牦牛源短小芽胞杆菌TS1的分离鉴定及其生物学特性研究

[目的]本试验旨在研究一株牦牛源短小芽孢杆菌(命名:TS1)的分离鉴定、抗逆性、对常用抗菌药物的耐药性以及对常见致病菌的抑菌活性,初步探索其作为益生菌添加剂在畜牧养殖中应用的可行性。[方法]对健康牦牛新鲜粪便进行100℃水浴处理10 min以筛选具有抗逆性的芽胞杆菌,进行形态学、生理生化特征及16S rRNA序列分析鉴定,通过测量生长曲线、耐酸、耐胆盐试验、药敏试验及抑菌试验等方法对其生物学特性进行研究,通过全基因测序分析其可能产生抗菌物质的类型。[结果]TS1为短小芽胞杆菌且具有耐酸、耐胆盐生长特性;TS1对于12种常见的抗菌药物敏感,且对大肠杆菌、沙门氏菌、金黄色葡萄球菌、猪链球菌具有抑菌作用。测序分析显示该菌株可编码与Amylocyclicin同源性达48.90%的一种新型抗菌肽,进而发挥其抗菌活性。[结论]短小芽胞杆菌TS1有一定的抗逆性和潜在的益生能力及广谱抗菌效果,并且在畜禽肠道微生物系统中对抗生素耐药性传递的风险较低,适合作为益生菌添加剂类抗生素替代产品。 展开更多

目的建立使用高效液相色谱法(HPLC)测定丙戊酸钠中醋酸含量的方法。方法色谱柱为Nano ChromCore C8(4.6 mm×150 mm,5μm);梯度洗脱,流动相A为磷酸水溶液(磷酸调节pH值至2.3),流动相B为甲醇;检测波长为210 nm;流速为0.5 mL/min;柱温... 目的建立使用高效液相色谱法(HPLC)测定丙戊酸钠中醋酸含量的方法。方法色谱柱为Nano ChromCore C8(4.6 mm×150 mm,5μm);梯度洗脱,流动相A为磷酸水溶液(磷酸调节pH值至2.3),流动相B为甲醇;检测波长为210 nm;流速为0.5 mL/min;柱温为40℃;进样体积10μL。结果醋酸在0.54~21.66 mg/L范围内线性关系良好(r=1.0000);重复性的平均回收率为100.1%,RSD为0.7%;低、中、高3个浓度的平均加样回收率为100.5%,RSD为1.1%;定量限为5.4 ng;检测限为1.6 ng。结论HPLC专属性好、灵敏度高,测得结果准确、可靠,可用于丙戊酸钠中醋酸含量的测定。 展开更多

Secreted proteins are important sources for early detection and diagnosis of disease, and as such have received considerable attention. The extraction of low concentration proteins from large volumes of culture media, which are rich in salts and other compounds that interfere with most proteomics techniques, presents a problem for secretome studies. Ultrafiltration, precipitation, and dialysis are three major extraction methods that can be used to overcome this problem. The present study for the first time, compared the merits and shortcomings of these three methods, without bias. Centrifugal ultrafiltration provided the best extraction efficiency, and precipitation provided the highest number of identifiable proteins. The three methods yielded closely related, but different, information on the secretome; thus, they should be considered complementary or, at least, supplementary methods. Three hundred and sixty unique proteins were identified, including 211 potential secreted proteins. Compared with previous studies, this study also identified 42 new secreted proteins. The present study not only offers a reference for the selection of secretome extraction methods, but also expands the secretome database for the investigation of hepatocellular carcinoma. 展开更多

Effects of galectin-3 inhibition on endometrial cell cycle and adhesion

Galectin-3(gal-3)and its ligands have been implicated in cell transformation and cancer metastasis.Gal-3 protein has been found in uterine epithelial cells adjacent to implanting blastocysts in different cell types.In order to investigate the role of gal-3 in the establishment of human endometrial receptivity,the expression of gal-3 in human endometrial cell line RL95-2 was silenced by RNA inter-ference technology using gal-3 specific small RNA.The expression of gal-3 was detected by the reverse transcriptase-polymerase chain reaction and Western blot analysis.After the suppression of gal-3,cell cycle changes and the expres-sion of integrin b1 were detected by flow cytometry.The adhesive ability of RL95-2 cells was analyzed by the adhesion test.Gal-3 siRNA transfection efficiency reached 70%–90%.The expression of gal-3 mRNA and protein in RL95-2 cells was strongly inhibited by 70%–90%after RNA interference.Inhibition of gal-3 expression decreased S-phase but increased G1 phase cells.Integrin b1 expression was down-regulated,and the adhesive ability of RL95-2 cells to fibronectin(FN)was significantly reduced.Gal-3 may be involved in the establishment of endometrial receptivity by regulating the proliferation and adhesion of endometrial cells.The influence on adhesion may be related with the integrin modulation. 展开更多
