Questionnaires were conducted with the general information questionnaire, Connor-Davidson Resilience Scale (CD-RISC) and Work-Related Quality of Life Scale-2 (WRQoL-2). <strong>Results:</strong> The total average score of the quality of work life of ICU nurses was (3.35 ± 0.44), which was at a medium level. Department, professional title, frequency of night shift, experience of workplace violence and psychological resilience were the influencing factors of the quality of work life of nurses in ICU, accounting for 39.4% of the variance. <strong>Conclusion:</strong> The quality of work life of ICU nurses needs to be improved. Nursing managers should pay attention to the work environment of ICU nurses, rationally allocate manpower, and improve the status of psychological resilience of nurses, so as to improve their quality of work life.展开更多

To relieve the suffering,...<strong>Background:</strong> Patients who had total laryngectomy not only have a pressure from the possible diagnosis of laryngeal cancer, but also suffer from unspeakable misery. To relieve the suffering, some patients choose esophageal speech training. However, most of the current researches focus solely on the results of the training with the negligence of the patients’ mental health and their life experience during the training. A qualitative research is highly sensitive to patients’ subjective feelings, and thus able to capture their subtle emotional changes. <strong>Objective:</strong> To explore deeply patients’ real experiences of esophageal speech training after total laryngectomy and reveal the public their training experience of the unique and special group. <strong>Methods:</strong> In May 2018, researchers conducted a two-month field observation on and in-depth interviews with 15 patients who were on esophageal speech training after total laryngectomy in the esophageal speech training room of the Eye and ENT Hospital of Fudan University. The research applied phenomenological research method and Colaizzi Data Analysis. <strong>Results:</strong> The data analysis indicated four main themes: changes of mindset;showing positive attitudes and behaviors in training;shift in social relations;and arduous training process. <strong>Conclusion:</strong> The study has the finding that patients on esophageal speech training had negative emotions in the early stage of the training and dissatisfaction with its teaching. The repercussion of the surgery brought them changes of social relations, social bias and prejudice ensued. It is, therefore, urgent and crucial to give the speech trainees psychological counseling in the early stage to improve their qualities of life, to mobilize social support to incentivize their training, to optimize esophageal speech training teaching, and to regulate the management of the esophageal speech training environment.展开更多

Design: Search the Cochrane Library, ProQuest Academic Journal Library, PubMed, Web of Science, Ovid, Embase, CINAHL complete (EESCO) and other related databases to determine the literature. Screen out articles consistent with this review and summarize them. Results: Since the establishment of the database, a total of 1463 articles have been retrieved. After reading the title, abstract and full text, and excluding non-related and duplicate articles, 22 reports were finally included. Among them, there are 8 articles using different medication methods to compare the effects of establishing a CP model. The included articles explored the effects of different animal models, drug types, and their dose, concentration, speed, and time on the CP model. Conclusion: The factors of dose, concentration and time were positively correlated with the incidence of CP. The effect of speed factors on CP and the results of different animal models are inconsistent. It requires further research in the future.展开更多

Methods: Sixty-three patients with active LN and 15 systemic lupus erythe...Objective: To evaluate the clinical significance of high mobility group box 1 (HMGB1) expression in nephridial tissues of lupus nephritis (LN). Methods: Sixty-three patients with active LN and 15 systemic lupus erythematosus (SLE) (combined without LN) were included. Renal biopsies were performed in the two groups. The biopsies were evaluated according to the World Health Organization (WHO) classification and renal disease activity was estimated using the British Isles lupus assessment group (BILAG) index. Serum levels of HMGB1 were analyzed by western blot. HMGB1 expression in renal tissue was assessed by immunohistochemistry in the two groups. The correlation between HMGB1 and renal active index (AI), chronicity index (CI), pathological type of LN was analyzed. Results: LN biopsies showed WHO class III, IV or V and all patients had high renal disease activity (BILAG A/B). The HMGB1 expression was higher in the LN groups than the control groups (t = 9.263, P < 0.05). It showed positive correlation between HMGB1 expression and SLE DAI classification (r = 0.579, P P < 0.05) and renal tubule interstitial (TIL) classification (r = 0.815, P < 0.05), and negative correlation between HMGB1 expression and CI classification (r = 0.582, P < 0.05). In all patients, serum levels of HMGB1 increased only slightly in the patients only with SLE;however, in patients with LN WHO class IV a significant decrease was observed (P = 0.02). Immunostaining revealed a pronounced extranuclear HMGB1 expression predominantly outlining the glomerular endothelium and in the mesangium. There were significant differences in HMGB1 expression between LN and control biopsies and it existed with apparent association to histopathological classification and clinical outcome. Conclusions: Renal tissue expression and serum levels of HMGB1 were elevated in LN. The unusual elevation of HMGB1 in serum and tissue in LN may reflect persistent inflammatory activity, which clearly indicates a role for HMGB1 in pathogenesis of LN.展开更多

Electrocatalytic nitrate(NO_(3)^(−))reduction to ammonia(NH_(3))offers a viable approach for sustainable NH_(3)production and environmental denitrification.Copper(Cu)possesses a distinctive electronic structure,which ...Electrocatalytic nitrate(NO_(3)^(−))reduction to ammonia(NH_(3))offers a viable approach for sustainable NH_(3)production and environmental denitrification.Copper(Cu)possesses a distinctive electronic structure,which can augment the reaction kinetics of NO_(3)^(−)and impede hydrogen evolution reaction(HER),rendering it a promising contender for the electrosynthesis of NH_(3)from NO_(3)^(−).Nevertheless,the role of Cu_(2)O in copper-based catalysts still requires further investigation for a more comprehensive understanding.Herein,the Cu_(2)O/Cu(OH)_(2)heterostructures are successfully fabricated through liquid laser irradiation using CuO nanoparticles as a precursor.Experimental and theoretical researches reveal that Cu_(2)O/Cu(OH)_(2)heterostructure exhibits enhanced electrocatalytic performance for NO_(3)^(−)to NH_(3)because Cu(OH)_(2)promotes electron transfer and reduces the valence state of Cu active site in Cu_(2)O.At−0.6 V(vs.reversible hydrogen electrode(RHE)),the NH_(3)yield reaches its maximum at 1630.66±29.72μg·h^(−1)·mgcat^(−1),while the maximum of Faraday efficiency(FE)is 76.95%±5.51%.This study expands the technical scope of copper-based catalyst preparation and enhances the understanding of the electrocatalytic mechanism of NO_(3)^(−)to NH_(3).展开更多
Background:The presence of fibrosis is a criterion for subtype classification in the newly updated hypersensitivity pneumonitis(HP)guidelines.The present study aimed to summarize differences in clinical characteristic...Background:The presence of fibrosis is a criterion for subtype classification in the newly updated hypersensitivity pneumonitis(HP)guidelines.The present study aimed to summarize differences in clinical characteristics and prognosis of non-fibrotic hypersensitivity pneumonitis(NFHP)and fibrotic hypersensitivity pneumonitis(FHP)and explore factors associated with the presence of fibrosis.Methods:In this prospective cohort study,patients diagnosed with HP through a multidisciplinary discussion were enrolled.Collected data included demographic and clinical characteristics,laboratory findings,and radiologic and histopathological features.Logistic regression analyses were performed to explore factors related to the presence of fibrosis.Results:A total of 202 patients with HP were enrolled,including 87(43.1%)NFHP patients and 115(56.9%)FHP patients.Patients with FHP were older and more frequently presented with dyspnea,crackles,and digital clubbing than patients with NFHP.Serum levels of carcinoembryonic antigen,carbohydrate antigen 125,carbohydrate antigen 153,gastrin-releasing peptide precursor,squamous cell carcinoma antigen,and antigen cytokeratin 21-1,and count of bronchoalveolar lavage(BAL)eosinophils were higher in the FHP group than in the NFHP group.BAL lymphocytosis was present in both groups,but less pronounced in the FHP group.Multivariable regression analyses revealed that older age,<20%of lymphocyte in BAL,and≥1.75%of eosinophil in BAL were risk factors for the development of FHP.Twelve patients developed adverse outcomes,with a median survival time of 12.5 months,all of whom had FHP.Conclusions:Older age,<20%of lymphocyte in BAL,and≥1.75%of eosinophil in BAL were risk factors associated with the development of FHP.Prognosis of patients with NFHP was better than that of patients with FHP.These results may provide insights into the mechanisms of fibrosis in HP.展开更多

In this paper,we investigate a two-dimensional avian influenza model with Allee effect and stochasticity.We first show that a unique global positive solution always exists to the stochastic system for any positive ini...In this paper,we investigate a two-dimensional avian influenza model with Allee effect and stochasticity.We first show that a unique global positive solution always exists to the stochastic system for any positive initial value.Then,under certain conditions,this solution is proved to be stochastically ultimately bounded.Furthermore,by constructing a suitable Lyapunov function,we obtain sufficient conditions for the existence of stationary distribution with ergodicity.The conditions for the extinction of infected avian population are also analytically studied.These theoretical results are conformed by computational simulations.We numerically show that the environmental noise can bring different dynamical outcomes to the stochastic model.By scanning different noise intensities,we observe that large noise can cause extinction of infected avian population,which suggests the repression of noise on the spread of avian virus.展开更多

Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability improvement.We obtained a novel anti-human programmed death 1(PD-1)antibody...Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability improvement.We obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology platform.We used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to PD-1.Then,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library construction.We identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key residues.The virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin G.Then,we used flow cytometry and sequencing approaches to sort and screen the candidates.Finally,we obtained a novel anti-human PD-1 antibody named FV78.FV78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to MIL75.Our results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy.展开更多
Objective:To explore the clinical role of contrast-enhanced ultrasound(CEUS)in a preoperative evaluation of early endometrial carcinoma.Methods:This study included 14 patients who underwent preoperative CEUS and contr...Objective:To explore the clinical role of contrast-enhanced ultrasound(CEUS)in a preoperative evaluation of early endometrial carcinoma.Methods:This study included 14 patients who underwent preoperative CEUS and contrast-enhanced magnetic resonance imaging(CEMRI)and were ultimately diagnosed with stage I endometrial carcinoma from December 2019 to December 2020.The parameters of the time-intensity curve(TIC)were compared with the endometrial carcinoma group,the invaded myometrium group,and the normal myometrium group.Results:TIC parameters between the endometrial carcinoma group and the invaded myometrium group were similar.Compared with the normal myometrium group,the time to peak(TTP)was significantly shorter and the ascending slope(AS)was significantly higher in the endometrial carcinoma group.The TTP of the invaded myometrium group was shorter than that of the normal myometrium group and the peak intensity(PI)was higher than that of the normal myometrium group.We then compared the TIC parameters between the endometrial carcinoma group and the invaded myometrium group after adjusting for the normal myometrium group,and the results still did not show any difference.Of the 14 cases of endometrial carcinoma,9 cases were diagnosed by CEMRI and were consistent with the pathology results,1 case was underestimated,and 4 cases were overestimated;while 11 cases diagnosed were diagnosed by CEUS and were consistent with the pathology results,1 case was underestimated,and 2 cases were overestimated.Conclusions:The contrast pulse sequencing technique used in the CEUS examinations performed well in evaluating the extent of the endometrial carcinoma.Future studies with larger sample sizes are needed to determine the applicability and value of this new procedure during preoperative assessments of early endometrial carcinoma.展开更多
Metal-free heteroatoms dual-doped carbon has been recognized as one of the most promising Pt/C-substitutes for oxygen reduction reaction(ORR).Herein,we optimize the preparation process by doping order of metal-free he...Metal-free heteroatoms dual-doped carbon has been recognized as one of the most promising Pt/C-substitutes for oxygen reduction reaction(ORR).Herein,we optimize the preparation process by doping order of metal-free heteroatoms to obtain the best electrocatalytic performance through three types of dual-doped carbon,including XC-N(first X doping then N doping),NC-X(first N doping then X doping) and NXC(N and X doping)(X=P,S and F).XC-N has more defect than the other two indicated by Raman spectra.X-ray photoelectron spectrom(XPS) measurements indicate that N and X have been dual-doped into the carbon matrix with different doping contents and modes,Electrocatalytic results,including the potential of ORR peak(Ep),the half-wave potential,the diffusion-limiting current density mainly follows the order of XC-N>NC-X> NXC,Furthermore,the synergistic effect of second atom doping are also compared with the single doped carbon(NC,PC,SC and FC).The differences in electronegativity and atomic radius of these metal-free heteroatoms can affect the defect degree,the doping content and mode of hete roatoms on carbon matrix,induce polarization effect and space effect to affect O2 adsorption and product desorption,ultimately to the ORR electrocatalytic performance.展开更多

To relieve the suffering, some patients choose esophageal speech training. However, most of the current researches focus solely on the results of the training with the negligence of the patients’ mental health and their life experience during the training. A qualitative research is highly sensitive to patients’ subjective feelings, and thus able to capture their subtle emotional changes. <strong>Objective:</strong> To explore deeply patients’ real experiences of esophageal speech training after total laryngectomy and reveal the public their training experience of the unique and special group. <strong>Methods:</strong> In May 2018, researchers conducted a two-month field observation on and in-depth interviews with 15 patients who were on esophageal speech training after total laryngectomy in the esophageal speech training room of the Eye and ENT Hospital of Fudan University. The research applied phenomenological research method and Colaizzi Data Analysis. <strong>Results:</strong> The data analysis indicated four main themes: changes of mindset;showing positive attitudes and behaviors in training;shift in social relations;and arduous training process. <strong>Conclusion:</strong> The study has the finding that patients on esophageal speech training had negative emotions in the early stage of the training and dissatisfaction with its teaching. The repercussion of the surgery brought them changes of social relations, social bias and prejudice ensued. It is, therefore, urgent and crucial to give the speech trainees psychological counseling in the early stage to improve their qualities of life, to mobilize social support to incentivize their training, to optimize esophageal speech training teaching, and to regulate the management of the esophageal speech training environment.
文摘Objective: This review aims to determine the impact of different drugs and methods on the successful establishment of an animal model for chemical phlebitis (CP). Design: Search the Cochrane Library, ProQuest Academic Journal Library, PubMed, Web of Science, Ovid, Embase, CINAHL complete (EESCO) and other related databases to determine the literature. Screen out articles consistent with this review and summarize them. Results: Since the establishment of the database, a total of 1463 articles have been retrieved. After reading the title, abstract and full text, and excluding non-related and duplicate articles, 22 reports were finally included. Among them, there are 8 articles using different medication methods to compare the effects of establishing a CP model. The included articles explored the effects of different animal models, drug types, and their dose, concentration, speed, and time on the CP model. Conclusion: The factors of dose, concentration and time were positively correlated with the incidence of CP. The effect of speed factors on CP and the results of different animal models are inconsistent. It requires further research in the future.

文摘Objective: To evaluate the clinical significance of high mobility group box 1 (HMGB1) expression in nephridial tissues of lupus nephritis (LN). Methods: Sixty-three patients with active LN and 15 systemic lupus erythematosus (SLE) (combined without LN) were included. Renal biopsies were performed in the two groups. The biopsies were evaluated according to the World Health Organization (WHO) classification and renal disease activity was estimated using the British Isles lupus assessment group (BILAG) index. Serum levels of HMGB1 were analyzed by western blot. HMGB1 expression in renal tissue was assessed by immunohistochemistry in the two groups. The correlation between HMGB1 and renal active index (AI), chronicity index (CI), pathological type of LN was analyzed. Results: LN biopsies showed WHO class III, IV or V and all patients had high renal disease activity (BILAG A/B). The HMGB1 expression was higher in the LN groups than the control groups (t = 9.263, P < 0.05). It showed positive correlation between HMGB1 expression and SLE DAI classification (r = 0.579, P P < 0.05) and renal tubule interstitial (TIL) classification (r = 0.815, P < 0.05), and negative correlation between HMGB1 expression and CI classification (r = 0.582, P < 0.05). In all patients, serum levels of HMGB1 increased only slightly in the patients only with SLE;however, in patients with LN WHO class IV a significant decrease was observed (P = 0.02). Immunostaining revealed a pronounced extranuclear HMGB1 expression predominantly outlining the glomerular endothelium and in the mesangium. There were significant differences in HMGB1 expression between LN and control biopsies and it existed with apparent association to histopathological classification and clinical outcome. Conclusions: Renal tissue expression and serum levels of HMGB1 were elevated in LN. The unusual elevation of HMGB1 in serum and tissue in LN may reflect persistent inflammatory activity, which clearly indicates a role for HMGB1 in pathogenesis of LN.
文摘The paper presents a description of the experience of the nursing for the Pseudomonas aeruginosa pneumonia patient with partial laryngectomy. The nursing of Pseudomonas aeruginosa pneumonia, conscious disturbance, electrolyte disorder, affection supports and mental nursing was provided in the process of the postoperative treatment. Our experience may provide useful insight for the nursing of laryngectomy patients secondary to pneumonia with conscious disturbance. A deeper knowledge about improving the quality of clinic special nursing for the partial or total laryngectomy patient is necessary, with the development of modern medical technologies and medicine.

文摘Electrocatalytic nitrate(NO_(3)^(−))reduction to ammonia(NH_(3))offers a viable approach for sustainable NH_(3)production and environmental denitrification.Copper(Cu)possesses a distinctive electronic structure,which can augment the reaction kinetics of NO_(3)^(−)and impede hydrogen evolution reaction(HER),rendering it a promising contender for the electrosynthesis of NH_(3)from NO_(3)^(−).Nevertheless,the role of Cu_(2)O in copper-based catalysts still requires further investigation for a more comprehensive understanding.Herein,the Cu_(2)O/Cu(OH)_(2)heterostructures are successfully fabricated through liquid laser irradiation using CuO nanoparticles as a precursor.Experimental and theoretical researches reveal that Cu_(2)O/Cu(OH)_(2)heterostructure exhibits enhanced electrocatalytic performance for NO_(3)^(−)to NH_(3)because Cu(OH)_(2)promotes electron transfer and reduces the valence state of Cu active site in Cu_(2)O.At−0.6 V(vs.reversible hydrogen electrode(RHE)),the NH_(3)yield reaches its maximum at 1630.66±29.72μg·h^(−1)·mgcat^(−1),while the maximum of Faraday efficiency(FE)is 76.95%±5.51%.This study expands the technical scope of copper-based catalyst preparation and enhances the understanding of the electrocatalytic mechanism of NO_(3)^(−)to NH_(3).

文摘Background:The presence of fibrosis is a criterion for subtype classification in the newly updated hypersensitivity pneumonitis(HP)guidelines.The present study aimed to summarize differences in clinical characteristics and prognosis of non-fibrotic hypersensitivity pneumonitis(NFHP)and fibrotic hypersensitivity pneumonitis(FHP)and explore factors associated with the presence of fibrosis.Methods:In this prospective cohort study,patients diagnosed with HP through a multidisciplinary discussion were enrolled.Collected data included demographic and clinical characteristics,laboratory findings,and radiologic and histopathological features.Logistic regression analyses were performed to explore factors related to the presence of fibrosis.Results:A total of 202 patients with HP were enrolled,including 87(43.1%)NFHP patients and 115(56.9%)FHP patients.Patients with FHP were older and more frequently presented with dyspnea,crackles,and digital clubbing than patients with NFHP.Serum levels of carcinoembryonic antigen,carbohydrate antigen 125,carbohydrate antigen 153,gastrin-releasing peptide precursor,squamous cell carcinoma antigen,and antigen cytokeratin 21-1,and count of bronchoalveolar lavage(BAL)eosinophils were higher in the FHP group than in the NFHP group.BAL lymphocytosis was present in both groups,but less pronounced in the FHP group.Multivariable regression analyses revealed that older age,<20%of lymphocyte in BAL,and≥1.75%of eosinophil in BAL were risk factors for the development of FHP.Twelve patients developed adverse outcomes,with a median survival time of 12.5 months,all of whom had FHP.Conclusions:Older age,<20%of lymphocyte in BAL,and≥1.75%of eosinophil in BAL were risk factors associated with the development of FHP.Prognosis of patients with NFHP was better than that of patients with FHP.These results may provide insights into the mechanisms of fibrosis in HP.

文摘Research on energy storage technology is a vital part of realizing the dual-carbon strategy at this stage.Aqueous zinc-ion batteries(AZIBs)are favorable competitors in various energy storage devices due to their high energy density,reassuring intrinsic safety,and unique cost advantages.The design of cathode materials is crucial for the large-scale development and application of AZIBs.Vanadium-based oxides with high theoretical capacity,diverse valence states,as well as high electrochemical activity,have been widely used as cathode materials for AZIBs.Unfortunately,there are some obstacles,including low electronic conductivity and sluggish kinetics,hindering their further application in AZIBs.In view of the above,this review will introduce a series of modification methods including morphology design,defect engineering,ingenious combination with conductive materials,and modification of electrolyte and zinc anode according to the intrinsic disadvantage of vanadium oxides and summarize the research progress of various modification methods including zinc storage performance and mechanism.Finally,several reasonable prospects will be proposed to appease the needs of basic research and practical applications according to the current status.

文摘In this paper,we investigate a two-dimensional avian influenza model with Allee effect and stochasticity.We first show that a unique global positive solution always exists to the stochastic system for any positive initial value.Then,under certain conditions,this solution is proved to be stochastically ultimately bounded.Furthermore,by constructing a suitable Lyapunov function,we obtain sufficient conditions for the existence of stationary distribution with ergodicity.The conditions for the extinction of infected avian population are also analytically studied.These theoretical results are conformed by computational simulations.We numerically show that the environmental noise can bring different dynamical outcomes to the stochastic model.By scanning different noise intensities,we observe that large noise can cause extinction of infected avian population,which suggests the repression of noise on the spread of avian virus.

文摘Using the axially deformed relativistic Hartree-Fock-Bogoliubov(D-RHFB)model,we explore the mechanism behind the parity inversion and halo occurrence in^(11)Be,which are well reproduced by the RHF Lagrangian PKA1.It is illustrated that evidently enhanced deformation effects by theπ-pseudo-vector andρ-tensor couplings in PKA1 are crucial for correctly describing both the even-parity ground state(GS)and the neutron halo of^(11)Be.Coupling with the deformation,the intrude 1d_(5/2)component largely enhances the couplings between the evenparity orbit 1/2_(2)^(+)and the nuclear core to ensure an even-parity GS,whereas the 2s_(1/2)component therein dominates the halo formation in^(11)Be.Moreover,the deformed halo in^(11)Be is found to be stabilized by the attractive inherent correlations between the 1d_(5/2)and 2s_(1/2)components of the halo orbit 1/2_(2)^(+),instead of pairing correlations,which paves a new way for understanding the halo pictures in deformed unstable nuclei.

文摘The Hippo signaling pathway has been established as a key regulator of organ size control,tumor suppression,and tissue regeneration in multiple organisms.Recently,emerging evidence has indicated that Hippo signaling might play an important role in regulating the immune system in both Drosophila and mammals.In particular,patients bearing a loss-of-function mutation of MST1 are reported to have an autosomal recessive primary immunodeficiency syndrome.MST1/2 kinases,the mammalian orthologs of Drosophila Hippo,may activate the non-canonical Hippo signaling pathway via MOB1A/B and/or NDR1/2 or cross-talk with other essential signaling pathways to regulate both innate and adaptive immunity.In this review,we present and discuss recent findings of cellular mechanisms/functions of Hippo signaling in the innate immunity in Drosophila and in mammals,T cell immunity,as well as the implications of Hippo signaling for tumor immunity.

文摘Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability improvement.We obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology platform.We used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to PD-1.Then,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library construction.We identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key residues.The virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin G.Then,we used flow cytometry and sequencing approaches to sort and screen the candidates.Finally,we obtained a novel anti-human PD-1 antibody named FV78.FV78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to MIL75.Our results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy.
文摘Objective:To explore the clinical role of contrast-enhanced ultrasound(CEUS)in a preoperative evaluation of early endometrial carcinoma.Methods:This study included 14 patients who underwent preoperative CEUS and contrast-enhanced magnetic resonance imaging(CEMRI)and were ultimately diagnosed with stage I endometrial carcinoma from December 2019 to December 2020.The parameters of the time-intensity curve(TIC)were compared with the endometrial carcinoma group,the invaded myometrium group,and the normal myometrium group.Results:TIC parameters between the endometrial carcinoma group and the invaded myometrium group were similar.Compared with the normal myometrium group,the time to peak(TTP)was significantly shorter and the ascending slope(AS)was significantly higher in the endometrial carcinoma group.The TTP of the invaded myometrium group was shorter than that of the normal myometrium group and the peak intensity(PI)was higher than that of the normal myometrium group.We then compared the TIC parameters between the endometrial carcinoma group and the invaded myometrium group after adjusting for the normal myometrium group,and the results still did not show any difference.Of the 14 cases of endometrial carcinoma,9 cases were diagnosed by CEMRI and were consistent with the pathology results,1 case was underestimated,and 4 cases were overestimated;while 11 cases diagnosed were diagnosed by CEUS and were consistent with the pathology results,1 case was underestimated,and 2 cases were overestimated.Conclusions:The contrast pulse sequencing technique used in the CEUS examinations performed well in evaluating the extent of the endometrial carcinoma.Future studies with larger sample sizes are needed to determine the applicability and value of this new procedure during preoperative assessments of early endometrial carcinoma.

文摘Metal-free heteroatoms dual-doped carbon has been recognized as one of the most promising Pt/C-substitutes for oxygen reduction reaction(ORR).Herein,we optimize the preparation process by doping order of metal-free heteroatoms to obtain the best electrocatalytic performance through three types of dual-doped carbon,including XC-N(first X doping then N doping),NC-X(first N doping then X doping) and NXC(N and X doping)(X=P,S and F).XC-N has more defect than the other two indicated by Raman spectra.X-ray photoelectron spectrom(XPS) measurements indicate that N and X have been dual-doped into the carbon matrix with different doping contents and modes,Electrocatalytic results,including the potential of ORR peak(Ep),the half-wave potential,the diffusion-limiting current density mainly follows the order of XC-N>NC-X> NXC,Furthermore,the synergistic effect of second atom doping are also compared with the single doped carbon(NC,PC,SC and FC).The differences in electronegativity and atomic radius of these metal-free heteroatoms can affect the defect degree,the doping content and mode of hete roatoms on carbon matrix,induce polarization effect and space effect to affect O2 adsorption and product desorption,ultimately to the ORR electrocatalytic performance.

文摘Radiometric normalization,as an essential step for multi-source and multi-temporal data processing,has received critical attention.Relative Radiometric Normalization(RRN)method has been primarily used for eliminating the radiometric inconsistency.The radiometric trans-forming relation between the subject image and the reference image is an essential aspect of RRN.Aimed at accurate radiometric transforming relation modeling,the learning-based nonlinear regression method,Support Vector machine Regression(SVR)is used for fitting the complicated radiometric transforming relation for the coarse-resolution data-referenced RRN.To evaluate the effectiveness of the proposed method,a series of experiments are performed,including two synthetic data experiments and one real data experiment.And the proposed method is compared with other methods that use linear regression,Artificial Neural Network(ANN)or Random Forest(RF)for radiometric transforming relation modeling.The results show that the proposed method performs well on fitting the radiometric transforming relation and could enhance the RRN performance.
