Chiroptical switches based on circularly polarized luminescence(CPL)have shown the promising applications in advanced information technologies.Herein,a pair of lanthanide coordination polymer enantiomers[Eu_(2)(L^(R))_(3)(BTFPO)_(2)]_(n) and[Eu_(2)(L^(S))_(3)(BTFPO)_(2)]_(n) with light-regulated CPL property are designed,which are assembled by a chiral binuclear triple-stranded Eu^(3+) helicates[Eu_(2)(L^(R/S))_(3)]coordinated with two photochromic triphenylphosphine oxides(BTFPO).Upon the alternative UV and 526 nm light irradiation,the complexes show the reversible photochromism,PL and CPL responses.Notably,the luminescence dissymmetry factor,g lum of ^(5)D_(0)→^(7)F_(1)(591 nm)transition shows an obvious increase from 0.19 to 0.29 before and after 275 nm light irradiation.Additionally,the emission from Eu^(3+) center is not completely quenched in closed-ring state due to the low photocyclization(Ф_(o-c))quantum yield of the polymer.The partial maintenance of emissive intensity is of essential importance for the monitor of CPL signal.More importantly,the CPL photo-switching property of the complexes in solid hybrid film is maintained,and still displays the enhanced CPL emission in photostationary state.Further,the potential applications of the doping film in logic gate and anti-counterfeiting were investigated. 展开更多

We examine whether the effectiveness of the monetary policy rate transmission differs before and after interest rate liberalization in China using the autoregressive distributed lag (ARDL) bound test and an error correction model (ECM). The results show that after liberalization the mark-up is lower, and both the Iong-run and shortrun interest rate pass-through has become faster and more complete. We attribute our findings to the ongoing reforms of China's banking system, which has improved the competitiveness of Chinese commercial banks. 展开更多

Hepatic cholesterol accumulation is an important contributor to hypercholesterolemia,which results in atherosclerosis and cardiovascular disease(CVD).ATP-citrate lyase(ACLY)is a key lipogenic enzyme that converts cytosolic citrate derived from tricarboxylic acid cycle(TCA cycle)to acetyl-CoA in the cytoplasm.Therefore,ACLY represents a link between mitochondria oxidative phosphorylation and cytosolic de novo lipogenesis.In this study,we developed the small molecule 326E with an enedioic acid structural moiety as a novel ACLY inhibitor,and its CoA-conjugated form 326E-CoA inhibited ACLY activity with an IC_(50)=5.31±1.2μmol/L in vitro.326E treatment reduced de novo lipogenesis,and increased cholesterol efflux in vitro and in vivo.326E was rapidly absorbed after oral administration,exhibited a higher blood exposure than that of the approved ACLY inhibitor bempedoic acid(BA)used for hypercholesterolemia.Chronic 326E treatment in hamsters and rhesus monkeys resulted in remarkable improvement of hyperlipidemia.Once daily oral administration of 326E for 24 weeks prevented the occurrence of atherosclerosis in ApoE^(-/-)mice to a greater extent than that of BA treatment.Taken together,our data suggest that inhibition of ACLY by 326E represents a promising strategy for the treatment of hypercholesterolemia. 展开更多

文中以大肠杆菌BL21(DE3)为宿主,构建两株分别共表达亮氨酸脱氢酶(LDH,来源蜡样芽孢杆菌)/甲酸脱氢酶(FDH,来源水生弯杆菌)和亮氨酸脱氢酶(LDH,来源蜡样芽孢杆菌)/醇脱氢酶(ADH,来源红球菌)的重组大肠杆菌。通过偶联两种不同NADH再生体... 文中以大肠杆菌BL21(DE3)为宿主,构建两株分别共表达亮氨酸脱氢酶(LDH,来源蜡样芽孢杆菌)/甲酸脱氢酶(FDH,来源水生弯杆菌)和亮氨酸脱氢酶(LDH,来源蜡样芽孢杆菌)/醇脱氢酶(ADH,来源红球菌)的重组大肠杆菌。通过偶联两种不同NADH再生体系,以L-苏氨酸为起始原料,利用苏氨酸脱氨酶(L-TD)与LDH-FDH或LDH-ADH一锅法合成L-2-氨基丁酸,并对LDH-FDH工艺和LDH-ADH工艺进行对比优化。LDH-FDH工艺的最适反应pH为7.5,最适反应温度为35℃,通过加入50 g/L甲酸铵、0.3 g/L NAD^+、10%LDH-FDH粗酶液(V/V)和7500 U/L的L-TD酶液,对L-苏氨酸进行分批补加,以便控制2-丁酮酸浓度小于15 g/L,反应28 h,实现了L-2-氨基丁酸的产量为161.8 g/L,产率97%。LDH-ADH工艺的最适pH为8.0,最适反应温度为35℃,通过加入0.3 g/L NAD^+、10%LDH-ADH粗酶液(V/V)及7500 U/L的L-TD酶液,分批补加L-苏氨酸及1.2倍摩尔量异丙醇,以便控制2-丁酮酸浓度小于15 g/L,且每生成约40 g/L的L-2-氨基丁酸,抽真空去除丙酮,反应24 h,实现了L-2-氨基丁酸的产量为119.6 g/L,产率98%。文中所采用的工艺及结果可为L-2-氨基丁酸的工业化提供一定的参考依据。 展开更多

Inner ear hair cells are important for maintaining hearing.Irreversible damage to hair cells is an important cause of sensorineural deafness.Electromagnetic radiation,especially high-power microwave,is an important threat to human health in modern society and war.However,it is not clear whether high-power microwave has an effect on cochlea hair cells.This study aimed to assess the effects of high-power microwave on cochlear hair cells in guinea pigs,and investigate the potential protection of these cells against high-power microwave-induced damage by recombinant adenovirus A20.Based on experimental results,a 65 W/cm^(2) irradiation density applied to guinea pigs in this study to establish a high-power microwave inner ear injury model.In addition,pAdEeay-1/A20 was injected via a round window into experimental guinea pig cochlea,whereas artificial perilymph was injected into the control group.Auditory function was assessed by testing the auditory brainstem response threshold,and damage to cochlear hair cells was investigated by cell counting and scanning electron microscopy observations of the basilar membrane.Inner ear injury was observed 6 hours after 65 W/cm^( 2 ) of irradiation and the auditory brainstem response threshold was significantly higher in the irradiation group(P<0.05)compared with other groups.Propidium iodide staining and scanning electron microscopy results indicated that significant morphological changes occurred after radiation,especially to inner hair cells,which exhibited remarkable damage and the presence of several unknown spherical substances.Auditory brainstem response threshold was decreased in the pAdEeay-1/A20 group compared with the artificial perilymph group;moreover,damage to hair cells was milder in the pAdEeay-1/A20 group compared with the control group(P<0.01).Thus,high-power microwave can cause damage to cochlear hair cells,as well as hearing loss with prolonged exposure and/or high dosage.In this regard,65 W/cm^( 2 ) of irradiation for 6 hours is a reliable target dose for observation of damage.The zinc finger protein A20 can protect cochlear hair cells from high-power microwave-induced damage and prevent further hearing loss.This study was approved by the Laboratory Animal Welfare and Ethics Committee of the Third Military Medical University,China on April 18,2017. 展开更多
