目的:观察FHIT及Ki67蛋白在膀胱尿路上皮癌组织中的表达及相关性。方法:采用免疫组织化SP法检测66例膀胱癌和42例正常膀胱组织中FHIT及Ki67蛋白表达情况,利用Spearman相关性分析对FHIT与Ki67蛋白进行相关性分析。结果:FHIT蛋白在正常膀... 目的:观察FHIT及Ki67蛋白在膀胱尿路上皮癌组织中的表达及相关性。方法:采用免疫组织化SP法检测66例膀胱癌和42例正常膀胱组织中FHIT及Ki67蛋白表达情况,利用Spearman相关性分析对FHIT与Ki67蛋白进行相关性分析。结果:FHIT蛋白在正常膀胱组织中的阳性率为92.85%,在膀胱尿路上皮癌中的阳性表达率为39.39%;而Ki67则相反;在膀胱尿路上皮癌中的表达率为90.91%;在正常膀胱组织中表达率为11.90%;FHIT和Ki67蛋白的异常表达与膀胱尿路上皮癌临床分期、分化程度有关。Spearman相关性分析显示,FHIT与Ki67蛋白表达呈负相关。结论:联合检测FHIT和Ki67,有助于提高膀胱尿路上皮癌侵袭能力的评估,有助于膀胱尿路上皮癌临床预后判断。 展开更多

目的全激素阻断治疗前列腺癌过程中对外周血辅助性T细胞17(Th17)和调节性T细胞(Treg)进行监测,探讨Th17/Treg免疫失衡与前列腺癌激素抵抗的评价。方法对全激素阻断治疗前列腺癌过程中54例发展为抵抗性的患者为治疗组,以一般状况匹配的3... 目的全激素阻断治疗前列腺癌过程中对外周血辅助性T细胞17(Th17)和调节性T细胞(Treg)进行监测,探讨Th17/Treg免疫失衡与前列腺癌激素抵抗的评价。方法对全激素阻断治疗前列腺癌过程中54例发展为抵抗性的患者为治疗组,以一般状况匹配的37例激素敏感性前列腺癌作为对照组,用流式细胞术检测激素难治组和激素敏感组治疗前、后患者外周血Th17和Treg细胞比例,酶联免疫吸附试验(ELISA)检测血清IL-10和IL-17,分析Th17与IL-17的相关性和Treg与IL-10的相关性以及Th17/Treg与前列腺癌控制相关性。结果治疗组外周血Th17、IL-10、IL-17水平均高于对照组(P<0.05);治疗组Treg比例低于对照组(P<0.05)。激素抵抗性前列腺癌患者经阿比特龙加泼尼松治疗3个月后,检测外周血Th17细胞比例、Treg细胞比例、IL-10、IL-17水平比较,2组差异均无统计学意义(P>0.05)。细胞因子IL-17、IL-10变化与Th17/Treg细胞变化一致且相关。结论Th17/Treg免疫平衡与前列腺癌激素抵抗性相关,Th17/Treg免疫平衡对前列腺癌激素治疗明显影响,Th17/Treg比值变化对前列腺癌激素治疗有评价意义。 展开更多

Background Nucleostemin is essential for the proliferation and survival of stem and cancer cells, but it is unknown whether this newly identified molecule is involved in prostate cancer pathogenesis.Methods Total RNA and protein were extracted from prostate cancer tissues and PC-3, LNCap and DU145 cell lines. The nucleostemin mRNA and protein expression were measured by RT-PCR and Western blot. Immunohistochemistry was also used to detect the nucleostemin protein expression in prostate cancer tissues and PC-3 cells. A nucleostemin specific, short hairpin RNA, expression plasmid was used to transfect PC-3 cells. The changes of nucleostemin gene were detected and the oroliferative capacitv of the cells was determined. Results Nucleostemin was highly expressed in prostate cancer tissues and cell lines. Nucleostemin expression level in the silencer group PC-3 cells remarkably reduced. The proliferation rate of silencer group PC-3 cells decreased and the percentage of G1 stage cells increased. The neoplasm forming capacity in nude mice of the silencer group PC-3 cells decreased significantly. Conclusions Nucleostemin is highly expressed in prostate cancer tissues and cell lines. The proliferative capacity of PC-3 cells is remarkably reduced after silencing nucleostemin gene expression. 展开更多
