Herbal extraction residues(HERs)cause serious environmental pollution and resource waste.In this study,a novel green route was designed for the comprehensive reutilization of all components in HERs,taking Magnolia off...Herbal extraction residues(HERs)cause serious environmental pollution and resource waste.In this study,a novel green route was designed for the comprehensive reutilization of all components in HERs,taking Magnolia officinalis residues(MOR)as an example.The reluctant structure of MOR was first destroyed by alkali pretreatment to release the functional ingredients(magnolol and honokiol)originally remaining in MOR and to make MOR more accessible for hydrolysis.A metal–organic frame material MIL-101(Cr)with a maximum absorption capacity of 255.64 mg g^(-1)was synthesized to absorb the released honokiol and magnolol from the pretreated MOR solutions,and 40 g L^(-1)reducing sugars were obtained with 81.8%enzymatic hydrolysis rate at 10%MOR solid loading.Finally,382 mg L-1β-amyrin was produced from MOR hydrolysates by an engineered yeast strain.In total,1 kg honokiol,8 kg magnolol,and 7.64 kg β-amyrin could produce from 1 ton MOR by this cleaner process with a total economic output of 170,700 RMB.展开更多

Background:Observational studies have shown that inflammatory bowel disease(IBD),such as ulcerative colitis(UC)and Crohn disease(CD),is associated with gingivitis and periodontal disease(GP).This study aims to investi...Background:Observational studies have shown that inflammatory bowel disease(IBD),such as ulcerative colitis(UC)and Crohn disease(CD),is associated with gingivitis and periodontal disease(GP).This study aims to investigate whether there is a causal relationship between IBD and GP.Methods:This study assessed the causal relationship between IBD and GP through a two-sample Mendelian randomization(MR)study.The required data were obtained through the IEU OpenGWAS project.Instrumental variable screening and the MR and sensitivity analyses were performed using the“TwoSampleMR”R package.Results:IBD,UC,and CD may have a causal effect on GP(IBD,inverse variance weighting[IVW]OR=1.05,95%CI=1.00–1.10,P=0.03;UC,IVWOR=1.05,95%CI=1.00–1.11,P=0.03;CD,weighted median OR=1.06,95%CI=1.00–1.13,P=0.04;simple mode OR=1.15,95%CI=1.02–1.31,P=0.03).Scatterplots,forest plots,and funnel plots showed a significant relationship between IBD and GP and confirmed the robustness of the model.In sensitivity testing,no horizontal pleiotropy or heterogeneity was found in this study.Conclusions:This study found a possible causal relationship between IBD(UC and CD)and GP,which deserves to be considered in clinical practice.展开更多
Previous studies have shown that resveratrol,a bioactive substance found in many plants,can reduce early brain injury after subarachnoid hemorrhage,but how it acts is still unclear.This study explored the mechanism us...Previous studies have shown that resveratrol,a bioactive substance found in many plants,can reduce early brain injury after subarachnoid hemorrhage,but how it acts is still unclear.This study explored the mechanism using the experimental subarachnoid hemorrhage rat model established by injecting autologous blood into the cerebellomedullary cistern.Rat models were treated with an intraperitoneal injection of 60 mg/kg resveratrol 2,6,24 and 46 hours after injury.At 48 hours after injury,their neurological function was assessed using a modified Garcia score.Brain edema was measured by the wet-dry method.Neuronal apoptosis in the prefrontal cortex was detected by terminal deoxyribonucleotidyl transferase-mediated biotin-16-dUTP nick-end labeling assay.Levels of reactive oxygen species and malondialdehyde in the prefrontal cortex were determined by colorimetry.CHOP,glucose-regulated protein 78,nuclear factor-erythroid2-related factor 2 and heme oxygenase-1 mRNA expression levels in the prefrontal cortex were measured by reverse transcription polymerase chain reaction.Tumor necrosis factor-alpha content in the prefrontal cortex was detected by enzyme linked immunosorbent assay.Immunohistochemical staining was used to detect the number of positive cells of nuclear factor-erythroid 2-related factor 2,heme oxygenase 1,glucose-regulated protein 78,CHOP and glial fibrillary acidic protein.Western blot assay was utilized to analyze the expression levels of nuclear factor-erythroid 2-related factor 2,heme oxygenase 1,glucose-regulated protein 78 and CHOP protein expression levels in the prefrontal cortex.The results showed that resveratrol treatment markedly alleviated neurological deficits and brain edema in experimental subarachnoid hemorrhage rats,and reduced neuronal apoptosis in the prefrontal cortex.Resveratrol reduced the levels of reactive oxygen species and malondialdehyde,and increased the expression of nuclear factor-erythroid 2-related factor 2,heme oxygenase-1 mRNA and protein in the prefrontal cortex.Resveratrol decreased glucose-regulated protein 78,CHOP mRNA and protein expression and tumor necrosis factor-alpha level.It also activated astrocytes.The results suggest that resveratrol exerted neuroprotective effect on subarachnoid hemorrhage by reducing oxidative damage,endoplasmic reticulum stress and neuroinflammation.The study was approved by the Animals Ethics Committee of Shandong University,China on February 22,2016(approval No.LL-201602022).展开更多

Diet-induced obese ratswere used as an insulin resistance model to study therelationship of resistin with insulin resistance.RESULTS: Resistin expression and secretion wereenhanced during differentiation 3T3-L1 preadipocytes.This cellular differentiation stimulated resistin expressionand secretion, but was suppressed by insulin. Resistinalso induced insulin resistance in H4IIE hepatocytes andL6 myoblasts. In diet-induced obese rats, serum resistinlevels were negatively correlated with insulin sensitivity,but not with serum insulin.CONCLUSION: Insulin can inhibit resistin expressionand secretion in vitro, but insulin is not a major regulatorof resistin in vivo . Hydrogen sulfide,which can be generated in the central nervous system from the sulfhydryl-containing amino acid,L-cysteine,by cystathionine-β-synthase,may exert protective effects in experimental subarachnoid hemorrh...Hydrogen sulfide,which can be generated in the central nervous system from the sulfhydryl-containing amino acid,L-cysteine,by cystathionine-β-synthase,may exert protective effects in experimental subarachnoid hemorrhage;however,the mechanism underlying this effect is unknown.This study explored the mechanism using a subarachnoid hemorrhage rat model induced by an endovascular perforation technique.Rats were treated with an intraperitoneal injection of 100 mM L-cysteine(30μL)30 minutes after subarachnoid hemorrhage.At 48 hours after subarachnoid hemorrhage,hematoxylin-eosin staining was used to detect changes in prefrontal cortex cells.L-cysteine significantly reduced cell edema.Neurological function was assessed using a modified Garcia score.Brain water content was measured by the wet-dry method.L-cysteine significantly reduced neurological deficits and cerebral edema after subarachnoid hemorrhage.Immunofluorescence was used to detect the number of activated microglia.Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the levels of interleukin 1β and CD86 mRNA in the prefrontal cortex.L-cysteine inhibited microglial activation in the prefrontal cortex and reduced the mRNA levels of interleukin 1βand CD86.RT-PCR and western blot analysis of the complement system showed that L-cysteine reduced expression of the complement factors,C1q,C3αand its receptor C3aR1,and the deposition of C1q in the prefrontal cortex.Dihydroethidium staining was applied to detect changes in reactive oxygen species,and immunohistochemistry was used to detect the number of NRF2-and HO-1-positive cells.L-cysteine reduced the level of reactive oxygen species in the prefrontal cortex and the number of NRF2-and HO-1-positive cells.Western blot assays and immunohistochemistry were used to detect the protein levels of CHOP and GRP78 in the prefrontal cortex and the number of CHOP-and GRP78-positive cells.L-cysteine reduced CHOP and GRP78 levels and the number of CHOP-and GRP78-positive cells.The cystathionine-β-synthase inhibitor,aminooxyacetic acid,significantly reversed the above neuroprotective effects of L-cysteine.Taken together,L-cysteine can play a neuroprotective role by regulating neuroinflammation,complement deposition,oxidative stress and endoplasmic reticulum stress.The study was approved by the Animals Ethics Committee of Shandong University,China on February 22,2016(approval No.LL-201602022).展开更多

We aimed to evaluate the levels of poly...Adverse birth outcomes are a leading cause of mortality in children in China, but the environmental influences of these conditions remain largely unexplained in this population. We aimed to evaluate the levels of polycyclic aromatic hydrocarbons (PAHs) in Chinese pregnant women and their newborns, and to examine the association between levels of PAHs and infant birth weight. We conducted a cross-sectional study including 81 pairs of mothers and newborns from four hospitals in four different cities in China. High Performance Liquid Chromatography was used to measure the concentration of nine PAHs in maternal and cord blood and multiple linear regression analyses were used to evaluate the associations of these PAHs with infant birth weight. Anthracene (ANT) had the highest average concentration and detection rate (geometric mean = 69.54 ng/g and 76.5%, respectively) in maternal serum samples, while fluoranthene (FLT) had the highest concentration and detection rate (geometric mean = 68.4 ng/g and 50.6%, respectively) in the cord blood. Most of the measured PAHs in maternal serum and three PAHs in cord blood were inversely but non-significantly associated with birth weight. The strongest associations were observed for higher concentrations of benzo (a) pyrene (BaP) in maternal serum (230.7 g decrease for levels > median vs. < LOD;p = 0.151) and for ANT in cord blood (153.1 g decrease for levels < median vs. < LOD;p = 0.208). Ant and FLT were the predominant PAHs in the maternal and cord blood serum. Serum concentrations of several measured PAHs were associated with a decreased birth weight, although not significantly, suggesting that further studies with larger sample sizes are needed to validate our findings.展开更多

Methods and Materials: Nine randoml...Purpose: To evaluate planning quality of Stereotactic body Radiotherapy (SBRT) with multiple lungmetastases generated by the Pinnacle and Tomotherapy planning systems, respectively. Methods and Materials: Nine randomly selected patients diagnosed with non-small cell lung carcinoma with multiple lesions were planned with Philips Pinnacle (version 9.2, Fitchburg, WI) and Tomotherapy (version 4.2, Madison, WI), respectively. Both coplanar and non-coplanar IMRT plans were generated on Pinnacle system. A total dose of 60 Gy was prescribed to cover 95% of Planning Target Volume (PTV) in 3 fractions based on the RTOG0236 protocol prescription [1]. All plans with single isocenter setting were used for multiple lesions planning. A set of nine static beams were used for Pinnacle plansusing Direct Machine Parameters Optimization (DMPO) algorithm of RTOT0236 dose constraints. Planning outcomes such as minimum and mean doses, V95, D95 (95% of target volume receivesprescription dose), D5, and D1 to PTV, maximum dose to heart, esophagus, cord, trachea, brachial plexus, rib, chest wall, and liver, mean dose toliver, total lung, right and left lung, volume of chest wall receives 30 Gy, volume of lungs receives 5 Gy and 20 Gy (V5 and V20), conformity index (CI) and heterogeneity index (HI) were all reported for evaluation. Results: Mean volume of PTV was 37.77 ± 23.4 cm3. D95 of PTV with Tomotherapy, coplanar, non-coplanar plan was 60.2 ± 0.3 Gy, 58.6 ± 1.2 Gy, and 59.1 ± 0.7 Gy, respectively. Various optimization methods, such as the particle swarm optimization (PSO), the genetic algorithm (GA), and a robust optimization method based on design for six sigma (DFSS), have been applied to realize uniform current distribution among the multilayer HTS conductors. The continuous and discrete variables, such as the winding angle, radius, and winding direction of each layer, are chosen as the design parameters. Under the constraints of the mechanical properties and critical current, PSO is proven to be a more powerful tool than GA for structural parameter optimization, and DFSS can not only achieve a uniform current distribution, but also improve significantly the reliability and robustness of the HTS cable quality.展开更多
The use of atmospheric rotating gliding arc(RGA)plasma is proposed as a facile,scalable and catalyst-free approach to synthesizing hydrogen(H2)and graphene sheets from coalbed methane(CBM).CH4 is used as a CBM surroga...The use of atmospheric rotating gliding arc(RGA)plasma is proposed as a facile,scalable and catalyst-free approach to synthesizing hydrogen(H2)and graphene sheets from coalbed methane(CBM).CH4 is used as a CBM surrogate.Based on a previous investigation of discharge properties,product distribution and energy efficiency,the operating parameters such as CH4 concentration,applied voltage and gas flow rate can effectively affect the CH4 conversion rate,the selectivity of H2 and the properties of solid generated carbon.Nevertheless,the basic properties of RGA plasma and its role in CH4 conversion are scarcely mentioned.In the present work,a 3D RGA model,with a detailed nonequilibrium CH4/Ar plasma chemistry,is developed to validate the previous experiments on CBM conversion,aiming in particular at the distribution of H2 and other gas products.Our results demonstrate that the dynamics of RGA is derived from the joint effects of electron convection,electron migration and electron diffusion,and is prominently determined by the variation of the gas flow rate and applied voltage.Subsequently,a combined experimental and chemical kinetical simulation is performed to analyze the selectivity of gas products in an RGA reaction,taking into consideration the formation and loss pathways of crucial targeted substances(such as CH4,C2H2,H2 and H radicals)and corresponding contribution rates.Additionally,the effects of operating conditions on the properties of solid products are investigated by scanning electron microscopy(SEM)and Raman spectroscopy.The results show that increasing the applied voltage and decreasing CH4 concentration will change the solid carbon from its initial spherical structure into folded multilayer graphene sheets,while the size of the graphene sheets is slightly affected by the change in gas flow rate.展开更多

In inertial confinement fusion(ICF),overlapping of laser beams is common.Owing to the effective high laser intensity of the overlapped beams,the collective mode of stimulated Brillouin scattering(SBS)with a shared sca...In inertial confinement fusion(ICF),overlapping of laser beams is common.Owing to the effective high laser intensity of the overlapped beams,the collective mode of stimulated Brillouin scattering(SBS)with a shared scattered light wave is potentially important.In this work,an exact analytic solution for the convective gain coefficient of the collective SBS modes with shared scattered wave is presented for two overlapped beams based on a linear kineticmodel.The effects of the crossing angle,polarization states,and finite beamoverlapping volume of the two laser beams on the shared light modes are analyzed for cases with zero and nonzero wavelength difference between the two beams.It is found that all these factors have a significant influence on the shared lightmodes of SBS.Furthermore,the out-of-plane modes,in which the wavevectors of daughter waves lie in different planes from the two overlapped beams,are found to be important for certain polarization states and especially for obtuse crossing angles.In particular,adjusting the polarization directions of the two beams to be orthogonal to each other or tuning the wavelength difference to a sufficiently large value(of the order of nanometers)are found to be effective methods to suppress the shared light modes of SBS.This work will be helpful for comprehending and suppressing collective SBS with shared scattered waves in ICF experiments.展开更多
Purpose: To evaluate planning quality and dosimetric differences of clinically deliverable 3D conformal plans generated from Tomotherapy with TomoDirectTM (TD) and conventional field-in-field approach in prone breast ...Purpose: To evaluate planning quality and dosimetric differences of clinically deliverable 3D conformal plans generated from Tomotherapy with TomoDirectTM (TD) and conventional field-in-field approach in prone breast treatment. Materials and methods: Total of twelve randomly selected early stage left breast patients who went through lumpectomy and were previously treated on traditional Linear Accelerator (LINAC) have been re-planned and tested on Tomotherapy TomoDirect module. Baseline prescription dose was chosen at 50.4 Gy (1.8 Gy × 28 fractions) to cover ≥95% of PTV for planning criteria with other critical structure dose constraints in the thoracic region. Planning outcomes such as D95 (95% of volume of PTV receiving the prescribed dose), D5 and D1, heart, both lungs as well as the contralateral breast were simultaneously evaluated. Conformity of the prescription isodose/volume to PTV was evaluated as conformity index (CI) and dose uniformity was also evaluated with homogeneity index (HI) in the same study series. All outcome parameters were analyzed and summarized to evaluate dosimetric impact of planning qualities between these two planning platforms. Results: The planning results indicate that CI, HI, D95, D5 and D1 of PTV, critical structures such as heart, ipsilateral and contralateral lungs as well as contralateral breast doses were comparable but with better overall statistical end points from TD plans. The D95, D5 and D1 of PTV for TD plans were superior in dosimetric analysis and more uniform than those plans generated from PinnacleTM field-in-field planning technique. Overall, TD plans have superior planning quality than the conventional method does, with straightforward and automated planning process once the beam delivery parameters were established. Conclusions: From the clinical treatment planning results, plans from TD in general achieved better uniform tumor coverage with fewer hot spots while sparing more critical structures were based upon isodose distribution and Dose Volume Histogram (DVH) analysis. Image guidance of TD delivery automates the setup within the treatment bore without tedious verification process compared to the process with LINAC. Though all plans are deliverable, TD planning possesses dosimetric advantages due to its modulated optimization pattern. However, TD did present a challenge during the simulation if a patient is oversized with long pendulant breast which is hard to fit into the Tomotherapy ring structure. From our analysis, TD plans reserve superior dosimetric outcome with CI, HI, D95, D5, and D1 of PTV, and better sparing contralateral lung and breast doses.展开更多
Objective:To observe the effect of interferonα-2b treatment on liver function,liver fibrosis,complement protein and oxidative stress in patients with hepatitis B.Methods:A total of 100 patients with hepatitis B in ou...Objective:To observe the effect of interferonα-2b treatment on liver function,liver fibrosis,complement protein and oxidative stress in patients with hepatitis B.Methods:A total of 100 patients with hepatitis B in our hospital were randomly divided into the control group and the observation group,with 50 cases in each group.After admission,patients in the control group were treated with entecavir,while patients in the observation group were treated with interferonα-2b combined with entecavir.The levels of serum total bilirubin(TBil),aspartate aminotransferase(AST),alanine aminotransferase(ALT),type III procollagen(PCIII),type IV collagen(CIV),complement C3 protein(C3),complement C4 protein(C4),malondialdehyde(MDA),superoxide dismutase(SOD)and nitric oxide(NO)were compared between the two groups before and after treatment.Results:After treatment,the levels of TBil,AST,ALT,PCIII,CIV,MDA and NO in serum of patients with hepatitis B in both groups were significantly lower than those before treatment,and the levels of C3,C4 and SOD were significantly higher than those before treatment(P<0.05).After treatment,the levels of TBil,AST,ALT,PC III,C IV,MDA and NO in serum of patients in the observation group were significantly lower than those in the control group,while the levels of C3,C4 and SOD in serum of patients in the observation group were significantly higher than those in the control group(P<0.05).Conclusions:The combination of interferonα-2b and entecavir has a good curative effect on hepatitis B.It can significantly improve the liver function and immune function of patients,delay the process of liver fibrosis and reduce oxidative stress injury.It is worthy of clinical promotion.展开更多

Serum PCT levels of both groups were detected, and the observation group were further divided into high PCT group and low PCT group (n=38) according to the median of PCT levels. Exploring the physiochemical properties and expanding the applications of actinide-containing materials is paramount to address the escalating challenge of radioactive waste accumulation.However,unlocking the full pot...Exploring the physiochemical properties and expanding the applications of actinide-containing materials is paramount to address the escalating challenge of radioactive waste accumulation.However,unlocking the full potential of these materials is largely crippled by the radiotoxicity of the actinides.We report here two porous and luminescent thorium-based metal-organic frameworks(Th-BITD-1 and Th-BITD-2)that serve as a bifunctional platform for sequencing and sensing of radioiodine,a much more radioactive fission product discharged during the nuclear fuel reprocessing.In particular,Th-BITD-1 displays better iodine uptake performance than Th-BITD-2 via the solution-based process and vapor diffusion with the maximum adsorption capacities of 831 and 1099 mg/g,respectively.Fur thermore,Th-BITD-1 can function as a highly sensitive luminescence sensor for iodate with a quenching constant(Ksv)of 6.6(5)×10^(3) M^(-1)and a detection limit of 2.02μM,respectively,outperforming 2.96(6)×10^(3)M^(-1)and 10.5μM of Th-BITD-2.Moreover,a positive correlation between the sensing efficacy and the iodate adsorption capacity has been revealed.This work highlights the opportunity in designing novel actinide-based MOFs for their potential applications in radiological fields,e.g.,radionuclide separation and detection.展开更多

文摘Herbal extraction residues(HERs)cause serious environmental pollution and resource waste.In this study,a novel green route was designed for the comprehensive reutilization of all components in HERs,taking Magnolia officinalis residues(MOR)as an example.The reluctant structure of MOR was first destroyed by alkali pretreatment to release the functional ingredients(magnolol and honokiol)originally remaining in MOR and to make MOR more accessible for hydrolysis.A metal–organic frame material MIL-101(Cr)with a maximum absorption capacity of 255.64 mg g^(-1)was synthesized to absorb the released honokiol and magnolol from the pretreated MOR solutions,and 40 g L^(-1)reducing sugars were obtained with 81.8%enzymatic hydrolysis rate at 10%MOR solid loading.Finally,382 mg L-1β-amyrin was produced from MOR hydrolysates by an engineered yeast strain.In total,1 kg honokiol,8 kg magnolol,and 7.64 kg β-amyrin could produce from 1 ton MOR by this cleaner process with a total economic output of 170,700 RMB.

文摘A new mechanism for the generation of high intensity speckles by coupling of overlapping beams is discovered and studied in detail.Using three-dimensional simulations,the coupling of overlapping beams smoothed by phase plates and by polarization smoothing are investigated in the regime relevant to inertial confinement fusion studies.It is found that the intensity distribution of the laser beam spot can be changed by nonuniform spatial phase modulation,and the speckles formed by the phase plate can be split into smaller speckles with higher intensities,which is favorable for the generation of laser plasma instabilities.Stimulated Brillouin scattering is compared in simulations with and without coupling of the overlapping incident beams,and the results confirm the enhancement of stimulated Brillouin scattering due to this mechanism.

文摘Background:Observational studies have shown that inflammatory bowel disease(IBD),such as ulcerative colitis(UC)and Crohn disease(CD),is associated with gingivitis and periodontal disease(GP).This study aims to investigate whether there is a causal relationship between IBD and GP.Methods:This study assessed the causal relationship between IBD and GP through a two-sample Mendelian randomization(MR)study.The required data were obtained through the IEU OpenGWAS project.Instrumental variable screening and the MR and sensitivity analyses were performed using the“TwoSampleMR”R package.Results:IBD,UC,and CD may have a causal effect on GP(IBD,inverse variance weighting[IVW]OR=1.05,95%CI=1.00–1.10,P=0.03;UC,IVWOR=1.05,95%CI=1.00–1.11,P=0.03;CD,weighted median OR=1.06,95%CI=1.00–1.13,P=0.04;simple mode OR=1.15,95%CI=1.02–1.31,P=0.03).Scatterplots,forest plots,and funnel plots showed a significant relationship between IBD and GP and confirmed the robustness of the model.In sensitivity testing,no horizontal pleiotropy or heterogeneity was found in this study.Conclusions:This study found a possible causal relationship between IBD(UC and CD)and GP,which deserves to be considered in clinical practice.

文摘Previous studies have shown that resveratrol,a bioactive substance found in many plants,can reduce early brain injury after subarachnoid hemorrhage,but how it acts is still unclear.This study explored the mechanism using the experimental subarachnoid hemorrhage rat model established by injecting autologous blood into the cerebellomedullary cistern.Rat models were treated with an intraperitoneal injection of 60 mg/kg resveratrol 2,6,24 and 46 hours after injury.At 48 hours after injury,their neurological function was assessed using a modified Garcia score.Brain edema was measured by the wet-dry method.Neuronal apoptosis in the prefrontal cortex was detected by terminal deoxyribonucleotidyl transferase-mediated biotin-16-dUTP nick-end labeling assay.Levels of reactive oxygen species and malondialdehyde in the prefrontal cortex were determined by colorimetry.CHOP,glucose-regulated protein 78,nuclear factor-erythroid2-related factor 2 and heme oxygenase-1 mRNA expression levels in the prefrontal cortex were measured by reverse transcription polymerase chain reaction.Tumor necrosis factor-alpha content in the prefrontal cortex was detected by enzyme linked immunosorbent assay.Immunohistochemical staining was used to detect the number of positive cells of nuclear factor-erythroid 2-related factor 2,heme oxygenase 1,glucose-regulated protein 78,CHOP and glial fibrillary acidic protein.Western blot assay was utilized to analyze the expression levels of nuclear factor-erythroid 2-related factor 2,heme oxygenase 1,glucose-regulated protein 78 and CHOP protein expression levels in the prefrontal cortex.The results showed that resveratrol treatment markedly alleviated neurological deficits and brain edema in experimental subarachnoid hemorrhage rats,and reduced neuronal apoptosis in the prefrontal cortex.Resveratrol reduced the levels of reactive oxygen species and malondialdehyde,and increased the expression of nuclear factor-erythroid 2-related factor 2,heme oxygenase-1 mRNA and protein in the prefrontal cortex.Resveratrol decreased glucose-regulated protein 78,CHOP mRNA and protein expression and tumor necrosis factor-alpha level.It also activated astrocytes.The results suggest that resveratrol exerted neuroprotective effect on subarachnoid hemorrhage by reducing oxidative damage,endoplasmic reticulum stress and neuroinflammation.The study was approved by the Animals Ethics Committee of Shandong University,China on February 22,2016(approval No.LL-201602022).

文摘AIM: To confirm whether insulin regulates resistinexpression and secretion during differentiation of 3T3-L1preadipocytes and the relationship of resistin with insulinresistance both in vivo and in vitro.METHODS: Supernatant resistin was measured duringdifferentiation of 3T3-L1 preadipocytes. L6 rat myoblastsand hepatoma cell line H4IIE were used to confirm thecellular function of resistin. Diet-induced obese ratswere used as an insulin resistance model to study therelationship of resistin with insulin resistance.RESULTS: Resistin expression and secretion wereenhanced during differentiation 3T3-L1 preadipocytes.This cellular differentiation stimulated resistin expressionand secretion, but was suppressed by insulin. Resistinalso induced insulin resistance in H4IIE hepatocytes andL6 myoblasts. In diet-induced obese rats, serum resistinlevels were negatively correlated with insulin sensitivity,but not with serum insulin.CONCLUSION: Insulin can inhibit resistin expressionand secretion in vitro, but insulin is not a major regulatorof resistin in vivo . Fat tissue mass affects insulinsensitivity by altering the expression and secretion ofresistin.

文摘Hydrogen sulfide,which can be generated in the central nervous system from the sulfhydryl-containing amino acid,L-cysteine,by cystathionine-β-synthase,may exert protective effects in experimental subarachnoid hemorrhage;however,the mechanism underlying this effect is unknown.This study explored the mechanism using a subarachnoid hemorrhage rat model induced by an endovascular perforation technique.Rats were treated with an intraperitoneal injection of 100 mM L-cysteine(30μL)30 minutes after subarachnoid hemorrhage.At 48 hours after subarachnoid hemorrhage,hematoxylin-eosin staining was used to detect changes in prefrontal cortex cells.L-cysteine significantly reduced cell edema.Neurological function was assessed using a modified Garcia score.Brain water content was measured by the wet-dry method.L-cysteine significantly reduced neurological deficits and cerebral edema after subarachnoid hemorrhage.Immunofluorescence was used to detect the number of activated microglia.Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the levels of interleukin 1β and CD86 mRNA in the prefrontal cortex.L-cysteine inhibited microglial activation in the prefrontal cortex and reduced the mRNA levels of interleukin 1βand CD86.RT-PCR and western blot analysis of the complement system showed that L-cysteine reduced expression of the complement factors,C1q,C3αand its receptor C3aR1,and the deposition of C1q in the prefrontal cortex.Dihydroethidium staining was applied to detect changes in reactive oxygen species,and immunohistochemistry was used to detect the number of NRF2-and HO-1-positive cells.L-cysteine reduced the level of reactive oxygen species in the prefrontal cortex and the number of NRF2-and HO-1-positive cells.Western blot assays and immunohistochemistry were used to detect the protein levels of CHOP and GRP78 in the prefrontal cortex and the number of CHOP-and GRP78-positive cells.L-cysteine reduced CHOP and GRP78 levels and the number of CHOP-and GRP78-positive cells.The cystathionine-β-synthase inhibitor,aminooxyacetic acid,significantly reversed the above neuroprotective effects of L-cysteine.Taken together,L-cysteine can play a neuroprotective role by regulating neuroinflammation,complement deposition,oxidative stress and endoplasmic reticulum stress.The study was approved by the Animals Ethics Committee of Shandong University,China on February 22,2016(approval No.LL-201602022).
文摘Adverse birth outcomes are a leading cause of mortality in children in China, but the environmental influences of these conditions remain largely unexplained in this population. We aimed to evaluate the levels of polycyclic aromatic hydrocarbons (PAHs) in Chinese pregnant women and their newborns, and to examine the association between levels of PAHs and infant birth weight. We conducted a cross-sectional study including 81 pairs of mothers and newborns from four hospitals in four different cities in China. High Performance Liquid Chromatography was used to measure the concentration of nine PAHs in maternal and cord blood and multiple linear regression analyses were used to evaluate the associations of these PAHs with infant birth weight. Anthracene (ANT) had the highest average concentration and detection rate (geometric mean = 69.54 ng/g and 76.5%, respectively) in maternal serum samples, while fluoranthene (FLT) had the highest concentration and detection rate (geometric mean = 68.4 ng/g and 50.6%, respectively) in the cord blood. Most of the measured PAHs in maternal serum and three PAHs in cord blood were inversely but non-significantly associated with birth weight. The strongest associations were observed for higher concentrations of benzo (a) pyrene (BaP) in maternal serum (230.7 g decrease for levels > median vs. < LOD;p = 0.151) and for ANT in cord blood (153.1 g decrease for levels < median vs. < LOD;p = 0.208). Ant and FLT were the predominant PAHs in the maternal and cord blood serum. Serum concentrations of several measured PAHs were associated with a decreased birth weight, although not significantly, suggesting that further studies with larger sample sizes are needed to validate our findings.
文摘Purpose: To evaluate planning quality of Stereotactic body Radiotherapy (SBRT) with multiple lungmetastases generated by the Pinnacle and Tomotherapy planning systems, respectively. Methods and Materials: Nine randomly selected patients diagnosed with non-small cell lung carcinoma with multiple lesions were planned with Philips Pinnacle (version 9.2, Fitchburg, WI) and Tomotherapy (version 4.2, Madison, WI), respectively. Both coplanar and non-coplanar IMRT plans were generated on Pinnacle system. A total dose of 60 Gy was prescribed to cover 95% of Planning Target Volume (PTV) in 3 fractions based on the RTOG0236 protocol prescription [1]. All plans with single isocenter setting were used for multiple lesions planning. A set of nine static beams were used for Pinnacle plansusing Direct Machine Parameters Optimization (DMPO) algorithm of RTOT0236 dose constraints. Planning outcomes such as minimum and mean doses, V95, D95 (95% of target volume receivesprescription dose), D5, and D1 to PTV, maximum dose to heart, esophagus, cord, trachea, brachial plexus, rib, chest wall, and liver, mean dose toliver, total lung, right and left lung, volume of chest wall receives 30 Gy, volume of lungs receives 5 Gy and 20 Gy (V5 and V20), conformity index (CI) and heterogeneity index (HI) were all reported for evaluation. Results: Mean volume of PTV was 37.77 ± 23.4 cm3. D95 of PTV with Tomotherapy, coplanar, non-coplanar plan was 60.2 ± 0.3 Gy, 58.6 ± 1.2 Gy, and 59.1 ± 0.7 Gy, respectively. Mean dose to PTV was lower for Tomotherapy (p 5 (p 1 (p = 0.001). CI was higher with Tomotherapyplans (p p 5 which needs more attention for toxicity analysis.
文摘In this paper, the design optimization of the structural parameters of multilayer conductors in high temperature superconducting (HTS) cable is reviewed. Various optimization methods, such as the particle swarm optimization (PSO), the genetic algorithm (GA), and a robust optimization method based on design for six sigma (DFSS), have been applied to realize uniform current distribution among the multilayer HTS conductors. The continuous and discrete variables, such as the winding angle, radius, and winding direction of each layer, are chosen as the design parameters. Under the constraints of the mechanical properties and critical current, PSO is proven to be a more powerful tool than GA for structural parameter optimization, and DFSS can not only achieve a uniform current distribution, but also improve significantly the reliability and robustness of the HTS cable quality.

文摘The use of atmospheric rotating gliding arc(RGA)plasma is proposed as a facile,scalable and catalyst-free approach to synthesizing hydrogen(H2)and graphene sheets from coalbed methane(CBM).CH4 is used as a CBM surrogate.Based on a previous investigation of discharge properties,product distribution and energy efficiency,the operating parameters such as CH4 concentration,applied voltage and gas flow rate can effectively affect the CH4 conversion rate,the selectivity of H2 and the properties of solid generated carbon.Nevertheless,the basic properties of RGA plasma and its role in CH4 conversion are scarcely mentioned.In the present work,a 3D RGA model,with a detailed nonequilibrium CH4/Ar plasma chemistry,is developed to validate the previous experiments on CBM conversion,aiming in particular at the distribution of H2 and other gas products.Our results demonstrate that the dynamics of RGA is derived from the joint effects of electron convection,electron migration and electron diffusion,and is prominently determined by the variation of the gas flow rate and applied voltage.Subsequently,a combined experimental and chemical kinetical simulation is performed to analyze the selectivity of gas products in an RGA reaction,taking into consideration the formation and loss pathways of crucial targeted substances(such as CH4,C2H2,H2 and H radicals)and corresponding contribution rates.Additionally,the effects of operating conditions on the properties of solid products are investigated by scanning electron microscopy(SEM)and Raman spectroscopy.The results show that increasing the applied voltage and decreasing CH4 concentration will change the solid carbon from its initial spherical structure into folded multilayer graphene sheets,while the size of the graphene sheets is slightly affected by the change in gas flow rate.
文摘Chondrosarcoma(CS) of larynx is uncommon and predominantly affects the cricoid cartilage.A 70% to 75% of these tumors arise on the endolaryngeal surface of the posterior lamina of the cricoid cartilage.The clinical presentation include hoarseness,stridor,dyspnea,or a neck mass etc.The diagnosis is suggested when calcification is identified in a laryngeal tumor.Computed tomography(CT) is the best noninvasive technique for imaging the lesion.Surgical extirpation is the standard therapy with no role for radiation therapy or chemotherapy.
文摘In inertial confinement fusion(ICF),overlapping of laser beams is common.Owing to the effective high laser intensity of the overlapped beams,the collective mode of stimulated Brillouin scattering(SBS)with a shared scattered light wave is potentially important.In this work,an exact analytic solution for the convective gain coefficient of the collective SBS modes with shared scattered wave is presented for two overlapped beams based on a linear kineticmodel.The effects of the crossing angle,polarization states,and finite beamoverlapping volume of the two laser beams on the shared light modes are analyzed for cases with zero and nonzero wavelength difference between the two beams.It is found that all these factors have a significant influence on the shared lightmodes of SBS.Furthermore,the out-of-plane modes,in which the wavevectors of daughter waves lie in different planes from the two overlapped beams,are found to be important for certain polarization states and especially for obtuse crossing angles.In particular,adjusting the polarization directions of the two beams to be orthogonal to each other or tuning the wavelength difference to a sufficiently large value(of the order of nanometers)are found to be effective methods to suppress the shared light modes of SBS.This work will be helpful for comprehending and suppressing collective SBS with shared scattered waves in ICF experiments.
文摘Purpose: To evaluate planning quality and dosimetric differences of clinically deliverable 3D conformal plans generated from Tomotherapy with TomoDirectTM (TD) and conventional field-in-field approach in prone breast treatment. Materials and methods: Total of twelve randomly selected early stage left breast patients who went through lumpectomy and were previously treated on traditional Linear Accelerator (LINAC) have been re-planned and tested on Tomotherapy TomoDirect module. Baseline prescription dose was chosen at 50.4 Gy (1.8 Gy × 28 fractions) to cover ≥95% of PTV for planning criteria with other critical structure dose constraints in the thoracic region. Planning outcomes such as D95 (95% of volume of PTV receiving the prescribed dose), D5 and D1, heart, both lungs as well as the contralateral breast were simultaneously evaluated. Conformity of the prescription isodose/volume to PTV was evaluated as conformity index (CI) and dose uniformity was also evaluated with homogeneity index (HI) in the same study series. All outcome parameters were analyzed and summarized to evaluate dosimetric impact of planning qualities between these two planning platforms. Results: The planning results indicate that CI, HI, D95, D5 and D1 of PTV, critical structures such as heart, ipsilateral and contralateral lungs as well as contralateral breast doses were comparable but with better overall statistical end points from TD plans. The D95, D5 and D1 of PTV for TD plans were superior in dosimetric analysis and more uniform than those plans generated from PinnacleTM field-in-field planning technique. Overall, TD plans have superior planning quality than the conventional method does, with straightforward and automated planning process once the beam delivery parameters were established. Conclusions: From the clinical treatment planning results, plans from TD in general achieved better uniform tumor coverage with fewer hot spots while sparing more critical structures were based upon isodose distribution and Dose Volume Histogram (DVH) analysis. Image guidance of TD delivery automates the setup within the treatment bore without tedious verification process compared to the process with LINAC. Though all plans are deliverable, TD planning possesses dosimetric advantages due to its modulated optimization pattern. However, TD did present a challenge during the simulation if a patient is oversized with long pendulant breast which is hard to fit into the Tomotherapy ring structure. From our analysis, TD plans reserve superior dosimetric outcome with CI, HI, D95, D5, and D1 of PTV, and better sparing contralateral lung and breast doses.

文摘Objective:To observe the effect of interferonα-2b treatment on liver function,liver fibrosis,complement protein and oxidative stress in patients with hepatitis B.Methods:A total of 100 patients with hepatitis B in our hospital were randomly divided into the control group and the observation group,with 50 cases in each group.After admission,patients in the control group were treated with entecavir,while patients in the observation group were treated with interferonα-2b combined with entecavir.The levels of serum total bilirubin(TBil),aspartate aminotransferase(AST),alanine aminotransferase(ALT),type III procollagen(PCIII),type IV collagen(CIV),complement C3 protein(C3),complement C4 protein(C4),malondialdehyde(MDA),superoxide dismutase(SOD)and nitric oxide(NO)were compared between the two groups before and after treatment.Results:After treatment,the levels of TBil,AST,ALT,PCIII,CIV,MDA and NO in serum of patients with hepatitis B in both groups were significantly lower than those before treatment,and the levels of C3,C4 and SOD were significantly higher than those before treatment(P<0.05).After treatment,the levels of TBil,AST,ALT,PC III,C IV,MDA and NO in serum of patients in the observation group were significantly lower than those in the control group,while the levels of C3,C4 and SOD in serum of patients in the observation group were significantly higher than those in the control group(P<0.05).Conclusions:The combination of interferonα-2b and entecavir has a good curative effect on hepatitis B.It can significantly improve the liver function and immune function of patients,delay the process of liver fibrosis and reduce oxidative stress injury.It is worthy of clinical promotion.
文摘Objective:To study the value of serum PCT levels for assessing the inflammatory response and RAS activation degree in patients with severe acute pancreatitis.Methods:A total of 76 patients with acute pancreatitis who were treated in our hospital between May 2013 and April 2016 were collected, and 50 patients with pure diarrhea who received infusion treatment in our hospital during the same period were selected as the control group. Serum PCT levels of both groups were detected, and the observation group were further divided into high PCT group and low PCT group (n=38) according to the median of PCT levels. Enzyme-linked immunosorbent assay (ELISA) was used to determine serum levels of pro-inflammatory factors, anti-inflammatory factors and RAS indexes.Results:Serum PCT level of observation group was higher than that of control group;serum pro-inflammatory factors IL-1β, IL-6, CRP and TNF-α levels of high PCT group and low PCT group were higher than those of control group, anti-inflammatory factors IL-4, IL-10 and IL-13 levels were higher than those of control group, and RAS indexes AngⅡ, RA and ALD levels were higher than those of control group;serum pro-inflammatory factors IL-1β, IL-6, CRP and TNF-α levels of high PCT group were higher than those of low PCT group, anti-inflammatory factors IL-4, IL-10 and IL-13 levels were higher than those of low PCT group, and the RAS indexes AngⅡ, RA and ALD levels were higher than those of low PCT group.Conclusion:The serum PCT levels can assess the inflammatory response and RAS activation degree in the patients with severe acute pancreatitis.

文摘Exploring the physiochemical properties and expanding the applications of actinide-containing materials is paramount to address the escalating challenge of radioactive waste accumulation.However,unlocking the full potential of these materials is largely crippled by the radiotoxicity of the actinides.We report here two porous and luminescent thorium-based metal-organic frameworks(Th-BITD-1 and Th-BITD-2)that serve as a bifunctional platform for sequencing and sensing of radioiodine,a much more radioactive fission product discharged during the nuclear fuel reprocessing.In particular,Th-BITD-1 displays better iodine uptake performance than Th-BITD-2 via the solution-based process and vapor diffusion with the maximum adsorption capacities of 831 and 1099 mg/g,respectively.Fur thermore,Th-BITD-1 can function as a highly sensitive luminescence sensor for iodate with a quenching constant(Ksv)of 6.6(5)×10^(3) M^(-1)and a detection limit of 2.02μM,respectively,outperforming 2.96(6)×10^(3)M^(-1)and 10.5μM of Th-BITD-2.Moreover,a positive correlation between the sensing efficacy and the iodate adsorption capacity has been revealed.This work highlights the opportunity in designing novel actinide-based MOFs for their potential applications in radiological fields,e.g.,radionuclide separation and detection.
