Objective:Diffuse large B-cell lymphoma(DLBCL)is an aggressive type of non-Hodgkin lymphoma.Due to its genetic heterogeneity and abnormal metabolism,many DLBCL patients have a poor prognosis.This study investigated th...Objective:Diffuse large B-cell lymphoma(DLBCL)is an aggressive type of non-Hodgkin lymphoma.Due to its genetic heterogeneity and abnormal metabolism,many DLBCL patients have a poor prognosis.This study investigated the key metabolism-related genes and potential mechanisms.Methods:Differentially expressed genes,differentially expressed transcription factors(TFs),and differentially expressed metabolism-related genes(DEMRGs)of glucose and lipid metabolic processes were identified using the edgeR package.Key DEMRGs were screened by Lasso regression,and a prediction model was constructed.The cell type identification by estimating relative subsets of RNA transcripts algorithm was utilized to assess the fraction of immune cells,and Gene Set Enrichment Analysis was used to determine immune-related pathways.A regulatory network was constructed with significant co-expression interactions among TFs,DEMRGs,immune cells/pathways,and hallmark pathways.Results:A total of 1551 DEMRGs were identified.A prognostic model with a high applicability(area under the curve=0.921)was constructed with 13 DEMRGs.Tumorigenesis of DLBCL was highly related to the neutrophil count.Four DEMRGs(PRXL2AB,CCN1,DECR2 and PHOSPHO1)with 32 TF-DEMRG,36 DEMRG-pathway,14 DEMRG-immune-cell,9 DEMRG-immune-gene-set,and 67 DEMRG-protein-chip interactions were used to construct the regulatory network.Conclusion:We provided a prognostic prediction model based on 13 DEMRGs for DLBCL.We found that phosphatase,orphan 1(PHOSPHO1)is positively regulated by regulatory factor X5(RFX5)and mediates MYC proto-oncogene(MYC)targeting the V2 pathway and neutrophils.展开更多
Background:To confirm that the four constitutions of the Korean medicine is closely related to the intestinal flora,and explore the influence of specific intestinal microbes on these four populations.Methods:We collec...Background:To confirm that the four constitutions of the Korean medicine is closely related to the intestinal flora,and explore the influence of specific intestinal microbes on these four populations.Methods:We collect stool samples of four different physiques of the Taeeum,Soeum,Taeyang,and Shaoyang.Based on 16S rDNA amplicon sequencing,SPSS 18.0 statistical software and GraphPad Prism 8.0 mapping software,the body weight,body mass index,cholesterol and intestinal microcommunity composition of the four groups were analyzed and compared.Results:The results found that the body weight and body mass index of the Taeeum group were significantly higher than the Shaoyang group and the Soeum group,the body weight,cholesterol and body mass index of the Taeyang group were significantly higher than the Soeum Group.In the analysis of gut microbiota,the richness and diversity of intestinal microorganisms and the abundance ratio of posterior firmicutes to bacteroides were all different in the four populations,and there were 24 gut microbiota with significant differences(P<0.05).Among them,the abundance of[Eubacterium]coprostanoligenes group of Taeyang people,Taeeum people,Shaoyang people,and Soeum people were 3.71,9.62,9.36 and 9.33,respectively.Conclusion:[Eubacterium]coprostanoligenes group may be related to the differences in cholesterol content of the four populations.Similarly,the firmicutes/bacteroidetes abundance ratio may also be related to the differences in body weight,blood glucose and blood lipid among the four groups.展开更多

Methods: C57BL/6 mice were selected as experimental animals, and the mouse model with transplanted tumor was established after non-small cell lung cancer cell line H358 was cultured;the mice with transplanted tumor were divided into the intervention group who received Corbrin Capsule therapy and the model group who received no drug therapy. 30 d after treatment, the expression of proliferation activity indexes, proliferation genes and invasion genes in transplanted tumor tissue were determined. Results: 30 d after treatment, the mRNA expression and protein expression of PCNA and Ki-67 in transplanted tumor lesion of intervention group were significantly lower than those of model group, and Notch-1, Bmi-1, MIF, Rap2a, NGAL and SIRT1 mRNA expression in transplanted tumor lesion were significantly lower than those of model group while PAQR3, LATS1, E-cadherin, ZO-1 and TES mRNA expression were significantly higher than those of model group. Conclusion: Cordycepin can inhibit the proliferation and invasive growth of non-small cell lung cancer cell line H358 in transplanted tumor tissue.展开更多
