elderly individuals with diabetes.Traditional Chinese medicine(TCM)prescriptions have shown remarkable effectiveness for treating DR.This study aimed to screen a novel TCM prescription against DR from patents and eluc... elderly individuals with diabetes.Traditional Chinese medicine(TCM)prescriptions have shown remarkable effectiveness for treating DR.This study aimed to screen a novel TCM prescription against DR from patents and elucidate its medication rule and molecular mechanism using data mining,network pharmacology,molecular docking and molecular dynamics(MD)simulation.Method:TCM prescriptions for treating DR was collected from patents and a novel TCM prescription was identified using data mining.Subsequently,the mechanism of the novel TCM prescription against DR was explored by constructing a network of core TCMs-core active ingredients-core targets-core pathways.Finally,molecular docking and MD simulation were employed to validate the findings from network pharmacology.Result:The TCMs of the collected prescriptions primarily possessed bitter and cold properties with heat-clearing and supplementing effects,attributed to the liver,lung and kidney channels.Notably,a novel TCM prescription for treating DR was identified,composed of Lycii Fructus,Chrysanthemi Flos,Astragali Radix and Angelicae Sinensis Radix.Twenty core active ingredients and ten core targets of the novel TCM prescription for treating DR were screened.Moreover,the novel TCM prescription played a crucial role for treating DR by inhibiting inflammatory response,oxidative stress,retinal pigment epithelium cell apoptosis and retinal neovascularization through various pathways,such as the AGE-RAGE signaling pathway in diabetic complications and the MAPK signaling pathway.Finally,molecular docking and MD simulation demonstrated that almost all core active ingredients exhibited satisfactory binding energies to core targets.Conclusions:This study identified a novel TCM prescription and unveiled its multi-component,multi-target and multi-pathway characteristics for treating DR.These findings provide a scientific basis and novel insights into the development of drugs for DR prevention and treatment. 展开更多

目的评估肾周脂肪粘连对后腹腔镜肾部分切除术(PN)的影响。方法回顾2013年10月-2015年12月收治行后腹腔镜PN术的患者共52例。通过术前CT图像和手术操作,依据患者肾周脂肪有无粘连,分成粘连组与无粘连组。比较两组患者的一般临床资料及... 目的评估肾周脂肪粘连对后腹腔镜肾部分切除术(PN)的影响。方法回顾2013年10月-2015年12月收治行后腹腔镜PN术的患者共52例。通过术前CT图像和手术操作,依据患者肾周脂肪有无粘连,分成粘连组与无粘连组。比较两组患者的一般临床资料及围手术期数据之间的差异。结果粘连组与无粘连组在体质指数(BMI)、肿瘤最大径、肾脏热缺血时间、住院时间、美国麻醉师协会评分(ASA)及RENAL评分的比较差异无统计学意义(P>0.05)。粘连组与无粘连组相比年龄大[(59.25±11.03)vs(49.71±11.86)岁,P=0.004],男性患者比例高(75.0%vs 46.4%,P=0.036),高血压患者多(62.5%vs 28.6%,P=0.014),糖尿病患者多(41.7%vs 14.3%,P=0.026),腹腔内脂肪(IAF)多[(2.02±0.47)vs(1.35±0.66)cm,P=0.000],手术时间长[(146.08±45.45)vs(119.32±28.83)min,P=0.017)],术中失血量多[(82.92±45.73)vs(51.79±25.10)ml,P=0.005)]。结论肾周脂肪粘连使后腹腔镜PN术的手术时间及出血量增加,但对肾脏热缺血时间及住院时间无明显影响。这在一定程度上增加了手术过程中分离的难度,但并不增加肿瘤切除的难度。 展开更多

电切镜联合输尿管镜在腺性膀胱炎中寻找困难输尿管开口的应用 被引量:9

目的探讨电切镜联合输尿管镜在腺性膀胱炎中寻找困难输尿管开口的方法,提高内镜下置入双J管的成功率。方法回顾性分析2015年3月-2016年5月,8例腺性膀胱炎合并有困难输尿管开口的患者的临床资料。均为男性患者,年龄38~64岁,平均44.3岁。... 目的探讨电切镜联合输尿管镜在腺性膀胱炎中寻找困难输尿管开口的方法,提高内镜下置入双J管的成功率。方法回顾性分析2015年3月-2016年5月,8例腺性膀胱炎合并有困难输尿管开口的患者的临床资料。均为男性患者,年龄38~64岁,平均44.3岁。电切膀胱内病变,切除深度为黏膜下层和肌层,将输尿管开口区域的坏死组织切除干净,切除深度能够显露肌层的纹理改变。进一步切除肌肉组织,透过薄层肌肉组织能够看见尿液。在导丝引导下置入输尿管镜,镜下观察证实为输尿管管腔。在导丝引导下留置双J管一根。结果 8例患者(16侧)均应用电切镜联合输尿管镜成功置入双J管。手术时间35~205 min,平均时间83.2 min,术中失血量约20~50 ml。术后3~5天拔除尿管。术后1或2天复查腹部平片,显示双侧双J管位置良好,没有出现异位、扭曲等现象。围手术期未出现并发症。结论应用电切镜联合输尿管镜在腺性膀胱炎中寻找困难输尿管开口,能够提高寻找输尿管开口成功的概率,此方法安全、创伤小、避免开放手术。 展开更多

Enhanced tolerance to drought in transgenic rice plants overexpressing C_4 photosynthesis enzymes 被引量:7

Maize-specific pyruvate orthophosphate dikinase(PPDK) was overexpressed in rice independently or in combination with the maize C4-specific phosphoenolpyruvate carboxylase(PCK). The wild-type(WT) cultivar Kitaake and transgenic plants were evaluated in independent field and tank experiments. Three soil moisture treatments,well-watered(WW), moderate drought(MD) and severe drought(SD), were imposed from 9d post-anthesis till maturity. Leaf physiological and biochemical traits, root activities,biomass, grain yield, and yield components in the untransformed WT and two transgenic rice lines(PPDK and PCK) were systematically studied. Compared with the WT, both transgenic rice lines showed increased leaf photosynthetic rate: by 20%–40% under WW, by45%–60% under MD, and by 80%–120% under SD. The transgenic plants produced 16.1%,20.2% and 20.0% higher grain yields than WT under the WW, MD and SD treatments,respectively. Under the same soil moisture treatments, activities of phosphoenolpyruvate carboxylase(PEPC) and carbonic anhydrase(CA) in transgenic plants were 3–5-fold higher than those in WT plants. Compared with ribulose-1,5-bisphosphate carboxylase, activities of PEPC and CA were less reduced under both MD and SD treatments. The transgenic plants also showed higher leaf water content, stomatal conductance, transpiration efficiency, and root oxidation activity and a stronger active oxygen scavenging system than the WT under all soil moisture treatments, especially MD and SD. The results suggest that drought tolerance is greatly enhanced in transgenic rice plants overexpressing C4photosynthesis enzymes. This study was performed under natural conditions and normal planting density to evaluate yield advantages on a field basis. It may open a new avenue to droughttolerance breeding via overexpression of C4enzymes in rice. 展开更多

The catalytic alcohol dehydrogenation to carboxylic acids has been demonstrated as an atom-economic and environmental-friendly synthetic method.However,the reported catalysts either require relatively complicated synthesis or exhibit high activity only in the presence of a solvent.Therefore,it is still in high demand to obtain catalysts that feature facile synthesis and perform well even without any solvent.Herein,a series of nanostructured bimetallic Zn/Co species embedded in N-doped hierarchical porous carbon were fabricated in a facile and green manner.More significantly,the best-performing material(Zn/Co@NC-800)was identified as the first heterogeneous catalyst to enable this transformation under solvent-free conditions.Moreover,the as-prepared composites were comprehensively investigated to de-termine their structural and morphological properties,which rationalized the superiority of Zn/Co@NC-800 over the other synthesized materials.Further studies signified that Co sites(both Co^(0)and Co^(2+))and N species(mainly pyridinic N)were probably involved in this process,functioning as the Lewis acid site and basic site,respectively.Hopefully,this study could provide a simple and sustainable method for carboxylic acid synthesis. 展开更多
