Reduced photosynthesis results directly from nitrogen or water deficiency in wheat plants,and leads to a decrease in grain yield.In this study,by measuring the effects of water and N deficiencies,both individually and combined,we characterized the responses of wheat(Triticum aestivum L.Yumai 49-198)plants to these two deficiencies using physiological measurements and comparative proteomics.Significant decreases in grain yield and leaf photosynthetic performance were observed in all deficiency conditions,and 106 photosynthetic proteins that showed responses were identified.Nitrogen deficiency induced the least change in photosynthetic proteins,and similar changes in most of these proteins were also observed for the combined nitrogen and water deficiencies.Water deficiency induced the largest change in photosynthetic proteins and resulted in the lowest 1000-kernel weight.Severe decreases in photosynthesis in both the water-deficiency and combined N and water deficiency groups were reflected mainly in an imbalanced ATP/NADPH ratio associated with the light reaction,which influences carbon metabolism in the Calvin cycle.Photorespiration was respectively stimulated or inhibited by N or water deficiency,while suppression of photorespiratory flux and activation of nitrogen recycling were observed in the combined N and water deficiency treatments.Comparison of photosynthetic proteins between experimental sites suggested that precipitation affected linear electron flow in the photoreaction,and thus photosynthetic efficiency.Our results provide a baseline for future studies of the roles of these photosynthetic proteins in the response to N or water deficiency and their effect on 1000-kernel weight. 展开更多

Spinal Cord Infarction with a Negative MRI Finding: A Case Report and Literature Review

Then abnormal signal wi... Introduction: Spinal cord infarction (SCI) is a relatively rare disease. Abnormality on magnetic resonance imaging (MRI) is an important diagnostic criterion. It may be normal on the first day. Then abnormal signal will appear a few days later. There were some reports with negative MRI findings at the beginning. However, those reports were neither found abnormalities on repeat MRI nor without repeat MRI. Case Presentation: We report a case of SCI presented as paralysis of lower limbs and chest pain with no abnormalities on MRI through the course of disease in a 74-year-old female. Anti-platelet aggregation drugs, stabilizing plaque treatments and rehabilitation exercise were performed. The patient recovered significantly. She could walk independently a month after the onset. Conclusions: Although MRI provides important evidence for diagnosis, clinical manifestations are the most important when diagnosing. For patients who were highly suspected of SCI clinically, treatment for vascular diseases should be applied as soon as possible even if no lesion is found on imaging examination. 展开更多

目的观察Apelin-13对血管性痴呆(VD)大鼠海马区氧自由基及自噬的作用,进一步探讨其对VD大鼠学习记忆功能的影响。方法取健康的雄性SD大鼠,随机分为假手术组、模型组和50μg/kg Apelin-13组和100μg/kg Apelin-13组。利用四血管阻断法建... 目的观察Apelin-13对血管性痴呆(VD)大鼠海马区氧自由基及自噬的作用,进一步探讨其对VD大鼠学习记忆功能的影响。方法取健康的雄性SD大鼠,随机分为假手术组、模型组和50μg/kg Apelin-13组和100μg/kg Apelin-13组。利用四血管阻断法建立血管性痴呆模型,给药组侧脑室注射135μL Apelin-13(剂量分别为50和100μg/kg)。造模24 h后取脑,用干湿重法测定大鼠脑含水量;WST-1法测定超氧化物歧化酶(SOD)的活力;TBA法检测丙二醛(MDA)含量;Morris水迷宫测定大鼠的学习记忆功能;Western blot法检测海马区Beclin-1、LC3蛋白水平。结果与模型组相比,50和100μg/kg Apelin-13的定位航行潜伏期缩短(P<0.05或P<0.01),空间探索实验的第1次穿台时间缩短(P<0.05或P<0.01),穿台次数增多(P<0.01)。50和100μg/kg Apelin-13组大鼠脑含水量降低(P<0.05),MDA含量降低(P<0.05或P<0.01),并具有剂量依赖性,SOD活力升高(P<0.05)。50和100μg/kg Apelin-13组Beclin-1蛋白表达降低(P<0.05或P<0.01),具有剂量依赖性;100μg/kg Apelin-13组LC3-Ⅱ/LC3-Ⅰ降低。结论 Apelin-13能够改善VD大鼠学习记忆功能,其机制可能与减少脑含水量,抗自由基损伤和抑制某些自噬蛋白有关。 展开更多

Bronchial asthma is a common chronic inflammatory disease characterized by airway hyperresponsiveness(AHR),inflammatory cell infiltration,and airway remodeling.F-box/WD repeat-containing protein 7(FBXW7),an E3 ubiquitin ligase,is required for various endothelial functions,such as cell migration,inflammation,and endothelial integrity.This study aimed to investigate the role of FBXW7 in lipopolysaccharide(LPS)-induced epithelial barrier impairment in bronchial epithelial cells in vitro.By using lentivirus-based technology,FBXW7 was overexpressed or silenced(24 h)in human bronchial epithelial(16HBE)cells,which were treated with LPS or not(24 h).Immunoprecipitation(IP)detection and Western blot analysis were used to evaluate the interaction of target proteins.Cell permeability was measured using transepithelial electrical resistance and FITC dextran flux(48 h).IL-1β,IL-18 and TNF-αin cell supernatants were measured using ELISA(48 h).The results showed that LPS stimulation suppressed FBXW7 expression in a time-and dose-dependent manner.LPS exposure decreased cell proliferation,elevated IL-1β,IL-18 and TNF-α,increased epithelial permeability,and p38 phosphorylation.These LPS-induced changes were partly compromised by FBXW7 overexpression.Similar to LPS stimulation,FBXW7 knockdown increased epithelial permeability and levels of inflammatory cytokines and p38 phosphorylation,which were,in part,blocked by apoptosis signal-regulating kinase(ASK)1 knockdown or p38 pathway inhibition.IP and Western blot analysis showed that FBXW7 interacted with ASK1.ASK1 expression was inversely associated with FBXW7 expression.FBXW7 overexpression markedly enhanced ASK1 ubiquitination.These data revealed that FBXW7 counter against inflammation and protects epithelial barrier integrity in bronchial epithelial cells by promoting ubiquitination-mediated degradation of ASK1 via the p38 pathway. 展开更多

Objective:To study the effect of intrapleural endostatin and mannatide infusion on malignant molecule expression in pleural fluid of malignant pleural effusion.Methods:Patients with lung cancer and malignant pleural effusion treated in our hospital between April 2013 and December 2015 were selected and randomly divided into two groups, the observation group received intrapleural endostatin and mannatide infusion treatment and control group accepted routine intrapleural infusion treatment. 4 weeks after treatment, the pleural fluid samples were collected to determine the levels of tumor markers, invasion-related molecules, VEGF-related molecules and anti-tumor cytokines.Results:4 weeks after treatment, CEA, CYFRA21-1, NSE, SCC-Ag, CXCL12, CXCR4, MMP2, MMP9, VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-R1, VEGF-R2 and VEGF-R3 levels in pleural fluid of observation group were significantly lower than those of control group while LASS2/TMSG-1, IFN-γ, IL-2, TNF-α, IL-17 and IL-23 levels were significantly higher than those of control group.Conclusion:Intrapleural endostatin and mannatide infusion treatment of malignant pleural effusion can more effectively kill cancer cells, inhibit cell invasion, angiogenesis and lymphangiogenesis, and enhance antitumor immune response. 展开更多

A simple quasi-distributed fiber sensing interrogation system based on random speckles is proposed for weak fiber Bragg gratings(WFBGs) in this work. Without using tunable lasers or spectrometers, a piece of multimode fiber is applied to interrogate the WFBGs relying on the wavelength sensitivity of speckles. Instead of the CCD sensor, an InGaAs quadrant detector serves as the receiver to capture the fast-changing speckle patterns. A supervised deep learning algorithm of the multilayer perceptron architecture is implemented to process speckle data and to interrogate temperature changes or dynamic strains. The proposed demodulation system is experimentally demonstrated for WFBGs with 0.1% reflectivity.The experimental results demonstrate that the new system is capable of measuring temperature change with an accuracy of 1℃ and achieving dynamic frequency of 100 Hz. This speckle-based interrogation system paves a new way for distributed WFBGs sensing with a simple design. 展开更多

Inactivated COVID-19 vaccines have been widely used to vaccinate the Chinese population.However,limited literature exists to explore the effect of obesity on the humoral and cellular immune response to these vaccines.In this study,132 high BMI(Body mass index)(obesity and overweight,BMI≥24 kg/m^(2))and 82 normal BMI(BMI<24 kg/m^(2))participants were enrolled.Adverse events(AEs),Spike receptor-binding domain IgG antibody(anti-RBD-IgG),neutralizing antibodies(NAbs),and specific B-cell and T-cell responses were evaluated 21–105 days after full-course inactivated COVID-19 vaccination.The overall incidence of AEs was similar in individuals with and without obesity/overweight.No serious vaccine-related AEs occurred.Individuals with obesity/overweight had a reduced seropositivity rate of NAbs compared to those with normal BMI.Anti-RBD-IgG and NAbs titers in the high BMI group were significantly lower than those in the normal BMI group.The frequencies of RBD-specific memory B cells(MBCs)and the numbers of spike-specific TNF-α+spot-forming cells(SFCs)in individuals with obesity/overweight were reduced compared with those noted in individuals without obesity/overweight.A similar trend of weakened humoral responses was also observed in individuals with central obesity.Our study results suggested that inactivated COVID-19 vaccines were safe and well tolerated but induced poor humoral and cellular immune responses in Chinese individuals with obesity/overweight. 展开更多

Polyneuropathy,organomegaly,endocrinopathy,M proteins,and skin changes(POEMS)syndrome is a multisystemic disorder that clinically manifests as paraneoplastic and monoclonal plasma cell dyscrasia.Its acronym is derived from its principal characteristics:polyneuropathy,organomegaly,endocrinopathy,M proteins,and skin changes.Here,the authors reported a case of POEMS syndrome that was also associated with Castleman disease.A 53-year-old female patient was admitted to our hospital with limb weakness,numbness,edema,abdominal distention,and fever.Physical examination revealed tetraplegia,paraesthesia,and hyporeflexia in all four limbs,in addition to lymphadenectasis,splenomegaly,skin hyperpigmentation,hypertrichosis,and pitting edema.Laboratory tests and imaging revealed thrombocytosis,hypothyroidism,diabetes,hydropericardium,hydrothorax,splenomegaly,and lymphadenectasis.Electromyography showed the characteristic patterns of both demyelinating disease and axonal degeneration.Serum protein electrophoresis revealed monoclonal immunoglobulin G-lambda paraproteins.Histological examination clearly diagnosed the disease as the hyaline vascular subtype.The final diagnosis in this case was POEMS syndrome in association with Castleman disease. 展开更多
