基于铜死亡相关基因构建肺腺癌预后模型和药物敏感性分析

背景与目的肺癌是全球最常见的恶性肿瘤之一,目前肺癌筛查和治疗策略不断完善,但其5年生存率仍然很低,严重危害人类的健康。因此探索新的生物标志物,提供个体化治疗并改善患者预后至关重要。铜死亡是一种新发现的细胞死亡类型,是由于细... 背景与目的肺癌是全球最常见的恶性肿瘤之一,目前肺癌筛查和治疗策略不断完善,但其5年生存率仍然很低,严重危害人类的健康。因此探索新的生物标志物,提供个体化治疗并改善患者预后至关重要。铜死亡是一种新发现的细胞死亡类型,是由于细胞内过量铜离子积累,最终导致细胞死亡,已有研究提示其与肺腺癌(lung adenocarcinoma,LUAD)的发生发展密切相关。本研究基于肿瘤基因组图谱(The Cancer Genome Atlas,TCGA)数据库,探究铜死亡相关基因(cuproptosis related gene,CRGs)与LUAD预后之间的关联,建立预后风险模型并分析CRGs与LUAD免疫细胞浸润之间的相互作用,为肺腺癌患者的治疗及预后提供参考。方法从TCGA数据库中下载LUAD组织和癌旁或正常肺组织的RNA-seq数据,从基因型-组织表达资料库(Genotype-tissue Expression,GTEx)下载正常肺组织的RNA-seq数据,并从基因表达综合数据库(Gene Expression Omnibus,GEO)下载462例肺腺癌数据作为验证,采用单因素Cox和Lasso-Cox回归分析构建评估预后的风险评分模型,用受试者工作特征(receiver operating characteristic,ROC)曲线及校准曲线评价模型的预测能力。并进一步对高、低风险组进行免疫相关和药物敏感性分析。结果共得到1656个CRGs,其中有1356个存在差异表达的CRGs,基于单因素Cox和Lasso-Cox回归分析筛选出13个CRGs构建预后风险模型,ROC曲线1、3、5年的曲线下面积(area under the curve,AUC)分别为0.749、0.740、0.689。进一步研究高、低风险组之间免疫相关功能和免疫检查点差异分析。高风险组对萨沃替尼、帕博西尼、阿糖胞苷等药物敏感性更高,更容易从免疫治疗中获益。结论基于13个CRGs构建的风险模型具有较好的预后价值,可以辅助LUAD患者进行个体化治疗,为LUAD的治疗及预后研究提供了重要理论依据。 展开更多

为了在间充质干细胞移植的各个阶段更好地获取移植干细胞的命运信息,制备了一种能同时实现荧光成像(FI)、磁共振成像(MRI)和正电子发射断层扫描(PET)的影像探针。首先制备了一种新型两亲性嵌段共聚物,第1嵌段为叔丁氧羰基保护的2-((叔... 为了在间充质干细胞移植的各个阶段更好地获取移植干细胞的命运信息,制备了一种能同时实现荧光成像(FI)、磁共振成像(MRI)和正电子发射断层扫描(PET)的影像探针。首先制备了一种新型两亲性嵌段共聚物,第1嵌段为叔丁氧羰基保护的2-((叔丁氧羰基)氨基)丙烯酸乙酯的均聚物,第2嵌段为短链聚乙二醇丙烯酸酯和五氟苯酚丙烯酸酯的无规共聚物,再通过氨解和酰胺化反应为聚合物修饰上双功能螯合剂DOTA和近红外染料Cy5.5,通过核磁共振和凝胶渗透色谱对聚合物结构及相对分子质量进行了表征和分析。聚合物在水中自组装并螯合Gd^(3+)得到1 mg/mL的纳米探针Gd-DOTA-Cy5.5-NP,动态光散射和透射电镜观察到探针为平均粒径30 nm的球形颗粒,并用电感耦合等离子体光谱法测得Gd^(3+)浓度为0.084 mmol/L。将纳米探针与间充质干细胞共孵育,通过Cell Counting Kit-8试剂验证1 mg/mL以下的Gd-DOTA-Cy5.5-NP相对细胞毒性较低,同时激光共聚焦显微镜在675 nm激发波长下观测细胞出现红色荧光,利用3.0 T磁共振成像系统测得探针纵向弛豫率为46.969 mmol/(L·s)。最后Gd-DOTA-Cy5.5-NP螯合^(68)Ga^(3+),得到三模态纳米探针^(68)Ga/Gd-DOTA-Cy5.5-NP,伽马计数仪测得间充质干细胞对该探针的摄取在90 min达到饱和。 展开更多

Different preparation routes for TiO_(2)-supported natural and synthetic clinoptilolite(TiO_(2)/CP)composites were thoroughly investigated on the basis of sol-gel,hydrothermal,and in-situ hydrothermal methods.The micro-structural features and physicochemical properties of resultant TiO_(2)/CPs were characterized via X-ray diffraction patterns,scanning(transmission)electron microscope images,Fourier transform infrared spectra,inductively coupled plasma-optical emission spectrometry methods,BET-isotherms,UV-visible spectra,and surface charge potential distributions.The results showed that in-situ hydrothermal method led to well dispersions of loaded-TiO_(2) particles on the surface of leaf-like CP,while obviously aggregated TiO_(2) on a relatively distorted structure of CP was obtained using sol-gel and hydrothermal methods.Their adsorptive and photocatalytic efficiencies for removal of crystal violet(CV)dye in aqueous solution were also explored under UV-irradiations.The results demonstrated that TiO_(2)/CPs synthesized via sol-gel and in-situ hydrothermal methods presented the excellent performances with 98% removal efficiencies as compare to the bare commercial TiO_(2) which achieved 53%removal of CV dye.While,the in-situ hydrothermally synthesized TiO_(2)/CPs were the best due to their moderate energy cost,highest adsorption capacities and removal efficiencies.Particularly,the synthetic CPs as supports exhibited synergetic photocatalytic activities for the degradation of CV dye,which is attributed to their high surface areas,better adsorption capability,and fine dispersion of TiO_(2) particles.Adsorption and degradation kinetics of CV dye were found to follow the pseudo-second and pseudo-first order models,respectively. 展开更多

[Objectives]To investigate the mechanism of miR-23a in the proliferation of human tongue squamous cell carcinoma cells.[Methods]Clinical tissue samples of tongue squamous cell carcinoma were collected and Tca8113 and CAL27 were cultured.Real-time quantitative PCR was performed to detect the expressions of miR-23a and PPP2R5E in clinical tissue samples of tongue squamous cell carcinoma.MTT assay,colony formation assay and growth curve assay were used to detect the effect of miR-23a and PPP2R5E on the proliferation of human tongue squamous carcinoma cell.Luciferase reporter assay verified the regulatory relationship between miR-23a and PPP2R5E.[Results]The expression of miR-23a in tongue squamous cell carcinoma was significantly up-regulated(P<0.01).miR-23a promoted the proliferation of tongue squamous cell carcinoma cells.Bioinformatics prediction and luciferin reporting experiments showed that PPP2R5E was a direct target gene of miR-23a.The expression of PPP2R5E was decreased in tongue squamous cell carcinoma.PPP2R5E inhibited the proliferation of tongue squamous cell carcinoma cells.Overexpression of PPP2R5E can reverse the proliferation promoting effect of miR-23a on tongue squamous cell carcinoma cells.[Conclusions]miR-23a can promote the proliferation of tongue squamous cell carcinoma cells through PPP2R5E and miR-23a plays an oncogene role in the occurrence and development of tongue squamous cell carcinoma. 展开更多

Purpose To evaluate the performance of diffusion-weighted imaging(DWI) and variable flip angle(VFA) T1 mapping as a supplement to image-guided biopsy in follow-up analysis of liver fibrosis. Materials and Methods This prospective study was approved by the institution's committee on human research, and written informed consent was provided from the enrolled patients. We investigated five MRI parameters of DWI and VFA T1 mapping, collected from 11 patients who underwent serial ultrasound image-guided biopsy with follow-up MRI within 1.5 years after treatment for liver fibrosis/cirrhosis. For each patient, four consecutive MRI examinations were conducted, including baseline MRI before treatment and three follow-up MRI examinations after treatment at each 0.5-year interval. ADC values at four b values and T1 relaxation times were correlated to pathology-confirmed liver fibrosis stages, which were subsequently divided into two groups, stages F2–3 and F4. The receiver operating characteristic(ROC) analysis and repeated measurement analysis of variance were used for statistical analysis. Results Among these ADC parameters, ADC value(b = 500 s/mm^2) was the most consistent in differentiating between stage F2–3 and F4 liver fibrosis. Repeated measurement analysis showed that the intra-group and inter-group differences were 0.447 and 0.024, respectively. T1 relaxation time could not consistently differentiate between the F2–3 and F4 groups; however, it was repeatable, and the intra-group and inter-group differences were 0.410 and 0.042, respectively. Conclusion MRI-ADC value at a b value of 500 s/mm^2 can be a promising biomarker for differentiating stages F2–3 and F4 liver fibrosis. A combination of this biomarker with repeatable T1 relaxation time may function as a non-invasive tool for follow-up liver fibrosis in patients who reject repeated image-guided biopsy. 展开更多

The effect of squeeze casting on microstructure and mechanical properties of hypereutectic Al-xS i alloys(x = 15, 17.5, 22 wt%) was investigated in this study. Results show that microstructure of the hypereutectic Al-x Si alloys was obviously improved by squeeze casting. The amount of coarse primary Si phase decreased, while that of fine primary α-Al dendrites increased with the increase of squeeze casting pressure. Due to the decrease of coarse primary Si particles, cracking of the matrix was reduced, whilst the fine microstructure, and mechanical properties of the squeeze casting alloys were improved. Compared with gravity casting alloys, mechanical properties of the hypereutectic Al-xS i alloys solidified at 600 MPa were improved significantly. Hardness of the squeeze casting hypereutectic Al-(15, 17.5, 22 wt%) Si alloys was improved by 15.91%, 12.23%, 17.48%, ultimate tensile strength was improved by 37.85%, 32.27%, 22.74%,and elongation was improved by 55.83%, 167.86%, 126.76%, respectively. Due to the uniform distribution of Si phases in squeeze casting Al-x Si alloys, their wear resistance was markedly enhanced. 展开更多

Ordered mesoporous Ba CO3/C composites were synthesized by a multi-component co-assembly method combined with a carbonization process using phenolic resol as carbon source, barium nitrate as barium precursor, and triblock copolymer Pluronic F127 as template. The synthesized materials were characterized by X-ray diffraction, transmission electron microscopy, N2 physical adsorption, thermogravimetric analysis, and temperature-programmed desorption of CO2. When Ba CO3 contents were increased from 9.1 wt% to 44.7 wt%, pore size increased from 3.1 to 4.3 nm and the BET(Brunauer-Emmett-Teller) surface area initially increased to a maximum value of 390 m2 g-1(at a Ba CO3 content of 18.5 wt%) before subsequently decreasing. Ba CO3 was well dispersed in the amorphous carbon framework, and no phase separation was observed. The mesoporous BaCO3/C composites exhibited high catalytic activities toward the transesterification of glycerol and dimethyl carbonate into glycerol carbonate. A glycerol conversion of 97.8% and a glycerol carbonate selectivity of 98.5% were obtained under the optimized reaction conditions. 展开更多

Genomic imprinting is an epigenetic modification of DNA,whereby gene expression is restricted to either maternally or paternally inherited alleles.Imprinted genes(IGs)in the placenta and embryo are essential for growth regulation and nutrient supply.However,despite being an important nutrition delivery organ,studies on mammary gland genomic imprinting remain limited.In this study,we found that both the number of IGs and their expression levels decreased during development of the mouse mammary gland.IG expression was lineage-specific and related to mammary gland development and lactation.Meta-analysis of single-cell RNA sequencing data revealed that mammary gland IGs were co-expressed in a network that regulated cell sternness and differentiation,which was confirmed by our functional studies.Accordingly,our data indicated that IGs were essential for the self-renewal of mammary gland stem cells and IG decline was correlated with mammary gland maturity.Taken together,our findings revealed the importance of IGs in a poorly studied nutrition-related organ,i.e.the mammary gland,thus providing a reference for further studies on genomic imprinting. 展开更多
