contains an epoxy and an active hydroxyl group, the Gly unit is difficult to introduce into certain polymeric chains in a controlled manner and usually yields hyperbranched polyglycidyl. Alternatively, the monomer 1-ethoxyethyl glycidyl ether(EEGE), derived from Gly and ethyl vinyl ether, has shown potential for application in polymer chemistry, and homopolymerization of this monomer directly produces linear poly(1-ethoxyethyl glycidyl ether) and further yields linear polyglycidyl. In this review, the initiation system of the EEGE monomer is first discussed in terms of chain transfer to monomers in ring-opening polymerization of epoxides with substituent groups. Then, random copolymerization of EEGE with other epoxides is considered. In addition, because the EEGE units on polymers can be transferred to Gly units and further used to construct copolymers with complicated architectures, the applications of EEGE monomers to block, graft, and hyperbranched copolymers are reviewed. Finally, the synthesis of main chain and terminal functional polyethers by transforming the hydroxyl groups at the polymer end or on the main chain into certain functional groups are also discussed. Chemistry based on EEGE has been proved to be an efficient, versatile route to constructing copolymers containing Gly units and ultimately yielding the target properties and applications.展开更多
