Methods: Retrospective analysis of 250 admitted type 2 diabetes inpatients admitted to the endocrinology department of qinghai provincial hospital of traditional chinese medicine from september 2022 to december 2023, collated and analysed the general data and laboratory indicators of the patient cases, and applied spss26.0 to process and analyse the data and explore the differences in the general conditions and commonly used clinical nutritional indicators of type 2 diabetes in patients with different 25-hydroxyVitamin D levels. The differences between the different 25-hydroxy Vitamin D levels in type 2 diabetes mellitus patients. Results: 1) A total of 250 inpatients with type 2 diabetes mellitus were included in this study, of which 56 cases (22.4%) were patients with 25 hydroxyvitamin D deficiency [25(OH)D P P P P > 0.05). 3) The distribution of some laboratory indexes among the three groups of patients was differentiated, with the average level of glycated haemoglobin in the lack group being significantly higher than that of the remaining two groups, the average level of albumin being significantly lower than that of the remaining two groups, and the average level of haemoglobin being significantly lower than that of the good group (P P > 0.05). Conclusion: The 25(OH)D level of type 2 diabetes mellitus inpatients is not optimistic. In clinical diagnosis and treatment, we should pay attention to the changes in 25 hydroxyvitamin D levels and other nutritional indexes of patients with type 2 diabetes mellitus, and focus on the control of blood glucose levels and timely supplementation of vitamins, proteins, and lipids, in order to improve the patients’ physical status, reduce the incidence of complications, and improve the clinical efficacy and the patients’ quality of life.展开更多

BACKGROUND RAS,BRAF,and mismatch repair(MMR)/microsatellite instability(MSI)are crucial biomarkers recommended by clinical practice guidelines for colorectal cancer(CRC).However,their characteristics and influencing f...BACKGROUND RAS,BRAF,and mismatch repair(MMR)/microsatellite instability(MSI)are crucial biomarkers recommended by clinical practice guidelines for colorectal cancer(CRC).However,their characteristics and influencing factors in Chinese patients have not been thoroughly described.AIM To analyze the clinicopathological features of KRAS,NRAS,BRAF,and PIK3CA mutations and the DNA MMR status in CRC.METHODS We enrolled 2271 Chinese CRC patients at the China-Japan Friendship Hospital.MMR proteins were tested using immunohistochemical analysis,and the KRAS/NRAS/BRAF/PIK3CA mutations were determined using quantitative polymerase chain reaction.Microsatellite status was determined using an MSI detection kit.Statistical analyses were conducted using SPSS software and logistic regression.RESULTS The KRAS,NRAS,BRAF,and PIK3CA mutations were detected in 44.6%,3.4%,3.7%,and 3.9% of CRC patients,respectively.KRAS mutations were more likely to occur in patients with moderate-to-high differentiation.BRAF mutations were more likely to occur in patients with right-sided CRC,poorly differentiated,or no perineural invasion.Deficient MMR(dMMR)was detected in 7.9% of all patients and 16.8% of those with mucinous adenocarcinomas.KRAS,NRAS,BRAF,and PIK3CA mutations were detected in 29.6%,1.1%,8.1%,and 22.3% of patients with dMMR,respectively.The dMMR was more likely to occur in patients with a family history of CRC,aged<50 years,right-sided CRC,poorly differentiated histology,no perineural invasion,and with carcinoma in situ,stage I,or stage II tumors.CONCLUSION This study analyzed the molecular profiles of KRAS,NRAS,BRAF,PIK3CA,and MMR/MSI in CRC,identifying key influencing factors,with implications for clinical management of CRC.展开更多
Objective:To analyze the enduring rotational steadiness of AcrySof IQ Toric intraocular lens(IOL)in cataract patients suffering from myopia in a long-term study.Methods:A retrospective study was conducted on a case se...Objective:To analyze the enduring rotational steadiness of AcrySof IQ Toric intraocular lens(IOL)in cataract patients suffering from myopia in a long-term study.Methods:A retrospective study was conducted on a case series involving 78 patients.A total of 120 eyes with an axial length(AL)ranging from 24-30 mm and corneal astigmatism≥1.50 D underwent implantation of AcrySof IQ Toric IOL guided by the version navigation system.The eyes were divided into two groups based on AL.Group A included 60 eyes with high myopia(AL≥26 mm),while Group B consisted of eyes with low to moderate myopia(24 mm≤AL<26 mm).Data on the preoperative AL were collected.Measurements were taken for residual astigmatism,the best corrected visual acuity(BCDVA),corneal astigmatism,and IOL rotation occurring between 24-and 48-months post-surgery.The percentage of eyes with an IOL rotation of under 5°and 10°was analyzed.Results:The mean length of follow-up times was recorded as 34.27±4.98,and the average rotation was 2.73±1.29°.Group A exhibited a slightly higher average rotation of 2.87±1.31°,compared to the rotation of 2.59±1.27°observed in Group B.At both the 24-36 month and 26-48 month post-operation marks,the degree of IOL rotation did not show a statistically significant difference between the two groups,with none of the patients experiencing a rotation exceeding 10°(P>0.05).The percentage of rotation degrees under 5°was recorded as 98.22%.After the procedure,the BCDVA was 0.1322±0.03 LogMAR.There was a substantial increase in theχvalue after the operation as compared to the pre-operativeχ^(2) value(χ^(2)=76.79).The standard deviation of preoperative corneal astigmatism was statistically significant(P<0.05)at 2.17±1.08 D.Following the surgical procedure,the remaining astigmatism was measured at 0.41±0.26 D.The data showed a notable gap in statistical significance(t=4.281,P<0.05).Conclusion:The AcrySof Toric IOL was a reliable solution for managing corneal astigmatism in cataract patients with myopia,demonstrating excellent long-term rotational stability.展开更多
Nitrogen(N)fertilization is necessary for obtaining high rice yield.But excessive N fertilizer reduces rice plant N efficiency and causes negative effects such as environmental pollution.In this study,we assembled key...Nitrogen(N)fertilization is necessary for obtaining high rice yield.But excessive N fertilizer reduces rice plant N efficiency and causes negative effects such as environmental pollution.In this study,we assembled key genes involved in different nodes of N pathways to boost nitrate and ammonium uptake and assimilation,and to strengthen amino acid utilization to increase grain yield and nitrogen use efficiency(NUE)in rice.The combinations OsNPF8.9a×OsNR2,OsAMT1;2×OsGS1;2×OsAS1,and OsGS2×OsAS2×OsANT3 optimized nitrate assimilation,ammonium conversion,and N reutilization,respectively.In co-overexpressing rice lines obtained by co-transformation,the tiller number,biomass,and grain yield per plant of the OsAMT1;2×OsGS1;2×OsAS1-overexpressing line exceeded those of wild-type ZH11,the OsNPF8.9a×OsNR2×OsGS1;2×OsAS1-overexpressing line,and the OsGS2×OsAS2×OsANT3-overexpressing line.The glutamine synthase activity,free amino acids,and nitrogen utilization efficiency(NUt E)of the OsAMT1;2×OsGS1;2×OsAS1-overexpressing line exceeded those of ZH11 and other lines that combined key genes.N influx efficiency was increased in the OsAMT1;2×OsGS1;2×OsAS1-overexpressing line and OsNPF8.9a×OsNR2×OsGS1;2×OsAS1-overexpressing line under a low ammonium and a low nitrate treatment,respectively.We propose that combining overexpression of OsAMT1;2,OsGS1;2,and OsAS1 is a promising breeding strategy for systematically increasing rice grain yield and NUE by focusing on key nodes in the N pathway.展开更多

Steganography techniques,such as audio steganography,have been widely used in covert communication.However,the deep neural network,especially the convolutional neural network(CNN),has greatly threatened the security o...Steganography techniques,such as audio steganography,have been widely used in covert communication.However,the deep neural network,especially the convolutional neural network(CNN),has greatly threatened the security of audio steganography.Besides,existing adversarial attacks-based countermeasures cannot provide general perturbation,and the trans-ferability against unknown steganography detection methods is weak.This paper proposes a cover enhancement method for audio steganography based on universal adversarial perturbations with sample diversification to address these issues.Universal adversarial perturbation is constructed by iteratively optimizing adversarial perturbation,which applies adversarial attack tech-niques,such as Deepfool.Moreover,the sample diversification strategy is designed to improve the transferability of adversarial perturbations in black-box attack scenarios,where two types of common audio-processing operations are considered,including noise addition and moving picture experts group audio layer III(MP3)compression.Furthermore,the perturbation ensemble method is applied to further improve the attacks’transferability by integrating perturbations of different detection networks with heterogeneous architec-tures.Consequently,the single universal adversarial perturbation can enhance different cover audios against a CNN-based detection network.Extensive experiments have been conducted,and the results demonstrate that the average missed-detection probabilities of the proposed method are higher than those of the state-of-the-art methods by 7.3%and 16.6%for known and unknown detection networks,respectively.It verifies the efficiency and transferability of the proposed methods for the cover enhancement of audio steganography.展开更多
BACKGROUND Hypertension is a common chronic disease that affects many people worldwide.Only a few reports related to the exploration of relevant indicators of the prethrombotic state in patients with primary hypertens...BACKGROUND Hypertension is a common chronic disease that affects many people worldwide.Only a few reports related to the exploration of relevant indicators of the prethrombotic state in patients with primary hypertension(PH)in clinical settings were available.AIM To detect prethrombotic state-related indicators in patients with PH and analyze their differences in different patient populations to provide a laboratory basis for the clinical prevention and control of hypertensive thrombotic diseases.METHODS The general data of patients with PH who attended the Department of Cardiovascular Medicine,The First Affiliated Hospital of Jiangxi Medical College,from January 2022 to December 2022 were collected retrospectively.The patients were divided into three groups of 40 patients each according to the Grade of PH:Grade 1,Grade 2,and Grade 3 hypertension experimental group.The baseline data of 40 volunteers,who underwent physical examination in our hospital but were not diagnosed with PH during the same period,were included in the control group.The relevant indicators of prethrombotic state of the participants were compared,and mainly included inflammation-related indicators,hemorheology-related indicators,and coagulation function related indicators.The relationship between the aforementioned indicators and the progression of PH was analyzed.RESULTS No significant differences were observed in age,sex,diabetes mellitus,smoking history,drinking history,body mass index,New York Heart Association functional classification,or the course of hypertension among the four groups(P>0.05).The expressions of high-sensitivity C-reactive protein(hs-CRP),thrombomodulin(TM),hematocrit(Hct),erythrocyte sedimentation rate(ESR),P-selectin on platelet surface(CD62P),and fibrinogen(FIB)in the control group were<Grade 1 hypertension group<Grade 2 hypertension group<Grade 3 hypertension group,and the expressions of platelet(PLT),activated partial thromboplastin time(APTT),prothrombin(PT),and plasma thrombin time(TT)in the control group was>Grade 1 hypertension group>Grade 2 hypertension group>Grade 3 hypertension group,and the difference was statistically significant(P<0.05).The results of the multivariate logistic regression model showed that the expression of hs-CRP,TM,Hct,ESR,CD62P,PLT,APTT,PT,TT,and FIB in the included participants was related to the progression of PH.Among these,high expression of hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT,and low expression of PLT and FIB were risk factors for PH(OR>1,P<0.05).The results of the receiver operating characteristic curve analysis showed that the area under the curve of hs-CRP,TM,ESR,CD62P,APTT,PT,TT,and FIB for the prediction of PH were>0.80,and the prediction value was ideal.Linear correlation analysis with bivariate Spearman showed that hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT were positively correlated with each other(r>0,P<0.05);PLT and FIB were negatively correlated with hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT(r<0,P<0.05);and PLT and FIB were positively correlated(r>0,P<0.05).Linear correlation analysis using bivariate Spearman showed that hs-CRP,TM,Hct,ESR,CD62P,and FIB were positively correlated with each other(r>0,P<0.05),whereas PLT,APTT,PT,and TT were negatively correlated with hs-CRP,TM,Hct,ESR,CD62P,and FIB(r<0,P<0.05).There was a positive correlation between PLT,APTT,PT,and TT(r>0,P<0.05).CONCLUSION The relevant indicators of the prethrombotic state in patients with PH,such as hs-CRP,TM,Hct,ESR,CD62P,PLT,APTT,PT,TT,and FIB,showed differences.High expression of hs-CRP,TM,Hct,ESR,CD62P,and FIB,and low expression of PLT,APTT,PT,and TT are the keys to the occurrence,progression,and thrombotic state of PH.Based on the above serum indicators’expression in patients,targeted interventions can be administered to patients with abnormal expression levels to control the progression of their disease and reduce the risk of developing a prethrombotic state.展开更多

Intracellular HBV DNA replication intermediates and ccc DNA were detected by Southern blot and real-time PCR, respectively. The acetylation levels of histones H3 and H4 were measured by Western blot. H3/H4-bound ccc DNA was detected by chromatin immunoprecipitation(Ch IP) assays. The deacetylase inhibitors trichostatin A and sodium butyrate were used to study the mechanism of action for curcumin. Additionally, short interfering RNAs(si RNAs) targeting HBV were tested along with curcumin.RESULTS Curcumin treatment led to time-and dose-dependent reductions in HBs Ag and HBe Ag expression and significant reductions in intracellular HBV DNA replication intermediates and HBV ccc DNA. After treatment with 20 μmol/L curcumin for 2 d, HBs Ag and ccc DNA levels in Hep G2.2.15 cells were reduced by up to 57.7%(P < 0.01) and 75.5%(P < 0.01), respectively, compared with levels in non-treated cells. Meanwhile, time-and dose-dependent reductions in the histone H3 acetylation levels were also detected upon treatment with curcumin, accompanied by reductions in H3-and H4-bound ccc DNA. Furthermore, the deacetylase inhibitors trichostatin A and sodium butyrate could block the effects of curcumin. Additionally, transfection of si RNAs targeting HBV enhanced the inhibitory effects of curcumin.CONCLUSION Curcumin inhibits HBV gene replication via downregulation of ccc DNA-bound histone acetylation and has the potential to be developed as a ccc DNA-targeting antiviral agent for hepatitis B.展开更多
We describe the computed tomography(CT)imaging findings in six cases(five males and one female;age range 61-78 years;mean age 67.3 years)with histologically proven hepatoid adenocarcinoma of the stomach(HAS).Five of t...We describe the computed tomography(CT)imaging findings in six cases(five males and one female;age range 61-78 years;mean age 67.3 years)with histologically proven hepatoid adenocarcinoma of the stomach(HAS).Five of the six patients had elevated serum alpha-fetoprotein levels.The most common type of gross appearance HAS on CT is a polypoid mass(83%,5/6).The most common contrast enhancement pattern was heterogeneous.All six patients had a regional lymphadenopathy larger than 6 mm in its short axis.Liver metastases(n=3)were noted.Venous tumor thrombosis was identified in the portal vein(n=2)of the regions near primary gastric tumors or metastatic masses.Our findings suggest in an elderly,male patients with a large heterogeneous enhancement tumor,the presence of distant metastases,regional lymphadenopathy and characteristically increased serum alphafetoprotein levels indicates a high likelihood of HAS.展开更多
AIM To detect abnormal micro RNA(miRNA) expression in type 1 gastric neuroendocrine neoplasms(g-NENs) and find potential target genes.METHODS Tumour tissues from patients with type 1 g-NENs were used as experimental s...AIM To detect abnormal micro RNA(miRNA) expression in type 1 gastric neuroendocrine neoplasms(g-NENs) and find potential target genes.METHODS Tumour tissues from patients with type 1 g-NENs were used as experimental samples, and gastric mucosal tissues from the same patients obtained during gastroscopy review after several months were used as control samples. miRNA expression was examined with Agilent human miRNA chips and validated via RTPCR. Three types of target gene prediction software(TargetScan, PITA, and microRNAorg) were used to predict potential target genes of the differentially expressed miRNAs, and a dual-luciferase reporter assay system was used for verification. RESULTS Six miRNAs were significantly upregulated or downregulated in the tumours compared to the control samples. Among them, miR-202-3 p was extraordinarily upregulated. RT-PCR of seven sample sets confirmed that miR-202-3 p was upregulated in tumour tissues. In total, 215 target genes were predicted to be associated with miR-202-3 p. Among them, dual-specificity phosphatase 1(DUSP1) was reported to be closely related to tumour occurrence and development. The dual-luciferase reporter assay showed that miR-202-3 p directly regulated DUSP1 in 293 T cells. CONCLUSION miR-202-3 p is upregulated in type 1 g-NEN lesions and might play important roles in the pathogenesis of type 1 g-NENs by targeting DUSP1.展开更多
BACKGROUND Poorly differentiated gastric neuroendocrine neoplasms(PDGNENs)include gastric neuroendocrine carcinoma(NEC)and mixed adenoneuroendocrine carcinoma,which are highly malignant and rare tumors,and their incid...BACKGROUND Poorly differentiated gastric neuroendocrine neoplasms(PDGNENs)include gastric neuroendocrine carcinoma(NEC)and mixed adenoneuroendocrine carcinoma,which are highly malignant and rare tumors,and their incidence has increased over the past few decades.However,the clinicopathological features and outcomes of patients with PDGNENs have not been completely elucidated.AIM To investigate the clinicopathological characteristics and prognostic factors of patients with PDGNENs.METHODS The data from seven centers in China from March 2007 to November 2019 were analyzed retrospectively.RESULTS Among the 232 patients with PDGNENs,191(82.3%)were male,with an average age of 62.83±9.11 years.One hundred and thirteen(49.34%)of 229 patients had a stage III disease and 86(37.55%)had stage IV disease.Three(1.58%)of 190 patients had no clinical symptoms,while 187(98.42%)patients presented clinical symptoms.The tumors were mainly(89.17%)solitary and located in the upper third of the stomach(cardia and fundus of stomach:115/215,53.49%).Most lesions were ulcers(157/232,67.67%),with an average diameter of 4.66±2.77 cm.In terms of tumor invasion,the majority of tumors invaded the serosa(116/198,58.58%).The median survival time of the 232 patients was 13.50 mo(7,31 mo),and the overall 1-year,3-year,and 5-year survival rates were 49%,19%,and 5%,respectively.According to univariate analysis,tumor number,tumor diameter,gastric invasion status,American Joint Committee on Cancer(AJCC)stage,and distant metastasis status were prognostic factors for patients with PDGNENs.Multivariate analysis showed that tumor number,tumor diameter,AJCC stage,and distant metastasis status were independent prognostic factors for patients with PDGNENs.CONCLUSION The overall prognosis of patients with PDGNENs is poor.The outcomes of patients with a tumor diameter>5 cm,multiple tumors,and stage IV tumors are worse than those of other patients.展开更多
Rye(Secale cereale L.)has been widely used to improve wheat(Triticum aestivum L.)cultivars.Oligo probes combined with non-denaturing fluorescence in situ hybridization(ND-FISH)technology provide a convenient and effic...Rye(Secale cereale L.)has been widely used to improve wheat(Triticum aestivum L.)cultivars.Oligo probes combined with non-denaturing fluorescence in situ hybridization(ND-FISH)technology provide a convenient and efficient way to identify individual rye chromosomes.However,suitable ND-FISH-positive oligo probes for recognizing specific segments of rye chromosomes are lacking.Five new ND-FISH-positive oligo probes:Oligo-5BL.46,Oligo-5A8080,Oligo-5A8080.1,Oligo-1AL.73,and Oligo-0R3,combined with two previously reported oligo probes,Oligo-44 and Oligo-45,were used in this study.Probes Oligo-5BL.46,Oligo-5A8080 and Oligo-44 produced signals only in intercalary regions of arms 1RS,5RS,and 5RL,respectively.Probe Oligo-5A8080.1 combined with probe Oligo-45 distinguished the intercalary regions of arms 1RS,5RS,and 6RS simultaneously.Oligo-5A8080 and Oligo-5A8080.1 revealed variation in the distribution of 5S rDNA sequences and polymorphism among 5R chromosomes.Probe Oligo-1AL.73 produced signals only on chromosomes 4R and 7R and contributed to the construction of an improved FISH map of chromosome 4RKu and to the confirmation of 4RLKu breakpoints in wheat-rye 4RLKu translocation chromosomes.Oligo-0R3 produced signals in the telomeric and subtelomeric regions of 14 rye chromosomes.These oligo probes also revealed five new tandem repeats in rye.Using the oligo probes reported in this study,the short arms of 1R,5R,and 6R and the long arms of 4R and 7R can be easily discriminated when these chromosomes are broken.展开更多
Monoclinic SrAl_(2)Si_(2)O_(8) ceramics for Sr immobilization were prepared by a liquid-phase sintering method.The sintering temperature,mineral phase composition,microstructure,flexural strength,bulk density,and Sr i...Monoclinic SrAl_(2)Si_(2)O_(8) ceramics for Sr immobilization were prepared by a liquid-phase sintering method.The sintering temperature,mineral phase composition,microstructure,flexural strength,bulk density,and Sr ion leaching characteristics of the SrAl_(2)Si_(2)O_(8) ceramics were investigated.A crystalline monoclinic SrAl_(2)Si_(2)O_(8) phase formed through liquid-phase sintering at 1223 K.The introduction of four flux agents(B_(2)O_(3),CaO·2B_(2)O_(3),SrO·2B_(2)O_(3),and BaO·2B_(2)O_(3))to the SrAl_(2)Si_(2)O_(8) ceramics not only reduced the densification temperature and decreased the volatilization of Sr during high-temperature sintering but also impacted the mechanical properties of the ceramics.Product consistency tests showed that the leaching concentration of Sr ions in the sample with flux agent B_(2)O_(3) was the lowest,whereas that of Sr ions in the sample with flux agent BaO·2B_(2)O_(3) was the highest.These results show that the leaching concentration of Sr ions depends largely on the amorphous phase in the ceramics.Meanwhile,the formation of mineral analog ceramics containing Sr is an important factor to improve Sr immobilization.展开更多
文摘Objective: To investigate the distribution of 25-hydroxy Vitamin D level status in type 2 diabetes mellitus inpatients, as well as the differences in general conditions and clinical indicators in patients with different Vitamin D status. Methods: Retrospective analysis of 250 admitted type 2 diabetes inpatients admitted to the endocrinology department of qinghai provincial hospital of traditional chinese medicine from september 2022 to december 2023, collated and analysed the general data and laboratory indicators of the patient cases, and applied spss26.0 to process and analyse the data and explore the differences in the general conditions and commonly used clinical nutritional indicators of type 2 diabetes in patients with different 25-hydroxyVitamin D levels. The differences between the different 25-hydroxy Vitamin D levels in type 2 diabetes mellitus patients. Results: 1) A total of 250 inpatients with type 2 diabetes mellitus were included in this study, of which 56 cases (22.4%) were patients with 25 hydroxyvitamin D deficiency [25(OH)D P P P P > 0.05). 3) The distribution of some laboratory indexes among the three groups of patients was differentiated, with the average level of glycated haemoglobin in the lack group being significantly higher than that of the remaining two groups, the average level of albumin being significantly lower than that of the remaining two groups, and the average level of haemoglobin being significantly lower than that of the good group (P P > 0.05). Conclusion: The 25(OH)D level of type 2 diabetes mellitus inpatients is not optimistic. In clinical diagnosis and treatment, we should pay attention to the changes in 25 hydroxyvitamin D levels and other nutritional indexes of patients with type 2 diabetes mellitus, and focus on the control of blood glucose levels and timely supplementation of vitamins, proteins, and lipids, in order to improve the patients’ physical status, reduce the incidence of complications, and improve the clinical efficacy and the patients’ quality of life.

文摘The poor interfacial stability not only deteriorates fibre lithium-ion batteries(FLBs)performance but also impacts their scalable applications.To efficiently address these challenges,Prof.Huisheng Peng team proposed a generalized channel structures strategy with optimized in situ polymerization technology in their recent study.The resultant FLBs can be woven into different-sized powering textiles,providing a high energy density output of 128 Wh kg^(-1) and simultaneously demonstrating good durability even under harsh conditions.Such a promising strategy expands the horizon in developing FLB with particular polymer gel electrolytes,and significantly ever-deepening understanding of the scaled wearable energy textile system toward a sustainable future.

文摘BACKGROUND RAS,BRAF,and mismatch repair(MMR)/microsatellite instability(MSI)are crucial biomarkers recommended by clinical practice guidelines for colorectal cancer(CRC).However,their characteristics and influencing factors in Chinese patients have not been thoroughly described.AIM To analyze the clinicopathological features of KRAS,NRAS,BRAF,and PIK3CA mutations and the DNA MMR status in CRC.METHODS We enrolled 2271 Chinese CRC patients at the China-Japan Friendship Hospital.MMR proteins were tested using immunohistochemical analysis,and the KRAS/NRAS/BRAF/PIK3CA mutations were determined using quantitative polymerase chain reaction.Microsatellite status was determined using an MSI detection kit.Statistical analyses were conducted using SPSS software and logistic regression.RESULTS The KRAS,NRAS,BRAF,and PIK3CA mutations were detected in 44.6%,3.4%,3.7%,and 3.9% of CRC patients,respectively.KRAS mutations were more likely to occur in patients with moderate-to-high differentiation.BRAF mutations were more likely to occur in patients with right-sided CRC,poorly differentiated,or no perineural invasion.Deficient MMR(dMMR)was detected in 7.9% of all patients and 16.8% of those with mucinous adenocarcinomas.KRAS,NRAS,BRAF,and PIK3CA mutations were detected in 29.6%,1.1%,8.1%,and 22.3% of patients with dMMR,respectively.The dMMR was more likely to occur in patients with a family history of CRC,aged<50 years,right-sided CRC,poorly differentiated histology,no perineural invasion,and with carcinoma in situ,stage I,or stage II tumors.CONCLUSION This study analyzed the molecular profiles of KRAS,NRAS,BRAF,PIK3CA,and MMR/MSI in CRC,identifying key influencing factors,with implications for clinical management of CRC.

文摘Objective:To analyze the enduring rotational steadiness of AcrySof IQ Toric intraocular lens(IOL)in cataract patients suffering from myopia in a long-term study.Methods:A retrospective study was conducted on a case series involving 78 patients.A total of 120 eyes with an axial length(AL)ranging from 24-30 mm and corneal astigmatism≥1.50 D underwent implantation of AcrySof IQ Toric IOL guided by the version navigation system.The eyes were divided into two groups based on AL.Group A included 60 eyes with high myopia(AL≥26 mm),while Group B consisted of eyes with low to moderate myopia(24 mm≤AL<26 mm).Data on the preoperative AL were collected.Measurements were taken for residual astigmatism,the best corrected visual acuity(BCDVA),corneal astigmatism,and IOL rotation occurring between 24-and 48-months post-surgery.The percentage of eyes with an IOL rotation of under 5°and 10°was analyzed.Results:The mean length of follow-up times was recorded as 34.27±4.98,and the average rotation was 2.73±1.29°.Group A exhibited a slightly higher average rotation of 2.87±1.31°,compared to the rotation of 2.59±1.27°observed in Group B.At both the 24-36 month and 26-48 month post-operation marks,the degree of IOL rotation did not show a statistically significant difference between the two groups,with none of the patients experiencing a rotation exceeding 10°(P>0.05).The percentage of rotation degrees under 5°was recorded as 98.22%.After the procedure,the BCDVA was 0.1322±0.03 LogMAR.There was a substantial increase in theχvalue after the operation as compared to the pre-operativeχ^(2) value(χ^(2)=76.79).The standard deviation of preoperative corneal astigmatism was statistically significant(P<0.05)at 2.17±1.08 D.Following the surgical procedure,the remaining astigmatism was measured at 0.41±0.26 D.The data showed a notable gap in statistical significance(t=4.281,P<0.05).Conclusion:The AcrySof Toric IOL was a reliable solution for managing corneal astigmatism in cataract patients with myopia,demonstrating excellent long-term rotational stability.

文摘Nitrogen(N)fertilization is necessary for obtaining high rice yield.But excessive N fertilizer reduces rice plant N efficiency and causes negative effects such as environmental pollution.In this study,we assembled key genes involved in different nodes of N pathways to boost nitrate and ammonium uptake and assimilation,and to strengthen amino acid utilization to increase grain yield and nitrogen use efficiency(NUE)in rice.The combinations OsNPF8.9a×OsNR2,OsAMT1;2×OsGS1;2×OsAS1,and OsGS2×OsAS2×OsANT3 optimized nitrate assimilation,ammonium conversion,and N reutilization,respectively.In co-overexpressing rice lines obtained by co-transformation,the tiller number,biomass,and grain yield per plant of the OsAMT1;2×OsGS1;2×OsAS1-overexpressing line exceeded those of wild-type ZH11,the OsNPF8.9a×OsNR2×OsGS1;2×OsAS1-overexpressing line,and the OsGS2×OsAS2×OsANT3-overexpressing line.The glutamine synthase activity,free amino acids,and nitrogen utilization efficiency(NUt E)of the OsAMT1;2×OsGS1;2×OsAS1-overexpressing line exceeded those of ZH11 and other lines that combined key genes.N influx efficiency was increased in the OsAMT1;2×OsGS1;2×OsAS1-overexpressing line and OsNPF8.9a×OsNR2×OsGS1;2×OsAS1-overexpressing line under a low ammonium and a low nitrate treatment,respectively.We propose that combining overexpression of OsAMT1;2,OsGS1;2,and OsAS1 is a promising breeding strategy for systematically increasing rice grain yield and NUE by focusing on key nodes in the N pathway.

文摘In order to improve the catalytic performance of the nitrobenzene hydrogenation rearrangement to prepare p-aminophenol,a bimetallic Pt-Ni/C(PNC)catalyst was synthesized.Taking advantage of the synergistic effect of Ni and Pt to enhance product selectivity and catalytic performance stability,the electrons in Ni are moved to Pt by the electron effect,which affects the catalyst’s ability to activate H_(2)as well as the amount of hydrogen activated.Furthermore,due to the strong Pt(5d)-Ni(3d)coupling effect,Ni can effectively maintain Pt stability in the acidic system and reduce Pt dissolution.The stability of the PNC can be found to be greatly enhanced compared to the Pt/C(PC)catalyst,and p-aminophenol selectivity is greatly enhanced,showing excellent catalytic performance.

文摘Steganography techniques,such as audio steganography,have been widely used in covert communication.However,the deep neural network,especially the convolutional neural network(CNN),has greatly threatened the security of audio steganography.Besides,existing adversarial attacks-based countermeasures cannot provide general perturbation,and the trans-ferability against unknown steganography detection methods is weak.This paper proposes a cover enhancement method for audio steganography based on universal adversarial perturbations with sample diversification to address these issues.Universal adversarial perturbation is constructed by iteratively optimizing adversarial perturbation,which applies adversarial attack tech-niques,such as Deepfool.Moreover,the sample diversification strategy is designed to improve the transferability of adversarial perturbations in black-box attack scenarios,where two types of common audio-processing operations are considered,including noise addition and moving picture experts group audio layer III(MP3)compression.Furthermore,the perturbation ensemble method is applied to further improve the attacks’transferability by integrating perturbations of different detection networks with heterogeneous architec-tures.Consequently,the single universal adversarial perturbation can enhance different cover audios against a CNN-based detection network.Extensive experiments have been conducted,and the results demonstrate that the average missed-detection probabilities of the proposed method are higher than those of the state-of-the-art methods by 7.3%and 16.6%for known and unknown detection networks,respectively.It verifies the efficiency and transferability of the proposed methods for the cover enhancement of audio steganography.

文摘BACKGROUND Hypertension is a common chronic disease that affects many people worldwide.Only a few reports related to the exploration of relevant indicators of the prethrombotic state in patients with primary hypertension(PH)in clinical settings were available.AIM To detect prethrombotic state-related indicators in patients with PH and analyze their differences in different patient populations to provide a laboratory basis for the clinical prevention and control of hypertensive thrombotic diseases.METHODS The general data of patients with PH who attended the Department of Cardiovascular Medicine,The First Affiliated Hospital of Jiangxi Medical College,from January 2022 to December 2022 were collected retrospectively.The patients were divided into three groups of 40 patients each according to the Grade of PH:Grade 1,Grade 2,and Grade 3 hypertension experimental group.The baseline data of 40 volunteers,who underwent physical examination in our hospital but were not diagnosed with PH during the same period,were included in the control group.The relevant indicators of prethrombotic state of the participants were compared,and mainly included inflammation-related indicators,hemorheology-related indicators,and coagulation function related indicators.The relationship between the aforementioned indicators and the progression of PH was analyzed.RESULTS No significant differences were observed in age,sex,diabetes mellitus,smoking history,drinking history,body mass index,New York Heart Association functional classification,or the course of hypertension among the four groups(P>0.05).The expressions of high-sensitivity C-reactive protein(hs-CRP),thrombomodulin(TM),hematocrit(Hct),erythrocyte sedimentation rate(ESR),P-selectin on platelet surface(CD62P),and fibrinogen(FIB)in the control group were<Grade 1 hypertension group<Grade 2 hypertension group<Grade 3 hypertension group,and the expressions of platelet(PLT),activated partial thromboplastin time(APTT),prothrombin(PT),and plasma thrombin time(TT)in the control group was>Grade 1 hypertension group>Grade 2 hypertension group>Grade 3 hypertension group,and the difference was statistically significant(P<0.05).The results of the multivariate logistic regression model showed that the expression of hs-CRP,TM,Hct,ESR,CD62P,PLT,APTT,PT,TT,and FIB in the included participants was related to the progression of PH.Among these,high expression of hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT,and low expression of PLT and FIB were risk factors for PH(OR>1,P<0.05).The results of the receiver operating characteristic curve analysis showed that the area under the curve of hs-CRP,TM,ESR,CD62P,APTT,PT,TT,and FIB for the prediction of PH were>0.80,and the prediction value was ideal.Linear correlation analysis with bivariate Spearman showed that hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT were positively correlated with each other(r>0,P<0.05);PLT and FIB were negatively correlated with hs-CRP,TM,Hct,ESR,CD62P,APTT,PT,and TT(r<0,P<0.05);and PLT and FIB were positively correlated(r>0,P<0.05).Linear correlation analysis using bivariate Spearman showed that hs-CRP,TM,Hct,ESR,CD62P,and FIB were positively correlated with each other(r>0,P<0.05),whereas PLT,APTT,PT,and TT were negatively correlated with hs-CRP,TM,Hct,ESR,CD62P,and FIB(r<0,P<0.05).There was a positive correlation between PLT,APTT,PT,and TT(r>0,P<0.05).CONCLUSION The relevant indicators of the prethrombotic state in patients with PH,such as hs-CRP,TM,Hct,ESR,CD62P,PLT,APTT,PT,TT,and FIB,showed differences.High expression of hs-CRP,TM,Hct,ESR,CD62P,and FIB,and low expression of PLT,APTT,PT,and TT are the keys to the occurrence,progression,and thrombotic state of PH.Based on the above serum indicators’expression in patients,targeted interventions can be administered to patients with abnormal expression levels to control the progression of their disease and reduce the risk of developing a prethrombotic state.

文摘AIM To investigate the potential effect of curcumin on hepatitis B virus(HBV) covalently closed circular DNA(ccc DNA) and the underlying mechanism.METHODS A Hep G2.2.15 cell line stably transfected with HBV was treated with curcumin, and HBV surface antigen(HBs Ag) and e antigen(HBe Ag) expression levels were assessed by ELISA. Intracellular HBV DNA replication intermediates and ccc DNA were detected by Southern blot and real-time PCR, respectively. The acetylation levels of histones H3 and H4 were measured by Western blot. H3/H4-bound ccc DNA was detected by chromatin immunoprecipitation(Ch IP) assays. The deacetylase inhibitors trichostatin A and sodium butyrate were used to study the mechanism of action for curcumin. Additionally, short interfering RNAs(si RNAs) targeting HBV were tested along with curcumin.RESULTS Curcumin treatment led to time-and dose-dependent reductions in HBs Ag and HBe Ag expression and significant reductions in intracellular HBV DNA replication intermediates and HBV ccc DNA. After treatment with 20 μmol/L curcumin for 2 d, HBs Ag and ccc DNA levels in Hep G2.2.15 cells were reduced by up to 57.7%(P < 0.01) and 75.5%(P < 0.01), respectively, compared with levels in non-treated cells. Meanwhile, time-and dose-dependent reductions in the histone H3 acetylation levels were also detected upon treatment with curcumin, accompanied by reductions in H3-and H4-bound ccc DNA. Furthermore, the deacetylase inhibitors trichostatin A and sodium butyrate could block the effects of curcumin. Additionally, transfection of si RNAs targeting HBV enhanced the inhibitory effects of curcumin.CONCLUSION Curcumin inhibits HBV gene replication via downregulation of ccc DNA-bound histone acetylation and has the potential to be developed as a ccc DNA-targeting antiviral agent for hepatitis B.
文摘We describe the computed tomography(CT)imaging findings in six cases(five males and one female;age range 61-78 years;mean age 67.3 years)with histologically proven hepatoid adenocarcinoma of the stomach(HAS).Five of the six patients had elevated serum alpha-fetoprotein levels.The most common type of gross appearance HAS on CT is a polypoid mass(83%,5/6).The most common contrast enhancement pattern was heterogeneous.All six patients had a regional lymphadenopathy larger than 6 mm in its short axis.Liver metastases(n=3)were noted.Venous tumor thrombosis was identified in the portal vein(n=2)of the regions near primary gastric tumors or metastatic masses.Our findings suggest in an elderly,male patients with a large heterogeneous enhancement tumor,the presence of distant metastases,regional lymphadenopathy and characteristically increased serum alphafetoprotein levels indicates a high likelihood of HAS.

文摘AIM To detect abnormal micro RNA(miRNA) expression in type 1 gastric neuroendocrine neoplasms(g-NENs) and find potential target genes.METHODS Tumour tissues from patients with type 1 g-NENs were used as experimental samples, and gastric mucosal tissues from the same patients obtained during gastroscopy review after several months were used as control samples. miRNA expression was examined with Agilent human miRNA chips and validated via RTPCR. Three types of target gene prediction software(TargetScan, PITA, and microRNAorg) were used to predict potential target genes of the differentially expressed miRNAs, and a dual-luciferase reporter assay system was used for verification. RESULTS Six miRNAs were significantly upregulated or downregulated in the tumours compared to the control samples. Among them, miR-202-3 p was extraordinarily upregulated. RT-PCR of seven sample sets confirmed that miR-202-3 p was upregulated in tumour tissues. In total, 215 target genes were predicted to be associated with miR-202-3 p. Among them, dual-specificity phosphatase 1(DUSP1) was reported to be closely related to tumour occurrence and development. The dual-luciferase reporter assay showed that miR-202-3 p directly regulated DUSP1 in 293 T cells. CONCLUSION miR-202-3 p is upregulated in type 1 g-NEN lesions and might play important roles in the pathogenesis of type 1 g-NENs by targeting DUSP1.

文摘BACKGROUND Poorly differentiated gastric neuroendocrine neoplasms(PDGNENs)include gastric neuroendocrine carcinoma(NEC)and mixed adenoneuroendocrine carcinoma,which are highly malignant and rare tumors,and their incidence has increased over the past few decades.However,the clinicopathological features and outcomes of patients with PDGNENs have not been completely elucidated.AIM To investigate the clinicopathological characteristics and prognostic factors of patients with PDGNENs.METHODS The data from seven centers in China from March 2007 to November 2019 were analyzed retrospectively.RESULTS Among the 232 patients with PDGNENs,191(82.3%)were male,with an average age of 62.83±9.11 years.One hundred and thirteen(49.34%)of 229 patients had a stage III disease and 86(37.55%)had stage IV disease.Three(1.58%)of 190 patients had no clinical symptoms,while 187(98.42%)patients presented clinical symptoms.The tumors were mainly(89.17%)solitary and located in the upper third of the stomach(cardia and fundus of stomach:115/215,53.49%).Most lesions were ulcers(157/232,67.67%),with an average diameter of 4.66±2.77 cm.In terms of tumor invasion,the majority of tumors invaded the serosa(116/198,58.58%).The median survival time of the 232 patients was 13.50 mo(7,31 mo),and the overall 1-year,3-year,and 5-year survival rates were 49%,19%,and 5%,respectively.According to univariate analysis,tumor number,tumor diameter,gastric invasion status,American Joint Committee on Cancer(AJCC)stage,and distant metastasis status were prognostic factors for patients with PDGNENs.Multivariate analysis showed that tumor number,tumor diameter,AJCC stage,and distant metastasis status were independent prognostic factors for patients with PDGNENs.CONCLUSION The overall prognosis of patients with PDGNENs is poor.The outcomes of patients with a tumor diameter>5 cm,multiple tumors,and stage IV tumors are worse than those of other patients.

文摘Rye(Secale cereale L.)has been widely used to improve wheat(Triticum aestivum L.)cultivars.Oligo probes combined with non-denaturing fluorescence in situ hybridization(ND-FISH)technology provide a convenient and efficient way to identify individual rye chromosomes.However,suitable ND-FISH-positive oligo probes for recognizing specific segments of rye chromosomes are lacking.Five new ND-FISH-positive oligo probes:Oligo-5BL.46,Oligo-5A8080,Oligo-5A8080.1,Oligo-1AL.73,and Oligo-0R3,combined with two previously reported oligo probes,Oligo-44 and Oligo-45,were used in this study.Probes Oligo-5BL.46,Oligo-5A8080 and Oligo-44 produced signals only in intercalary regions of arms 1RS,5RS,and 5RL,respectively.Probe Oligo-5A8080.1 combined with probe Oligo-45 distinguished the intercalary regions of arms 1RS,5RS,and 6RS simultaneously.Oligo-5A8080 and Oligo-5A8080.1 revealed variation in the distribution of 5S rDNA sequences and polymorphism among 5R chromosomes.Probe Oligo-1AL.73 produced signals only on chromosomes 4R and 7R and contributed to the construction of an improved FISH map of chromosome 4RKu and to the confirmation of 4RLKu breakpoints in wheat-rye 4RLKu translocation chromosomes.Oligo-0R3 produced signals in the telomeric and subtelomeric regions of 14 rye chromosomes.These oligo probes also revealed five new tandem repeats in rye.Using the oligo probes reported in this study,the short arms of 1R,5R,and 6R and the long arms of 4R and 7R can be easily discriminated when these chromosomes are broken.

文摘Monoclinic SrAl_(2)Si_(2)O_(8) ceramics for Sr immobilization were prepared by a liquid-phase sintering method.The sintering temperature,mineral phase composition,microstructure,flexural strength,bulk density,and Sr ion leaching characteristics of the SrAl_(2)Si_(2)O_(8) ceramics were investigated.A crystalline monoclinic SrAl_(2)Si_(2)O_(8) phase formed through liquid-phase sintering at 1223 K.The introduction of four flux agents(B_(2)O_(3),CaO·2B_(2)O_(3),SrO·2B_(2)O_(3),and BaO·2B_(2)O_(3))to the SrAl_(2)Si_(2)O_(8) ceramics not only reduced the densification temperature and decreased the volatilization of Sr during high-temperature sintering but also impacted the mechanical properties of the ceramics.Product consistency tests showed that the leaching concentration of Sr ions in the sample with flux agent B_(2)O_(3) was the lowest,whereas that of Sr ions in the sample with flux agent BaO·2B_(2)O_(3) was the highest.These results show that the leaching concentration of Sr ions depends largely on the amorphous phase in the ceramics.Meanwhile,the formation of mineral analog ceramics containing Sr is an important factor to improve Sr immobilization.
