支链氨基酸(branched-chainaminoacids,BCAA)可通过蛋鸡肝脏的脂肪酸代谢作用调控卵黄脂蛋白的产生,因而日粮中BCAA的含量以及不同BCAA之间的配比对蛋鸡拥有理想的产蛋性能极为重要。然而,文献报道的蛋鸡最适支链氨基酸的需要量和不同B... 支链氨基酸(branched-chainaminoacids,BCAA)可通过蛋鸡肝脏的脂肪酸代谢作用调控卵黄脂蛋白的产生,因而日粮中BCAA的含量以及不同BCAA之间的配比对蛋鸡拥有理想的产蛋性能极为重要。然而,文献报道的蛋鸡最适支链氨基酸的需要量和不同BCAA之间的合适配比并不统一。综合各类研究发现,日粮中异亮氨酸和缬氨酸+亮氨酸的含量对蛋鸡生产性能的影响存在互作效应,当缬氨酸+亮氨酸的浓度低时,增加异亮氨酸的含量可以降低蛋鸡的蛋重和增重;而当缬氨酸+亮氨酸的浓度高时,高水平的异亮氨酸只能使蛋鸡的生产性能出现略微的下降。另外,日粮中异亮氨酸的含量在一定水平上可以使蛋鸡获得最大的氮沉积量,超过一定的浓度范围会降低蛋鸡的体重,超过幅度更大时会降低蛋鸡的产蛋量。 展开更多

支链氨基酸(branched-chain amino acids,BCAA;主要为亮氨酸、异亮氨酸和缬氨酸)除了维持动物机体能量的稳态和为转氨基作用提供氮底物和碳框架外,还是蛋白激酶B(protein kinase B,AKT)调节葡萄糖、脂质和蛋白质合成的信号分子,也是蛋... 支链氨基酸(branched-chain amino acids,BCAA;主要为亮氨酸、异亮氨酸和缬氨酸)除了维持动物机体能量的稳态和为转氨基作用提供氮底物和碳框架外,还是蛋白激酶B(protein kinase B,AKT)调节葡萄糖、脂质和蛋白质合成的信号分子,也是蛋白激酶B-哺乳动物雷帕霉素靶点(mammalian targetofrapamycin,mTOR)信号通路的靶点。谷物普遍含有较高水平的亮氨酸,蛋白质含量低的日粮(俗称“低蛋白日粮”)中这三种BCAA的比例不平衡,会对家禽的生长性能产生负面影响。这是由于这三种BCAA具有相似的结构,会引发代谢竞争,并会干扰酶降解。最新的证据表明,日粮中加入BCAA对家禽的先天性和适应性免疫系统维持正常功能以及肠道黏膜保持完整性至关重要。至于家禽的BCAA需要量,NRC(1994)给出了推荐值,商品肉鸡和蛋鸡的营养标准也明确了各自的推荐量。综合文献资料可知,家禽的BCAA需要量受日粮类型以及家禽的品种和年龄的影响。值得注意的是,与饲料原料成分中的BCAA(即结合型BCAA)相比,针对非结合型BCAA的添加比例的研究有限,而这将会影响家禽在不同疾病挑战条件下对BCAA的动态利用率,特别是那些会影响肠道消化糜排空速率的疾病。 展开更多

Geographically replicating objects across multiple data centers improves the performance and reliability of cloud storage systems.Maintaining consistent replicas comes with high synchronization costs,as it faces more expensive WAN transport prices and increased latency.Periodic replication is the widely used technique to reduce the synchronization costs.Periodic replication strategies in existing cloud storage systems are too static to handle traffic changes,which indicates that they are inflexible in the face of unforeseen loads,resulting in additional synchronization cost.We propose quantitative analysis models to quantify consistency and synchronization cost for periodically replicated systems,and derive the optimal synchronization period to achieve the best tradeoff between consistency and synchronization cost.Based on this,we propose a dynamic periodic synchronization method,Sync-Opt,which allows systems to set the optimal synchronization period according to the variable load in clouds to minimize the synchronization cost.Simulation results demonstrate the effectiveness of our models.Compared with the policies widely used in modern cloud storage systems,the Sync-Opt strategy significantly reduces the synchronization cost. 展开更多

为构建一个能够快速检测凋亡的含荧光素酶报告基因(Fluc)的真核表达质粒,采用PCR的方法将Caspase-3的识别基序Asp-Glu-Val-Asp (DEVD)四个氨基酸引入至Fluc基因的C端和N端之间;同时选取Asp-Glu-Val-Gly (DEVG)四个氨基酸作为阴性对照。... 为构建一个能够快速检测凋亡的含荧光素酶报告基因(Fluc)的真核表达质粒,采用PCR的方法将Caspase-3的识别基序Asp-Glu-Val-Asp (DEVD)四个氨基酸引入至Fluc基因的C端和N端之间;同时选取Asp-Glu-Val-Gly (DEVG)四个氨基酸作为阴性对照。随后重组片段分别克隆至分离型内含肽的N端和C端之间,并命名为pFluc-DEVD和pFluc-DEVG。将该表达质粒与海肾荧光素酶报告质粒(RLUC)同时转染至HeLa细胞,通过双荧光素酶报告基因和Western blotting实验检测细胞凋亡水平。双荧光素酶报告基因系统实验结果显示,当发生凋亡时,pFluc-DEVD质粒组表达的萤火虫荧光素酶的含量高出pFluc-DEVG质粒组约3倍。Western blotting检测结果显示,转染pFluc-DEVD质粒的细胞在凋亡药物刺激后,Fluc活化的蛋白显著增多,且Caspase-3 的活化程度与Fluc的表达呈正相关,并有显著的统计学差异。以上结果表明,pFluc-DEVD真核表达质粒表达的萤火虫荧光素酶蛋白能被细胞内的Caspase-3酶裂解,且该质粒能够精确地反映出细胞的凋亡水平,为后续进行凋亡定量检测提供了一个可借鉴的方法。 展开更多

Chronic obstructive pulmonary disease(COPD)is emphysema and/or chronic bronchitis characterised by long-term breathing problems and poor airflow.The prevalence of COPD has increased over the last decade and the drugs most commonly used to treat it,such as glucocorticoids and bronchodilators,have significant therapeutic effects;however,they also cause side effects,including infection and immunosuppression.Here we reviewed the pathogenesis and progression of COPD and elaborated on the effects and mechanisms of newly developed molecular targeted COPD therapeutic drugs.Among these new drugs,we focussed on thioredoxin(Trx).Trx effectively prevents the progression of COPD by regulating redox status and protease/anti-protease balance,blocking the NF-κB and MAPK signalling pathways,suppressing the activation and migration of inflammatory cells and the production of cytokines,inhibiting the synthesis and the activation of adhesion factors and growth factors,and controlling the cAMP-PKA and PI3K/Akt signalling pathways.The mechanism by which Trx affects COPD is different from glucocorticoid-based mechanisms which regulate the inflammatory reaction in association with suppressing immune responses.In addition,Trx also improves the insensitivity of COPD to steroids by inhibiting the production and internalisation of macrophage migration inhibitory factor(MIF).Taken together,these findings suggest that Trx may be the ideal drug for treating COPD. 展开更多

This study aims to evaluate the effects of aflatoxins on growth performance and skeletal muscle of Cherry Valley meat male ducks as they grow and develop. One-day-old healthy meat male ducks(n = 180) were randomly divided into 2 groups; there were 6 replicates in each group and 15 ducks in each replicate. The control group was fed a basic diet, and the experimental group was fed a mold-exposed cottonseed meal diet containing aflatoxins instead of normal cottonseed meal. The experimental period was 35 days, and divided into two stages of 1 to 14 days(early stage) and 15 to 35 days(late stage). During the experimental period, live weight, breast muscle weight and thigh muscle weight of meat male ducks were measured weekly. Results showed as follows: 1) aflatoxins contained in the mold-exposed diet significantly reduced daily weight gain and feed intake, and increased feed-to-gain ratio of meat male ducks at different ages(P < 0.05); 2) the Gompertz equation(W_t = W_m exp {-exp [-B(t-t~*)]}) could successfully fit the growth curve and growth and developmental patterns of skeletal muscles of Cherry Valley meat male ducks(R^2≥0.97); 3) the relationship between chest muscle and live weight was the best described by a power regression and polynomial regression(R^2 = 0.99); the relationship between live weight and thigh muscle weight was the best described by linear regression, polynomial regression, and power regression(R^2 = 0.99); 4) aflatoxins in the mold-exposed diet significantly reduced live weight, breast muscle weight and thigh muscle weight of Cherry Valley meat male ducks at various ages; and 5) aflatoxins delayed the age at peak in growth of meat male ducks, and reduced weights at the peak for breast muscle, thigh muscle and whole body as well as the maximal daily weight gain. In summary,aflatoxins delayed growth of Cherry Valley meat male ducks and development of skeletal muscle. 展开更多

A new cyclometalated iridium(Ⅲ) complex [Ir(2-pq)2(HPIP)]C1 (lrl, 2-pq=3-phenylisoquinoline, HPIP= 2-(2-hydroxyphenyl)imidazo[4,5-f] 1,10-phenanthroline) was synthesized and applied to image mitochondria in living cells. Irl displayed excellent ability to selectively accumulate in mitochondria of live cells with no requirement of replacement of the culture medium. Due to Irl exhibiting better photostability than the commercially available mitochondrial trackers, this complex could be applicable for the imaging and tracking of the mitochondrial mor- phological changes over long periods of time. In addition, in vitro cytotoxicity investigation revealed that it! showed negligible cytotoxicity at the concentrations employed.
