BACKGROUND F-box and leucine-rich repeat 6(FBXL6)have reportedly been associated with several cancer types.However,the role and mechanisms of FBXL6 in gastric cancer(GC)require further elucidation.AIM To investigate t...BACKGROUND F-box and leucine-rich repeat 6(FBXL6)have reportedly been associated with several cancer types.However,the role and mechanisms of FBXL6 in gastric cancer(GC)require further elucidation.AIM To investigate the effect of FBXL6 in GC tissues and cells and the underlying mechanisms.METHODS TCGA and GEO database analysis was performed to evaluate the expression of FBXL6 in GC tissues and adjacent normal tissues.Reverse transcription-quantitative polymerase chain reaction,immunofluorescence,and western blotting were used to detect the expression of FBXL6 in GC tissue and cell lines.Cell clone formation,5-ethynyl-2’-deoxyuridine(EdU)assays,CCK-8,transwell migration assay,and wound healing assays were performed to evaluate the malignant biological behavior in GC cell lines after transfection with FBXL6-shRNA and the overexpression of FBXL6 plasmids.Furthermore,in vivo tumor assays were performed to prove whether FBXL6 promoted cell proliferation in vivo.RESULTS FBXL6 expression was upregulated more in tumor tissues than in adjacent normal tissues and positively associated with clinicopathological characteristics.The outcomes of CCK-8,clone formation,and Edu assays demonstrated that FBXL6 knockdown inhibited cell proliferation,whereas upregulation of FBXL6 promoted proliferation in GC cells.Additionally,the transwell migration assay revealed that FBXL6 knockdown suppressed migration and invasion,whereas the overex pression of FBXL6 showed the opposite results.Through the subcutaneous tumor implantation assay,it was evident that the knockdown of FBXL6 inhibited GC graft tumor growth in vivo.Western blotting showed that the effects of FBXL6 on the expression of the proteins associated with the epithelial-mesenchymal transition-associated proteins in GC cells.CONCLUSION Silencing of FBXL6 inactivated the EMT pathway to suppress GC malignancy in vitro.FBXL6 can potentially be used for the diagnosis and targeted therapy of patients with GC.展开更多

Background: The prognosis of patients with tumors is partly influenced by the stage when patients are diagnosed. Therefore, the time of confirmed diagnosis after onset can be considered as a prognostic predictor. Alth...Background: The prognosis of patients with tumors is partly influenced by the stage when patients are diagnosed. Therefore, the time of confirmed diagnosis after onset can be considered as a prognostic predictor. Although a few of studies work on it, factors that influence the duration remain poorly understood in different areas. Methods: This retrospective study involved patients with gastric cancer. Participants completed questionnaires by either phone or face-to-face interviews. Results: Females are with a longer interval than males. Farms lived in rural area and with New Rural Cooperative Medical Scheme are associated with a longer time interval. Patients whose income is mainly from their spouses also have a longer interval. Abdominal pain seems to be associated with longer interval time, while vomiting leads to a shorter time interval. Taking medicine after symptoms appear is associated with longer time intervals, especially those take medicine nonstandardly and more than 2 weeks. Conclusions: The results demonstrate that the medical resource in rural area should still be enhanced;meanwhile taking medicine without doctor’s advice should be cut down. Additional efforts must be made to devise strategies to properly guide patients’ behavior.展开更多
The Sulige gas field is a typical low-pressure low-permeability tight sandstone gas reservoir. The reservoir has poor seepage capacity, strong heterogeneity, high mineralization of formation water and extremely scarce...The Sulige gas field is a typical low-pressure low-permeability tight sandstone gas reservoir. The reservoir has poor seepage capacity, strong heterogeneity, high mineralization of formation water and extremely scarce water resources on the site. These unfavorable factors have brought great difficulties to the on-site mining process. Now, a nano-composite green environmental protection slick water fracturing fluid system CQFR can be quickly dissolved because of the larger specific surface area, and the small molecular size makes the damage to the reservoir less than 5%, and the average drag reduction effect can reach more than 73%. It can quickly and well dissolve and maintain performance under high salinity conditions and fracturing flowback fluids. It responds well to the complex reservoir conditions on the construction site and makes the flowback fluid recyable, which greatly reduces the consumption of water resources on the construction site and effectively improves the construction efficiency and economic benefits.展开更多
