BACKGROUND Type 1 diabetes(T1D),a chronic metabolic and autoimmune disease,seriously endangers human health.In recent years,mesenchymal stem cell(MSC)transplantation has become an effective treatment for diabetes.Mens...BACKGROUND Type 1 diabetes(T1D),a chronic metabolic and autoimmune disease,seriously endangers human health.In recent years,mesenchymal stem cell(MSC)transplantation has become an effective treatment for diabetes.Menstrual bloodderived endometrial stem cells(MenSC),a novel MSC type derived from the decidual endometrium during menstruation,are expected to become promising seeding cells for diabetes treatment because of their noninvasive collection procedure,high proliferation rate and high immunomodulation capacity.AIM To comprehensively compare the effects of MenSC and umbilical cord-derived MSC(UcMSC)transplantation on T1D treatment,to further explore the potential mechanism of MSC-based therapies in T1D,and to provide support for the clinical application of MSC in diabetes treatment.METHODS A conventional streptozotocin-induced T1D mouse model was established,and the effects of MenSC and UcMSC transplantation on their blood glucose and serum insulin levels were detected.The morphological and functional changes in the pancreas,liver,kidney,and spleen were analyzed by routine histological and immunohistochemical examinations.Changes in the serum cytokine levels in the model mice were assessed by protein arrays.The expression of target proteins related to pancreatic regeneration and apoptosis was examined by western blot.RESULTS MenSC and UcMSC transplantation significantly improved the blood glucose and serum insulin levels in T1D model mice.Immunofluorescence analysis revealed that the numbers of insulin+and CD31+cells in the pancreas were significantly increased in MSC-treated mice compared with control mice.Subsequent western blot analysis also showed that vascular endothelial growth factor(VEGF),Bcl2,Bcl-xL and Proliferating cell nuclear antigen in pancreatic tissue was significantly upregulated in MSC-treated mice compared with control mice.Additionally,protein arrays indicated that MenSC and UcMSC transplantation significantly downregulated the serum levels of interferonγand tumor necrosis factorαand upregulated the serum levels of interleukin-6 and VEGF in the model mice.Additionally,histological and immunohistochemical analyses revealed that MSC transplantation systematically improved the morphologies and functions of the liver,kidney,and spleen in T1D model mice.CONCLUSION MenSC transplantation significantly improves the symptoms in T1D model mice and exerts protective effects on their main organs.Moreover,MSC-mediated angiogenesis,antiapoptotic effects and immunomodulation likely contribute to the above improvements.Thus,MenSC are expected to become promising seeding cells for clinical diabetes treatment due to their advantages mentioned above.展开更多

Porous carbon nanofibers (PCNFs) were prepared through electrospinning, pre-oxidation and carbonization with polyacrylonitrile (PAN) as carbon precursor and polymethyl methacrylate (PMMA), CaCO3 as pore-forming agents...Porous carbon nanofibers (PCNFs) were prepared through electrospinning, pre-oxidation and carbonization with polyacrylonitrile (PAN) as carbon precursor and polymethyl methacrylate (PMMA), CaCO3 as pore-forming agents. The structure, morphology, specific surface area and electrochemical performance of the carbon nanofibers were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), N2 adsorption/desorption method and electrochemical tests. Compared with PCNFs without CaCO3, PCNFs(CaCO3 1%) had higher specific surface area, better dispersion of Pt nanoparticles, and the particle size become smaller, which was corresponding with the results of electrochemical performance test. It could be seen in cyclic voltammetry (CV) and linear sweep voltammetry (LSV) test, ECSA of Pt/PCNFs (CaCO3 1%) attained 82 m2?g?1, while that of JM20 and Pt/PCNFs without CaCO3 were 77 m2?g?1 and 60 m2?g?1, respectively. These results revealed that CaCO3 as the second pore-forming agent can further increase the mesoporous number and specific surface area of nanofibers, and can improve the electrochemical properties of Pt catalyst as the support.展开更多

In this paper, nitrogen-doped porous nanofibers (N-PCNF) were fabricated via cost effective electrospinning technique by blending the PI and PAN as precursors, followed by heat treatment procedures. The N-PCNFs were used as support to prepare platinum (Pt) catalyst (Pt/N-PCNFs). SEM figure indicated that the porous structures not only existed on the surface but also in the cross session of the fibers. XPS and TEM displayed that with the help of heteroatoms nitrogen, the fiber had rougher surface and more defective structure, contributing to the dispersion of Pt nanoparticles. The catalytic performance for ORR was evaluated by cyclic voltammetry (CV) and liner sweep voltammetry (LSV) with a rotating disk electrode (RDE). According to the results, Pt/N-PCNF exhibited superior property (more positive onset potential and half-wave potential) than that of JM20. The excellent ORR activity of Pt/N-PCNF was attributed to the enriched nitrogen heteroatoms coordinated within the microstructure which increased the exposure of more active sites and dispersion of Pt nanoparticles.展开更多

Some new sixth-order compact finite difference schemes for Poisson/Helmholtz equations on rectangular domains in both two-and three-dimensions are developed and analyzed.Different from a few sixth-order compact finite...Some new sixth-order compact finite difference schemes for Poisson/Helmholtz equations on rectangular domains in both two-and three-dimensions are developed and analyzed.Different from a few sixth-order compact finite difference schemes in the literature,the finite difference and weight coefficients of the new methods have analytic simple expressions.One of the new ideas is to use a weighted combination of the source term at staggered grid points which is important for grid points near the boundary and avoids partial derivatives of the source term.Furthermore,the new compact schemes are exact for 2D and 3D Poisson equations if the solution is a polynomial less than or equal to 6.The coefficient matrices of the new schemes are M-matrices for Helmholtz equations with wave number K≤0,which guarantee the discrete maximum principle and lead to the convergence of the new sixth-order compact schemes.Numerical examples in both 2D and 3D are presented to verify the effectiveness of the proposed schemes.展开更多
