BACKGROUND Gastrointestinal stromal tumor(GIST)is a common neoplasm with high rates of recurrence and metastasis,and its therapeutic efficacy is still not ideal.There is an unmet need to find new molecular therapeutic...BACKGROUND Gastrointestinal stromal tumor(GIST)is a common neoplasm with high rates of recurrence and metastasis,and its therapeutic efficacy is still not ideal.There is an unmet need to find new molecular therapeutic targets for GIST.TATA-boxbinding protein-associated factor 15(TAF15)contributes to the progress of various tumors,while the role and molecular mechanism of TAF15 in GIST progression are still unknown.AIM To explore new molecular therapeutic targets for GIST and understand the biological role and underlying mechanisms of TAF15 in GIST progression.METHODS Proteomic analysis was performed to explore the differentially expressed proteins in GIST.Western blotting and immunohistochemical analysis were used to verify the expression level of TAF15 in GIST tissues and cell lines.Cell counting kit-8,colony formation,wound-healing and transwell assay were executed to detect the ability of TAF15 on cell proliferation,migration and invasion.A xenograft mouse model was applied to explore the role of TAF15 in the progression of GIST.Western blotting was used to detect the phosphorylation level and total level of RAF1,MEK and ERK1/2.RESULTS A total of 1669 proteins were identified as differentially expressed proteins with 762 upregulated and 907 downregulated in GIST.TAF15 was selected for the further study because of its important role in cell proliferation and migration.TAF15 was significantly over expressed in GIST tissues and cell lines.Overexpression of TAF15 was associated with larger tumor size and higher risk stage of GIST.TAF15 knockdown significantly inhibited the cell proliferation and migration of GIST in vitro and suppressed tumor growth in vivo.Moreover,the inhibition of TAF15 expression significantly decreased the phosphorylation level of RAF1,MEK and ERK1/2 in GIST cells and xenograft tissues,while the total RAF1,MEK and ERK1/2 had no significant change.CONCLUSION TAF15 is over expressed in GIST tissues and cell lines.Overexpression of TAF15 was associated with a poor prognosis of GIST patients.TAF15 promotes cell proliferation and migration in GIST via the activation of the RAF1/MEK/ERK signaling pathway.Thus,TAF15 is expected to be a novel latent molecular biomarker or therapeutic target of GIST.展开更多
Background:Bupleuri Radix is a common Chinese medicinal material in traditional Chinese medicine.Currently,the therapeutic effect of treating schizophrenia is relatively well understood.However,there are fewer studies...Background:Bupleuri Radix is a common Chinese medicinal material in traditional Chinese medicine.Currently,the therapeutic effect of treating schizophrenia is relatively well understood.However,there are fewer studies examining the underlying mechanisms of its treatment.The objective of the study was to investigate the primary mechanisms of Bupleuri Radix in treating schizophrenia through network pharmacology and clinical validation.Method:Network pharmacology revealed possible molecular mechanisms,followed by clinical verification.Sixty-seven schizophrenia patients undergoing treatment at the Hunan Brain Hospital between October and November 2022 were recruited and randomly divided into the olanzapine group and the olanzapine+Bupleuri Radix group.Additionally,32 healthy people undergoing physical examinations during the same period were included as the control group.The patient’s positive and negative symptom scale scores were compared.qPCR was used to detect the mRNA expression levels of ESR1,mTOR,EIF4E,and SMAD4 in peripheral blood.Results:Through network pharmacological analysis,it was concluded in this study that Bupleuri Radix might regulate the mTOR,PI3K-Akt,and HIF-1 signaling pathways.Clinical experiments indicated that compared with before treatment,the positive and negative symptom scale scores and total scores of the two treatment groups were significantly decreased after treatment(P<0.01).In addition,the positive and negative symptom scale scores and total scores in the olanzapine+Bupleuri Radix group were significantly decreased(P<0.01)compared to the olanzapine group after treatment.Before treatment,ESR1 mRNA expression levels in peripheral blood were significantly higher in the two treatment groups than in the control group,whereas the mRNA expression levels of mTOR,EIF4E,and SMAD4 in peripheral blood were significantly lower(P<0.01).The mRNA expression levels of mTOR,EIF4E,and SMAD4 in peripheral blood were significantly higher after therapy than before treatment,whereas the mRNA expression levels of ESR1 in peripheral blood were significantly lower(P<0.01).After therapy,the olanzapine+Bupleuri Radix group’s mRNA expression levels of mTOR,EIF4E,and SMAD4 were significantly higher than those of the olanzapine group,whereas the mRNA expression levels of ESR1 were significantly lower(P<0.01).Conclusion:The mechanism of Bupleuri Radix’s therapeutic efficacy in schizophrenia may involve the up-regulation of mTOR,EIF4E,and SMAD4 mRNA expression and the down-regulation of ESR1 mRNA expression in peripheral blood.展开更多

Tectonic processes involving amalgamations of microblocks along zones of ocean closure represented by granite-greenstone belts(GGB) were fundamental in building the Earth's early continents. The crustal growth and...Tectonic processes involving amalgamations of microblocks along zones of ocean closure represented by granite-greenstone belts(GGB) were fundamental in building the Earth's early continents. The crustal growth and cratonization of the North China Craton(NCC) are correlated to the amalgamation of microblocks welded by 2.75-2.6 Ga and ~2.5 Ga GGBs. The lithological assemblages in the GGBs are broadly represented by volcano-sedimentary sequences, subduction-collision related granitoids and bimodal volcanic rocks(basalt and dacite) interlayered with minor komatiites and calc-alkalic volcanic rocks(basalt, andesite and felsic rock). The geochemical features of meta-basalts in the major GGBs of the NCC display affinity with N-MORB, E-MORB, OIB and calc-alkaline basalt, suggesting that the microblocks were separated by oceanic realm. The granitoid rocks display arc signature with enrichment of LILE(K,Rb, Sr, Ba) and LREE, and depletion of HFSE(Nb, Ta, Th, U, Ti) and HREE, and fall in the VAG field. The major mineralization includes Neoarchean BIF-type iron and VMS-type Cu-Zb deposits and these,together with the associated supracrustal rocks possibly formed in back-arc basins or arc-related oceanic slab subduction setting with or without input from mantle plumes. The 2.75-2.60 Ga TTG rocks,komatiites, meta-basalts and metasedimentary rocks in the Yanlingguan GGB are correlated to the upwelling mantle plume with eruption close to the continental margin within an ocean basin. The volcanosedimentary rocks and granitoid rocks in the late Neoarchean GGBs display formation ages of 2.60-2.48 Ga, followed by metamorphism at 2.52-2.47 Ga, corresponding to a typical modern-style subduction-collision system operating at the dawn of Proterozoic. The late Neoarchean komatiite(Dongwufenzi GGB), sanukitoid(Dongwufenzi GGB and Western Shandong GGB), BIF(Zunhua GGB) and VMS deposit(Hongtoushan-Qingyuan-Helong GGB) have closer connection to a combined process of oceanic slab subduction and mantle plume. The Neoarchean cratonization of the NCC appears to have involved two stages of tectonic process along the 2.75-2.6 Ga GGB and ~2.5 Ga GGBs, the former involve plume-arc interaction process, and the latter involving oceanic lithospheric subduction, with or without arcplume interaction.展开更多

The Southern Great Xing’an Range (SGXR) which forms part of the eastern segment of the Central Asian Orogenic Belt (CAOB) is known as one of the most important Cu-Mo-Pb-Zn-Ag-Au metallogenic belts in China,hosting a ...The Southern Great Xing’an Range (SGXR) which forms part of the eastern segment of the Central Asian Orogenic Belt (CAOB) is known as one of the most important Cu-Mo-Pb-Zn-Ag-Au metallogenic belts in China,hosting a number of porphyry Mo (Cu),skarn Fe (Sn),epithermal Au-Ag,and hydrothermal veintype Ag-Pb-Zn ore deposits.Here we investigate the Bianjiadayuan hydrothermal vein-type Ag-Pb-Zn ore deposit in the southern part of the SGXR.Porphyry Sn ±Cu ± Mo mineralization is also developed to the west of the Ag-Pb-Zn veins in the ore field.We identify a five-stage mineralization process based on field and petrologic studies including (i) the early porphyry mineralization stage,(ii) main porphyry mineralization stage,(iii) transition mineralization stage,(iv) vein-type mineralization stage and (v) late mineralization stage.Pyrite is the predominant sulfide mineral in all stages except in the late mineralization stage,and we identify corresponding four types of pyrites: Py1 is medium-grained subhedral to euhedral occurring in the early barren quartz vein;Py2 is medium- to fine-grained euhedral pyrite mainly coexisting with molybdenite,chalcopyrite,minor sphalerite and galena;Py3 is fine-grained,subhedral to irregular pyrite and displays cataclastic textures with micro-fractures;Py4 occurs as euhedral microcrystals and forms irregularly shaped aggregate with sphalerite and galena.LA-ICP-MS trace element analyses of pyrite show that Cu,Pb,Zn,Ag,Sn,Cd and Sb are partitioned into pyrite as structurally bound metals or mineral micro/nano-inclusions,whereas Co,Ni,As and Se enter the lattice via isomorphism in all types of pyrite.The Cu,Zn,Ag,Cd concentrations gradually increase from Py1 to Py4,which we correlate with cooling and mixing of ore-forming fluid with meteoric water.Py2 contains the highest contents of Co,Ni,Se,Te and Bi,suggesting high temperature conditions for the porphyry mineralization stage.Ratios of Co/Ni (0.03-10.79,average 2.13) and sulphur isotope composition of sulfide indicate typical hydrothermal origin for pyrites.The δ^34SCDT values of Py1 (0.42‰-1.61‰,average 1.16‰),Py2 (-1.23‰ to 0.82‰,average 0.35‰),Py3 (-0.36‰ to 2.47‰,average 0.97‰),Py4 (2.51‰-3.72‰,average 3.06‰),and other sulfides are consistent with those of typical porphyry deposit (-5‰ to 5‰),indicating that the Pb-Zn polymetallic mineralization in the Bianjiadayuan deposit is genetically linked to the Yanshanian (JurassiceCretaceous) magmatic-hydrothermal events.Variations of d34S values are ascribed to the changes in physical and chemical conditions during the evolution and migration of the ore-forming fluid.We propose that the high Sn content of pyrite in the Bianjiadayuan hydrothermal vein-type PbeZn polymetallic deposit can be used as a possible pathfinder to prospect for Sn mineralization in the surrounding area or deeper level of the ore field in this region.展开更多
Post-stroke spasticity is associated with restriction in the range of motion of the shoulder.Reducing muscular dystrophy may help relieve muscular dysfunction in patients with post-stroke shoulder spasticity.Dry needl...Post-stroke spasticity is associated with restriction in the range of motion of the shoulder.Reducing muscular dystrophy may help relieve muscular dysfunction in patients with post-stroke shoulder spasticity.Dry needle therapy is a method of needling the trigger points using a syringe needle without the use of a drug.Dry needle therapy is commonly used for pain at the shoulder,neck,waist,and back.In this case study,a 62-year-old male patient affected with cerebral hemorrhage of the right frontal lobe had received rehabilitative treatment for 12 years.However,he still experienced shoulder spasticity.The patient received daily dry needling at the trigger points of infraspinatus,teres minor,posterior deltoid,and pectoralis major on 9 days.After the first and ninth treatment,the Modified Ashworth Scale and the passive range of motion of the shoulder was used to assess the effect of the treatment.The spasticity and range of motion of the shoulder showed obvious improvement.These results indicate that dry needling at the myofascial trigger points can effectively treat chronic poststroke shoulder spasticity.展开更多

Focal cerebral ischemia and repe...This study examines the neuroprotective effects and mechanisms of action of total saponins from Rubus parvifolius L. (TSRP) on focal cerebral ischemia and reperfusion injury in rats. Focal cerebral ischemia and reperfusion injury was performed in rats using the suture method. The results indicate that intragastric injection of TSRP, at 5, 10 and 20 mg/kg, could decrease neurological impairment, reduce cerebral infarct volume, diminish pathological changes, and significantly inhibit the apoptosis of neurons surrounding the ischemic area. In addition, TSRP upregulated the expression of the anti-apoptotic factor Bcl-2, at the protein and mRNA levels, and it downregulated the expression of the pro-apoptotic factor Bax, at the protein and mRNA levels. These findings indicate that TSRP protects against cerebral ischemia/reperfusion injury, and that it may do so by regulating the expression of Bcl-2 and Bax.展开更多

Retinal flat mounts and sections were double-labeled by fluorogold(FG) and β-III-Tubulin antibody or single-labeled by β-III-Tubulin antibody,then RGCs were counted and compared respectively.· RESULTS: IOP of the injected eyes were elevated significantly and reached the peak at 22.8 ±0.7 mm Hg by day 14 after injection, then dropped to 11.3 ±0.7 mm Hg by day 28. RGC numbers counted by FG labeling and β-III- Tubulin antibody labeling were 64 807 ± 4930 and64 614 ±5054 respectively in the control group, with no significant difference. By day 14, RGCs in the experimental group decreased significantly compared to the control group, but there was no significant difference between the FG labeling counting and the β-III-Tubulin antibody labeling counting either in the experimental group or in the control group. The result was similar by day 28, with further RGC loss.·CONCLUSION: Our result suggested that the β-III-Tubulin protein was not affected by IOP elevation and can be used as a reliable marker for RGC in experimental models of glaucoma.展开更多

文摘Tectonic processes involving amalgamations of microblocks along zones of ocean closure represented by granite-greenstone belts(GGB) were fundamental in building the Earth's early continents. The crustal growth and cratonization of the North China Craton(NCC) are correlated to the amalgamation of microblocks welded by 2.75-2.6 Ga and ~2.5 Ga GGBs. The lithological assemblages in the GGBs are broadly represented by volcano-sedimentary sequences, subduction-collision related granitoids and bimodal volcanic rocks(basalt and dacite) interlayered with minor komatiites and calc-alkalic volcanic rocks(basalt, andesite and felsic rock). The geochemical features of meta-basalts in the major GGBs of the NCC display affinity with N-MORB, E-MORB, OIB and calc-alkaline basalt, suggesting that the microblocks were separated by oceanic realm. The granitoid rocks display arc signature with enrichment of LILE(K,Rb, Sr, Ba) and LREE, and depletion of HFSE(Nb, Ta, Th, U, Ti) and HREE, and fall in the VAG field. The major mineralization includes Neoarchean BIF-type iron and VMS-type Cu-Zb deposits and these,together with the associated supracrustal rocks possibly formed in back-arc basins or arc-related oceanic slab subduction setting with or without input from mantle plumes. The 2.75-2.60 Ga TTG rocks,komatiites, meta-basalts and metasedimentary rocks in the Yanlingguan GGB are correlated to the upwelling mantle plume with eruption close to the continental margin within an ocean basin. The volcanosedimentary rocks and granitoid rocks in the late Neoarchean GGBs display formation ages of 2.60-2.48 Ga, followed by metamorphism at 2.52-2.47 Ga, corresponding to a typical modern-style subduction-collision system operating at the dawn of Proterozoic. The late Neoarchean komatiite(Dongwufenzi GGB), sanukitoid(Dongwufenzi GGB and Western Shandong GGB), BIF(Zunhua GGB) and VMS deposit(Hongtoushan-Qingyuan-Helong GGB) have closer connection to a combined process of oceanic slab subduction and mantle plume. The Neoarchean cratonization of the NCC appears to have involved two stages of tectonic process along the 2.75-2.6 Ga GGB and ~2.5 Ga GGBs, the former involve plume-arc interaction process, and the latter involving oceanic lithospheric subduction, with or without arcplume interaction.

文摘The Southern Great Xing’an Range (SGXR) which forms part of the eastern segment of the Central Asian Orogenic Belt (CAOB) is known as one of the most important Cu-Mo-Pb-Zn-Ag-Au metallogenic belts in China,hosting a number of porphyry Mo (Cu),skarn Fe (Sn),epithermal Au-Ag,and hydrothermal veintype Ag-Pb-Zn ore deposits.Here we investigate the Bianjiadayuan hydrothermal vein-type Ag-Pb-Zn ore deposit in the southern part of the SGXR.Porphyry Sn ±Cu ± Mo mineralization is also developed to the west of the Ag-Pb-Zn veins in the ore field.We identify a five-stage mineralization process based on field and petrologic studies including (i) the early porphyry mineralization stage,(ii) main porphyry mineralization stage,(iii) transition mineralization stage,(iv) vein-type mineralization stage and (v) late mineralization stage.Pyrite is the predominant sulfide mineral in all stages except in the late mineralization stage,and we identify corresponding four types of pyrites: Py1 is medium-grained subhedral to euhedral occurring in the early barren quartz vein;Py2 is medium- to fine-grained euhedral pyrite mainly coexisting with molybdenite,chalcopyrite,minor sphalerite and galena;Py3 is fine-grained,subhedral to irregular pyrite and displays cataclastic textures with micro-fractures;Py4 occurs as euhedral microcrystals and forms irregularly shaped aggregate with sphalerite and galena.LA-ICP-MS trace element analyses of pyrite show that Cu,Pb,Zn,Ag,Sn,Cd and Sb are partitioned into pyrite as structurally bound metals or mineral micro/nano-inclusions,whereas Co,Ni,As and Se enter the lattice via isomorphism in all types of pyrite.The Cu,Zn,Ag,Cd concentrations gradually increase from Py1 to Py4,which we correlate with cooling and mixing of ore-forming fluid with meteoric water.Py2 contains the highest contents of Co,Ni,Se,Te and Bi,suggesting high temperature conditions for the porphyry mineralization stage.Ratios of Co/Ni (0.03-10.79,average 2.13) and sulphur isotope composition of sulfide indicate typical hydrothermal origin for pyrites.The δ^34SCDT values of Py1 (0.42‰-1.61‰,average 1.16‰),Py2 (-1.23‰ to 0.82‰,average 0.35‰),Py3 (-0.36‰ to 2.47‰,average 0.97‰),Py4 (2.51‰-3.72‰,average 3.06‰),and other sulfides are consistent with those of typical porphyry deposit (-5‰ to 5‰),indicating that the Pb-Zn polymetallic mineralization in the Bianjiadayuan deposit is genetically linked to the Yanshanian (JurassiceCretaceous) magmatic-hydrothermal events.Variations of d34S values are ascribed to the changes in physical and chemical conditions during the evolution and migration of the ore-forming fluid.We propose that the high Sn content of pyrite in the Bianjiadayuan hydrothermal vein-type PbeZn polymetallic deposit can be used as a possible pathfinder to prospect for Sn mineralization in the surrounding area or deeper level of the ore field in this region.
文摘Post-stroke spasticity is associated with restriction in the range of motion of the shoulder.Reducing muscular dystrophy may help relieve muscular dysfunction in patients with post-stroke shoulder spasticity.Dry needle therapy is a method of needling the trigger points using a syringe needle without the use of a drug.Dry needle therapy is commonly used for pain at the shoulder,neck,waist,and back.In this case study,a 62-year-old male patient affected with cerebral hemorrhage of the right frontal lobe had received rehabilitative treatment for 12 years.However,he still experienced shoulder spasticity.The patient received daily dry needling at the trigger points of infraspinatus,teres minor,posterior deltoid,and pectoralis major on 9 days.After the first and ninth treatment,the Modified Ashworth Scale and the passive range of motion of the shoulder was used to assess the effect of the treatment.The spasticity and range of motion of the shoulder showed obvious improvement.These results indicate that dry needling at the myofascial trigger points can effectively treat chronic poststroke shoulder spasticity.

文摘This study examines the neuroprotective effects and mechanisms of action of total saponins from Rubus parvifolius L. (TSRP) on focal cerebral ischemia and reperfusion injury in rats. Focal cerebral ischemia and reperfusion injury was performed in rats using the suture method. The results indicate that intragastric injection of TSRP, at 5, 10 and 20 mg/kg, could decrease neurological impairment, reduce cerebral infarct volume, diminish pathological changes, and significantly inhibit the apoptosis of neurons surrounding the ischemic area. In addition, TSRP upregulated the expression of the anti-apoptotic factor Bcl-2, at the protein and mRNA levels, and it downregulated the expression of the pro-apoptotic factor Bax, at the protein and mRNA levels. These findings indicate that TSRP protects against cerebral ischemia/reperfusion injury, and that it may do so by regulating the expression of Bcl-2 and Bax.

文摘·AIM: To evaluate the reliability of β-III-Tubulin protein as a retinal ganglion cell(RGC) marker in the experimental glaucoma model.·METHODS: Glaucoma mouse models were established by injecting polystyrene microbeads into the anterior chamber of C57BL/6J mice, then their retinas were obtained 14 d and 28 d after the intraocular pressure(IOP)was elevated. Retinal flat mounts and sections were double-labeled by fluorogold(FG) and β-III-Tubulin antibody or single-labeled by β-III-Tubulin antibody,then RGCs were counted and compared respectively.· RESULTS: IOP of the injected eyes were elevated significantly and reached the peak at 22.8 ±0.7 mm Hg by day 14 after injection, then dropped to 11.3 ±0.7 mm Hg by day 28. RGC numbers counted by FG labeling and β-III- Tubulin antibody labeling were 64 807 ± 4930 and64 614 ±5054 respectively in the control group, with no significant difference. By day 14, RGCs in the experimental group decreased significantly compared to the control group, but there was no significant difference between the FG labeling counting and the β-III-Tubulin antibody labeling counting either in the experimental group or in the control group. The result was similar by day 28, with further RGC loss.·CONCLUSION: Our result suggested that the β-III-Tubulin protein was not affected by IOP elevation and can be used as a reliable marker for RGC in experimental models of glaucoma.
