姜黄素抑制肺癌细胞A549上皮间质转化的机制研究 被引量:2

目的探讨姜黄素对肺癌细胞A549上皮间质转化的影响。方法姜黄素诱导肺癌细胞A54924 h,CCK-8法检测A549细胞活性,Transwell实验检测A549细胞迁移、侵袭的能力,Western blotting检测上皮间质转化标志蛋白的相对表达量,实时荧光定量聚合酶... 目的探讨姜黄素对肺癌细胞A549上皮间质转化的影响。方法姜黄素诱导肺癌细胞A54924 h,CCK-8法检测A549细胞活性,Transwell实验检测A549细胞迁移、侵袭的能力,Western blotting检测上皮间质转化标志蛋白的相对表达量,实时荧光定量聚合酶链反应(qRT-PCR)检测HOTAIR、E-cadherin、N-cadherin及Snail mRNA的相对表达量,测序方法检测姜黄素诱导后lncRNA表达的变化,实验验证介导姜黄素发挥作用的具体非编码RNA,分析HOTAIR与临床病理参数之间的关系。结果CCK-8法检测结果显示,姜黄素诱导肺癌细胞A54924 h后,不同浓度组肺癌细胞A549的OD值比较,差异有统计学意义(P<0.05),随着姜黄素浓度的升高,OD值降低。Transwell结果显示,姜黄素诱导肺癌细胞A54924 h后,姜黄素诱导组抑制肺癌细胞A549的侵袭、迁移,与对照组比较,差异有统计学意义(P<0.05)。Western blotting和qRT-PCR检测结果显示,姜黄素诱导组的间质表型基因和蛋白(E-cadherin、N-cadherin、Snail)与对照组比较,差异有统计学意义(P<0.05),N-cadherin、Snail相对表达量下降,E-cadherin相对表达量升高。癌旁正常组和肺癌组中HOTAIR的相对表达量比较,差异有统计学意义(P<0.05),肺癌组高于癌旁正常组。测序检测和qRT-PCR结果显示,姜黄素抑制HOTAIR的表达。分析HOTAIR在肺癌组织中的表达及其与临床病理参数之间的关系显示,HOTAIR的表达与TNM分期、分化程度、淋巴结转移密切相关。对照组、姜黄素诱导组、姜黄素诱导+HOTAIR过表达组A549细胞OD值比较,差异有统计学意义(P<0.05)。结论姜黄素有效抑制肺癌细胞A549的增殖、侵袭及迁移,姜黄素抑制lncRNA HOTAIR表达,姜黄素抑制肺癌细胞A549上皮间质转化是通过HOTAIR实现的。 展开更多

Cemented tailings backfill(CTB) have increasingly been used in recent years to improve the stability of mining stopes in deep underground mines. Deep mining processes are often associated with rock bursting and high-speed dynamic loading conditions. Therefore, it is important to investigate the characteristics and dynamic mechanical behavior of CTB. This paper presents the results of dynamic tests on CTB specimens with different cement and solid contents using a split Hopkinson pressure bar(SHPB). The results showed that some CTB specimens exhibited one to two lower stress peaks after reaching dynamic peak stress before they completely failed. The greater the cement-to-tailings ratio is, the more obvious the strain reaction. This property mainly manifested as follows. First,the dynamic peak stress increased with the increase of the cement-to-tailings ratio when the impact velocity was fixed. Second, the dynamic peak stress had a quadratic relationship with the average stress rate. Third, the cement-to-tailings ratio could enhance the increase rate of dynamic peak stress with strain rate. In addition, the dynamic strength enhancement factor K increased with the increase of strain rate, and its value was larger than that of the rock samples. The failure modes of CTB specimens under low-speed impact were tensile failure and X conjugate shear failure, where were nearly the same as those under static uniaxial and triaxial compression. The CTB specimens were crushed and broken under critical strain, a failure mode similar to that of low-strength concrete. The results of the experimental research can improve the understanding of the dynamic mechanical properties of CTB and guide the strength design of deep mining backfills. 展开更多

However, the exact molecular mechanism remains unclear. In this study, muscarinic acetylcholine receptor M1 (CHRM1) expression was measured via immunohistochemical analysis in human prostate cancer tissue array slides. PC-3, LNCaP, and A549 cells were treated with pirenzepine or carbachol, and the cell migration and invasion abilities were evaluated. Western blotting and quantitative real-time PCR were performed to measure GLI family zinc finger 1 (GLI1), patched 1 (PTCH1), and sonic hedgehog (SHH) expression levels. High expression of CHRM1 was found in early-stage human prostate cancer tissues. In addition, the selective CHRM1 antagonist pirenzepine inhibited PC-3, LNCaP, and A549 cell migration and invasion, but the agonist carbachol promoted the migration and invasion of these three cell lines. Muscarinic signaling can be relayed by hedgehog signaling. These data show that CHRM1 is involved in the regulation of prostate cancer migration and invasion through the hed^eho~ si^nalin~ ~athwav. 展开更多
