However, the clinical impact of HH activation in HCC patients is still unclear. This study was conducted to confirm whether the expression of HH pathway components was associated with HCC progression and clinical outcome. Methods: This study was a sample-expanded and prolonged follow up of one of our previous studies. It included 46 HCC patients who underwent surgical treatment from 2002 to 2005. The expression of sonic HH (SHIq), patched-1 (PTCHI), smoothened (SMOH) and glioma-associated oncogene-1 (GLI1) genes in tumor and adjacent normal tissues extracted from the patients were examined by reverse transcription- polymerase chain reaction (RT-PCR) to explore the relationship between these genes and the clinical prognosis of HCC. Results: The expression levels ofSHH, PTCH1, SMOH and GLI1 in HCC tissues were 60.87%, 50.00%, 32.61% and 54.35%, respectively. The expression levels of SHH-related molecules were relatively intense in cancer tissue, but insignificantly correlated with any clinicopathological factors of tumor. Transcriptional factor GLI1 was the only molecule associated with poor prognosis among the HCC patients. The expression of GLI1 gene in tumor tissues was significantly related with disease-free survival (DFS) (P=0.042) and overall survival (OS) (P=0.030). The simultaneous expression of GLI1 in tumor and adjacent normal liver tissues correlated with DFS (P〈0.029) and OS (P〈0.025). Conclusions: HH signaling activation is an important event in the development of human HCC. The expression of GLI1 in SHH pathway is possibly involved in HCC progression, which may be a useful prognostic indicator of HCC.展开更多

Here we investigated how neurons in the inferior colliculus respond to the paired sounds that produce precedence-effect illusions, and whether their firing behavior can be modulated through inhibition with gamma-aminobutyric acid (GABA). We recorded extracellularly from 36 neurons in rat inferior colliculus under three conditions: no injection, injection with saline, and injection with gamma-aminobutyric acid. The paired sounds that produced precedence effects were two identical 4-ms noise bursts, which were delivered contralaterally or ipsilaterally to the recording site. The normalized neural responses were measured as a function of different inter-stimulus delays and half-maximal interstimulus delays were acquired. Neuronal responses to the lagging sounds were weak when the inter-stimulus delay was short, but increased gradually as the delay was lengthened. Saline injection produced no changes in neural responses, but after local gamma-arninobutyric acid application, responses to the lagging stimulus were suppressed. Application of gamma-aminobutyric acid affected the normalized response to lagging sounds, independently of whether they or the paired sounds were contralateral or ipsilateral to the recording site. These observations suggest that local inhibition by gamma-aminobutyric acid in the rat inferior colliculus shapes the neural responses to lagging sounds, and modulates the precedence effect.展开更多

We aim to determine whether HER2-specific T cells generated from dendritic cells (DCs) modified with HER2 gene could effectively kill the HER2-positive breast cancer cells, especially the trastuzumab-resistant cells. Methods: The peripheral blood mononuclear cells (PBMCs) from healthy donors, whose HLA haplotypes were compatible with the tumor cell lines, were transfected with reconstructive human adeno-association virus (rhAAV/HER2) to obtain the specific killing activities of T cells, and were evaluated by lactate dehydrogenase (LDH) releasing assay. Results: Trastuzumab produced a significant inhibiting effect on SK-BR-3, the IC50 was 100ng/ml. MDA-MB-453 was resistant to trastuzumab even at a concentration of 10,000 ng/ml in vitro. HER2-specific T lymphocytes killed effectively SK-BR-3 [(69.86±13.41)%] and MDA-MB-453 [(78.36±10.68)%] at 40:1 (effector:target ratio, E:T), but had no significant cytotoxicity against HER2-negative breast cancer cell lines MDA-MB-231 or MCF-7 (less than 10%). Conclusion: The study showed that HER2-specific T lymphocytes generated from DCs modified by rhAAV/HER2 could kill HER2-positive breast cancer cell lines in a HER2-dependent manner, and result in significantly high inhibition rates on the intrinsic trastuzumab-resistant cell line MDA-MB-453 and the tastuzumab-sensitive cell line SK-BR-3. These results imply that this immunotherapy might be a potential treatment to HER2-positive breast cancer.展开更多

The value and using time of these kinds of loads are uncontrollable, which lead to the three-phase imbalance in low-voltage distribution system, and seriously affect the quality of power supply. A new type of the commutation system and an improved quantum genetic algorithm (IQGA) are proposed in the paper. At last, the rationality and the efficiency of the method are verified by a practical example.展开更多

Sample simulations were implemented for mold filling processes of precision titanium castings under gravity and different centrifugal casting techniques. The computation results show that the alloy melt has a much stronger mold filling ability for thin section castings under a centrifugal force field than that only under the gravity. A 'return back' mold filling manner is showed to be a reasonable technique for centrifugal casting processes, especially for thin section precision castings.展开更多

A force model of Al alloy melt in the traveling magnetic field is founded by analyzing traveling magnetic field carefully. Numerical model of Al alloy mold-filling is founded based on N-S equation, which was suitable for traveling magnetic field. By using acryl glass mold with indium as alloy melt, the experiment testified the filling state of alloy in traveling magnetic field. The results of numerical simulation indicate that the mold-filling ability of gallium melt increases continually with the increase of the input ampere turns.展开更多

However once its forward and backward steps were determined during iteration, it would make computing time increased and affected the reconstruction efficiency. This paper presents a algorithm called forward-backward pursuit algorithm based on weak selection (SWFBP) by introducing threshold strategy into FBP algorithm, and in view of that in the first few iterations, most of the atoms which are selected are right, so this part of atoms are directly incorporated into support set instead of using backward strategy to reduce them. Flexible forward and backward steps accelerate the speed of atom selecting and improve the reconstruction accuracy. We compared SWFBP and FBP algorithm via one-dimensional signal and two-dimensional image reconstruction experiments. The simulation results demonstrate that compared with FBP, SWFBP algorithm has superior performance, including higher PSNR, faster computing speed and lower recovery time.展开更多

Obtaining the dump resistance low limit according to the temperature resistance allows, and calculating the optimal value by drop voltage in the DC-Bus during the fault. The feasibility of the proposed algorithm is verified by simulation results.

文摘Low-voltage distribution systems in our country are mostly used in agricultural loads and household loads. The value and using time of these kinds of loads are uncontrollable, which lead to the three-phase imbalance in low-voltage distribution system, and seriously affect the quality of power supply. A new type of the commutation system and an improved quantum genetic algorithm (IQGA) are proposed in the paper. At last, the rationality and the efficiency of the method are verified by a practical example.
文摘Objective:To study the correlation of serum cyclophilin A (CyPA) and monocyte chemoattractant protein-1 (MCP-1) levels with carotid atherosclerosis in patients with acute cerebral infarction. Methods: 106 patients with acute cerebral infarction who were hospitalized in our hospital between July 2011 and August 2015 were selected as observation group, and 50 cases of healthy persons who received physical examination in our hospital during the same period were selected as normal control group. The serum CyPA and MCP-1 contnets in two groups were determined. According to the median of CyPA and MCP-1 contents in observation group, they were divided into high CyPA group and low CyPA group as well as high MCP-1 group and low MCP-1 group, 53 cases in each group. Contents of lipid metabolism indexes and carotid atherosclerosis illness-related indicators were compared between acute cerebral infarction patients with different CyPA and MCP-1 contents. Results:Serum CyPA and MCP-1 contents in observation group were significantly higher than those in control group. Serum TC, LP(a) and LDL-C contents in high CyPA group and high MCP-1 group were higher than those in low CyPA group and low MCP-1 group while HDL-C contents were lower than those in low CyPA group and low MCP-1 group. Serum CysC, Hcy and UA contents in high CyPA group and high MCP-1 group were higher than those in low CyPA group and low MCP-1 group. Conclusion: Serum CyPA and MCP-1 contents in patients with acute cerebral infarction are higher than those in normal population, and the contents of CyPA and MCP-1 are positively correlated with the degree of carotid atherosclerosis.
文摘Computer simulation codes were developed based on a proposed mathematical model for centrifugal mold filling processes and previous computer software for 3D mold filling and solidification of castings (CASM-3D for Windows). Sample simulations were implemented for mold filling processes of precision titanium castings under gravity and different centrifugal casting techniques. The computation results show that the alloy melt has a much stronger mold filling ability for thin section castings under a centrifugal force field than that only under the gravity. A 'return back' mold filling manner is showed to be a reasonable technique for centrifugal casting processes, especially for thin section precision castings.
文摘The numerical simulation for mold-filling of thin-walled aluminum alloy castings in horizontal traveling magnetic field is performed. The results of numerical simulation indicate that the mold-filling ability of gallium melt increases continually with the increase of the input ampere turns.

文摘We theoretically investigate the frequency-nondegenerate and frequency degenerate squeezed lights with a four-wave mixing process(4WM)driven by two pump fields crossing at a small angle.Different from a 4WM process driven by a single pump field, the refractive index of the corresponding probe field,np,can be converted to a value that is greater than 1 or less than 1 by an angle adjustment. In the new region with np<1,the bandwidth of the gain is relatively large due to the slow change in the refractive index with the two-photon detuning. In this region with an exchange of the roles of the pump and probe beams, the frequency degenerate and spatial nondegenerate twin beams can be generated, which has potential application in quantum information and quantum metrology.

文摘BACKGROUND Hemophilic pseudotumor(HP)is a rare complication in patients with hemophilia. The lesion most frequently occurs in the long bones, pelvis, small bones of the hands and feet,or rarely in the maxillofacial region.Postoperative changes in HP are seldom arrested, whereas angiogenesis characterized by disturbed wound healing in HP may cause vascular malformations.CASE SUMMARY We report the case of an 11-year-old boy who was affected by maxillary intraosseous venous malformation.Enucleation of an HP without factor replacement was performed initially on the right side of the maxilla 3 years ago.The patient was referred to us because of painless swelling in the same location.Factor replacement and subtotal maxillectomy were performed.Pathological examinations revealed intraosseous venous malformation.CONCLUSION This study is the first to document the development of intraosseous venous malformation after enucleation of an HP in the maxillofacial region.Angiogenesis characterized by disturbed wound healing in patients with hemophilia may be pivotal in the pathogenesis of this condition.
文摘Au-Ft,as a green synthesized nanoparticle,is composed of a ferritin nanocage enclosing a pair of Au nanoclusters inside. Our previous study has demonstrated that Au-Ft can be an excellentfluorescent probe for whole body imaging of mice with kidney specific targeting. But,the accuratelocalization of Au-Ft in kidney is still absent. In the current study,we detected and assessed the cellular and subcellular localization of Au-Ft in renal cortex and medulla of nu/nu mice after tailvein injection by using Nuance optical system(CRi,Woburn,USA)and inForm intelligent image analysis soft ware based on single cell segmentation. We obtained the fluorescence intensity and cellular location of kidney-targeting Au-Ft probe in particular cell of renal glomerulus or renaltubules, which provided valuable proofs to clarify the mechanism of Au-Ft selective enrichment in kidney and the associated metabolic processes.

文摘With the development of the greenhouse grape cultivation area, the supporting greenhouse grape varieties appear to be single. At present, there are few researches on the selection of grape varieties suitable for two-harvest-a-year cultivation in greenhouses. In order to meet the needs of fruit growers for the diversity of greenhouse grape varieties, experimental studies had been carried out.Through three consecutive years of two-harvest-a-year production of six grape varieties of‘Jingxiangyu’,‘Ruidukemei’,‘Xiangfei’,‘Jingyan’,‘Zaomanao’and‘Hanxiangmi’,the first crop of greenhouse grapes matured from May to July, and the second crop matured during the New Year s Day and Spring Festival.Through the investigation and analysis of their phenological periods, fruit economic characteristics and economic benefits,it was found that the four varieties of‘Jingxiangyu’,‘Ruidukemei’,‘Xiangfei’and‘Jingyan’performed well under greenhouse cultivation conditions, and thus can be used as supporting fine varieties for large-scale promotion.
文摘Forward-backward pursuit (FBP) algorithm is a novel two-stage greedy approach. The simulation results demonstrate that compared with FBP, SWFBP algorithm has superior performance, including higher PSNR, faster computing speed and lower recovery time.
