为了明确NtPDF2在烟草中的生物学功能及其调控腺毛分化发育的作用机制,同源克隆了烟草NtPDF2基因,并对其序列、基因结构、进化关系以及表达模式进行分析。结果表明:NtPDF2 CDS全长为2 199 bp,编码732个氨基酸残基。烟草PDF2蛋白质序列... 为了明确NtPDF2在烟草中的生物学功能及其调控腺毛分化发育的作用机制,同源克隆了烟草NtPDF2基因,并对其序列、基因结构、进化关系以及表达模式进行分析。结果表明:NtPDF2 CDS全长为2 199 bp,编码732个氨基酸残基。烟草PDF2蛋白质序列高度保守,尤其是C端序列,均含有3个保守结构域(Homeobox domain,START domain和SAD domain)和1个保守LZ(Leucine zipper)元件。烟草PDF2基因结构高度保守,均由10个外显子和9个内含子组成。进化分析表明,普通烟草NtPDF2基因是由林烟草ML1基因进化而来。表达分析显示,烟草PDF2基因表达具有明显的时空特异性。干旱、黑暗和磷饥饿胁迫可显著下调NtPDF2基因的表达,而盐胁迫对其没有明显影响;NtPDF2基因的表达在赤霉素(GA)、茉莉酸甲酯(Me JA)、脱落酸(ABA)和打顶处理条件下均显著上调,这暗示烟草NtPDF2基因参与了调控多种非生物胁迫和激素应答过程。 展开更多

【目的】以小兴安岭林区兴安落叶松人工林为研究对象,基于林分空间结构参数和经营密度表确定采伐强度及采伐木,对林分空间结构进行优化,分析优化效果,为人工林空间结构优化提供理论依据。【方法】在黑龙江省伊春市南岔县设置4块面积为0.... 【目的】以小兴安岭林区兴安落叶松人工林为研究对象,基于林分空间结构参数和经营密度表确定采伐强度及采伐木,对林分空间结构进行优化,分析优化效果,为人工林空间结构优化提供理论依据。【方法】在黑龙江省伊春市南岔县设置4块面积为0.2 hm^(2)的兴安落叶松人工林固定样地,选择常用林分空间结构参数角尺度、大小比数、竞争指数和开阔比,并构建空间结构评价指数,结合兴安落叶松经营密度表确定采伐强度和采伐木,并分析间伐前后林分空分间结构变化。【结果】样地中的林木分别属于聚集分布、随机分布和均匀分布,中小径级林木较多,处于劣势状态,林木的生长空间处于严重不足状态和不足状态,林木竞争压力较大,各样地评价等级都为2级。根据经营密度表对监测样地2和监测样地4进行采伐,采伐株数强度分别为20.7%和12.7%,采伐之后角尺度分别降低3.9%和增加3.8%,大小比数分别降低0.5%和0.4%,竞争指数分别降低16.8%和5.2%,开阔比分别增加24.5%和4.2%,空间结构评价指数分别增加27.8%和7.2%。【结论】采用林分空间结构参数结合经营密度表进行林分抚育间伐可以有效改善林分空间结构,提升林木生长空间和优势程度,并减小了竞争压力。 展开更多

建立了气相色谱-串联质谱同时检测卷烟滤嘴中15种多环芳烃的方法。卷烟滤嘴用二氯甲烷振荡萃取后,经0.22μm有机相滤膜过滤,采用DB-5MS色谱柱(30 m×0.25 mm,0.25μm)进行分离,电子轰击源、正离子模式下以多反应监测模式进行检测,... 建立了气相色谱-串联质谱同时检测卷烟滤嘴中15种多环芳烃的方法。卷烟滤嘴用二氯甲烷振荡萃取后,经0.22μm有机相滤膜过滤,采用DB-5MS色谱柱(30 m×0.25 mm,0.25μm)进行分离,电子轰击源、正离子模式下以多反应监测模式进行检测,内标法进行定量。15种多环芳烃(苊烯、苊、芴、菲、蒽、荧蒽、芘、苯并[a]蒽、屈、苯并[b]荧蒽、苯并[k]荧蒽、苯并[a]芘、二苯并[a,h]蒽、苯并[g,h,i]苝和茚并[1,2,3-c,d]芘)的线性关系良好,相关系数(R^2)为0.991 4~0.999 9。15种多环芳烃在低、中、高3个添加水平下的平均回收率为81.6%~111.2%;除了芴在低添加水平时相对标准偏差为19.2%外,其他相对标准偏差均小于16%。15种多环芳烃的检出限为0.02~0.24 ng/滤嘴,定量限为0.04~0.80 ng/滤嘴。方法前处理简便,具有快速、准确、灵敏度高及重复性好的优点,适用于卷烟滤嘴中多环芳烃的分析。 展开更多

Some stress response-related genes have been identified in Ustilaginoidea virens,but it is not clear whether and how defects of stress responses affect the pathogenesis processes of U.virens.In this study,we identified a general stress response factor UvWHI2 as a homolog of Saccharomyces cerevisiae Whi2 in U.virens.The relative expression level of Uv Whi2 was significantly up-regulated during infection,suggesting that UvWHI2 may be involved in pathogenesis.Furthermore,knockout of Uv Whi2 showed decreased mycelial growth,increased conidiation in the potato sucrose medium and a defect in pathogenicity.In addition,the RNA-Seq and phenotypic analysis showed that UvWHI2 was involved in response to oxidative,hyperosmotic,cell wall stress and nutrient limitation.Further studies revealed that the defects of stress responses of the?Uvwhi2 mutant affected the formation of secondary spores on the nutrient limited surface and the rice surface,resulting in a significant reduction of pathogenicity of U.virens.Our results suggest that UvWHI2 is necessary for fungal growth,stress responses and the formation of secondary spores in U.virens.In addition,the defects of stress responses can affect the formation of secondary spores on the rice surface,and then compromise the pathogenicity of U.virens. 展开更多

A novel recipe is proposed to generate nonclassical vibrational states of the center-of mass motions of ions in a quantized trap,based on the quantum conversion between the quantum cavity fields and the quantized trap.It is shown that,when an ion trap system interacts with the eigen mode of a single-mode Fabry-Pérot cavity where the trap was set in,and a particularly selected external laser pulse through Raman transitions,the interchange of the quantum features between the quantum light states and the quantized trap states occurs.This kind of quantum conversion can be used to prepare some nonclassical trap states in an ion trap as well as to measure the properties of initial nonclassical vibrational states. 展开更多

枝下高是反映树冠特征的重要指标,准确预测枝下高对森林的经营管理和提高林分生产具有重要意义。本研究采用非线性回归构建枝下高广义基础模型,再进一步扩展到混合效应模型和分位数回归模型,通过“留一法”检验对模型的预测能力进行评... 枝下高是反映树冠特征的重要指标,准确预测枝下高对森林的经营管理和提高林分生产具有重要意义。本研究采用非线性回归构建枝下高广义基础模型,再进一步扩展到混合效应模型和分位数回归模型,通过“留一法”检验对模型的预测能力进行评价和比较。此外,使用4种抽样设计和不同抽样大小对枝下高模型进行校正,选择最佳的模型校正方案。结果表明:基于包含树高、胸径、林分每公顷断面积和优势木平均高的枝下高广义模型、扩展后的混合效应模型以及三分位数组合模型的预测精度均显著提高,混合效应模型略优于三分位数组合模型,最佳抽样校正方案为抽5株平均木。因此,推荐在实践应用中使用混合效应模型,抽5株样地平均木校正预测枝下高。 展开更多

稻叶黑粉病是由担子菌Entyloma oryzae侵染水稻叶片或叶鞘引起的真菌病害,但对病原菌与寄主互作的细胞学机制一直缺少了解。本文对采自田间自然发病叶片上的病斑进行了初步的细胞学分析,结果发现,病原菌侵染后,寄主病斑部位的表皮细胞... 稻叶黑粉病是由担子菌Entyloma oryzae侵染水稻叶片或叶鞘引起的真菌病害,但对病原菌与寄主互作的细胞学机制一直缺少了解。本文对采自田间自然发病叶片上的病斑进行了初步的细胞学分析,结果发现,病原菌侵染后,寄主病斑部位的表皮细胞外部形态基本保持完整;病原菌主要在寄主叶肉细胞部位产生大量的厚垣孢子并逐渐取代叶肉细胞;病原菌菌丝在寄主胞外扩展,未见其穿透寄主细胞壁进入细胞内,也没有产生典型的真菌吸器。靠近病原菌菌丝的寄主各种细胞内的细胞器均发生降解,降解产生的脂类物质凝聚成了体积较大的脂质球。寄主维管束组织的细胞壁一直保持完整,未发现病原菌菌丝进入维管束,病原菌菌丝和孢子被限制在寄主相邻两个维管束之间。在发病后期,由于寄主叶片表皮结构整体性破坏导致大量细菌进入,加速了叶片的衰老死亡。本研究结果表明,稻叶黑粉病菌的侵染模式为胞外侵染,类似于活体营养真菌;但病原菌的侵染导致附近寄主细胞降解死亡,类似于腐生营养真菌。 展开更多

许多病原菌能够通过分泌不同的效应蛋白以调控植物的防御以及胞内进程,从而助其有效入侵植物组织。稻瘟病菌的效应蛋白根据其不同的定位特点,被分为细胞质效应蛋白与质外体效应蛋白两类。在侵染过程中,细胞内的侵染菌丝被源于宿主植物... 许多病原菌能够通过分泌不同的效应蛋白以调控植物的防御以及胞内进程,从而助其有效入侵植物组织。稻瘟病菌的效应蛋白根据其不同的定位特点,被分为细胞质效应蛋白与质外体效应蛋白两类。在侵染过程中,细胞内的侵染菌丝被源于宿主植物的交界面菌丝膜(extrainvasive hyphal membrane, EIHM)包围,由EIHM与侵染菌丝细胞壁形成的质外体隔间是效应蛋白分泌的"必经通道"。此外,在稻瘟病菌侵染过程中会形成一个高度局部化的结构—活体营养表面复合体(biotrophic interfacial complex, BIC),它能够聚集由侵染菌丝分泌的细胞质效应蛋白。该文综述了稻瘟病菌效应蛋白的功能及其迁移过程,介绍了BIC的两个重要的形成阶段,阐明了不同效应蛋白的定位特点以及胞间转运的动态过程,揭示了效应蛋白分泌、转运至水稻细胞质以及在水稻细胞之间移动的分子机制。 展开更多
