Hepatocellular carcinoma(HCC)is a common malignancy that is driven by multiple genes and pathways.The aim of this study was to investigate the role and specific mechanism of the actin-interacting protein zyxin(ZYX)in ...Hepatocellular carcinoma(HCC)is a common malignancy that is driven by multiple genes and pathways.The aim of this study was to investigate the role and specific mechanism of the actin-interacting protein zyxin(ZYX)in HCC.We found that the expression of ZYX was significantly higher in HCC tissues compared to that in normal liver tissues.In addition,overexpression of ZYX in hepatoma cell lines(PLC/PRF/5,HCCLM3)enhanced their proliferation,migration and invasion,whereas ZYX knockdown had the opposite effects(SK HEP-1,Huh-7).Furthermore,the change in the expression levels of ZYX also altered that of proteins related to cell cycle,migration and invasion.Similar results were obtained with xenograft models.The AKT/mTOR signaling pathway is one of the key mediators of cancer development.While ZYX overexpression upregulated the levels of phosphorylated AKT/mTOR proteins,its knockdown had the opposite effect.In addition,the AKT inhibitor MK2206 neutralized the pro-oncogenic effects of ZYX on the HCC cells,whereas the AKT activator SC79 restored the proliferation,migration and invasion of HCC cells with ZYX knockdown.Taken together,ZYX promotes the malignant progression of HCC by activating AKT/mTOR signaling pathway,and is a potential therapeutic target in HCC.展开更多
The Advanced Geosynchronous Radiation Imager(AGRI)is a mission-critical instrument for the Fengyun series of satellites.AGRI acquires full-disk images every 15 min and views East Asia every 5 min through 14 spectral b...The Advanced Geosynchronous Radiation Imager(AGRI)is a mission-critical instrument for the Fengyun series of satellites.AGRI acquires full-disk images every 15 min and views East Asia every 5 min through 14 spectral bands,enabling the detection of highly variable aerosol optical depth(AOD).Quantitative retrieval of AOD has hitherto been challenging,especially over land.In this study,an AOD retrieval algorithm is proposed that combines deep learning and transfer learning.The algorithm uses core concepts from both the Dark Target(DT)and Deep Blue(DB)algorithms to select features for the machinelearning(ML)algorithm,allowing for AOD retrieval at 550 nm over both dark and bright surfaces.The algorithm consists of two steps:①A baseline deep neural network(DNN)with skip connections is developed using 10 min Advanced Himawari Imager(AHI)AODs as the target variable,and②sunphotometer AODs from 89 ground-based stations are used to fine-tune the DNN parameters.Out-of-station validation shows that the retrieved AOD attains high accuracy,characterized by a coefficient of determination(R2)of 0.70,a mean bias error(MBE)of 0.03,and a percentage of data within the expected error(EE)of 70.7%.A sensitivity study reveals that the top-of-atmosphere reflectance at 650 and 470 nm,as well as the surface reflectance at 650 nm,are the two largest sources of uncertainty impacting the retrieval.In a case study of monitoring an extreme aerosol event,the AGRI AOD is found to be able to capture the detailed temporal evolution of the event.This work demonstrates the superiority of the transfer-learning technique in satellite AOD retrievals and the applicability of the retrieved AGRI AOD in monitoring extreme pollution events.展开更多
Organic electrochemical transistors(OECTs) exhibit significant potential for applications in healthcare and human-machine interfaces, due to their tunable synthesis, facile deposition, and excellent biocompatibility. ...Organic electrochemical transistors(OECTs) exhibit significant potential for applications in healthcare and human-machine interfaces, due to their tunable synthesis, facile deposition, and excellent biocompatibility. Expanding OECTs to the fexible devices will significantly facilitate stable contact with the skin and enable more possible bioelectronic applications. In this work,we summarize the device physics of fexible OECTs, aiming to offer a foundational understanding and guidelines for material selection and device architecture. Particular attention is paid to the advanced manufacturing approaches, including photolithography and printing techniques, which establish a robust foundation for the commercialization and large-scale fabrication. And abundantly demonstrated examples ranging from biosensors, artificial synapses/neurons, to bioinspired nervous systems are summarized to highlight the considerable prospects of smart healthcare. In the end, the challenges and opportunities are proposed for fexible OECTs. The purpose of this review is not only to elaborate on the basic design principles of fexible OECTs, but also to act as a roadmap for further exploration of wearable OECTs in advanced bio-applications.展开更多
In general,self-healing dielectric composites are mainly composed of polar hydrogen bonds,which have high hydrophilicity and are unsuitable for humid environment.Dielectric composite with Diels-Alder(D-A)bond contains...In general,self-healing dielectric composites are mainly composed of polar hydrogen bonds,which have high hydrophilicity and are unsuitable for humid environment.Dielectric composite with Diels-Alder(D-A)bond contains covalent bonds,it can be adopted as an efficient self-healing material.Here,we construct self-healing barium titanate(BT)/polyurethane(PU)dielectric composites by adopting PU with D-A bond as matrix(BT/PU-DA).The prepared 10%BT/PU-DA composite exhibits superior self-healing ability than that of PUDA.Moreover,its dielectric constant can reach 9.3 with a loss of only 0.04 at 1000 Hz and maintain 93%repair efficiency of tensile strength.The experimental analysis suggests the introduction of D-A bond can enhance the thermostability and self-healing ability of BT/PU-DA composite.In addition,the incorporation of BT nanoparticles and D-A bond in the self-healing composite contributes to the lower dielectric loss and excellent tensile strength after healing.The adopted strategy is a promising and facile approach to develop highly efficient selfhealing dielectric material,which will be conducive to reuse and sustainable development of the electronic packaging material in aqueous medium or wet environment.展开更多

The program is highly scalable and cost-effective. This paper developed the home server-side application based on MFC technology and the mobile terminal application. The mobile client can remotely control home devices and query the running state, electric energy information and historical data of home devices. At the same time, the home server-side application can store electric energy information and electricity consumption of home devices. Combined with household distributed photovoltaic generation system, the system can be applied to home energy management system. Through running tests and application, the results show that the system has realized basic functions of smart home and achieved the desired design goals.展开更多
Hypertriglyceridemia(HTG)is the third most common cause of acute pancreatitis(AP),and the incidence of HTG-induced AP(HTG-AP)is rising worldwide as a result of changes in lifestyle and dietary habits.In China,HTG acco...Hypertriglyceridemia(HTG)is the third most common cause of acute pancreatitis(AP),and the incidence of HTG-induced AP(HTG-AP)is rising worldwide as a result of changes in lifestyle and dietary habits.In China,HTG accounts for 10%to 20%of the causes of AP,even becoming the second leading cause of AP.The presentation and diagnosis of HTG-AP is similar to that of AP from other causes,however,the complication rates and severe AP are significantly higher in patients with HTG-AP than in patients with other etiologies.Regarding the management of HTG-AP,the initial management is similar to that of AP from other causes and mainly includes fluid resuscitation,pain control,and nutritional support.In addition to supportive care,it is necessary to take appropriate measures to decrease serum TG levels,in order to alleviate the progression of AP and prevent recurrence.This review aims to summarize existing clinical and basic research evidence of HTG-AP and seeks to highlight the epidemiology,definition,pathogenesis,clinical course,diagnosis,management,prevention of recurrence,and specific clinical scenarios,such as HTG-AP during pregnancy,HTG-AP with diabetic ketoacidosis.展开更多
Bluetongue (BT) is a serious hemorrhagic disease of ruminants caused by bluetongue virus (BTV). Inactive BTV vaccines have been successful in field trials in some areas, and inactivated vaccines are considered safer. ...Bluetongue (BT) is a serious hemorrhagic disease of ruminants caused by bluetongue virus (BTV). Inactive BTV vaccines have been successful in field trials in some areas, and inactivated vaccines are considered safer. However, information about the effect of the viral antigen level on the serological response and efficiency of the inactive BTV-16 vaccine is lacking. In the present study, the serological response and efficiency of the viral antigen concentration in the binary ethylenimine-inactivated Chinese BTV serotype-16 vaccine were investigated. The viral antigens in the viral suspension (VS) were quantified using a modified BTV AC-ELISA method. Four batches of vaccine containing 1, 5, 10, and 50 μg/ml of viral antigen were generated from the VS. Four groups of naive Chinese sheep were vaccinated with the different vaccine batches, and the serological response and vaccine efficiency were investigated before and after challenge infection. The vaccines containing 10 and 50 μg/ml of viral antigen induced significant ELISA and neutralizing antibody titers 14 days after vaccination, whereas the vaccines containing 1 and 5 μg/ml of viral antigen did not have these effects. A booster immunization at 21 days enhanced all groups’ antibody titers;however, the increased titer was related to the viral antigen level. In contrast to the serological response, the viral antigen level of the vaccines did not have a significant effect on the vaccine efficiency. With the exception of one sheep from the 5 μg/ml viral antigen group, all vaccinated sheep from the four antigen level groups showed strong resistance to infection based on their clinical symptoms, rectal temperatures and viremia. Collectively, these data suggested that viral antigen levels from 1 to 50 μg/ml had a significant effect on the serological response of the animals but a limited effect on the vaccine efficiency. The BTV-16 vaccine containing 1 μg/ml of viral antigen was sufficient to achieve high efficiency, but only the vaccines with more than 10 μg/ml of antigen induced a significant antibody response. To obtain a better serological response, we suggest the use of vaccines with more than 10 μg/ml of viral antigen. The findings in the study will be useful for BTV vaccine production.展开更多
Background:Compatibility is a characteristic of the clinical application of traditional Chinese medicine,often leading to enhanced therapeutic effects.In the treatment of cerebral ischemia,blood-activating and open or...Background:Compatibility is a characteristic of the clinical application of traditional Chinese medicine,often leading to enhanced therapeutic effects.In the treatment of cerebral ischemia,blood-activating and open orifices herbs are frequently used individually;however,their combination is not commonly practiced.This study aims to investigate the impact of combining safflower and borneol as examples of open orifices herbs and blood-activating herbs on the neurovascular unit in rats with ischemic stroke.The objective is to determine whether this combination exhibits superior therapeutic efficacy compared to using borneol or safflower alone while exploring its underlying mechanism.These findings may provide novel insights for clinical treatments.Methods:SD male rats were randomly divided into 6 groups:sham operation group,model group,borneol group(0.1 g/kg),safflower group(5 g/kg),borneol combined with safflower group(0.1 g/kg+5 g/kg)and nimodipine group(0.01 g/kg).The middle cerebral artery cerebral ischemia(MCAO)model were prepared after continuous intragastric administration for 7 days in each group,the neurological function of each group were scored 24h after operation,and water content in brain tissue were measured by weighing method.The activity of superoxide dismutase(SOD)and the contents of nitric oxide(NO)and malondialdehyde(MDA)in brain tissue and serum were determined by spectrophotometry,and the mRNA expressions of matrix metalloproteinase 2(MMP-2),tight junction protein 1(ZO-1),vascular endothelial growth factor(VEGF)and brain-derived neurotrophic factor(BDNF)were detected by Real time PCR.Result:Compared with the model group,the group treated with borneol combined with safflower exhibited a significant decrease in the neural function score of MCAO rats(P<0.01).Additionally,it led to a reduction in brain tissue water content(P<0.01),elevated SOD activity,and reduced levels of NO and MDA in both serum and brain tissue(P<0.01 or P<0.05).Moreover,this treatment resulted in a decrease in the mRNA expression of MMP-2 and an increase in ZO-1 in brain tissue,along with an increase in the mRNA expression of VEGF and BDNF(P<0.01).Conclusion:Borneol combined with safflower demonstrates a protective effect on the neurovascular unit in rats with ischemic stroke.This effect is likely associated with increased SOD activity,reduced MDA and NO content in both serum and brain tissue of MCAO rats,and a decrease in MMP-2 mRNA expression in brain tissue,coupled with an increase in ZO-1,VEGF,and BDNF mRNA expression.These effects were superior to those observed with borneol or safflower administered alone.展开更多

The viral antigens in the viral suspension (VS) were quantified using a modified BTV AC-ELISA method. Four batches of vaccine containing 1, 5, 10, and 50 μg/ml of viral antigen were generated from the VS. Four groups of naive Chinese sheep were vaccinated with the different vaccine batches, and the serological response and vaccine efficiency were investigated before and after challenge infection. The vaccines containing 10 and 50 μg/ml of viral antigen induced significant ELISA and neutralizing antibody titers 14 days after vaccination, whereas the vaccines containing 1 and 5 μg/ml of viral antigen did not have these effects. A booster immunization at 21 days enhanced all groups’ antibody titers;however, the increased titer was related to the viral antigen level. In contrast to the serological response, the viral antigen level of the vaccines did not have a significant effect on the vaccine efficiency. With the exception of one sheep from the 5 μg/ml viral antigen group, all vaccinated sheep from the four antigen level groups showed strong resistance to infection based on their clinical symptoms, rectal temperatures and viremia. Collectively, these data suggested that viral antigen levels from 1 to 50 μg/ml had a significant effect on the serological response of the animals but a limited effect on the vaccine efficiency. The BTV-16 vaccine containing 1 μg/ml of viral antigen was sufficient to achieve high efficiency, but only the vaccines with more than 10 μg/ml of antigen induced a significant antibody response. To obtain a better serological response, we suggest the use of vaccines with more than 10 μg/ml of viral antigen. The findings in the study will be useful for BTV vaccine production.

文摘Background:Compatibility is a characteristic of the clinical application of traditional Chinese medicine,often leading to enhanced therapeutic effects.In the treatment of cerebral ischemia,blood-activating and open orifices herbs are frequently used individually;however,their combination is not commonly practiced.This study aims to investigate the impact of combining safflower and borneol as examples of open orifices herbs and blood-activating herbs on the neurovascular unit in rats with ischemic stroke.The objective is to determine whether this combination exhibits superior therapeutic efficacy compared to using borneol or safflower alone while exploring its underlying mechanism.These findings may provide novel insights for clinical treatments.Methods:SD male rats were randomly divided into 6 groups:sham operation group,model group,borneol group(0.1 g/kg),safflower group(5 g/kg),borneol combined with safflower group(0.1 g/kg+5 g/kg)and nimodipine group(0.01 g/kg).The middle cerebral artery cerebral ischemia(MCAO)model were prepared after continuous intragastric administration for 7 days in each group,the neurological function of each group were scored 24h after operation,and water content in brain tissue were measured by weighing method.The activity of superoxide dismutase(SOD)and the contents of nitric oxide(NO)and malondialdehyde(MDA)in brain tissue and serum were determined by spectrophotometry,and the mRNA expressions of matrix metalloproteinase 2(MMP-2),tight junction protein 1(ZO-1),vascular endothelial growth factor(VEGF)and brain-derived neurotrophic factor(BDNF)were detected by Real time PCR.Result:Compared with the model group,the group treated with borneol combined with safflower exhibited a significant decrease in the neural function score of MCAO rats(P<0.01).Additionally,it led to a reduction in brain tissue water content(P<0.01),elevated SOD activity,and reduced levels of NO and MDA in both serum and brain tissue(P<0.01 or P<0.05).Moreover,this treatment resulted in a decrease in the mRNA expression of MMP-2 and an increase in ZO-1 in brain tissue,along with an increase in the mRNA expression of VEGF and BDNF(P<0.01).Conclusion:Borneol combined with safflower demonstrates a protective effect on the neurovascular unit in rats with ischemic stroke.This effect is likely associated with increased SOD activity,reduced MDA and NO content in both serum and brain tissue of MCAO rats,and a decrease in MMP-2 mRNA expression in brain tissue,coupled with an increase in ZO-1,VEGF,and BDNF mRNA expression.These effects were superior to those observed with borneol or safflower administered alone.
