Potential relevance between the placental inflammation and CMV-related autism has been reported by clinical observation. Meanwhile, abnormal expression of Toll-like receptor 2 (TLR2) and TLR4 in placenta of patients with chorioamnionitis was observed in multiple studies. IL-6 and IL- 10 are two important maternal inflammatory mediators involved in neurodevelopmental disorders. To investigate whether murine CMV (MCMV) infection causes alterations in placental IL-6/10 and TLR2/4 levels, we analyzed the dynamic changes in gene expression of TLR2/4 and IL-6/10 in placentas following acute MCMV infection. Mouse model of acute MCMV infection during pregnancy was created, and pre-pregnant MCMV infected, lipopolysaccharide (LPS)-treated and uninfected mice were used as controls. At E13.5, E 14.5 and E 18.5, placentas and fetal brains were harvested and mRNA expression levels of placental TLR2/4 and IL-6/10 were analyzed. The results showed that after acute MCMV infection, the expression levels of placental TLR2/4 and IL-6 were elevated at E13.5, accompanied by obvious placental inflammation and reduction of placenta and fetal brain weights. However, LPS 50 ktg/kg could decrease the IL-6 expression at E13.5 and E14.5. This suggests that acute MCMV infection during pregnancy could up-regulate the gene expression of TLR2/4 in placental trophoblasts and activate them to produce more pro- inflammatory cytokine IL-6. High dose of LPS stimulation (50 gg/kg) during pregnancy can lead to down-regulation of IL-6 levels in the late stage. Imbalance of IL-6 expression in placenta might be associated with the neurodevelopmental disorders in progeny.展开更多

The differences in the levels of hk10 and DJ-1 in preoperative serum as well as the expression of ovarian cancer-related proliferation genes and invasion genes in intraoperative lesion tissue were compared between the two groups. Correlation analysis was used to assess the inner link of serum hk10 and DJ-1 levels with tumor malignancy in patients with ovarian cancer. Results: Serum hk10 and DJ-1 levels of ovarian cancer group were higher than those of ovarian cyst group;proliferation genes Clusterin, FUNDC1, AKT, TNFAIP8 and HBXIP mRNA expression in lesion tissues were higher than those of ovarian cyst group;invasion genes Beclin1, BRMS1 and SIRT2 mRNA expression in lesion tissues were lower than those of ovarian cyst group whereas EFEMP1 mRNA expression was higher than that of ovarian cyst group. The Pearson test showed that serum hk10 and DJ-1 levels in ovarian cancer were directly correlated with the expression of proliferation genes and invasion genes. Conclusion: hk10 and DJ-1 are highly expressed in serum of patients with ovarian cancer, and their levels are directly correlated with the proliferation and invasion activity of cancer cells.展开更多

Therapeutic benefits of using RDS for both disease control and prevention, in the effort to contain the coronavirus disease 2019(COVID-19), have been shown. However, the biochemically active constituents of RDS and their mechanisms of action are still unclear. The goal of the present study is to clarify the material foundation and action mechanism of RDS.Methods: To conduct an analysis of RDS, an integrative analytical platform was constructed, including target prediction, protein–protein interaction(PPI) network, and cluster analysis;further, the hub genes involved in the disease-related pathways were identified, and the their corresponding compounds were used for in vitro validation of molecular docking predictions. The presence of these validated compounds was also measured in samples of the RDS formula to quantify the abundance of the biochemically active constituents. In our network pharmacological study, a total of 26 bioinformatic programs and databases were used, and six networks, covering the entire Zang-fu viscera, were constructed to comprehensively analyze the intricate connections among the compounds-targets-disease pathways-meridians of RDS.Results: For all 1071 known chemical constituents of the nine ingredients in RDS, identified from established TCM databases, 157 passed drug-likeness screening and led to 339 predicted targets in the constituent–target network. Forty-two hub genes with core regulatory effects were extracted from the PPI network, and 134 compounds and 29 crucial disease pathways were implicated in the target–constitu ent–disease network. Twelve disease pathways attributed to the Lung–Large Intestine meridians, with six and five attributed to the Kidney–Urinary Bladder and Stomach–Spleen meridians, respectively. Onehundred and eighteen candidate constituents showed a high binding affinity with SARS-coronavirus-23-chymotrypsin-like protease(3 CLpro), as indicated by molecular docking using computational pattern recognition. The in vitro activity of 22 chemical constituents of RDS was validated using the 3 CLproinhibition assay. Finally, using liquid chromatography mass spectrometry in data-independent analysis mode, the presence of seven out of these 22 constituents was confirmed and validated in an aqueous decoction of RDS, using reference standards in both non-targeted and targeted approaches. Conclusion: RDS acts primarily in the Lung–Large Intestine, Kidney–Urinary Bladder and Stomach–Spleen meridians, with other Zang-fu viscera strategically covered by all nine ingredients. In the context of TCM meridian theory, the multiple components and targets of RDS contribute to RDS’s dual effects of healthstrengthening and pathogen-eliminating. This results in general therapeutic effects for early COVID-19 control and prevention.展开更多

The ELF3 activity is subjected to attenuation in response to warm temperature;however, how the protein level of ELF3 is regulated at warm temperature remains less understood. Here, we report that the E3 ligase XB3 ORTHOLOG 5 IN ARABIDOPSIS THALIANA, XBAT35, mediates ELF3 degradation. XBAT35 interacts with ELF3 and ubiquitinates ELF3. Loss-of-function mutation of XBAT35 increases the protein level of ELF3 and confers a short-hypocotyl phenotype under warm temperature conditions. Thus, our findings establish that XBAT35 mediates ELF3 degradation to lift the inhibition of ELF3 on PIF4 for promoting thermoresponsive hypocotyl growth in plants.展开更多

The calcium-sensing receptor (CaSR) is expressed in testicular tissues and plays a pivotal role in calcium homeostasis by activating cellular signaling pathways, but its role in testicular damage induced by diabetes remains unclear. A diabetic model was established by a single intraperitoneal injection of streptozotocin (STZ, 40 mg kg-1) in Wistar rats. Animals then received GdCl3 (an agonist of CaSR, 8.67 mg kg-1), NPS-2390 (an antagonist of CaSR, 0.20 g kg-~), or a combination of both 2 months after STZ injection. Diabetic rats had significantly lower testes weights and serum levels of testosterone compared to healthy rats, indicating testicular damage and dysfunction in STZ-induced diabetic rats. Compared with healthy controls, the testicular tissues of diabetic rats overexpressed the CaSR protein and had higher levels of malondialdehyde (MDA), lower superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity, and higher numbers of apoptotic germ cells. The testicular tissues from diabetic rats also expressed lower levels of Bcl-2 and higher levels of Bax and cleaved caspase-3 in addition to higher phosphorylation rates of c-Jun NH2-terminal protein kinase (JNK), p38, and extracellular signaling-regulated kinase (ERK) 1/2. The above parameters could be further increased or aggravated by the administration of GdCI3, but could be attenuated by injection of NPS-2390. In conclusion, the present results indicate that CaSR activation participates in diabetes-induced testicular damage, implying CaSR may be a potential target for protective strategies against diabetes-induced testicular damage and could help to prevent infertility in diabetic men.展开更多

Imbalance of IL-6 expression in placenta might be associated with the neurodevelopmental disorders in progeny.

文摘It is important for intelligent orchards to be able to achieve automatic monitoring of fruit growth information within a natural growing environment. The issue of how to track green and oscillating fruits under the influence of wind and farming operations is a key aspect of monitoring of the growth state of the fruit. In order to realize the accurate tracking of green fruit targets,a new method based on target tracking is proposed.First,an optical flow method is applied to realize the automatic detection of green fruit targets, and this lays the foundation for the accurate and automatic tracking of these targets. Then,Kalman and kernelized correlation filter(KCF)algorithms are applied to achieve multi-target tracking and prediction. In order to verify the performance of these different algorithms on various types of green fruit targets, experiments were carried out based on nine video sequences. The experimental results for the tracking of single, double and triple green fruit targets show that the average tracking success rates of the Kalman algorithm are 88.15%,82.30%and 53.10%,respectively,and those of the KCF algorithm are 94.07%,87.35%and 61.46%,respectively,meaning that the average tracking results from KCF are 5.92%,5.05%and 8.36%higher than those from the Kalman algorithm. The time consumed is also reduced by 35.40%,36.27%and 40.86%,respectively.The results show that it is feasible to apply the KCF algorithm to the tracking of green fruit targets.
文摘Male infertility caused by testicular damage is one of the complications of diabetes mellitus. In conclusion, the present results indicate that CaSR activation participates in diabetes-induced testicular damage, implying CaSR may be a potential target for protective strategies against diabetes-induced testicular damage and could help to prevent infertility in diabetic men.

文摘Objective The amount of nuclear DNA(C-value)is a key biodiversity character that provides strong unifying elements in revealing the phylogenetic regularity and relationship between genome size and functional traits for plant resource. The estimation of C-values could primarily extend our knowledge on the genetic background and genome diversity for medicinal plants, and thereby the variation of pharmacological constituents and phylogenetic mechanism of medicinal plant taxa will be revealed. However,a large number of medicinal plants(e.g.Cornus officinalis)typically contain a series of secondary metabolites, especially tannic acid, which would significantly affect the estimation of DNA content by flow cytometry(FCM).Methodological discussions and improvement need to be made to solve this problem. Methods Two isolation buffers LB01 and Otto 1 were selected to prepare nuclear suspension with additional treatments of pre-soaking and centrifugation combination of gradient centrifugal force and duration. The best isolation and estimation methods were determined by FCM measurement in C.officinalis.Results The dry leaves were pre-soaked in Otto I buffer for 15 min and the Otto I nuclear suspension was centrifugated at 1.0×103 g for 2 min.The results showed that debris and nuclei were better separated and the scatterplots of good quality were obtained with low coefficient of variation(CV).Contrarily, the nuclear DNA content of C.officinalis could not be accurately estimated for nuclei extracted by LB01 buffer.Finally,2C-value and genome size of C.officinalis were first estimated as 5.92 pg and 2893 Mbp,respectively. Conclusion The new methods proposed here are able to accurately estimate DNA content of C.officinalis, which provides valuable references for the estimation of genome size in other tannin-rich medicinal plants.
