However, the precise role of conophylline in the growth and survival of immortalized pancreatic mesenchymal stem cells (iPMSCs) derived from fetal porcine pancreas were not understood at present. To determine whether this molecule is involved in controlling the proliferation of iPMSCs, we examined the effects of conophylline on iPMSCs. We found that conophylline can robustly stimulate iPMSCs proliferation, even promote their potential differentiation into islet-like clusters analyzed by cell counting, morphology, RT-PCR and real-time PCR, Western blotting, glucose-stimulated insulin release and insulin content analysis. The effects of conophylline were inhibited by LY294002, which is the inhibitor of the PI3K pathway. These results suggest that conophylline plays a key role in the regulation of cell mass proliferation, maintenance of the undifferentiated state of iPMSCs and also promotes iPMSCs differentiated into insulin-producing cells.展开更多
有别于传统分布式系统,实时协同编辑系统强调自然和谐的人人交互和人机交互,允许不同地点的协同用户同时编辑同一共享对象.为了给协同用户提供良好的响应性,实时协同编辑系统有必要采用全复制式体系结构,但又给共享对象的一致性维护问...有别于传统分布式系统,实时协同编辑系统强调自然和谐的人人交互和人机交互,允许不同地点的协同用户同时编辑同一共享对象.为了给协同用户提供良好的响应性,实时协同编辑系统有必要采用全复制式体系结构,但又给共享对象的一致性维护问题带来巨大挑战,这一直是协同计算学科的研究热点.近年研究重点逐步从结果一致性发展到操作意图一致性.该文以操作意图一致性为主线,对支持操作意图一致性的实时协同编辑算法进行深入和全面的比较、分析和总结.首先,以Lamport事件偏序关系为起点,对协同编辑系统的因果关系、简单并发关系和偏并发关系概念进行整理.同时,给出实时协同编辑系统中全序关系的分类,操作的全序和操作对象的全序.在对全序关系和优先级进行剖析的基础上,阐述了实时协同编辑系统中的三类一致性模型:CC(Causality-preservation,Convergence)模型、CCI(Causality-preservation,Convergence,Intention-preservation)模型和CA(Causalitypreservation,Admissibility)模型.特别地,文中按照因果一致性、结果一致性和操作意图一致性的分类,给出操作意图一致性的维护路线图.然后,分别综述了各类实时协同编辑算法的研究进展和现状,包括OT(Operational Transformation)算法、AST(Address Space Transformation)算法和CRDT(Commutative Replicated Data Type)算法.进一步,文中给出了各类操作意图一致性算法的基本原理和执行框架,并基于一个代表性协同工作场景和算例对典型算法进行详细解析.接下来,文中从操作意图一致性、支持操作意图一致性的典型算法、操作转换函数和算法时间复杂度这4个关键方面对实时协同编辑算法进行归纳和对比.最后对全文小结,并指出为了进一步发展实时协同编辑系统,未来还需要在优先级策略、算法伸缩性、复杂操作语义类型、粗粒度操作对象以及选择性撤销机制等方面深入开展一些研究工作.展开更多
目的系统评价来曲唑(LE)与克罗米芬(CC)对多囊卵巢综合征(PCOS)患者的疗效,为临床用药提供依据。方法全面检索中国知网(CNKI)、万方(Wanfang)、维普(VIP)、中医生物医学文献数据库(CBM)、PubMed、Embase、Cochrane Library、Web of Scie...目的系统评价来曲唑(LE)与克罗米芬(CC)对多囊卵巢综合征(PCOS)患者的疗效,为临床用药提供依据。方法全面检索中国知网(CNKI)、万方(Wanfang)、维普(VIP)、中医生物医学文献数据库(CBM)、PubMed、Embase、Cochrane Library、Web of Science数据库,检索时间为建库至2020年2月,检索内容为LE与CC治疗PCOS的临床随机对照试验(RCT),根据纳入及排除标准进行筛选,并由2名评价员独立对纳入研究进行资料提取及方法学质量评估,采用Cochrane系统评价软件RevMan5.3进行Meta分析,采用GRADEprofiler软件对证据质量等级进行分级。结果本研究纳入22篇RCT研究,共计PCOS患者3506例。Meta分析结果显示,LE能显著提高PCOS患者的排卵率[RR=1.14,95%CI(1.05,1.24),P=0.003]、妊娠率[RR=1.37,95%CI(1.24,1.51),P<0.00001]和活产率[RR=1.55,95%CI(1.31,1.84),P<0.00001],增加HCG日子宫内膜厚度[MD=1.75,95%CI(0.46,3.05),P=0.008],降低OHSS发生率[RR=0.18,95%CI(0.06,0.56),P=0.003];LE组的成熟卵泡数[MD=-0.55,95%CI(-0.79,-0.31),P<0.00001]及HCG日E 2水平[MD=-185.84,95%CI(-233.60,-138.08),P<0.00001]相较于CC组更低,差异均有统计学意义(P<0.05);两组在流产率、多胎妊娠率及子宫动脉血流指数方面差异无统计学意义(P>0.05)。根据GRADE标准,活产率、OHSS发生率为中等质量证据,妊娠率、多胎妊娠率、流产率、成熟卵泡数为低质量证据,排卵率、HCG日子宫内膜厚度、HCG日E 2水平、子宫动脉阻力指数(RI)、子宫动脉搏动指数(PI)为极低质量证据。结论LE的促排卵疗效总体优于CC,但需要大规模,设计更严谨的多中心临床试验加以验证。展开更多
