Maintaining glutamate homeostasis after hypoxic ischemia is important for synaptic function and neural cell activity,and regulation of glutamate transport between astrocyte and neuron is one of the important modalities for reducing glutamate accumulation.However,further research is needed to investigate the dynamic changes in and molecular mechanisms of glutamate transport and the effects of glutamate transport on synapses.The aim of this study was to investigate the regulatory mechanisms underlying Notch pathway mediation of glutamate transport and synaptic plasticity.In this study,Yorkshire neonatal pigs(male,age 3 days,weight 1.0–1.5 kg,n=48)were randomly divided into control(sham surgery group)and five hypoxic ischemia subgroups,according to different recovery time,which were then further subdivided into subgroups treated with dimethyl sulfoxide or a Notch pathway inhibitor(N-[N-(3,5-difluorophenacetyl-l-alanyl)]-S-phenylglycine t-butyl ester).Once the model was established,immunohistochemistry,immunofluorescence staining,and western blot analyses of Notch pathway-related proteins,synaptophysin,and glutamate transporter were performed.Moreover,synapse microstructure was observed by transmission electron microscopy.At the early stage(6–12 hours after hypoxic ischemia)of hypoxic ischemic injury,expression of glutamate transporter excitatory amino acid transporter-2 and synaptophysin was downregulated,the number of synaptic vesicles was reduced,and synaptic swelling was observed;at 12–24 hours after hypoxic ischemia,the Notch pathway was activated,excitatory amino acid transporter-2 and synaptophysin expression was increased,and the number of synaptic vesicles was slightly increased.Excitatory amino acid transporter-2 and synaptophysin expression decreased after treatment with the Notch pathway inhibitor.This suggests that glutamate transport in astrocytes-neurons after hypoxic ischemic injury is regulated by the Notch pathway and affects vesicle release and synaptic plasticity through the expression of synaptophysin. 展开更多

Several consecutive extreme cold events impacted China during the first half of winter 2020/21,breaking the low-temperature records in many cities.How to make accurate climate predictions of extreme cold events is still an urgent issue.The synergistic effect of the warm Arctic and cold tropical Pacific has been demonstrated to intensify the intrusions of cold air from polar regions into middle-high latitudes,further influencing the cold conditions in China.However,climate models failed to predict these two ocean environments at expected lead times.Most seasonal climate forecasts only predicted the 2020/21 La Niña after the signal had already become apparent and significantly underestimated the observed Arctic sea ice loss in autumn 2020 with a 1-2 month advancement.In this work,the corresponding physical factors that may help improve the accuracy of seasonal climate predictions are further explored.For the 2020/21 La Niña prediction,through sensitivity experiments involving different atmospheric-oceanic initial conditions,the predominant southeasterly wind anomalies over the equatorial Pacific in spring of 2020 are diagnosed to play an irreplaceable role in triggering this cold event.A reasonable inclusion of atmospheric surface winds into the initialization will help the model predict La Niña development from the early spring of 2020.For predicting the Arctic sea ice loss in autumn 2020,an anomalously cyclonic circulation from the central Arctic Ocean predicted by the model,which swept abnormally hot air over Siberia into the Arctic Ocean,is recognized as an important contributor to successfully predicting the minimum Arctic sea ice extent. 展开更多

Skyrmion bags are spin structures with arbitrary topological charges, each of which is composed of a big skyrmion and several small skyrmions. In this work, by using an in-plane alternating current(AC) magnetic field, we investigate the spinwave modes of skyrmion bags, which behave differently from the clockwise(CW) rotation mode and the counterclockwise(CCW) rotation mode of skyrmions because of their complex spin topological structures. The in-plane excitation power spectral density shows that each skyrmion bag possesses four resonance frequencies. By further studying the spin dynamics of a skyrmion bag at each resonance frequency, the four spin-wave modes, i.e., a CCW-CW mode, two CW-breathing modes with different resonance strengths, and an inner CCW mode, appear as a composition mode of outer skyrmion–inner skyrmions. Our results are helpful in understanding the in-plane spin excitation of skyrmion bags, which may contribute to the characterization and detection of skyrmion bags, as well as the applications in logic devices. 展开更多

目的通过对宫颈癌根治术患者的用血情况及相关因素进行分析,探讨宫颈癌根治术中输血的风险因素,为术前评估术中用血风险提供参考。方法选取2013年7月1日—2016年11月30日中国医科大学附属盛京医院行宫颈癌根治术且进行术前交叉配型的患... 目的通过对宫颈癌根治术患者的用血情况及相关因素进行分析,探讨宫颈癌根治术中输血的风险因素,为术前评估术中用血风险提供参考。方法选取2013年7月1日—2016年11月30日中国医科大学附属盛京医院行宫颈癌根治术且进行术前交叉配型的患者427例,排除记录不全47例,将剩余380例患者按术中是否输注滤白红细胞分为输血组(53例)和未输血组(327例),比较两组患者基本信息、术前实验室检查、手术相关因素、肿瘤相关因素及术前合并疾病等差异,并进行二元Logistic回归分析。结果各类因素均进行组间比较,其中是否合并化疗、术前血红蛋白、术前血小板、手术方式及肿瘤分期在两组间的差异有统计学意义(P <0.05)。多因素分析结果显示,是否化疗、术前血红蛋白和血小板水平是术中输血的危险因素(P <0.05)。结论影响宫颈癌根治术术中用血的相关危险因素主要有术前是否化疗、术前血红蛋白及血小板水平。 展开更多

BACKGROUND Penetrating neck injuries require prompt recognition,diagnosis and management of critical airways.This case demonstrates an emergent situation that a“medical negligence”was avoided with the aid of end-tidal carbon dioxide(ETCO2)waveform.CASE SUMMARY We report a case of malposition of the endotracheal tube into the right hemithoracic cavity for cervical knife trauma,resulting in pneumothorax.Tube placement was not confirmed during emergency airway management,and the patient was directly transferred to the emergency operation room.Assisted by ETCO2 and imaging examinations,the anesthetist timely noticed the absence of ETCO2 waveform and resolved this urgent situation before anesthesia induction.CONCLUSION This case emphasizes the necessity of ETCO2 waveform and/or X-ray confirmation of endotracheal intubation even in emergent situations. 展开更多

Preserving GABAergic interneurons in acute brain slices of mice using the N-methyl-D-glucamine-based artificial cerebrospinal fluid method

Defects in the function and development of GABAergic interneurons have been linked to psychiatric disorders, so preservation of these interneurons in brain slices is important for successful electrophysiological recording in various ex vivo methods. However, it is difficult to maintain the activity and morphology of neurons in slices from mice of >30 days old. Here we evaluated the N-methyl-D-glucamine(NMDG)-based artificial cerebrospinal fl uid(a CSF) method for the preservation of interneurons in slices from mice of up to ~6 months old and discussed the steps that may affect their quality during slicing. We found that the NMDG-a CSF method rescued more cells than sucrose-a CSF and successfully preserved different types of interneurons including parvalbumin- and somatostatin-positive interneurons. In addition, both the chemical and electrical synaptic signaling of interneurons were maintained. These results demonstrate that the NMDG-a CSF method is suitable for the preservation of interneurons, especially in studies of gap junctions. 展开更多

近年来,在水处理领域中,利用高级氧化技术去除水中有机污染物引起了研究者的广泛关注.双氧水、过一硫酸盐(PMS)和过二硫酸盐(PDS)等氧化剂可以被含Fe,Mn,Cu和Co等过渡金属的催化剂活化,并通过产生的活性氧物质、高价金属或直接电子转移... 近年来,在水处理领域中,利用高级氧化技术去除水中有机污染物引起了研究者的广泛关注.双氧水、过一硫酸盐(PMS)和过二硫酸盐(PDS)等氧化剂可以被含Fe,Mn,Cu和Co等过渡金属的催化剂活化,并通过产生的活性氧物质、高价金属或直接电子转移等途径实现污染物高效降解.其中,Co被认为是活化过硫酸盐最有效的物种之一.粉体ZIF-67和ZIF-9等Co基金属有机框架(Co-MOFs)在水体污染物去除方面得到了广泛关注,但面临稳定性差及回收困难等问题.经熔融-淬火工艺所制备的无定形MOF玻璃颗粒克服了粉体材料难以回收再利用的缺点,同时保持了其中催化位点的高活性.本文利用MOF玻璃作为非均相催化剂实现高级氧化降解水体微污染物,研究了其催化活化性能及反应机理.本文采用熔融-淬火方式将ZIF-62(Co)晶体转化成ZIF-62(Co)玻璃(ag-ZIF-62(Co)),并研究了ag-ZIF-62(Co)活化PMS降解磺胺甲恶唑(SMX)等微污染物的性能.粉末X射线衍射、傅里叶变换红外光谱、X射线光电子能谱(XPS)等表征结果表明,熔融-淬火过程使结构有序的ZIF-62(Co)晶体变成无定形的玻璃态ag-ZIF-62(Co),但并未破坏其中的有机配体和Co-N键.ag-ZIF-62(Co)可以在较宽的溶液pH范围(3.0-9.0)内高效催化活化PMS降解SMX等污染物.由于ag-ZIF-62(Co)玻璃活化PMS产生的SO_(4^(•-))自由基有可能被溶液中碳酸氢根消耗,所以溶液中的碳酸氢根浓度越高,SMX的降解效率越差.水中的氯离子会诱导产生HClO,从而提高ag-ZIF-62(Co)活化PMS降解SMX的效率.其他阴离子(如硝酸根、硫酸根与磷酸二氢根等)对ag-ZIF-62(Co)活化PMS降解SMX的效率无明显抑制或促进作用.利用自来水配制模拟含SMX废水,研究其中存在的多重阴离子和阳离子对ag-ZIF-62(Co)活化PMS降解SMX性能的影响.结果表明,ag-ZIF-62(Co)在30 min内对低浓度(1.0 mg/L)和高浓度(5.0 mg/L)SMX的降解效率均保持较高的水平.捕捉实验、电子自旋共振和自由基定量等实验证明了硫酸根自由基与单线态氧是降解SMX的主要活性物种.结合电喷雾质谱结果提出了SMX的降解路径,并评估了中间产物的毒性.利用XPS分析了ag-ZIF-62(Co)反应前后Co的价态变化,并结合其他实验结果阐明了硫酸根自由基与单线态氧的产生路径.为探究ag-ZIF-62(Co)在实际应用中的潜在价值,搭建了固定床反应器并填充ag-ZIF-62(Co).结果表明,ag-ZIF-62(Co)在反应器中能连续120 h活化PMS并高效降解SMX.此外,ag-ZIF-62(Co)还可以高效降解多种微污染物(如阿特拉津、双酚A、磷酸氯喹、氧氟沙星和罗丹明B等).ag-ZIF-62(Co)玻璃在水体系中活化PMS时,Co离子溶出低于GB 25467-20标准要求,有效地避免了Co离子带来的环境压力.综上所述,本文报道了ag-ZIF-62(Co)活化PMS降解水中SMX的性能,分析了不同因素对SMX降解效率的影响,并探究了活化PMS降解SMX的机理.通过搭建了固定床反应器,实现了对SMX的长时间高效降解,显示了ag-ZIF-62(Co)在AOPs中的应用潜力.本研究不仅为MOF玻璃在环境污染控制中的应用提供了新思路,也为后续MOF玻璃作为催化剂在环境催化领域的发展提供参考. 展开更多

背景:一些观察性研究显示,肝肾综合征(HRS)患者在经过药物治疗后再行原位肝移植,可以获得与非HRS患者相似的移植效果。方法:检索PubMed、Elsevier、Medline及Cochrane数据库1999年1月1日至2018年2月24日间的相关文献。主要结局指标是HR... 背景:一些观察性研究显示,肝肾综合征(HRS)患者在经过药物治疗后再行原位肝移植,可以获得与非HRS患者相似的移植效果。方法:检索PubMed、Elsevier、Medline及Cochrane数据库1999年1月1日至2018年2月24日间的相关文献。主要结局指标是HRS的逆转,次要结局指标是血肌酐和血钠的变化。我们基于Bayesian方法学通过网络荟萃分析评估了不同药物治疗方案。结果:共计24篇文献1419例病例纳入分析,评估了7种HRS的药物治疗方案。对于逆转HRS最有效的治疗方案是特利加压素+白蛋白,去甲肾上腺素+白蛋白,以及特利加压素单药治疗,其累计排名曲线下面积(SUCRA)分别为0.086、0.151和0.451。对于降低血肌酐最有效的两种治疗方案是多巴胺+呋塞米+白蛋白(等级概率0.620)和特利加压素+白蛋白(等级概率0.570)。对于提高血钠水平,最理想的治疗方案是奥曲肽+米多君+白蛋白(等级概率0.800),其次是特利加压素+白蛋白(等级概率0.544)。结论:降低HRS患者的血肌酐水平,可优先考虑特利加压素+白蛋白和多巴胺+呋塞米+白蛋白这两种治疗方案;而奥曲肽+米多君+白蛋白则是提高其血钠水平的最有效方案。特利加压素+白蛋白显示出了既能降低血肌酐又能提高血钠的综合疗效。 展开更多
