OBJECTIVE This study aimed to optimize polysaccharides extraction from Urena lobata L.and investigate its antioxidant activity.METHODS The mathematical model was established by re.sponse surface method(RSM) based on the results of single factor experiments,using polysaccha.rides extraction rate as response value,and using the ratio of water to material,cellulase concentra.tion,extraction temperature and time as experimental factors,which was used to screen optimum poly.saccharide extraction conditions from Urena lobata L..Antioxidant activity of polysaccharides was stud.ied by DPPH and ·OH free radical elimination method.RESULTS The optimum conditions obtained by RSM were as follows:the cellulase level was 10.8 g·L^(-1),extraction time duration was 72 min,the ra.tio of water to feedstock was 7 mL·g^(-1),extraction temperature was 43℃,the pH value was 5.0.Under the optimal conditions,there was a difference of less than 5% between predicted extraction rate 13.37% and experimental extraction rate 13.32%.The polysaccharide yield was most significantly af.fected by cellulase concentration,followed by extraction time,water to material ratio and extraction tem.perature.IC50 of DPPH and ·OH were 1.082 g·L^(-1) and 3.202 mg·L^(-1),respectively.Antioxidant activity of sample polysaccharides was weaker than those of vitamin C.CONCLUSION The polysaccharide extraction process from Urena lobata L.by cellulase enzymolysis approach was obtained,which was convenient and feasible,and extracted polysaccharides had good free radical scavenging activity. 展开更多

Nondestructive determination of paper properties of Populus × euramericana with near infrared spectroscopy

Five Populus x euramericana wood samples representing three different sites were selected and nearinfrared (NIR) spectra were obtained. For these sections, basis weight, brightness and three mechanical properties (tensile index, tearing index and bursting index) were determined by standard analytical methods. Calibrations were developed for each paper property using the NIR spectra, data on paper properties, using partial least squares (PLS) regression. The results show that the coefficients of correlation of calibration and validation for basis weight were 0.8824 and 0.8299, respectively; the standard error of calibration (SEC) and prediction (SEP) were 1.150 and 1.170, respectively. In testing for brightness, the correlation coefficient of calibration was 0.9621 and for validation 0.9612, while the SEC and SEP were 0.997 and 1.300, respectively; paper brightness and NIR spectroscopy were highly correlated. NIR spectroscopy can be used to predict tensile, tearing and bursting indices of paper samples rapidly. We found that the paper properties fitted by NIR and GB methods were highly correlated. The coefficients of correlation of calibration and validation for basis weight exceeded 0.8000, while the SEC and SEP were very small. These results reveal that the five paper properties of Populus x euramericana and those predicted by the NIR model were highly correlated. We conclude that the NIR models can be used for the prediction of paper properties. 展开更多

In this study,a natural cotton thread immunoassay device combined with gold nanorod(GNR) reporter probe is developed for the rapid,sensitive and quantitative electrochemical determination of human ferritin,a lung cancer related biomarker.Human ferritin as an analyte and a pair of monoclonal antibodies are used to demonstrate the proof-of-concept on the cotton thread immunoassay device.An enhancement of the sensitivity is achieved by using gold nanorod as an electroactive report probe compared with a traditional gold nanoparticle(GNP) report probe.The device was capable of measuring 1.58 ng/mL ferritin in 30 min by anodic stripping voltammetry(ASV) testing,which meet the requirement for clinical diagnosis. 展开更多
