A premature senescence and death 128 (psd128) mutant was isolated from an ethyl methane sulfonate-induced rice IR64 mutant bank. The premature senescence phenotype appeared at the six-leaf stage and the plant died at the early heading stage, psd128 exhibited impaired chloroplast develop- ment with significantly reduced photosynthetic ability, chlorophyll and carotenoid contents, root vigor, soluble protein content and increased malonaldehyde content. Furthermore, the expression of senescence-related genes was significantly altered in psd128. The mutant trait was controlled by a single recessive nuclear gene. Using map- based strategy, the mutation Oryza sativa cell division cycle 48 (OsCDC48) was isolated and predicted to encode a putative AAA-type ATPase with 809 amino-acid residuals. A single base substitution at position C2347T in psd128 resulted in a premature stop codon. Functional complementation could rescue the mutant phenotype. In addition, RNA interference resulted in the premature senescence and death phenotype. OsCDC48 was expressed constitutively in the root, stem, leaf and panicle. Subcellular analysis indicated that OsCDC48:YFP fusion proteins were located both in the cytoplasm and nucleus. OsCDC48 was highly conserved with more than 90% identity in the protein levels among plant species. Our results indicated that the impaired function of OsCDC48 was responsible for the premature senescence and death phenotype. 展开更多

Control charts are commonly used tools in statistical process control for the detection of shifts in process parameters. Shewhart-type charts are efficient for large shift values, whereas cumulative sum(CUSUM) charts ... Control charts are commonly used tools in statistical process control for the detection of shifts in process parameters. Shewhart-type charts are efficient for large shift values, whereas cumulative sum(CUSUM) charts are effective in detecting medium and small shifts. Control chart use commonly assumes that data are free of outliers and parameters are known or correctly estimated based on an in-control process. In practice, these assumptions are not often true because some processes occasionally have outliers. Monitoring the location parameter is usually based on mean charts, which are seriously affected by violations of these assumptions. In this paper we propose several CUSUM median control charts based on auxiliary variables, and offer comparisons with their corresponding mean control charts. To monitor the location parameter, we examined the performance of mean and median control charts in the presence and absence of outliers. Both symmetric and non-symmetric processes were studied to examine the properties of the proposed control charts to monitor the location parameter using CUSUM control charts. We used different run length measures to study in-control and out-of-control performances of CUSUM charts. Results revealed that our proposed control charts perform much better than the traditional charts in the presence of outliers. A real application of our study was provided using data on concrete compressive strength as it relates to the quality of cement manufacturing. 展开更多

A problem that frequently occurs in biological experiments with laboratory animals is that some subjects are less susceptible to the treatment group than others. Finite mixture models have traditionally been used to describe the distribution of responses in treated subjects for such studies. In this paper, we first study the mixture normal model with multi-levels and multiple mixture sub-populations under each level, with particular attention being given to the model in which the proportions of susceptibility are related to dose levels, then we use EM-algorithm to find the maximum likelihood estimators of model parameters. Our results are generalizations of the existing results. Finally, we illustrate realistic significance of the above extension based on a set of real dose-response data. 展开更多
