Resolvin D1 Induces mTOR-independent and ATG5-dependent Autophagy in BV-2 Microglial Cells

Objective The activation state of microglia is known to occupy a central position in the pathophysiological process of cerebral inflammation.Autophagy is a catabolic process responsible for maintaining cellular homeostasis.In recent years,autophagy has been demonstrated to play an important role in neuroinflammation.Resolvin D1(RvD1)is a promising therapeutic mediator that has been shown to exert substantial anti-inflammatory and proresolving activities.However,whether RvD1-mediated resolution of inflammation in microglia is related to autophagy regulation needs further investigation.The present study aimed to explore the effect of RvD1 on microglial autophagy and its corresponding pathways.Methods Mouse microglial cells(BV-2)were cultured,treated with RvD1,and examined by Western blotting,confocal immunofluorescence microscopy,transmission electron microscopy,and flow cytometry.Results RvD1 promoted autophagy in both BV-2 cells and mouse primary microglia by favoring the maturation of autophagosomes and their fusion with lysosomes.Importantly,RvD1 had no significant effect on the activation of mammalian target of rapamycin(mTOR)signaling.Furthermore,RvD1-induced mTOR-independent autophagy was confirmed by observing reduced cytoplasmic calcium levels and suppressed calcium/calmodulin-dependent protein kinase II(CaMK II)activation.Moreover,by downregulating ATG5,the increased phagocytic activity induced by RvD1 was demonstrated to be tightly controlled by ATG5-dependent autophagy.Conclusion The present work identified a previously unreported mechanism responsible for the role of RvD1 in microglial autophagy,highlighting its therapeutic potential against neuroinflammation. 展开更多

目的通过检测股骨头坏死患者血清Dickkopf-1(DKK-1)蛋白含量变化,探究其在早期诊断股骨头坏死及病情监测中的意义。方法选取2016年5月—2016年11月于广州中医药大学第一附属医院住院的股骨头坏死患者67例(ARCO分期:Ⅱ期13例、Ⅲ期27例... 目的通过检测股骨头坏死患者血清Dickkopf-1(DKK-1)蛋白含量变化,探究其在早期诊断股骨头坏死及病情监测中的意义。方法选取2016年5月—2016年11月于广州中医药大学第一附属医院住院的股骨头坏死患者67例(ARCO分期:Ⅱ期13例、Ⅲ期27例、Ⅳ期27例)作为实验组,并随机选取67例同期在该院体检中心体检正常者作为对照组,采用ELISA法检测两组患者的血清DKK-1蛋白浓度,比较并分析实验组与对照组、实验组内不同病因、不同ARCO分期患者的血清DKK-1蛋白浓度差异。结果实验组血清DKK-1蛋白浓度高于对照组(P <0.05);塌陷后患者血清DKK-1蛋白浓度高于塌陷前(P <0.05);不同病因的患者血清DKK-1蛋白浓度比较,差异无统计学意义(P>0.05);实验组ARCOⅡ、Ⅲ及Ⅳ期的患者血清DKK-1蛋白浓度比较,差异有统计学意义(P<0.05)。ROC曲线分析提示血清DKK-1蛋白在诊断股骨头坏死塌陷时具有一定的敏感性和显著的特异性,分别为57.40%和100.00%。血清DKK-1蛋白浓度与ARCO分期呈正相关(P<0.05)。结论血清DKK-1蛋白浓度与股骨头坏死患者的病情进展密切相关,对早期诊断股骨头坏死及监测患者病情变化具有一定的参考意义,可作为临床诊断及治疗的参考指标之一。 展开更多

Efficient capture,safe storage and release of tritium from the international thermonuclear experimental reactor(ITER) reaction exhaust gas is a perplexing problem,and the development of an efficient tritium-getter material with ultra-low hydrogenation equilibrium pressure is considered as a reliable way.In this work,Zr_(2)Co alloy was selected as a tritium-getter material and prepared through induction levitation melting.Fundamental performance test results show that Zr_(2)Co exhibits an ultra-low hydrogenation equilibrium pressure of 3.22 × 10^(-6) Pa at 25℃ and excellent hydriding kinetics under a low hydrogen pressure of 0.005 MPa.Interestingly,unique phase transition behaviors were presented in Zr_(2)Co-H system.Specifically,Zr_(2)CoH_(5) formed by Zr_(2)Co hydrogenated at room temperature is initially decomposed into ZrH_(2) and ZrCoH_(3) at200 ℃.With the temperature increasing to 350 ℃,ZrCoH_(3)is dehydrogenated to ZrCo,and then ZrCo further reacts with ZrH_(2) at 650 ℃ to reform Zr_(2)Co and hydrogen.Among the staged phase transition pathways during dehydrogenation,the decomposition of Zr_(2)CoH_(5) occurs preferentially,which is well accordance with both the smallest reaction energy barrier and the maximum reaction spontaneity that are determined respectively from kinetics activation energy and thermodynamics Gibbs free energy.Furthermore,first principles calculation results indicate that the stronger binding of hydrogen in interstitial environments of ZrCoH_(3)and ZrH_(2) triggers the hydrogen-stabilized phase transformation of Zr_(2)CoH_(5).The unique phase transition mechanisms in Zr_(2)Co-H system can shed light on the further exploration and regulation of analogous staged phase transition of hydrogen storage materials. 展开更多

利用血液中含量丰富的糖蛋白—纤维蛋白原,通过纤维蛋白原与还原剂三(2-羧乙基)磷盐酸盐(TCEP)反应,制备了基于纤维蛋白原类淀粉样聚集的纳米薄膜.利用荧光光谱、远紫外-圆二色谱等光学表征手段,探究了纤维蛋白原与TCEP反应的动力学过... 利用血液中含量丰富的糖蛋白—纤维蛋白原,通过纤维蛋白原与还原剂三(2-羧乙基)磷盐酸盐(TCEP)反应,制备了基于纤维蛋白原类淀粉样聚集的纳米薄膜.利用荧光光谱、远紫外-圆二色谱等光学表征手段,探究了纤维蛋白原与TCEP反应的动力学过程及其二级结构的变化;通过透射电子显微镜对薄膜结构进行了表征;用原子力显微镜和光学椭偏仪探讨了反应时间、纤维蛋白原浓度、TCEP溶液pH值以及浓度对薄膜厚度的影响,通过原子力显微镜表征了其在不同溶剂中浸泡的稳定性;最后通过薄膜的血小板吸附实验和对蛋白质的吸附实验,表征了其抗污能力.实验结果表明:通过反应条件的控制,实现了对薄膜厚度的可控,且薄膜在不同的环境中表现出优异的稳定性;薄膜表现出了对血小板、蛋白质、生物体液(如胎牛血清、牛奶等)等非特异性吸附的良好抵抗能力,有望作为新一代生物基抗污材料运用于表/界面改性领域. 展开更多

Nanoscale CeAl_4 was directly synthesized by the thermal reaction between CeH_2 and nano-aluminum at300℃.Then nano CeAl_4-doped sodium alanate(NaAlH_4)was synthesized by ball milling NaH/Al with 0.04CeAl_4under hydrogen atmosphere at room temperature,and the catalytic efficiency of nanoscale CeAl_4 for hydrogen storage of NaAlH_4 was systematically investigated.It is shown that CeAl_4 can effectively improve the dehydrogenation properties of sodium alanate system.The 0.04CeAl_4-doped NaAlH_4 system starts to release hydrogen below 80℃,completes dehydrogenation within 10 min at 170℃,and exhibits good cycling de/hydrogenation kinetics at relatively lower temperature(100-140℃).Apparent activation energy of the dehydrogenation of NaAlH_4 can be effectively reduced by addition of CeAl_4,resulting in the decrease in desorption temperatures.Moreover,by analyzing the reaction kinetics of nano CeAl_4-doped NaAlH_4sample,both of the decomposition steps are conformed to a two-dimensional phase-boundary growth mechanism.The mechanistic investigations gained here can help to understand the de-/rehydrogenation behaviors of catalyzed complex metal hydride systems. 展开更多

Objective To establish a rapid method for detecting acetylbritannilactone(ABL)by online sweeping-micellar electrokinetic chromatography(MEKC)and to elevate the sensitivity of the detection.Methods The combination of online sweeping technique with MEKC was used to determine the content of ABL in the extract of Inula britannica in plasma of rats.Results ABL was completely separated within 15 min in running buffer and sample buffer.The optimal conditions were as follows:on uncoated fused quartz silica capillary,with separation voltage of 23 kV,capillary temperature of 25 oC,and detection wavelength of 195 nm.The regression equations revealed good linear relationships between the peak area and concentration of ABL(r=0.998),with the detection limits of 0.005–0.15 mg/mL.The relative standard deviations of migration time and peak areas for intra-and inter-batch were<2.45%and<2.26%,respectively.The recovery rate of this method was 96.3%–97.2%.Conclusion This method provides some advantages in separation speed,testing sensitivity,and operating convenience,with low sample and reagent consumption.The online sweeping-MEKC is an effective method for pharmacokinetic study and analysis on tracing biological samples. 展开更多

Electrocatalytic water splitting coupled with sustainable energies is identified as an environmentally friendly and renewable strategy to generate high-quality hydrogen for the fuel cells.However,the main challenge is to develop high performance,low cost and chemically stable electrocatalysts to decline the energy barriers and enhance the sluggish kinetics of hydrogen evolution reaction(HER).Herein,a three-dimensional hierarchically ordered macroporous Ru-CoP@NC electrocatalyst(3DOM Ru-CoP@NC)derived from ordered macro-microporous metal-organic frameworks has been prepared using the precursor@template and double-solvent methods.The prepared 3DOM Ru-CoP@NC catalyst exhibits an overpotential of 15 mV(j=10 mA·cm^(-2))and a reaction Tafel slope of 38 mV·dec^(-1)in alkaline electrolyte,which are superior to commercial Pt@C catalyst.Additionally,the overpotential and reaction Tafel slope of this catalyst in acidic media are 45 mV and 50 mV·dec^(-1),respectively.The outstanding HER activities of 3DOM Ru-CoP@NC catalysts are ascribed to the 3D highly interconnectedreticular nanospaces that can increase effective reaction active sites.The N dope d carbon framework improves the electronic properties and conductivity.Moreover,the strong interaction of Ru and CoP nanoparticles also boosts the HER process.These results indicate that 3DOM Ru-CoP@NC catalysts with high catalytic activities have a broad application prospect in the future. 展开更多
