Background: The nutritional value of rapeseed meal may be variable due to the variation of its chemical composition. And a precise understandin9 of the nutritional value of an ingredient is beneficial for the accurate diet formulation and reduction of feed costs. This study was conducted to determine the chemical composition, dicjestible energy (DE) and metabolizable enercjy (ME) content, and apparent ileal digestibility (AID) and standardized ileal dicjestibility (SID) of amino acids (AA) for growincj pigs. Thirteen solvent-extracted double-low rapeseed meal (DLIRSM) samples were obtained from the main double-low rapeseed producing areas in China. Methods: The DE and ME contents of the 13 DLRSM samples were measured in cjrowin9 pigs (six pigs per DLRSM sample, average initial body weicjht (BW) =48.3 kg). The AID and SID of AA of 10 DLRSM samples were determined in 12 crossbred barrows (average initial BW = 35.3 kcj) by using two 6 x 6 Latin square designs. Each Latin square comprised one N-flee diet and 5 DLRSM test diets. Results: The chemical composition of DLRSM varied among samples, and the coefficient of variation was greater than ]0 % for ether extract (EE), neutral detergent fiber (NDF), acid detergent fiber (ADF), calcium (Ca), and total 91ucosinolates. The AA content of DLRSM varied among samples especially for lysine (Lys) and methionine (Met). On a dry matter (DM) basis, the apparent total tract digestibility (ATFD) of gross energy (GE), the DE and ME and the ME:DE ratio of DLRSM averaged 62.39 %, 2862 kcal/k9 and 2723 kcal/kcj, and 94.95 %, respectively. The mean value of S1D of Lys was 70.52 % which varied from 66.54-76.54 %. The SID of crude protein (CP), Met, and threonine (Thr) averaged 72.81%, 82.41%, and 69.76 %, respectively. Conclusions: There was great variability in chemical composition especially in the concentration of EE, NDF and ADF, but no significant differences in energy content of the DLIRSM samples were observed. In addition, the AID and SID of all AA were relatively similar amoncj DLRSM samples except for that of Lys. 展开更多

The effects and the mechanisms of AA in LNCaP cells (... Anacardic acid (AA) is a mixture of 2-hydroxy-6-alkylbenzoic acid homologs. It is widely regarded as a non-specific histone acetyltransferase inhibitor of p300. The effects and the mechanisms of AA in LNCaP cells (prostate cancer cells) remain unknown. To investigate the effect of AA on LNCaP cells, we had carried out several experiments and found that AA inhibits LNCaP cell proliferation, induces G1/S cell cycle arrest and apoptosis of LNCaP cell. The mechanisms via which AA acts on LNCaP cells may be due to the following aspects. First, AA can regulate p300 transcription and protein level except for its mechanisms regulating function of p300 through post-translational modification in LNCaP cells. Second, AA can activate p53 through increasing the phosphorylation of p53 on Serl 5 in LNCaP cells. AA can selectively activate p21 (target genes of p53). Third, AA can down-regulates androgen receptor (All) through supressing p300. Our study suggests that AA has multiple anti-tumor activities in LNCaP cells and warrants further investigation. 展开更多

The newly discovered early Paleozoic Delenuoer ophiolite,in the western margin of the Central Qilian Shan,is composed of serpentinized peridotite,cumulate gabbro,diabase,massive basalt,and pillow basalt.This study presents geochronological and geochemical data for the cumulate gabbro and basalt.LA-ICP-MS U-Pb dating of zircons from the cumulate gabbro yielded a magmatic crystallization age of 472±4 Ma.The basalts have normal mid-ocean ridge basalt(N-MORB)compositions and a narrow range ofεNd(t)values(+4.5 to+5.3),which indicates they were derived from a depleted mantle source.On the basis of regional geological constraints,it is proposed that the Delenuoer ophiolite is a westward extension of the South Ophiolite Belt(Yushigou-Youhulugou-Donggou-Dongcaohe Ophiolite Belt)in the North Qilian Shan.The Delenuoer ophiolite,along with the Gulangxia-Delenuoer fault,defines the westernmost part of the tectonic boundary between the North and Central Qilian Shan.This ophiolite may have formed during southward subduction of the Qilian Ocean slab during the early Paleozoic. 展开更多

Background: Dietary fiber can be fermented in gut of pigs and the end products of fermentation were short-chain fatty acids(SCFA). The SCFA had positive effects on gut bacteria and host immune system. In addition, SCFA can provide a part of available energy for pigs. However, there were limited reports on the relationship between dietary fiber, gut bacteria, and energy metabolism. Therefore, this study investigated how dietary fiber and enzyme addition impacted energy metabolism by acting on the microbial community and SCFA.Methods: Wheat bran(WB) was added to the corn-soybean meal-based diet at the levels of 12% and 27%, and oat bran(OB) at 15% and 36%. One of each diet was supplemented with or without 5000 U/kg feed of xylanase, so a total of 10 diets were allotted to 60 growing pigs(initial body weight: 27.2 ± 1.2 kg) using a randomized complete block design. The experiment was conducted in 10 consecutive periods using 6 similar open-circuit respiration chambers. Each pig was used for one 20-day period. During each period, six pigs were allowed 14 d to adapt to the diets in metabolic cages followed by 6 d(from d 15 to d 20) in respiration chambers to measure heat production(HP).Results: Pigs fed 36% OB diets had greater(P < 0.05) nutrient digestibility and net energy(NE) values compared to those fed 27% WB diets. Apparent digestibility coefficients of dry matter(DM) and crude protein(CP) were lower(P < 0.05) in pigs fed 27% WB diets compared with those fed 12% WB diets. Enzyme addition improved(P < 0.05)the NE values(11.37 vs. 12.43 MJ/kg DM) in diets with 27% WB. Supplementation of xylanase did not affect NE values for basal diets, OB diets and 12%WB diets. Compared with diets with 36% OB, pigs fed 27% WB-based diets excreted more total SCFA, acetate and propionate(expressed as g/kg feed DM) in fecal samples of pigs(P < 0.05).Pigs in the WB diets had greater proportion of phylum Bacteroidetes while phylum Firmicutes were greater in pigs fed OB diets(P < 0.05). Pigs fed WB diets had greater(P < 0.05) abundance of Succinivibrio and Prevotella, which were associated with fiber degradation and SCFA production.Conclusion: Our results indicated diets supplied by high level of OB or WB promote the growth of fiber-degrading bacteria. The differences in fiber composition between WB and OB led to differences in nutrient digestibility and bacterial communities, which were ultimately reflected in energy metabolism. Enzyme supplementation improved nutrient digestibility as well as NE values for 27% WB diets but not for other diets, which indicated that effects of enzyme were related to type and level of dietary fiber in diets. 展开更多

We demonstrate a low-noise,high-gain,and large-dynamic-range photodetector(PD)based on a junction field-effect transistor(JFET)and a charge amplifier for the measurement of quantum noise in Bell-state detection(BSD).Particular photodiode junction capacitance allows the silicon N-channel JFET 2sk152 to be matched to the noise requirement for charge amplifier A250.The electronic noise of the PD is effectively suppressed and the signal-to-noise ratio(SNR)is up to 15 dB at the analysis frequency of 2.75 MHz for a coherent laser power of 50.08µW.By combining of the inductor and capacitance,the alternating current(AC)and direct current(DC)branches of the PD can operate linearly in a dynamic range from 25.06µW to 17.50 mW.The PD can completely meet the requirements of SNR and dynamic range for BSD in quantum optics experiments. 展开更多

The effects of Lactobacillus plantarum in microencapsulation(LPM)on intestinal development in layer chicks were investigated in this study,as well as the colonization of L.plantarum in the gut.A total of 480 healthy Hy-Line Brown layer chicks at 0 d old were randomly divided into 4 groups(8 replicates each treatment),and the diets of these birds were supplemented with nothing(control),L.plantarum(0.02 g/kg feed;10^(9) CFU/kg feed),LPM(1.0 g/kg feed;109 CFU/kg feed)and wall material of LPM(WM;0.98 g/kg feed),respectively.Compared to control,LPM improved growth performance and intestinal development of layer chicks,evidenced by significantly increased body weight,average daily gain,average daily feed intake,villus height,villus height/crypt depth,as well as weight and length of the duodenum,jejunum and ileum(P<0.05).These results could be attributed to the increased colonization of L.plantarum in the gut,which was verified by significant increases in lactic acid content,viable counts in chyme and mucosa(P<0.05),as well as a visible rise in number of strains labeled with fluorescein isothiocyanate.Meanwhile,the relative abundances of Lactobacillus and Bifidobacterium significantly increased in response to microencapsulated L.plantarum supplementation(P<0.05),accompanied by the significant up-regulation of colonization related genes(P<0.05),encoding solute carrier family,monocarboxylate transporter,activin A receptor,succinate receptor and secretogranin II.To sum up,microencapsulated L.plantarum supplementation promoted intestinal development,which could be attributed to the enhancement of L.plantarum colonization in the intestine through the mutual assistance of Bifidobacterium and interactions with colonization related transmembrane proteins. 展开更多

Oil sorbents are an attractive option for oil-spill cleanup as they may be used for collection and complete removal of oil without adversely affecting the environment. However, traditional oil sorbents exhibit low oil/water separation efficiency and/or low oil-sorption capacity. In this study, an ultra-high performance graphene/polyurethane (PU) sponge has been successfully obtained by in situ polymerization in the presence of graphene dispersed in N-methylpyrrolidone (NMP). During polymerization, the NMP/graphene dispersion not only serves as a weak amine catalyst for the formation of the sponge, but promotes fixation of the graphene sheets in the framework of the PU sponge owing to the strong dipole interaction between NMP and graphene. The as-prepared graphene/PU sponge was used as an absorbing material for the continuous removal of oil from oil-spill water. The graphene/PU sponge can continuously and rapidly remove oils from immiscible oil/water mixtures in corrosive solutions, including strong acids and bases, hot water, and ice water, with an excellent separation efficiency of above 99.99%. In addition, the as-prepared graphene/PU sponge was effective in separating surfactant-stabilized emulsions with a high separation efficiency of 〉99.91%. 展开更多

Background:Nucleic acid amplification enhancers suitable for use in a recombinase-aided amplification(RAA)assay were studied for the first time,and amplification of a long-fragment(509 bp)was initially explored.Methods:Using recombinant plasmids and clinical samples,RAA fluorescence and basic methods were used to evaluate the efficacy.The fluorescence method was evaluated by threshold time and fluorescence value,and the basic method was characterized by 2%agarose gel electrophoresis.Results:Taking a previously established RAA assay for HPV18 as an example,we demonstrated that the addition of 0.2 M,0.4 M,and 0.6 M betaine and 10%pullulan could enhance the RAA.The new RAA assays with betaine and pullulan were named B-RAA and P-RAA,respectively.Using the B-RAA and P-RAA fluorescence methods,the threshold time values could be shortened by 1.72-2.32 minutes and 2.60 minutes,respectively,and the fluorescence values could be enhanced by 8847.25-9094.37 mv and 5250 mv,respectively.Using the basic method,the sensitivity could be increased 10-fold.We successfully amplified a long-fragment of 509 bp using a P-RAA assay with a sensitivity of 102 copies/μL(compared with 103 copies/μL in the RAA assay).Conclusions:Thus,we concluded that betaine and pullulan are effective additives to enhance the sensitivity of RAA assays. 展开更多

ABSTRACT Background:COVID-19 infection is a major public health problem worldwide,and the D614G mutation enhances the infectivity of COVID-19.Methods:A probe-directed recombinase amplification(PDRA)assay was discussed to detect the D614G mutation at 39℃for 30 min.The sensitivity,specificity,and reproducibility of the PDRA were evaluated by D614 and G614 recombinant plasmids.The clinical performance of PDRA assay was validated by testing of 53 previously confirmed COVID-19 positive RNAs and 10 negative samples.Direct sequencing was carried out in parallel for comparison.Result:With good reproducibility and specificity,the PDRA assay worked well with the concentration in the range of 103–107 copies/reaction.Compared with direct sequencing as a reference,the recombinase-aided amplification(RAA)assay obtained 100%sensitivity and 100%specificity using clinical samples. 展开更多

The Paleo-Asian Ocean (Southern Mongolian Ocean) ophiolitic belts and massive granitoids are exposed in the Alxa block, in response to oceanic subduction processes. In this work, we report petrographic, geochem- ical, and zircon U-Pb age data of some granitoid intrusions from the northern Alxa. Zircon U-Pb dating for the quartz diorite, tonalite, monzogranite, and biotite granite yielded weighted mean 206pb/238U ages o f 302 ±9.2 Ma, 246.5±54.6 Ma, 235±4.4 Ma, and 229.5±5.6 Ma, respectively. The quartz diorites (-302 Ma) exhibit geochemical similarities to adakites, likely derived from partial melting of the initially subducted Chaganchulu back-arc oceanic slab. The tonalites (-246.5 Ma) display geochemical affinities of I-type granites. They were probably derived by fractional crystallization of the modified lithospheric mantle-derived basaltic magmas in a volcanic arc setting. The monzo- granites (-235 Ma) are characterized by low A1203, but high Y and Yb with notably negative Eu anomalies. In contrast, the biotite granites (-229.5 Ma) show high A1203 but low Y and Yb with steep HREE patterns and the absence of negative Eu anomalies. Elemental data suggested that the biotite granites were likely derived from a thickened lower crust, but the monzogranites originated from a thin crust. Our data suggested that the initial subduction of the Chaganchulu oceanic slab towards the Alxa block occurred at - 302 Ma. This subduction process continued to the Early Triassic (-246 Ma) and the basin was finally closed before the Middle Triassic (-235 Ma). Subsequently, the break-off of the subducted slab triggered asthenosphere upwelling (240-230 Ma). 展开更多
