蛋白质的翻译后修饰(post-translational modifications,PTMs)可通过在前体蛋白的氨基酸残基上添加化学基团来改变其结构和功能,在信号转导、表观遗传和疾病等过程中发挥重要作用。乳酸化修饰(lactylation)是一种新发现的PTM,由于其与... 蛋白质的翻译后修饰(post-translational modifications,PTMs)可通过在前体蛋白的氨基酸残基上添加化学基团来改变其结构和功能,在信号转导、表观遗传和疾病等过程中发挥重要作用。乳酸化修饰(lactylation)是一种新发现的PTM,由于其与糖酵解代谢终产物乳酸密切相关,为理解细胞代谢重编程和表观遗传调控之间的联系提供了新视角。研究表明乳酸化修饰在肿瘤进展中发挥重要作用,并与临床不良预后相关。组蛋白的乳酸化异常可以影响肿瘤细胞和免疫细胞中的基因表达,从而进一步调控肿瘤的进展和免疫抑制。非组蛋白的乳酸化也可以调节肿瘤的增殖和耐药等过程。本文对乳酸化修饰领域的最新研究进展及其在肿瘤发生发展、微环境和免疫抑制中的作用和机制进行综述,并对其在肿瘤靶向治疗及联合免疫治疗方面的应用价值进行探讨。 展开更多

肿瘤的发生是个复杂的过程,不仅取决于本身基因的突变或缺失,还受表观遗传调控的影响。近年来研究表明甲基化修饰在转录调控、异染色质的形成、X染色体失活、DNA损伤、肿瘤的发生发展等众多过程中均发挥着重要作用。含SET域赖氨酸甲基... 肿瘤的发生是个复杂的过程,不仅取决于本身基因的突变或缺失,还受表观遗传调控的影响。近年来研究表明甲基化修饰在转录调控、异染色质的形成、X染色体失活、DNA损伤、肿瘤的发生发展等众多过程中均发挥着重要作用。含SET域赖氨酸甲基转移酶7(SET domain containing lysine methyltransferase 7,SETD7)是体内一种重要的赖氨酸甲基转移酶,可甲基化组蛋白,也可使非组蛋白甲基化。这种修饰调节方式一旦发生紊乱可直接导致细胞异常,从而引起多种疾病的发生。研究表明SETD7与多种肿瘤的发生发展具有相关性,但是有关SETD7甲基化底物位点及其在相应肿瘤中的调控机制并未完全阐明。本文将就SETD7对组蛋白及非组蛋白的甲基化修饰在肿瘤中发挥的作用及调控分子机制等方面的研究进展加以概述,以期为肿瘤的发病机制及诊疗研究提供新的治疗靶点。 展开更多

Resonant four-photon photoemission from SnSe_(2)(001)

High-order nonlinear multiphoton absorption is usually inefficient,but can be enhanced by designing resonant excitations between occupied and unoccupied energy levels.We conducted angle-resolved multi-photon photoemission(mPPE)studies on the SnSe_(2)(001)surfaces excited by ultrashort laser pulses.By tuning photon energy and light polarization,we demonstrate the presence of a resonant four-photon photoemission(4PPE)process involving the occupied valence band(VB),the unoccupied second conduction band(CB2)and the unoccupied image-potential state(IPs)of SnSe_(2).In this 4PPE process,VB electrons of SnSe_(2) are resonantly excited into CB2 by adsorbing two photons,followed by the adsorption of another photon to populate the n=1 IPs before being emitted out to the vacuum by adsorbing one more photon.This results in a double-resonant 4PPE process,which exhibits approximately a 40 times enhancement in photoemission yields compared to cases where one of the resonant pathways,CB2→IPs,is inhibited by involving a virtual state instead of the IPs in the 4PPE.The double-resonant 4PPE process efficiently excite the bulk VB electrons outside the vacuum,like taking advantage of resonant“ladders”through two real empty electronic states of SnSe_(2).Our results highlight the important applications of mPPE in probing the band-structure,particularly the unoccupied states,of recently emerging main group dichalcogenide semiconductors.Furthermore,the discovered resonant mPPE process contributes to the exploration of their promising optoelectronic applications. 展开更多

The quantum harmonic oscillator(QHO),one of the most important and ubiquitous model systems in quantum mechanics,features equally spaced energy levels or eigenstates.Here we present a new class of nearly ideal QHOs formed by hydrogenic substitutional dopants in an AlGaAs/GaAs heterostructure.On the basis of model calculations,we demonstrate that,when aδ-doping Si donor substitutes the Ga/Al lattice site close to AlGaAs/GaAs heterointerface,a hydrogenic Si QHO,characterized by a restoring Coulomb force producing square law harmonic potential,is formed.This gives rise to QHO states with energy spacing of~8–9 meV.We experimentally confirm this proposal by utilizing gate tuning and measuring QHO states using an aluminum single-electron transistor(SET).A sharp and fast oscillation with period of~7–8 mV appears in addition to the regular Coulomb blockade(CB)oscillation with much larger period,for positive gate biases above 0.5 V.The observation of fast oscillation and its behavior is quantitatively consistent with our theoretical result,manifesting the harmonic motion of electrons from the QHO.Our results might establish a general principle to design,construct and manipulate QHOs in semiconductor heterostructures,opening future possibilities for their quantum applications. 展开更多

已经研究和报道较多的三维狄拉克节线半金属,具有新颖的准粒子激发现象及有趣的输运性质.然而,在强自旋轨道耦合作用下仍具有稳定的狄拉克线的二维材料却罕有报道.本文发现一个具体的有分数平移的C_(2v)×Z_(2)^(T)对称点群,在强自... 已经研究和报道较多的三维狄拉克节线半金属,具有新颖的准粒子激发现象及有趣的输运性质.然而,在强自旋轨道耦合作用下仍具有稳定的狄拉克线的二维材料却罕有报道.本文发现一个具体的有分数平移的C_(2v)×Z_(2)^(T)对称点群,在强自旋轨道耦合下也能保护稳定的二维狄拉克线.这种对称性可以存在于铋的一种新的同素异形体(砖相)中,也可以推广到具有相同对称性下的其他奇数层以及与铋同族的其他元素,如磷、砷、锑.三层铋砖相结构的价电子总数为30,能够被2整除,但不能被4整除,这正好保证了狄拉克线处于半填充状态而穿过费米能级.有趣的是,三层铋砖相结构的费米能级处除了狄拉克线之外不存在其他多余的态,使得铋砖相结构中出现了干净的狄拉克线.费米能级处干净的狄拉克线不受其他杂态的干扰,这更有利于实验上探测由狄拉克线诱导的相关输运性质. 展开更多

Promoters are critical elements to control gene expression but could behave differently under various growth conditions. Here we report the construction of a genome-wide promoter library, in which each native promoter in Saccharomyces cerevisiae was cloned upstream of a yellow fluorescent protein (YFP) reporter gene. Nine libraries were arbitrarily defined and assembled in bacteria. The resulting pools of promoters could be prepared and transformed into a yeast strain either as centromeric plasmids or integrated into a genomic locus upon enzymatic treatment. Using fluorescence activated cell sorting, we classified the yeast strains based on YFP fluorescence intensity and arbitrarily divided the entire library into 12 bins, representing weak to strong promoters. Several strong promoters were identified from the most active bins and their activities were assayed under different growth conditions. Finally, these promoters were applied to drive the expression of genes in xylose utilization to improve fermentation efficiency. Together, this library could provide a quick solution to identify and utilize desired promoters under user-defined growth conditions. 展开更多
