Background:LL-37 peptide is a member of the human cathelicidin family,and has been shown to promote the healing of pressure ulcers.However,the low stability of this peptide within the wound environment limits its clinical use.Chitosan(CS)hydrogel is commonly used as a base material for wound dressing material.Methods:CS hydrogel(2.5%w/v)was encapsulated with LL-37.Cytotoxicity of the product was examined in cultured NIH3 T3 fibroblasts.Effects on immune response was examined by measuring tumor necrosis factor-α(TNF-α)release from RAW 264.7 macrophages upon exposure to lipopolysaccharides.Antibacterial activity was assessed using Staphylococcus aureus.Potential effect on pressure ulcers was examined using a mouse model.Briefly,adult male C57 BL/6 mice were subjected to skin pressure using magnets under a 12/12 h schedule for 21 days.Mice were randomized to receive naked LL-37(20μg),chitosan gel containing 20μg LL-37(LL-37/CS hydrogel)or hydrogel alone under the ulcer bed(n=6).A group of mice receiving no intervention was also included as a control.Results:LL-37/CS hydrogel did not affect NIH3 T3 cell viability.At a concentration of 1–5μg/ml,LL-37/CS inhibited TNF-αrelease from macrophage.At 5μg/ml,LL-37/CS inhibited the growth of Staphylococcus aureus.The area of the pressure ulcers was significantly lower in mice receiving LL-37/CS hydrogel in comparison to all other 3 groups on days 11(84.24%±0.25%),13(56.22%±3.91%)and 15(48.12%±0.28%).Histological examination on days 15 and 21 showed increased epithelial thickness and density of newly-formed capillary with naked LL-37 and more so with LL-37/CS.The expression of key macromolecules in the process of angiogenesis(i.e.,hypoxia inducible factor-1α(HIF-1α)and vascular endothelial growth factor-A(VEGF-A))in wound tissue was increased at both the mRNA and protein levels.Conclusion:Chitosan hydrogel encapsulated with LL-37 is biocompatible and could promote the healing of pressure ulcers. 展开更多

AIM:To evaluate volume differences between anterior and posterior orbit and demographic characteristics of Chinese patients with congenital microphthalmia.METHODS:A retrospective cohort study,involving 169 unilateral congenital microphthalmia patients aged between 1 and 57 years old was conducted.Three-dimensional images of the orbit were generated from past CT scans,and digital orbital volume comprehensive measurement was done.The measured data included orbital volume(OBV),posterior orbital volume(POV),orbital width(OBW),orbital height(OBH),orbital depth(OBD),and posterior orbital area ratio.RESULTS:Significant differences were observed among OBV,POV,OBW,OBH,and OBD of the affected and unaffected eyes in different age-based groups(all P<0.001).Among them,OBH had the greatest different.The mean microphthalmic to contralateral ratio(MCR) of OBV,POV,OBW,and OBH continuously increased from 1 to 3 years old,whereas the MCR of POV decreased from 3 to 17 years old.The MCR of OBD was not found to be correlated to age.There was no significant difference between OBV,POV,OBW,and OBH in ages from 13 years old to adulthood(all P>0.05).The difference in posterior orbital area ratio between the affected and unaffected groups was not statistically significant(P>0.05).CONCLUSION:OBH is maximally affected,whereas OBD is minimally affected by microphthalmia.Posterior orbital retardation began 2y prior to orbital retardation and occurred at 3 years old in the affected eye,suggesting that intervention therapy should be done before the age of 4. 展开更多

BACKGROUND Sporadic cases of rheumatoid arthritis(RA)due to unsatisfactory responses to Abatacept(ABT)have been reported;however,the rescue therapy has not been finalized.Here,we present a case with difficult-to-treat RA(D2T RA)that was resistant to either a single ABT or a Janus kinase(JAK)inhibitor(Tofacitinib),but improved with a combination of ABT and JAK inhibitor(Baricitinib,BAT).CASE SUMMARY A 46-year-old Chinese woman who had RA for ten years that was resistant to Tocilizumab,Etanercept,Adalimumab,and ABT.According to the European League Against Rheumatism definition,the patient was diagnosed with D2T RA.It was then improved with a combination of ABT and a JAK inhibitor BAT.CONCLUSION ABT combined with BAT may be an acceptable strategy for treating D2T RA. 展开更多

目的研究个性化干预对高龄直肠癌腹会阴联合直肠癌根治术(Miles术)后结肠造口患者的临床效果。方法研究对象选取该院2014年6月-2016年1月收治的行直肠癌Miles术后结肠造口的高龄患者114例,采用随机数字法将其分为对照组和观察组,每组各5... 目的研究个性化干预对高龄直肠癌腹会阴联合直肠癌根治术(Miles术)后结肠造口患者的临床效果。方法研究对象选取该院2014年6月-2016年1月收治的行直肠癌Miles术后结肠造口的高龄患者114例,采用随机数字法将其分为对照组和观察组,每组各57例,对照组患者给予常规干预,观察组患者则联合个性化干预,比较两组患者的各种并发症发生率、干预满意度及生存质量评分。同时比较两组患者干预前自护能力评分。结果观察组的总并发症发生率(5.25%)明显低于对照组(22.80%)(χ~2=8.36,P=0.000),观察组的总干预满意度(91.23%)明显高于对照组(75.44%)(χ~2=6.60,P=0.010),干预后,观察组的自护能力评分和生存质量评分明显高于对照组(P<0.01)。结论采用个性化干预可减少高龄直肠癌Miles术后结肠造口患者各种并发症发生,对患者的生存质量及自护能力均有明显提高,且可有效缓解负面情况,提高患者对干预的满意度,具有较高的开展价值,值得在临床推广。 展开更多

目的:分析葡萄膜黑色素瘤的形状对视网膜结构的影响。方法:回顾性分析葡萄膜黑色素瘤患者的病理切片和临床资料,分析肿瘤形状、大小、高度-基底比值、视网膜薄变和浸润的差异。结果:本研究包括102例患者(102眼),平均年龄45.6±12.4... 目的:分析葡萄膜黑色素瘤的形状对视网膜结构的影响。方法:回顾性分析葡萄膜黑色素瘤患者的病理切片和临床资料,分析肿瘤形状、大小、高度-基底比值、视网膜薄变和浸润的差异。结果:本研究包括102例患者(102眼),平均年龄45.6±12.4岁。其中葡萄膜黑色素瘤76眼(75%)为梭形细胞型,6眼(6%)为上皮样细胞型,16眼(16%)为混合细胞型,其它4眼(4%)。按肿瘤部位分:睫状体黑色素瘤3眼(2.9%),睫状体和脉络膜黑色素瘤28眼(27.5%),脉络膜黑色素瘤71眼(69.6%)。视网膜组织的分析中,43眼(43%)发现视网膜变薄,86眼(84%)发现视网膜浸润。肿瘤的高度在1.2~15.6(平均8.3±3.6) mm,肿瘤基底直径为4.8~31.2(平均17.3±6.5) mm。肿瘤峰高度-基底R值为0.53±0.32。视网膜薄变组(n=43)的平均R值显着高于无视网膜薄变组(n=59)(0.69±0.31与0.42±0.27;P<0.01)。视网膜肿瘤浸润组(n=86)的平均R值比无视网膜浸润组(n=16)略高(0.56±0.33比0.41±0.25;P=0.09),但无统计学意义。结论:高R值的肿瘤与其邻近视网膜薄变有关,与视网膜内肿瘤细胞浸润相关性不大。葡萄膜黑色素瘤的切面形状和邻近视网膜组织的受累情况关系紧密。 展开更多

AIM:To investigate the feasibility of corneal anterior lamellar reconstruction with human corneal epithelial cells and fibroblasts,and an acellular porcine cornea matrix(APCM) in vitro.·METHODS:The scaffold was prepared from fresh porcine corneas which were treated with 0.5%sodium dodecyl sulfate(SDS)solution and the complete removal of corneal cells was confirmed by hematoxylin-eosin(HE)staining and 4’,6-diamidino-2-phenylindole(DAPI)staining.Human corneal fibroblasts and epithelial cells were cultured with leaching liquid extracted from APCM,and then cell proliferative ability was evaluated by MTT assay.To construct a human corneal anterior lamellar replacement,corneal fibroblasts were injected into the APCM and cultured for 3d,followed by culturing corneal epithelial cells on the stroma construction surface for another 10d.The corneal replacement was analyzed by HE staining,and immunofluorescence staining.·R ESULTS:Histological examination indicated that there were no cells in the APCM by HE staining,and DAPI staining did not detect any residual DNA.The leaching liquid from APCM had little influence on the proliferation ability of human corneal fibroblasts and epithelial cells.At 10d,a continuous 3 to 5 layers of human corneal epithelial cells covering the surface of the APCM was observed,and the injected corneal fibroblasts distributed within the scaffold.The phenotype of the construction was similar to normal human corneas,with high expression of cytokeratin 12 in the epithelial cell layer and high expression of Vimentin in the stroma.·CONCLUSION:Corneal anterior lamellar replacement can be reconstructed in vitro by cultivating human corneal epithelial cells and fibroblasts with an acellular porcine cornea matrix.This laid the foundation for the further transplantation in vitro. 展开更多

The castrated male... It is known that human benign prostatic hyperplasia might arise from an estrogen/androgen (E/T) imbalance. We studied the response of castrated rat prostate to different ratios of circulating E/T. The castrated male Wistar rats were randomly injected with E/T at different ratios for 4 weeks. The prostates of E/T (1:100) group showed a distinct prostatic hyperplasia response by prostatic index, hematoxylin and eosin staining, and quantitative immunohistochemical analysis of a-smooth muscle actin (SMA). In this group, cells positive for Vimentin, non-muscle myosin heavy chain (NMMHC) and proliferating cell nuclear antigen (PCNA) increased in the stroma and epithelium. Furthermore, the mRNA levels of smooth muscle myosin heavy chain (SMMHC) and NMMHC increased. So E/T at a ratio of 1:100 can induce a stromal hyperplastic response in the prostate of castrated rats. The main change observed was an increase of smooth muscle cells, whereas some epithelial changes were also seen in the rat prostates. 展开更多

Objective Isocitrate dehydrogenase gene(IDH)mutations are associated with tumor angiogenesis and therefore play an important role in glioma management.This study compared the performance of tumor blood vessels counted from contrast-enhanced 3D brain volume(3D-BRAVO)sequence and dynamic contrast-enhanced(DCE)MRI in differentiating IDH1 status in gliomas.Methods Forty-four glioma patients[16 with IDH1 mutant-type(IDH1-MT),28 with IDH1 wild-type(IDH1-WT)]were retrospectively analyzed.A blood vessel entering a tumor was defined as an intratumoral vessel;a blood vessel adjacent to the edge of a tumor was defined as a peritumoral vessel.Combined vessels were defined as the sum of the intratumoral and peritumoral vessels.DCE-derived metrics of tumor were normalized to the contralateral normal-appearing white matter.Results Intratumoral,peritumoral,and combined tumor blood vessels were all significantly different between IDH1-MT and IDH1-WT gliomas,and the range of area under curves(AUCs)was 0.816–0.855.For DCE-derived parameters,cerebral blood volume,cerebral blood flow,mean transit time,and volume transfer constant were significantly different between IDH1-MT and IDH1-WT gliomas,and the range of AUCs was 0.703–0.756.Combined vessels possessed the best performance for identifying IDH1 mutations in gliomas(AUC:0.855,sensitivity:0.857,specificity:0.812,P<0.001).Conclusion The number of tumor blood vessels has comparable diagnostic performance with DCE-derived parameters for differentiating IDH1 mutations and can serve as a potential imaging biomarker to reflect IDH1 mutations in gliomas. 展开更多

AIM: To detect whether there is Helicobacter pylori (Hpylori) colonization in the pharynx mucous membrane of healthy people and whether chronic pharyngitis is related to Hpylori infection. METHODS: Fifty cases of chronic pharyngitis refractory over three months were prospectively studied from March 2004 to August 2004 in the otolaryngology outpatient department of the Second Hospital of Xi'an Jiaotong University. Template-directed dye-terminator incorporated with fluorescence polarization detection (TDI-FP) and modified Giemsa stain were used to examine pharynx mucous membrane tissue for H pylori colonization in the patients with chronic pharyngitis and the healthy people as a control group. RESULTS: In the control group, no people were detected to have Hpylori in the pharynx. In contrast, in 50 cases with chronic pharyngitis, 19 (38.0%) cases were H pylori positive with a TDI-FP assay and 4 (8%) cases were TDI-FP positive with Giemsa staining in the pharynx. Sixteen of the 50 pharyngitis cases had stomach ailment history, 11 cases (68.8%) of these 16 patients were determined to be H pylori positive in the pharynx with the TDI-FP assay. 2,2 test showed that this infection rate was remarkably higher (P= 0.0007) than that in the cases without stomach ailment history. Giemsa staining showed that 3 cases (18.8%) of the patients with stomach ailment history were infected with H pylori in the pharynx, which was remarkably higher (P = 0.042) than that in the patients without stomach ailment history (1 case, which was 2.9%). CONCLUSION: H pylori may not be detected in the pharynx of healthy people. Chronic pharyngitis may be related to H pylori infection. The infection rate with Hpylori in the pharynx is higher in patients with stomach ailment histories than in patients without stomach ailment histories, suggesting that chronic pharyngitis may be related to stomach ailment history. 展开更多

Regulators of the estrogen receptor (ER) are tissue- and cell-specific. We evaluated the effect of estrogen antagonist, raloxifene (Ral), on the prevention and treatment of BPH by investigating its effect on the proliferation of two different prostate cell lines: a stromal cell line, WPMY-1, and a benign prostatic hyperplasia epithelial cell line, BPH-1. We additionally evaluated its effect on prostatic hyperplasia induced by estrogen and androgen in a rat model. The effect of Ral on the prevention of prostatic hyperplasia was analyzed by haematoxylin and eosin staining and quantitative immunohistochemistry (IHC) for proliferating cell nuclear antigen and α-smooth muscle actin. In vitro and in vivo, tamoxifen (Tam), another anti-estrogen drug, and finasteride (Fin), a drug for the clinical treatment of BPH, served as efficacy controls. The in vitro data showed that neither Ral nor Tam alone affected the proliferation of WPMY-1 and BPH-1, but both antagonized the effect of oestradiol in promoting the proliferation of the two cells. Results from the IHC staining of the rat prostates indicated that, similar to Tam and Fin, Ral inhibited the proliferation of stromal cells in vivo. Interestingly, in contrast to Tam, both Ral and Fin inhibited the proliferation of epithelial cells. Furthemore, Ral treatment much strongly decreased the number of prostatic acini and the surrounding layers of smooth muscle cells than Fin (P 〈 0.05). Our data showed for the first time that Ral may have a role in the response of the rat prostate to selective ER modulators. 展开更多

AIM: To clone human liver special F protein and to express it in a prokaryotic system. METHODS: Total RNA was isolated from human liver tissue and first-strand cDNA was reverse transcribed using the PCR reverse primer. Following this, cDNA of the F protein was ligated into the clone vector pUCm-T. The segment of F protein's cDNA was subdoned into the expression vector pET-15b and transformed into E. coli BL21 (DE3) pLyss. Isopropy-β-D-thiogalactoside (IPTG) was then used to induce expression of the target protein. RESULTS: The cDNA clone of human liver special F protein (1134bp) was successfully produced, with the cDNA sequence being published in Gene-bank: DQ188836. We confirmed the expression of F protein by Western blot with a molecular weight of 43 kDa. The expressed protein accounted for 40% of the total protein extracted. CONCLUSION: F protein expresses cDNA clone in a prokaryotic system, which offers a relatively simple way of producing sufficient quantities of F protein and contributes to understanding the principal biological functions of this protein. 展开更多

Objectives:To examine the effects of finger-movement exercises and finger weight-lift training on handgrip strength and Activities of Daily Living Scale(ADLS)values.Methods:A total of 80 very elderly adults(aged80 years)were assigned to either an intervention group(n?40)or a control group(n?40).Subjects in the intervention group performed finger-movement exercises and weight-lift training for a period of 3 months,while subjects in the control group received no intervention,and were unaware of the interventions received in the other group.Results:After completing 3 months of finger-movement exercises and weight-lift training,the average handgrip strength of the 40 participants in the intervention group had increased by 2.1 kg,whereas that in the control group decreased by 0.27 kg(P<0.05).After receiving intervention,the number of subjects in the intervention group with an ADLS score>22 points decreased by 7.5%(P<0.05,vs.pre-intervention).Conclusions:The combined use interventionwith finger-movement exercises and proper finger weight-lift training improved the handgrip strength andADLS values of very elderly individuals.These rehabilitation exercisesmay be used to help the elderlymaintain their self-care abilities. 展开更多

OBJECTIVE Recent studies have shown that hepsin, a type of transmembrane serine protease, is highly upregulated in prostate cancer, but, little is known about its role in progression and invasion of this cancer. We constructed a hepsin-expressing plasmid and transfected it into PC-3 cells to investigate the effect of the hepsin gene on the biological behavior of the PC-3 cells. METHODS Plasmid pHepsin-IRES2 was transfected into prostate cancer PC-3 cells using Fugene6, and the cells with stable hepsin expression were screened and selected with Zeocin (600 mg/L). The hepsin mRNA level was measured by real-time PCR and the growth curve of the PC-3-transfected cells assessed using MTT and BrdU assays. A Boyden chamber was used to examine the difference in invasion and metastases between transfected and non-transfected cells. RESULTS The hepsin mRNA level in pHepsin-IRES2 transfected -PC-3 cells was significantly higher than that found in the control PC -3 cells. While the growth curve of the hepsin gene transfected PC -3 cells showed that there was no significant effect on proliferation, the invasive ability of the pHepsin-IRES2 transfected PC-3 cells, as compared with control cells, was significantly increased (P<0.05). CONCLUSION The results suggest that even though hepsin has no effect on the proliferation of prostate cancer PC-3 cells, it does promote cellular invasion and metastasis.Therefore hepsin may have a role in the development of prostate cancer. 展开更多

Due to its highly insidious and rapid progression,deep tissue pressure injury(DTPI)is a clinical challenge.Our previous study found that DTPI may be a skeletal muscle injury dominated by macrophage immune dysfunction due to excessive iron accu-mulation.Decellularized extracellular matrix(dECM)hydrogel promotes skeletal muscle injury repair.However,its role in po-larizing macrophages and regulating iron metabolism in DTPI remains unclear.Here,porcine dECM hydrogel was prepared,and its therapeutic function and mechanism in repairing DTPI were investigated.The stimulus of dECM hydrogel toward RAW264.7 cells resulted in a significantly higher percentage of CD2o6+macrophages and notably decreased intracellular divalent iron levels.In mice DTPI model,dECM hydrogel treatment promoted M1 to M2 macrophage conversion,improved iron metabolism and reduced oxidative stress in the early stage of DTPI.In the remodeling phase,the dECM hydrogel remarkably enhanced revascularization and accelerated skeletal muscle repair.Furthermore,the immunomodulation of dEcM hydrogels in vivo was mainly involved in the P13k/Akt signaling pathway,as revealed by GO and KEGG pathway analysis,which may ameliorate the iron deposition and promote the healing of DTPI.Our findings indicate that dECM hydrogel is promising in skeletal muscle repair,inflammation resolution and tissue injury healing by effectively restoring macrophage immune homeostasis and normalizing iron metabolism. 展开更多

Excessive reactive oxygen species(ROS)at severe burn injury sites may promote metabolic reprogramming of macrophages to induce a deteriorative and uncontrolled inflammation cycle,leading to delayed wound healing and regeneration.Here,a novel bioactive,anti-fouling,flexible polyzwitterionic hydrogel encapsulated with epigallocatechin gallate(EGCG)-copper(Cu)capsules(termed as EGCG-Cu@CBgel)is engineered for burn wound management,which is dedicated to synergistically exerting ROS-scavenging,immune metabolic regulation and pro-angiogenic effects.EGCG-Cu@CBgel can scavenge ROS to normalize intracellular redox homeostasis,effectively relieving oxidative damages and blocking proinflammatory signal transduction.Importantly,EGCG-Cu can inhibit the activity of hexokinase and phosphofructokinase,alleviate accumulation of pyruvate and convert it to acetyl coenzyme A(CoA),whereby inhibits glycolysis and normalizes tricarboxylic acid(TCA)cycle.Additionally,metabolic reprogramming of macrophages by EGCG-Cu downregulates M1-type polarization and the expression of proinflammatory cytokines both in vitro and in vivo.Meanwhile,copper ions(Cu^(2+))released from the hydrogel facilitate angiogenesis.EGCG-Cu@CBgel significantly accelerates the healing of severe burn wound via promoting wound closure,weakening tissue-damaging inflammatory responses and enhancing the remodeling of pathological structure.Overall,this study demonstrates the great potential of bioactive hydrogel dressing in treating burn wounds without unnecessary secondary damage to newly formed skin,and highlights the importance of immunometabolism modulation in tissue repair and regeneration. 展开更多

AIM:To conduct a bibliometric analysis of studies on microphthalmos and anophthalmos(M/A),explore research hotspots,and provide information on future research interests in this field to benefit clinicians and researchers.METHODS:Totally 751 publications related to M/A from the year 2004 to 2023 were collected from the Web of Science Core Collection database.These publications consist of both original and review articles,that are composed in English.The contributions of different countries,institutions,journals,and authors were analyzed,and network analysis was conducted by using Microsoft Excel 2021,VOSviewer,and R Studio to visualize research hotspots.RESULTS:Among all publications included,the highest number of publications came from USA(218,29.03%).China followed with 99 publications(13.18%),and England with 86 publications(11.45%).The publications from the USA had the highest frequency of citations,with 16699 citations,and the highest H-index of 49.The American Journal of Medical Genetics Part A(43,5.73%)published the largest number of papers,and the University of London had the most publications(41,5.46%).The genetic and molecular mechanisms of M/A were still unclear and the clinical intervention for M/A had gained a lot of attention as an emerging area of interest.CONCLUSION:Data have been gathered on the yearly count of published materials and citations,as well as the rise in publication trends,the efficiency of regions or countries,authors,journals,and organizations,along with the high-cited publications in M/A.The recent trend of research has shifted from genetic mechanisms to different clinical phenotypes and corresponding clinical interventions,which can give direction to future research. 展开更多
