In order to utilize mulberry branches efficiently and reduce environment pollution, a strain named A1 was isolated from the intestinal tract of Acr/da Chinensis using Congo red staining method on the plate of carboxymethyl cellulose sodium (CMC-Na) medium and mulberry branch powder culture medium. The strain A1 was mutagenized by uhraviolet radiation and the strain Z3 with a significantly improved specific activity of filter paper enzyme was screened. Enzymatic characteristics, degradation ability of cellulose and taxonomic status of A1 and Z3 were also studied in this paper. The results showed that filter paper enzyme specific activity of Z3 increased 74.47% than that of A1 and the property of enzyme production was steady. The optimum pH values of filter paper enzyme produced by A1 and Z3 were 4. 6 and 3.8 respectively and the optimum temperatures were 45 and 50 ℃, respectively. When CMC-Na was used as substrate, Vmax of the two strains was similar while Km of Z3 was lower than that of A1 , which indicated that Z3 had a strong affinity for the substrate. Enzymes produced by Z3 were more resistant to high temperature. Strains A1 and Z3 could grow well on the culture mediums with mulberry branch powder as carbon source, but Z3 grew better than A1. A1 and Z3 were preliminarily identified as AspergiUus according to the morphological identification. 展开更多

Background: Production of chimeric mice is a useful tool for the elucidation of gene function. After successful isolation of embryonic stem (ES) cell lines, there are many methods for producing chimeras, including co-culture with the embryos, microinjection of the ES ceils into pre-implantation embryos, and use of tetraploid embryos to generate the full ES-derived transgenic mice. Here, we aimed to generate the transgenic ES cell line, compare the production efficiency of chimeric mice and its proportion to yield the male chimeric mice by microinjected ES cells into 4- to 8-cell and blastocysts embryos with the application of Piezo-Micromanipulator (PMM), and trace the fate of the injected ES cells. Results: We successfully generated a transgenic ES cell line and proved that this cell line still maintained pluripotency. Although we achieved a satisfactory chimeric mice rate, there was no significant difference in the production of chimeric mice using the two different methods, but the proportion of the male chimeric mice in the 4- to 8-cell group was higher than in the blastocyst group. We also found that there was no tendency for ES cells to aggregate into the inner cell mass using in vitro culture of the chimeric embryos, indicating that they aggregated randomly. Conclusions: These results showed that the PMM method is a convenient way to generate chimeric mice and microinjection of ES cells into 4- to 8-cell embryos can increase the chance of yielding male chimeras compared to the blastocyst injection. These results provide useful data in transgenic research mediated by ES cells. 展开更多

Diphtheria toxin A (DTA), a segment of the diphtheria toxin (tox), inhibits protein synthesis in cells. When released from a cell, DTA is nontoxic and cannot enter other cells independently without the help of diphtheria toxin B. In this study, we artificially synthesized the DTA gene sequence and cloned it into pEGFP-N1 to generate the recombinant vector pEGFP-N1-DTA. This recombinant vector was then transfected into 293T cells to observe the effect of DTA protein expression on enhanced green fluorescent protein (EGFP) protein expression and the proliferation of 293T cells. After 48 h, high levels of EGFP expression were seen in control pEGFP-Nl-transfected cells, whereas very low levels were seen in cells transfected with pEGFP-N1-DTA. Reverse transcription polymerase chain reaction confirmed the expression of the DTA gene in cells transfected with pEGFP-N^-DTA. Further, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (M-I-I-) assay revealed a significant difference in cell proliferation between the control group and the pEGFP-N1-DTA-transfected group. Using the expression of EGFP expression as an indicator, this study revealed that DTA expression can inhibit intracellular protein synthesis and cell proliferation. 展开更多

[Results] Eucalyptol had good control effect on E. pirisuga , with slow effect but long duration. It was harmless to natural enemies in tea gardens, with high safety and no chemical injury. The XRD and HRTEM results show that YAG crystal structure has little change when Eu^(3+), Ce^(3+) are doped. Absorption spectra and photoluminescence spectra show that the Ce,Eu:YAG single crystal can effectively absorb the 460 nm blue light to form a broadband emission center at 530 nm, decay curves of the crystal show that the energy transfer between Ce^(3+) and Eu^(3+) is highly suppressed. Compared with commercial Ce:YAG phosphor, Ce,Eu:YAG exhibits better thermal stability. 展开更多

A simple biomolecule-assisted synthetic route has been successfully developed to prepare bismuth sulfide(Bi 2 S 3 ) nanorods under solvothermal conditions.In the synthetic system,pentahydrate bismuth nitrate was employed to supply Bi source and L-cystine was used as sulfide source and complexing agent.The morphology,structure,and phase composition of the as-prepared Bi 2 S 3 products were characterized by X-ray diffraction(XRD),energy dispersion spectroscopy(EDS),X-ray photoelectron spectroscopy(XPS),field-emission scanning electron microscopy(FESEM),transmission electron microscopy(TEM),selected area electron diffraction(SAED),and high-resolution transmission electron microscopy(HRTEM).The experimental results show that the nanorods have uniform diameter of 100-200 nm and length of 2-4 μm.The possible formation mechanism for the bismuth sulfide nanorods was discussed. 展开更多

In general, MCI beams are largely produced by electron beam ion source (EBIS) [1], laser ion source (LIS) [2], or high-frequency (mostly >5 GHz) electron cyclotron resonance (ECR) ion source [3]. Among these, only ECR ion source can operate in the continuous wave (CW) mode, while EBIS and LIS only support pulses. In addition, ECR ion source with lower frequency (mostly 2.45 GHz) are required primarily for generating single charge state ions, because the corresponding ECR field (875 Gs) is not sufficiently strong for MCI generation [4]. 展开更多

A quartz-chamber 2.45 GHz electron cyclotron resonance ion source(ECRIS) was designed for diagnostic purposes at Peking University [Patent Number: ZL 201110026605.4]. This ion source can produce a maximum 84 m A hydrogen ion beam at 50 k V with a duty factor of 10%. The root-mean-square(RMS) emittance of this beam is less than 0.12π mm mrad. In our initial work,the electron temperature and electron density inside the plasma chamber had been measured with the line intensity ratio of noble gases. Based on these results, the atomic and molecular emission spectra of hydrogen were applied to determine the dissociation degree of hydrogen and the vibrational temperature of hydrogen molecules in the ground state, respectively. Measurements were performed at gas pressures from 4×10^(-4) to 1×10^(-3) Pa and at input peak RF power ranging from 1000 to 1800 W. The dissociation degree of hydrogen in the range of 0.5%-10% and the vibrational temperature of hydrogen molecules in the ground state in the range of 3500-8500 K were obtained. The plasma processes inside this ECRIS chamber were discussed based on these results. 展开更多
